Green synthesis of polydopamine functionalized magnetic mesoporous biochar for lipase immobilization and its application in interesterification for novel structured lipids production

Junxin Zhao,Maomao Ma,Xianghui Yan,Guohua Zhang,Jiaheng Xia,Zheling Zeng,Ping Yu,Qiang Deng,Deming Gong

In this study, the polydopamine functionalized magnetic mesoporous biochar (MPCB-DA) was prepared for immobilization of Bacillus licheniformis lipase via covalent immobilization. Under optimized immobilization conditions, the maximum immobilization yield, efficiency and immobilized lipase amount were found to be 45%, 54% and 36.9 mg/g, respectively. The immobilized lipase, MPCB-DA-Lipase showed good thermal stability and alkali resistance. The MPCB-DA-Lipase retained 56% initial activity after 10 reuse cycles, with more than 85% relative activity after 70 days’ storage at 4 or 25 °C. The MPCB-DA-Lipase was efficiently applied in the interesterification of Cinnamomum camphora seed kernel oil and perilla seed oil, with maximum interesterification efficiency of 46%. The produced structured lipids belong to the S 2 U and U 2 S triacylglycerols, a novel medium-and long-chain triacylglycerol. These results demonstrated that the MPCB-DA-Lipase may be used as an efficient biocatalyst in lipid processing applications of food industries.

One-pot green synthesis of poly(hexamethylenediamine-tannic acid)-bacterial cellulose composite for the reduction, immobilization, and recovery of Cr(VI)

Kaihua Su,Dongmin Zhao,Ai Lu,Chaoran Zhong,Xing-Can Shen,Changping Ruan

Hexavalent chromium Cr(VI) is one of the most toxic heavy metal pollutants in water due to its high water solubility, high toxicity, strong mutagenicity and carcinogenicity . To enable efficient Cr(VI) removal from wastewater, it remains difficult to fabricate advanced adsorbents with good selectivity and high adsorption capacity in large scale. Herein, a poly(hexamethylenediamine-tannic acid)-bacterial cellulose (denoted as HTA-BC) composite was prepared by using natural raw materials with abundance, low cost and nontoxicity in a facile one-step method and applied for the decontamination of highly toxic Cr(VI). The as-synthesized HTA-BC functionalized with abundant amino and phenolic hydroxyl groups had demonstrated high capacity (534.8 mg g −1 ) and good selectivity for Cr(VI) scavenging. Notably, the treatment of real water samples with the HTA-BC composite could efficiently reduce the concentration of the spiked Cr(VI) below the acceptable levels for drinking water . Moreover, high-purity Cr 2 O 3 was obtained by calcination of the Cr species-laden HTA-BC to ensure that the toxic Cr(VI) residing in the spent adsorbents can’t leak out into the surroundings, thus avoiding the secondary pollution. The natural abundance, low cost, easy availability, and sustainability of the raw materials, combined with the facile and green synthesis endowed the HTA-BC composite with great potential for not only decontaminating Cr(VI) wastewater, but also recycling Cr by calcination of the Cr-laden HTA-BC. Moreover, no secondary pollution would be brought during the synthesis and the post-treatment after the Cr(VI) removal, thus it is expected to be a green and sustainable approach for the purification of Cr(VI) containing wastewater.

Covalent Immobilization of Phytosynthesized Silver Nanoparticles and Quaternary Ammonium Chitosan on Cotton Fabrics: Enhanced Antimicrobial Durability, Antioxidant Capacity, and Natural Dye Affinity

Hegui Yang,Wei Li,Di Xu,Changhai Xu,Jinmei Du,Dagang Miao,Guowei Xiao,Xiaodong Jiang,Yang Jiang

This study developed a green multifunctional modification strategy for cotton fabrics through covalent coimmobilization of phytosynthesized silver nanoparticles (AgNPs) and N-(2-hydroxy)-propyl-3-trimethylammonium chitosan (HTCC) using dimethyloldihydroxyethyleneurea (DMDHEU). AgNPs were synthesized using Aloe vera extract with an average size of 81.96 nm (PDI 0.191) and integrated with HTCC via a pad-cure process. The cotton fabric modified with DMDHEU, HTCC, and AgNPs (Cotton-DHA, where “DHA” denotes the three modifying agents: DMDHEU, HTCC, and AgNPs) demonstrated synergistic antibacterial activity against Escherichia coli (E. coli) (99.81%) and Staphylococcus aureus (S. aureus) (99.29%), with over 90.00% inhibition retained after 20 washing cycles, indicating robust durability. The Cotton-DHA also exhibited significantly enhanced antioxidant capacity, with an ABTS free radical scavenging rate of 72.51%, in stark contrast to the 4.20% rate of pristine cotton. Natural dye affinity was substantially improved, as evidenced by the increase in K/S value from 0.30 to 2.58. The covalent bonding mediated by DMDHEU ensured durable immobilization, where HTCC provided primary antibacterial action (>94.00%) and AgNPs enhanced the efficacy to >99.00%. This synergistic system provides a sustainable approach to developing high-performance textiles with integrated antibacterial, antioxidant, and dyeing properties.

Immobilization of cross-linked In-doped Mo(O,S)2 on cellulose nanofiber for effective organic-compound degradation under visible light illumination

Hairus Abdullah,Chi-Ning Hsu,Hardy Shuwanto,Noto Susanto Gultom,Worku Lakew Kebede,Chang-Mou Wu,Chiu-Chun Lai,Ri-Ichi Murakami,Masayuki Hirota,Antonio Norio Nakagaito,Dong-Hau Kuo

Relatively small amounts of In-doped Mo(O,S) 2 (IMS) catalysts (10%, 20%, and 30%) were deposited on cellulose nanofiber (CNF) by cross-linking them with functional groups of siloxane and epoxy to form CNF-IMS hybrid composites. The as-prepared hybrid composites were characterized and tested their performances toward the photo degradations of cationic (MB and RhB) and anionic (MO) dyes. As indium was doped into Mo(O,S) 2 lattice to form solid-solution, the charge transfer and photocarrier separation during the catalytic reaction were simultaneously enhanced as probed with electrochemical impedance spectroscopy and photoluminescence measurements, respectively. To ensure the catalyst on CNF was well deposited and recyclable, the hybrid composite was evaluated with a reusability experiment to show the stability performance in degrading organic dyes. It was found the environmentally friendly CNF-IMS hybrid composite was relatively stable during the reusability experiment, indicating no catalyst powder was leached out during the photocatalytic reaction. The photoreaction mechanism was convinced by radical-scavenging experiments to show that hydroxyl and superoxide played essential roles for the organic dye degradations in visible-light illuminated conditions. The cross-linked organic/inorganic hybrid catalyst showed a prospective visible light active material not only to solve the environmental issue due to the leaching of nanoparticles but also to lower the post-treatment/recycling cost of photocatalyst in industrial application.

Differential immobilization mechanisms of calcium silicate-based materials toward Pb(Ⅱ) and Cd(Ⅱ): From adsorption characteristics to long-term leaching stability

Wenjun Fu,Jiangjie Fang,Yongdian Yue,Xinzhuang Fu,Chuanxi Peng,Di Chen,Youchuan Chen,Tong Yue

Calcium silicate-based materials hydrate to form calcium silicate hydrate (C-S-H), a reactive phase capable of immobilizing heavy metals. However, the mechanisms governing element-specific long-term stability remain unclear. In this study, dicalcium silicate (C 2 S) and tricalcium silicate (C 3 S) were used to compare the removal behavior, aging stability, and microscopic immobilization mechanisms of Pb(Ⅱ) and Cd(Ⅱ) in aqueous systems. Both minerals efficiently removed Pb(Ⅱ) and Cd(Ⅱ), while C 3 S showed stronger early Pb(Ⅱ) removal, possibly related to faster hydration-product formation under the tested conditions. Cd(Ⅱ) removal remained above 99% in both systems. Apparent isotherm and kinetic analyses indicated that metal removal was governed by a coupled apparent process involving interfacial adsorption, surface complexation, hydration-product formation, and precipitation, rather than by physical adsorption alone. Long-term aging and acidic leaching tests revealed contrasting stability: Cd-bearing products exhibited strong leaching resistance, whereas Pb-bearing products showed a potential risk of re-release after aging. XRD, XPS, TEM, and TEM-EDS analyses indicated that Pb and Cd were closely associated with C-S-H-related hydration products, suggesting possible Ca-associated binding or partial structural incorporation. The stronger acid-leaching resistance of Cd-bearing products implies more stable association with C-S-H-related phases than that of Pb-bearing products. These findings clarify the element-specific immobilization mechanisms of C 2 S/C 3 S and provide guidance for calcium silicate-based treatment of heavy-metal-contaminated water.

Integrating enzymatic activity into highly emissive metal-organic frameworks derived from AIE ligands via horseradish peroxidase immobilization for dual-mode optical sensing

Zuyao Fu,Lingfeng Yang,Yuanyuan Zhang,Xuanjun Zhang,Jing Xie,Zhaoyang Ding

HRP was covalently immobilized on MAF to form a biocomposite with high activity and fluorescence. • HRP@MAF exhibited a remarkable fluorescence quantum yield of 97.77%. • HRP@MAF showed superior substrate affinity with a Km value 25-fold lower than HRP. • A dual-mode colorimetric/ratiometric fluorescent sensor was developed for histamine. Integrating strong fluorescence and efficient biocatalysis within a single metal-organic framework (MOF) is attractive for advanced optical sensing. Herein, a highly emissive metal-organic framework (denoted as MAF) was constructed using ligands with aggregation-induced emission (AIE) properties, followed by horseradish peroxidase (HRP) immobilization to yield HRP@MAF with high catalytic activity and strong fluorescence. The resulting HRP@MAF exhibited a fluorescence quantum yield of 97.77%. Benefiting from the high surface area and porous structure of MAF, HRP@MAF showed markedly improved stability and substrate affinity toward 3,3′,5,5′-tetramethylbenzidine (TMB), with a K m value 25-fold lower than that of free HRP. Based on its catalytic and optical properties, a colorimetric/ratiometric fluorescence sensor was developed for histamine detection with detection limits of 1.71 μM and 0.56 μM, respectively. In addition, smartphone-integrated on-site detection was achieved with linear ranges of 10–1000 μM for the colorimetric mode and 1–1000 μM for the ratiometric fluorescence mode. This work establishes an effective strategy for constructing multifunctional sensing platforms by coupling AIEgens derived highly emissive MOFs with immobilized enzymes. Multi-module immobilization of Gluconobacter frateurii alcohol dehydrogenase and Lactobacillus brevis NADH oxidase with tethered nicotinamide adenine dinucleotide for D-allulose biosynthesis

Jiaying Ai,Jiajun Yan,Yuxin Zhang,Fuping Lu,Shuhong Mao,Yixun Jiang,Lei Li,Hui-Min Qin

Chemical tethering NAD + to GfADH enhances enzyme-cofactor spatial proximity. • The application of conjugate and NOX reduced the NAD + demand by 13-fold. • The NAD + -GfADH conjugate and NOX were co-fixed via sequential recruitment. • Co-immobilization enhanced enzyme stability with improved cofactor regeneration. The green biomanufacturing of high-value compounds is limited by enzyme instability and cofactor dependence. Herein, we developed a tailored multi-module immobilization strategy that integrated cofactor tethering with sequential multi-enzyme co-immobilization to facilitate the efficient conversion of allitol into D-allulose catalyzed by the Gluconobacter frateurii -derived dehydrogenase (GfADH). In this strategy, nicotinamide adenine dinucleotide (NAD + ) was covalently tethered to the surface of GfADH via a flexible linker, creating an NAD + -GfADH conjugate. Coupling the conjugate with NADH oxidase (NOX) established an efficient cofactor self-regenerating system, reducing the external NAD + demand by 13-fold. Furthermore, this conjugate was encapsulated within hybrid nanoflowers, thereby improving its reusability. To facilitate cofactor transfer, NOX was directionally recruited and colocalized with GfADH using the SpyTag/SpyCatcher system. The co-immobilized enzyme exhibited a high D-allulose yield of 88.7% from 110 mM allitol and retained 85.0% of its initial activity after 6–8 cycles. This work provides a valuable integrated strategy for cofactor regeneration and enzyme stabilization in cofactor-dependent biosynthesis. Aptamer-Driven Assembly of Enzyme Immobilization on Ag-HMPB@UiO-66 MOF-on-MOF Nanostructures for High-sensitivity Electrochemical Biosensing of Trichlorfon

Zongda Li,Yukun Zhang,Youning Zhang,Yu Zhang,Jiangxia Xu,Fanxing Meng,Minwei Zhang

Combining high-electroactive nanomaterial with enzyme immobilization, the development of the electrochemical biosensor is a frontier approach to improve the sensitivity and selectivity for the detection of organophosphorus pesticides (OPs). In this study, a core–shell nanostructure (Ag-HMPB@UiO-66) was prepared with the Ag-HMPB as a core and a UiO-66 forming the shell as an advanced platform for acetylcholinesterase (AChE) immobilization. The innovation lies in the synergistic integration of the excellent electron transfer capability of Ag-HMPB with the good structural stability of UiO-66. Based on Lewis acid–base theory, the form of Zr─O─P coordination bond could improve the efficiency of aptamer (Apt) immobilization (131.65 nM mg −1 ) compared with physical adsorption. Electrochemical analysis revealed that the as-prepared biosensor exhibited exceptional substrate affinity, with a low K m of 0.234 mM. Under optimized conditions, the biosensor exhibited sensitive detection of trichlorfon over a wide range (1 × 10 −13 to 2.5 × 10 −9 g L −1 ), with a low LOD of 0.44 pg L −1 (3σ/slope). Additionally, the biosensor can also be applied to the cucumber with good recovery (97.34% to 103.35%), as well as good storage stability and reproducibility. This provides a reliable and powerful tool for the trace detection of OPs in complex agricultural products.

Construction of a chitosan/γ-poly(glutamic acid) composite hydrogel crosslinked by ZIF-8 for enhanced α-glucosidase immobilization and inhibitor screening

Zhentao Chen,Lijing Lei,Xiangze Fan,Jinhua Zhu,Haodong Zhou,Yun Zhang,Fanyi Ma,Xiuhua Liu,Md. Zaved H. Khan

ZIF-8 as multi-dentate crosslinking hub converts weak CS/γ-PGA complexes into robust dual-network hydrogel. • Ultra-large pores eliminate diffusion limitation (Da ≈ 0.001) with K m increase of only 2.2-fold. • HSP regression (R 2  = 0.76, p ≈ 0.0002) validates synergistic thermodynamic exclusion and size-sieving mechanism. • Immobilized α-Glu@PC/ZIF-8 enables ligand fishing of two α-glucosidase inhibitors via capture-wash-elute workflow. Herein, a composite hydrogel based on two natural macromolecules—chitosan (CS) and γ-poly(glutamic acid) (γ-PGA)—was developed, crosslinked by zeolitic imidazolate framework-8 (ZIF-8) nanoparticles, which serve as reactive crosslinking hubs to covalently connect the biopolymer chains for efficient immobilization of α-glucosidase (α-Glu). A carboxyl-terminated poly(ethylene glycol)–poly(glutamic acid) copolymer was first synthesized and its activated carboxyl groups underwent amidation with amino groups of CS to rapidly form a three-dimensional PC/ZIF-8 hydrogel. The effects of ZIF-8 content and CS concentration on micro-morphology, swelling behavior, and rheological properties were systematically investigated. ZIF-8-mediated crosslinking was essential for gel formation, enabling effective modulation of network density and pore architecture. The optimized hydrogel showed no inhibitory effect on α-Glu. Immobilized α-Glu@PC/ZIF-8 exhibited enhanced pH stability (retaining 53% and 70% activity at pH 4.0 and 9.0), outstanding storage stability (>90% after 4 weeks), excellent organic solvent tolerance (>90% in most solvents), and good reusability (near 100% activity over 6 cycles). Mass transfer studies confirmed that the designed crosslinking density provided efficient substrate/product diffusion channels, minimizing product inhibition. Hansen solubility parameter analysis revealed a synergistic “thermodynamic exclusion + size-sieving” mechanism for solvent tolerance. Coupled with HPLC, the immobilized enzyme successfully identified two α-glucosidase inhibitors—batatasin I and 2,4-dimethoxy-6,7-dihydroxyphenanthrene—from Dioscorea opposita Thunb. peel. This work demonstrates that rational chemical modification of natural macromolecular networks via MOF crosslinkers provides an effective strategy for constructing high-performance enzyme immobilization platforms. Cascade utilization of phosphate-loaded low-temperature pyrolysis gypsum board for Cd(Ⅱ) and Pb(Ⅱ) immobilization from aqueous and soil systems

Tao Chang,Qin Wang,Shijia Wu,Zhimiao Zhao,Yinjiang Zhang,Xian Lu,Yunhui Zhang

Phosphate-loaded pyrolyzed gypsum board was innovatively reused for Cd(Ⅱ)/Pb(Ⅱ) remediation. • Single and coexisting heavy metal systems were systematically evaluated in both aqueous and soil environments. • The recycled material achieved enhanced Cd(Ⅱ) adsorption while retaining high Pb(Ⅱ) removal capacity. • Phosphate-driven mineralization and ion exchange between hydroxyapatite and heavy metals enhance the immobilization. This study proposed a cascade utilization strategy for waste gypsum board by reusing phosphate-loaded pyrolyzed gypsum board (PGB150P) for Cd(Ⅱ) and Pb(Ⅱ) remediation in aqueous and soil environments. The results showed that PGB150 exhibited excellent Pb(Ⅱ) affinity (q max  = 1396.14 mg/g) but limited Cd(Ⅱ) adsorption capacity (161.21 mg/g). PGB150P significantly enhanced Cd(Ⅱ) adsorption (403.84 mg/g) while retaining high Pb(Ⅱ) adsorption performance (1271.13 mg/g). In the Cd(Ⅱ)–Pb(Ⅱ) system, the adsorption capacities of Cd(Ⅱ) and Pb(Ⅱ) decreased to 170.17 and 980.05 mg/g, respectively, with Pb(Ⅱ) showing a stronger competitive advantage. Adsorption followed the Langmuir model and was spontaneous endothermic chemisorption controlled by film and intraparticle diffusion. PGB150P exhibited a broad pH adaptability and strong resistance to common coexisting ions. Mechanistic quantification revealed that Cd(Ⅱ) removal by PGB150P was dominated by precipitation (58.56%) and ion exchange (34.7%), while Pb(Ⅱ) removal was also dominated by precipitation (62.41%) and ion exchange (31.7%). Soil immobilization experiments showed that after 28 days treatment, PGB150P increased the residual fractions of Cd and Pb to 29% and 50%, respectively. In the Cd-Pb system, the residual fractions were increased to 15% and 49%, respectively. After five adsorption–desorption cycles, PGB150P retained about 71% of its Cd(Ⅱ) capacity and 93% of its Pb(Ⅱ) capacity. Cost analysis indicated a total cost of only 39–54 USD/t, substantially lower than that of adsorbents prepared with exogenous phosphate or commercial hydroxyapatite. This study provides theoretical support for the high-value utilization of construction solid waste and the remediation of multi-media heavy metal pollution. Efficient removal and immobilization of arsenic from smelting wastewater through the formation of amorphous ferric arsenate minerals under ambient-pressure hydrothermal conditions

Yongkui Li,Qiqi Gong,Nianyi Zao,Guizhi Yan,Guohua Li,Xianjin Qi

Arsenic removal from waste acid using ferric oxide as iron source. • Amorphous ferric arsenate was formed under ambient-pressure hydrothermal conditions. • Prospective process for the effective treatment of arsenic-containing wastewater. Owing to its high toxicity and environmental mobility, high‑arsenic smelting wastewater (HSW) poses a persistent risk to ecological stability and human health. In this study, an effective method was proposed to remove and immobilize arsenic from HSW by using ferric oxide as a solid iron source to form amorphous ferric arsenate (AFA). Initially, ferric oxide was pre-dissolved in HSW to release Fe 3+ ions, while simultaneously removing 35.5% of arsenic. During the ambient-pressure hydrothermal precipitation process, the dissolved Fe 3+ reacted with arsenic to form AFA precipitate. Notably, additional H + ions generated during this reaction promoted the continuous dissolution of ferric oxide, establishing a self-sustaining dissolution-precipitation cycle. Under optimized conditions, including a pre-dissolution time of 300 min, a Fe/As molar ratio of 4, an initial pH of 1.5, a reaction temperature of 90 °C, and a reaction time of 24 h, the arsenic concentration decreased from an initial 8276 mg/L to 585 mg/L, corresponding to a removal efficiency of 92.9%. A subsequent treatment process, involving adsorption onto fresh ferric oxide followed by lime neutralization of the post-precipitation supernatant, successfully reduced the final arsenic concentration to below 0.08 mg/L and ensured that all other heavy metal ions met industrial wastewater discharge standards. Moreover, compared with direct one-step neutralization, the proposed method first removed most arsenic as an AFA precipitate under acidic conditions, thereby reducing the amount and hazard potential of the final neutralization sludge. This study demonstrates that the hydrothermal method holds significant potential for the large-scale treatment of HSW. Download: Download high-res image (273KB) Download: Download full-size image

High-Value Utilization of Phosphate Tailings: Phosphorus Recovery and In-Situ Synthesis of Layered Double Hydroxides for Stable Immobilization of Heavy Metals

Jinjia Du,Bin Dong,Ping Ning,Zuxin Xu

Acid leaching-precipitation-synthesis enables high-value utilization of PT. • 99% of P in the leachate was recovered as white hydroxyapatite via pH adjustment. • PLDH exhibits exceptional mineralization capacities for Cd 2+ , Cu 2+ , Zn 2+ , and Pb 2+ . • Mineralization mechanism involves isomorphic substitution and chemical precipitation. • PLDH could effectively reduce the risks of Cd, Cu, Zn, and Pb contamination in soil. High-value utilization of phosphate tailings (PT) is of great significance for addressing environmental issues while advancing circular economy goals. This study proposed an integrated acid leaching–precipitation–synthesis strategy for high-value utilization of PT. After recovering phosphorus (P) via acid leaching and precipitation, a functional layered double hydroxide (PLDH) was successfully synthesized by recycling the remaining leachate. Results showed that the leaching efficiencies of P, calcium (Ca), and magnesium (Mg) reached approximately 100% at 25 °C using 0.4 M HCl at a liquid-to-solid ratio of 50 mL/g for 60 min. The recovered P primarily occurred as hydroxyapatite, with P 2 O 5 content as high as 31.2%. The synthesized PLDH exhibited excellent mineralization performance toward cadmium, copper, zinc, and lead ions (Cd 2+ , Cu 2+ , Zn 2+ , and Pb 2+ , respectively) in aqueous solutions, with maximum capacities of 561.28, 349.65, 340.14, and 775.54 mg/g, respectively, surpassing those of most reported sorbents. Mechanistic investigations revealed that Cd 2+ , Cu 2+ , and Zn 2+ were mineralized through isomorphic substitution at Ca 2+ sites within the PLDH lattice, whereas Pb 2+ was mineralized through precipitation to form Pb 3 (CO 3 ) 2 (OH) 2 . An exploratory soil column experiment further suggested that PLDH could effectively limit heavy metal migration under soil-column conditions. After 42 d of soil remediation, 4% PLDH reduced heavy-metal bioavailability by 59.9–77.3% and TCLP-leachable concentrations by 49.5–83.6%. Wheat growth experiments further showed no apparent phytotoxicity of 4% PLDH under the tested conditions. Lastly, techno-economic analysis indicated the economic potential of the proposed strategy, providing strong support for industrial applications.
