A dual stimulus-responsive NIR-II photoacoustic probe for precise diagnosis of colon cancer and photothermal-starvation synergistic therapy

Yang Wang,Shulong Wang,Min He,Yanli Luo,Fanggui Ye,Jianniao Tian,Shulin Zhao

Integrating imaging-guided diagnoses with therapeutic strategies holds significant potential in the treatment of cancer and other diseases. Herein, we developed a dual stimulus-responsive nanoagent that is responsive to two specific stimuli, and demonstrated its application to the diagnosis and photothermal-starvation therapy of colon cancer. The elevated levels of H 2 S and glucose in colon cancer cells are used as targets for a dual stimulus-responsive, self-assembled copper-albumin-glucose oxidase@polyaniline (Cu 2 + -BSA-GO x @PANI) nanoplatform. Upon entering colon cancer cells, GO x oxidizes glucose to produce gluconic acid, which leads to the protonation of PANI and the red-shifting of the wavelength of maximum absorbance of the nano-platform from 680 nm to 780 nm. Concurrently, Cu 2+ reacts with H 2 S to form CuS, which further induces a redshift from 780 nm to 830 nm, and significantly enhancing the near-infrared II region absorption intensity and enabling photoacoustic imaging under 1064 nm laser irradiation. Additionally, the consumption of glucose within cancer cells inhibits the generation of ATP, achieving starvation therapy while reducing the expression of tumor heat-shock protein 90 to weaken tumoral thermo-tolerance, resulting in a synergistic anti-tumor effect. In vitro and in vivo analyses indicate that the Cu 2+ -BSA-GOx@PANI nano-platform possesses specific visual diagnostic and therapeutic capabilities for colon cancer, providing a novel strategy for the design of highly effective nanodiagnosis-treatment agents.

Cocktail strategy-based nanomedicine: A synergistic cascade of starvation, NIR-II photothermal, and gas therapy for enhanced tumor immunotherapy

Lianying Zhang,Xiaotong Chen,Beixian Zhou,Wei Meng,Haifeng Zeng,Yongjian Chen,Guoqin Huang,Yingshan Zhang,Huimin Wang,Ming Chen,Jinxiang Chen

Immunotherapy has emerged as a highly promising strategy in the realm of cancer treatment, wherein immunogenic cell death (ICD) is considered a potential trigger for anti-tumor immunity by inducing adaptive immunity to dying cell antigens. This process is often accompanied by the exposure, active secretion, or passive release of a large number of damage-associated molecular patterns (DAMPs), which activate dendritic cells (DCs) and enhance their antigen-presenting capacity. Subsequently, it promotes the recruitment and activation of cytotoxic T lymphocytes, ultimately leading to tumor growth inhibition. In addition, polarizing the M2 phenotype of tumor-associated macrophages (TAMs) to the M1 phenotype is another way to activate anti-tumor immunity, which can further enhance the effect of anti-tumor immunotherapy. In this study, we engineered a composite nanoparticle of UiO-66-NH 2 @Gold nanoshells@GOx-P-Arg (denoted as UGsGP). The gold nano shells in UGsGP exhibit a broad Near-Infrared-II (NIR-II) absorption to give a high photothermal conversion efficiency and achieve photothermal therapy (PTT). The GOx in UGsGP involves the breakdown of glucose, which results in a decrease in ATP levels and an inhibition of HSP90 and HSP70 production, ultimately enhancing the heat sensitivity of the tumor for PTT. In addition, GOx-mediated starvation therapy by glucose exhaustion produces a substantial amount of hydrogen peroxide (H 2 O 2 ), which can then react with P-Arg to produce intratumoral NO Thus, the synergistic effect of PTT resensitization, the photothermally-enhanced GOx-mediated starvation, and NO-based gas therapy promote the induction of ICD and the polarization of TAMs. The combination therapy exhibits significant antitumor effects both in vitro and in vivo. Statement of significance (1) Gold nanoshells on the surface of UiO-66-NH 2 display a broad absorption spectrum ranging from 900 to 1700 nm, combined with a high photothermal conversion efficiency of 74.0 %, demonstrating their remarkable ability to harness and convert light energy into heat for effective tumor ablation. (2) Under laser irradiation, GOx within the UGsGPs effectively consumes glucose, increasing intratumoral H 2 O 2 levels, which then reacts with P-Arg to produce NO within the tumor. Concurrently, the reduction in ATP levels suppresses HSP90 and HSP70 production, thereby enhancing the tumor's sensitivity to photothermal therapy. (3) The synergistic combination of NO gas therapy, starvation therapy, and PTT promotes ICD induction and TAM polarization, thereby improving the therapeutic outcomes for primary and distant tumors.

PEGMa modified molybdenum oxide as a NIR photothermal agent for composite thermal/pH-responsive p(NIPAM-co-MAA) microgels

Xiaofang Qi,Jing Peng,Dongyan Tang,Ning Wang,Hongyun Zou

Molybdenum oxide nanomaterials have received increasing attention in recent years for their strong localized surface plasmon resonance (LSPR) absorption in the near-infrared (NIR) region and pH-dependent degradability. However, the rapid degradation of MoOx at physiological pH values would decrease the NIR photothermal effect. Here, we designed and prepared PEGMa–MoOx/p(NIPAM-co-MAA) microgels (NCs) exhibiting adjustable degradation. For that purpose, PEGMa (Mn = 400) modified MoOx was synthesized through a simple hydrothermal process. Then, PEGMa–MoOx with existing CC groups was continually polymerized with NIPAM and MAA through emulsion polymerization. MAA was introduced to provide an acidic microenvironment (pH at 3–5) to control the degradation rate of PEGMa–MoOx. The photothermal efficiency of the NCs could be maintained at 54% even after 14 days. Meanwhile, the NCs also displayed a pH-dependent thermal response. Moreover, PEGMa was added again in the emulsion polymerization process to elevate the photothermal efficiency, and the irradiation result indicated that the NCs could increase the temperature by 27 °C. The complex microgel integrates pH/temperature/photothermal triple response characteristics and exhibits huge potential for chemo-photothermal combination cancer therapy.

CuS@mSiO2-PEG core–shell nanoparticles as a NIR light responsive drug delivery nanoplatform for efficient chemo-photothermal therapy

Xijian Liu,Qilong Ren,Fanfan Fu,Rujia Zou,Qian Wang,Guobing Xin,Zhiyin Xiao,Xiaojuan Huang,Qian Liu,Junqing Hu

We report a facile and low-cost approach to design a difunctional nanoplatform (CuS@mSiO2-PEG) as a near-infrared (NIR) light responsive drug delivery system for efficient chemo-photothermal therapy. The nanoplatform demonstrated good biocompatibility and colloidal stability, as well as high loading capacity for the anticancer drug (26.5 wt% for doxorubicin (DOX)). The CuS nanocrystals (core) within these CuS@mSiO2-PEG core–shell nanoparticles can effectively absorb and convert NIR light to fatal heat under NIR light irradiation for photothermal therapy, and the release of DOX from the mesoporous silica (shell) can be triggered by pH and NIR light for chemotherapy. When the CuS@mSiO2-PEG/DOX nanocomposites were irradiated by 980 nm light, both chemotherapy and photothermal therapy were simultaneously driven, resulting in a synergistic effect for killing cancer cells. Importantly, compared with chemotherapy or photothermal treatment alone, the combined therapy significantly improved the therapeutic efficacy.

Development of a pH/NIR/temperature-responsive drug delivery system using AuNRs@ZnO@mPDA nanoparticles for synergistic cancer therapy

Zhen Ma,Zehua Han,Menghui Wan,Zhiqiang Wang,Minjie Gao,Yanbao Zhao,Lei Sun

In recent years, there has been a notable focus on the development and design of multifunctional drug delivery systems tailored for photothermal, photodynamic, and chemotherapy triple-modality cancer treatment. The paper presents the utilization of gold nanorods (AuNRs) for photothermal effect, zinc oxide (ZnO) for photodynamic effect, mesoporous polydopamine (mPDA) loaded with curcumin (Cur), and 1-tetradecanol (TD) as a temperature-sensitive gating switch to achieve integration of functional organic/inorganic nanomaterials. The core-shell structural nanocarriers (AuNRs@ZnO@mPDA-Cur/TD) achieved a drug loading rate of 178 mg/g. It is important to note that the pH-responsive release properties of Cur have been observed due to the acid response of mPDA. Furthermore, the photothermal effect of AuNRs and mPDA facilitates the thermal-induced melting of TD and subsequent release of Cur upon near-infrared (NIR) irradiation, thereby achieving an effective integration of photothermal therapy and chemotherapy. Simultaneously, under NIR excitation, AuNRs inject thermal electrons into the conduction band of ZnO, leading to the generation of singlet oxygen species that confer remarkable photodynamic healing capabilities to the as-prepared carriers. The combined treatment involving photothermal, photodynamic, and chemotherapy demonstrated synergistic effects and superior efficacy compared to individual treatments. The core-shell nanodrug carrier AuNRs@ZnO@mPDA developed in this study exhibits pH-, NIR-, and temperature-responsive drug release properties, highlighting the synergistic impact of photothermal, photodynamic, and chemotherapy on tumor therapy. This research provides a novel concept and preparation strategy for a multifunctional smart drug delivery platform with integrated therapeutic efficacy.

Generating Immunological Memory Against Cancer by Camouflaging Gold-Based Photothermal Nanoparticles in NIR-II Biowindow for Mimicking T-Cells

Xuefan Xiong,Ying Zhang,Xinqi Huang,Shusheng Zhang,Qiong Li

Photothermal therapy (PTT) against cancer not only directly ablates tumors but also induces tumor immunogenic cell death (ICD). However, the antitumor immune response elicited by ICD is insufficient to prevent relapse and metastasis because of the immunosuppressive tumor microenvironment (TME). A biomimetic nanoplatform (bmNP) mimicking cytotoxic lymphocytes (CTLs) for combinational photothermal-immunotherapy to effectively regulate the immunosuppressive TME is reported here. The bmNP is constructed by wrapping the T -cell membrane onto a new type of photothermal agents, spherical Au-based PNCs (sAuPNCs). Similar to T -cells, the bmNP enhanced accumulation at the tumor site by targeting the tumor via adhesion proteins on T -cell membrane. The obtained sAuPNCs have a wide absorption band in the second near-infrared (NIR-II) region with a high photothermal conversion efficiency (PCE) up to about 75% and excellent photostability. The bmNP with a smaller size is more superior compete with T -cells to bond with tumor cells via PD-1/PD-L1 interaction to effectively block the PD-1 checkpoint of T -cells for preventing T -cell exhaustion. Furthermore, in vivo studies reveal the immunological memory effect is significantly elicited in mice received bmNPs therapy. Collectively, bmNPs show great potential in photothermal-enhanced immunotherapy.

A novel KSrScSi2O7:xCr silicate phosphor, luminescence/chromaticity, and applications in NIR pc-LED and stable green ceramic pigments

Sihan Yang,Xuyan Xue,Feng Jiang,Liang Dong,Qi Zhu,Xuejiao Wang,Ji-Guang Li

The emission wavelength of current near-infrared phosphors activated by Cr 3+ is generally smaller than 900 nm in near-infrared (NIR) I region, and it is extremely challenging and is of great practical significance to realize emission towards NIR-II region. In this study, a novel Cr 3+ excited KSrScSi 2 O 7 silicate broad-band phosphor was prepared using the traditional solid-state method. Cr 3+ resides in a weak crystal field in the KSrScSi 2 O 7 lattice and exhibits broad-band near-infrared emission at 984 nm, longer than those of most Cr 3+ activated phosphors, under 493 nm blue light excitation. This is due to the strong charge polarization caused by the unique local coordination environment of the silicate matrix, which leads to a reduction in the crystal field splitting energy of the [ScO 6 ] octahedron where Cr 3+ is located and the downward shift of the 4 T 2 energy level. The optimal doping concentration of Cr 3+ is found to be 2 mol%, and the quenching mechanism is dipole-dipole interaction. Compared to the phosphors with similar emission wavelengths ( λ em > 900 nm), KSrScSi 2 O 7 :Cr 3+ demonstrates outstanding advantages in various aspects of luminescent performance. The fabricated phosphor-converted light-emitting diode (pc-LED) is shown to have the potential for night vision and non-invasive imaging. Novel application of KSrScSi 2 O 7 : x Cr 3+ as stable green ceramic pigments is also explored. The KSrScSi 2 O 7 : x Cr 3+ powders show a bright yellowish green appearance, and the KSrScSi 2 O 7 :0.02Cr 3+ typical composition has chromaticity values of L* = 82.73, a* = –8.53, b* = 7.97. Remarkably, the glazing samples using KSrScSi 2 O 7 : x Cr 3+ as pigments well retain the bright color after 1200 °C sintering in different atmospheres. Therefore, multi-functional applications of KSrScSi 2 O 7 : x Cr 3+ for near-infrared spectroscopy and as ceramic pigments are achieved in this work.

Platinum Nanoparticles Regulated V2C MXene Nanoplatforms with NIR-II Enhanced Nanozyme Effect for Photothermal and Chemodynamic Anti-infective Therapy

Xiaojun He,Ya Lv,Yanling Lin,Hong Yu,Yipiao Zhang,Yuhua Tong,Chunwu Zhang

Given the challenge of multidrug resistance in antibiotics, non-antibiotic-dependent antibacterial strategies show promise for anti-infective therapy. V 2 C MXene-based nanomaterials have demonstrated strong biocompatibility and photothermal conversion efficiency (PCE) for photothermal therapy (PTT). However, the limitation of V 2 C MXene's laser irradiation to the near-infrared region I (NIR-I) restricts tissue penetration, making it difficult to achieve complete bacterial eradication with single-effect therapeutic strategies. To address this, Pt nanoparticles (Pt NPs) were attached to V 2 C, forming artificial nanoplatforms (Pt@V 2 C). Pt@V 2 C exhibited enhanced PCE (59.6%) and a longer irradiation laser (NIR-II) due to the surface plasmon resonance effect of Pt NPs and V 2 C. Notably, Pt@V 2 C displayed dual enzyme-like activity with chemodynamic therapy (CDT) and NIR-II enhanced dual enzyme-like activity. The biocatalytic mechanism of Pt@V 2 C was elucidated using density functional theory. In an in vivo animal model, Pt@V 2 C effectively eliminated methicillin-resistant Staphylococcus aureus from deep-seated tissues in subcutaneous abscesses and bacterial keratitis environments, accelerating abscess resolution and promoting wound and cornea healing through the synergistic effects of PTT/CDT. Transcriptomic analysis revealed that Pt@V 2 C targeted inflammatory pathways, providing insight into its therapeutic mechanism. This study presents a promising therapeutic approach involving hyperthermia-amplified biocatalysis with Pt NPs and MXene nanocomposites. This article is protected by copyright. All rights reserved

Noninvasive Detection and Monitoring of the Integrity of the Intestinal Barrier through NIR-II Fluorescence Imaging and Colorimetric Urinalysis

Li Zhang,Lei Feng,Lin Yang,Hao Wan

The variation of the integrity of the intestinal barrier is closely related to the occurrence and progression of various diseases, making its accurate detection and monitoring essential to provide reliable information for guiding the refinement of treatment scenarios. Herein, we developed a strategy utilizing glutathione-capped gold clusters (Au-GSH) to achieve simultaneous in vivo second near-infrared (NIR-II) fluorescence imaging and in vitro colorimetric urinalysis for noninvasive detection and monitoring of intestinal barrier integrity. After oral administration, Au-GSH can demonstrate different distribution behaviors in terms of the variation of intestinal barrier integrity. Specifically, Au-GSH could selectively permeate through compromised intestinal barrier to be distributed into the bladder in view of its ultrasmall size below the glomerular filtration cutoff, leading to the rapid excretion through urine. Such process can be visually profiled by collecting NIR-II fluorescence (>1100 nm) emitting from Au-GSH in a noninvasive real-time manner. By virtue of the peroxidase-like activity of Au-GSH, the simple colorimetric urinalysis was further established for evaluating the integrity of the intestinal barrier, advancing the detection and monitoring performance. As a result, our developed strategy successfully detected the damage of intestinal barrier integrity in ulcerative colitis (UC) mice, a common disease characterized by compromised intestinal barrier integrity. Excitingly, the monitoring of restoration of intestinal barrier integrity in both therapeutic and preventative modes for UC mice was also realized by our strategy, making it a promising diagnosis scenario for diseases related to the variation of intestinal barrier integrity.

NIR-II-amplify high-entropy MXene-based sonosensitizer as sonodynamic therapy promotes methicillin-resistant Staphylococcus aureus–Infected wound healing

Wei Song,Danyan Wang,Shuai Xiao,Xiaojun He,Wei Xiong,Jianliang Shen

The growing pace of two-dimensional (2D) transitional metal carbides and nitrides (MXenes) has become a spotlight for recent research. Similarly, the need to close the gap left by conventional antibiotics, fighting off bacterial infections, and drug resistance has become eminent. Especially, MXenes are widely used as excellent photothermal agents. However, there are limited reports on MXenes as sonosensitizer. Herein, a high-entropy MXene (HEM), TVNMC was fabricated for the generation of reactive oxide species (ROS) against Methicillin-resistance Staphylococcus aureus (MRSA). Interestingly the energy storage and conversion from the diverse composition of the TVNMC MXenes allowed the generation of ROS under US irradiation and even enhanced under Photothermal therapy. This combinatorial therapy undertook a near-total kill and eradication of MRSA with its biofilm. Moreover, TVNMC MXene simultaneously eliminated MRSA abscesses while promoting wound healing with negligible anomaly to murine tissues under sonodynamic and photothermal therapy, making the TVNMC MXene a promising sonosensitizer for possible biomedical usage. Overall, this system provides great potential for the use of high-entropy MXenes as a new type of sonosensitizer in anti-infection therapy.

NIR-triggering cobalt single-atom enzyme switches off-to-on for boosting the interactive dynamic effects of multimodal phototherapy

Dai Hao,Han Ali,Wang Xijun,Zhu Peng,Wang Dingsheng,Wang Yuguang

Noninvasive phototherapy with functional preservation is considered to be a promising cancer therapeutic method. However, the clinical application of tumor phototherapy is severely restrained by the lack of appropriate multimodal phototherapy agents exhibiting an ideal tissue penetration depth to maximize the antitumor efficiency as well as to maintain important tissue functions. Herein, an innovative near-infrared ray (NIR)-triggered photodynamic-photocatalytic-photothermal therapy (PDT-PCT-PTT) agent based on an atomically dispersed cobalt single-atom enzyme (Co-SAE) anchored on hollow N-doped carbon sphere (HNCS) has been strategically developed. Reactive oxygen species (ROS) are highly activated and amplified through both the photogenerated electrons and the photothermal conversion induced by NIR irradiation, as systematically demonstrated by the experimental and density functional theory (DFT) calculation results. Mild hyperthermia is eventually achieved through apoptosis and ferroptosis caused by ROS, significantly boosting the interaction of ROS dynamic effects and thermodynamic effects in the tumor microenvironment (TME). More importantly, Co-SAEs/HNCS not only causes multimodal damage through limited TME products but also preserves important organ functions by the induction of mild local hyperthermia. This work expands the biomedical application field of SAEs and presents an innovative all-in-one, multimodal concept for the noninvasive treatment of head and neck cancer.

Phototheranostic LPP-QDs-IR-820 Nanocomposites for Specific NIR-II Imaging of Lymphatic and Photothermal Therapy of Cervical Tumors

Lijun Zhu,Zhong Du,Jiabao Xiong,Hui Li,Chi Zhang,Xueliang Zhang,Nuernisha Alifu,Biao Dong

Precise theranostics of tumors is intricately linked to the early detection and monitoring of lymph nodes (LN) and metastases, making the targeted localization of LNs essential for tumor identification. However, designing LN-targeting probes remains a significant challenge due to issues such as lymphatic uptake, biocompatibility, and fluorescence stability. To address these challenges, near-infrared II (NIR-II) fluorescence probes are developed through meticulous analysis of LN physiological structure and passive targeting strategy for LN detection and tumor therapy. An LPP-QDs-IR-820 nanocomposite (NCs) is engineered, comprising the IR-820 molecules and ultrabright PbS@CdS quantum dots (QDs), which are encapsulated within a liposome-SH-mPEG 2000 polymer matrix. These NCs demonstrates remarkable lymphatic enrichment, facilitating real-time tracking of LN via electrostatic repulsion and extracellular matrix effects. Importantly, the NCs exhibit negligible in vivo toxicity and high biocompatibility. The intense NIR-II fluorescence emissions of IR-820 and PbS@CdS QDs confer upon the NCs a high NIR-II fluorescence quantum yield (6%). The cervical tumors and their deep microvessels are clearly observed via NIR-II fluorescence imaging. Moreover, the photothermal properties of IR-820 enable the NCs to achieve a photothermal conversion efficiency of 36.56%, leading to effective photothermal therapy in cervical tumor mice.
