Curcumin (CUR) exhibits potential inhibitory effects on tumor growth; however, its hydrophobicity and instability limit its clinical applications. In the present study, we developed CUR nanoparticles (CUR-NPs) and evaluated their biochemical characteristics. Cell uptake and proliferation were assessed using scratch and Transwell assays, respectively. Western blotting was performed to investigate the expression levels of proteins related to the NF-κB/PRL-3 signaling pathway, inflammatory response, cell proliferation, and cell migration in SKOV3 cells. Our findings showed that the blank vector was not cytotoxic to cells, allowing us to disregard any effects caused by the vector itself. CUR-NPs exhibited concentration- and time-dependent inhibitory effects on cell proliferation, surpassing those of CUR alone. Increasing the concentration of CUR-NPs resulted in a reduced cell scratch-healing ability and lower chamber migration capacity. Compared to the control group, expression levels of proteins associated with NF-κB/PRL-3 signaling pathway, inflammatory response (TNF-α and IL-6), cell proliferation (cyclin E1 and cyclin A1), as well as cell migration (N-cadherin and vimentin) were significantly elevated in the lipopolysaccharide (LPS) stimulation and NF-κB p65 overexpression groups. Conversely, E-cadherin expression was significantly decreased under these conditions. However, treatment with high concentrations of CUR-NPs effectively reversed these changes. These results highlight the significant ability of CUR-NPs to inhibit human ovarian cancer cell proliferation and migration, while suppressing inflammatory responses through the regulation of the NF-κB/PRL-3 signaling pathway.

In Ca 2+ -induced emulsion gels, rapid Ca 2+ release leads to uneven gel structures, impairing curcumin (Cur) protection and delivery. To overcome this issue, a novel approach combining low-temperature heating (40 °C) and mild acidic environments (pH 6.5) was proposed to gradually releas Ca 2+ from CaCO 3 . This strategy significantly improves the mechanical properties and stability of flaxseed protein (FP)-chitosan (CS) emulsion gels by forming a uniform and dense network structure. At a CaCO 3 concentration of 15 mg/mL, the gel exhibited optimal performance, increasing Cur's photostability and storage stability by 81.3 %, 61.6 %, and 54.8 %, respectively, compared to the control (Cur-oil). Increasing the CaCO 3 concentration also inhibited the release of Cur and free fatty acids, reducing bioaccessibility during in vitro digestion. Overall, this study provides new insights into the stability of bioactive compounds and expands the application of CaCO 3 -induced emulsion gels as delivery systems in the food industry.

Curcumin (CUR) is a natural polyphenol that holds promise for treating ulcerative colitis (UC), yet oral administration of CUR exhibits limited bioavailability and existing formulations for oral delivery of CUR often suffer from unsatisfactory loading capacity. This study presents hydroxyethyl starch-curcumin microspheres (HC-MSs) with excellent CUR loading capacity (54.52 %), and the HC-MSs can further encapsulate anti-inflammatory drugs dexamethasone (DEX) to obtain a combination formulation (DHC-MSs) with high DEX loading capacity (19.91 %), for combination therapy of UC. The microspheres were successfully engineered, retaining the anti-oxidative and anti-inflammatory activities of parental CUR and demonstrating excellent biocompatibility and controlled release properties, notably triggered by α-amylase, facilitating targeted drug delivery to inflamed sites. In a mouse UC model induced by dextran sulfate sodium, the microspheres effectively accumulated in inflamed colons and both HC-MSs and DHC-MSs exhibited superior therapeutic efficacy in alleviating UC symptoms compared to free DEX. Moreover, mechanistic exploration uncovered the multifaceted therapeutic mechanisms of these formulations, encompassing anti-inflammatory actions, mitigation of spleen enlargement, and modulation of gut microbiota composition. These findings underscore the potential of HC-MSs and DHC-MSs as promising formulations for UC, with implications for advancing treatment modalities for various inflammatory bowel disorders.

High internal phase emulsions (HIPEs) are widely used to encapsulate unsaturated fats and fat-soluble bioactive ingredients in food, nutraceutical, cosmetic, and pharmaceutical applications. This study successfully developed stable emulsion gels by substituting water with glycerol in the whey protein isolate (WPI) aqueous phase, using glycerol mass percentages of 30%, 60%, and 90%. Increasing the glycerol ratio resulted in aggregation and an increase in particle size of whey isolate proteins, as well as enhanced protein hydrophobicity. Notably, the interfacial tension between the WPI/glycerol mixture and fish oil decreased with higher glycerol content. While a higher proportion of glycerol resulted in increased adsorption of interfacial proteins in HIPEs, it also led to a decrease in the concentration of interfacially adsorbed proteins due to the larger surface area. Additionally, higher glycerol ratios improved the thermal, centrifugal, and freeze-thaw stability of the HIPEs. The HIPEs fabricated with the WPI/glycerol aqueous phase exhibited reduced oxidation of unsaturated oils and enhanced gastrointestinal stability and bioaccessibility of loaded curcumin compared to pure fish oil. The study offers a novel approach for the straightforward preparation of stable protein-based HIPEs, providing a new strategy for oxidative protection of unsaturated fats and enhancing the bioaccessibility of fat-soluble bioactive ingredients.

This study developed a stable nanoparticle (CUR-SPIH/SSPS) using soybean protein isolate hydrolysate (SPIH) and soybean polysaccharides (SPSS) to protect curcumin (CUR) from degradation during storage and exposure to light and heat conditions, achieving controlled release. The SPIH to SPSS mass ratio of 5:1 gave the CUR-SPIH/SPSS nanoparticles with the highest CUR encapsulation efficiency (95.60 ± 3.00 %) and the strongest antioxidant capacity (90.26 ± 2.42 % and 66.78 ± 1.89 % for ABTS •+ and DPPH radical scavenging ability, respectively), and CUR was successfully encapsulated within the CUR-SPIH/SPSS as evidenced by X-ray diffraction. FTIR and fluorescence spectroscopy analysis confirmed that the interactions in CUR-SPIH/SPSS are primarily driven by electrostatic, hydrogen bonding, and hydrophobic interactions. Moreover, the CUR-SPIH/SPSS nanoparticles significantly enhanced CUR's thermal and UV light stability. The UV degradation kinetics showed that the half-life of CUR-SPIH/SPSS (247.55 min) was 1.61 times longer than that of free CUR (154.03 min). The release rate of CUR incorporated into CUR-SPIH/SPSS was significantly delayed during in vitro gastrointestinal digestion. This study introduces an innovative nanoparticle strategy for the stable delivery of lipophilic compounds.

The use of antibacterial hydrogel with biocompatibility has gained attention in wound healing due to the proliferation of drug-resistant microorganisms caused by antibiotic misuse, which slows down the healing process of skin wounds. This research employed a hydrogel that responds to both pH and near-infrared (NIR) in order to facilitate the healing of infected wounds. The dual-network hydrogel was successfully produced by utilizing the freeze–thaw method, which consists of secure components like ammonium alginate (AA) and polyvinyl alcohol (PVA). Polydopamine-curcumin (PDA-Cur) nanoparticles were prepared and loaded into the hydrogel. The medications could come out of the AA/PVA@PDA-Cur hydrogel (Gel-PC) when required under NIR laser irradiation. The incorporation of Curcumin provided the hydrogel with excellent antibacterial capabilities. Meanwhile, NIR radiation activated the photothermal PDA nanoparticles in the Gel-PC, generating localized thermal therapy to eliminate bacteria. In vivo treatment of mice on a Staphylococcus aureus -infected alloplastic defect model demonstrated that the Gel-PC had excellent antibacterial and wound-healing properties, leading to a reduction in inflammation. Gel-PC stimulated angiogenesis, collagen deposition, and the development of granulation tissue. Consequently, Gel-PC offered a tactical alternative for creating antibacterial wound dressings in conjunction with photothermal therapy and may find use in the treatment of infected wounds.

This study aimed to investigate the protective effect of curcumin against carbon tetrachloride (CCl4)-induced acute liver injury in a mouse model, and to explain the underlying mechanism. Curcumin at doses of 50, 100 and 200 mg/kg/day were administered orally once daily for seven days prior to CCl4exposure. At 24 h, curcumin-attenuated CCl4induced elevated serum transaminase activities and histopathological damage in the mouse’s liver. Curcumin pre-treatment at 50, 100 and 200 mg/kg significantly ameliorated CCl4-induced oxidative stress, characterized by decreased malondialdehyde (MDA) formations, and increased superoxide dismutase (SOD), catalase (CAT) activities and glutathione (GSH) content, followed by a decrease in caspase-9 and -3 activities. Curcumin pre-treatment significantly decreased CCl4-induced inflammation. Furthermore, curcumin pre-treatment significantly down-regulated the expression of TGF-β1 and Smad3 mRNAs (bothp< 0.01), and up-regulated the expression of nuclear-factor erythroid 2-related factor 2 (Nrf2) and HO-1 mRNA (bothp< 0.01) in the liver. Inhibition of HO-1 attenuated the protective effect of curcumin on CCl4-induced acute liver injury. Given these outcomes, curcumin could protect against CCl4-induced acute liver injury by inhibiting oxidative stress and inflammation, which may partly involve the activation of Nrf2/HO-1 and inhibition of TGF-β1/Smad3 pathways. Keywords:curcumin;acute liver injury;Nrf2/HO-1 pathway;oxidative stress;TGF-β1/Smad3 pathway

The purpose of this study was to fabricate composite nanoparticles using soy protein isolate (SPI) and sorghum bran arabinoxylan (AX) for the delivery of curcumin (Cur). The influences of AX concentrations on the physicochemical characteristic, stability and bioaccessibility of curcumin were investigated. The findings showed that the encapsulation efficiency of curcumin obviously increased upon incorporating AX in comparison to SPI-Cur particles. Hydrogen bonds and hydrophobic interactions were the primary driving forces for the formation of SPI-Cur-AX nanoparticles (SCA). SCA nanoparticles with 1.00 % AX exhibited a uniform size with orderly distribution, suggesting its remarkable physical stability due to the strengthened electrostatic repulsion. However, excessive AX led to aggregation of particles, a noticeable increase in size, and subsequently, a reduction in stability. Due to the heightened free radical scavenging capacity of sorghum AX, SCA nanoparticles exhibited superior antioxidant capabilities. Compared to free curcumin, encapsulation within composite particles significantly enhanced the retention rate and bioaccessibility of curcumin. This improvement was attributed to the potent emulsification ability of AX, which coordinated with bile salt to promote the transfer of curcumin into micelles. The research provides an effective strategy for developing food-grade delivery carriers aimed at enhancing dispersibility, stability and bioaccessibility of the fat-soluble bioactives.

Curcumin, a natural phenolic compound with various bioactivities, is easily decomposed into stability ferulic acid and vanillin molecules. Whether the functional equivalence of decomposition products can match that of curcumin is a matter worth attention. In this work, the inhibition and interaction of curcumin and its decomposition products on pepsin were studied. The inhibition by curcumin and ferulic acid belonged to competitive inhibition, the quenching types were static quenching with spontaneous process. Binding energies of curcumin, ferulic acid and vanillin were −8.33 ± 0.14, −7.88 ± 0.09 and −6.12 ± 0.08 kcal/mol, respectively, through H-bonds and hydrophobic interactions. All molecules significantly changed the secondary structure of pepsin and reduced the deformation temperature about 15 °C. The results show that better inhibition on pepsin from curcumin than ferulic acid, whereas vanillin did not show inhibition. The results provided the experimental basis for curcumin as potential inhibitors of pepsin in special diets.

Curcumin (Cur) exhibits diverse natural pharmacological activities, despite its limited water solubility (hydrophobicity) and low bioavailability. In this investigation, a valine-curcumin conjugate (Val-Cur) was synthesized through amino acid side chain modification, and its solubility increased to 1.78 mg/mL. In vitro experimental findings demonstrated that the antibacterial activity of Val-Cur against Escherichia coli , Staphylococcus aureus , Aeromonas hydrophila , and Vibrio parahaemolyticus was significantly superior to that of Cur. The inhibition rate of Val-Cur against HepG2 (human hepatocellular carcinoma) cells was higher than that of Cur at low concentrations (below 25 μmol/L), although the IC 50 value of Val-Cur did not differ significantly from that of Cur. In vivo biological effects of Val-Cur were assessed by adding it into the feed (150 mg/kg) of American eels ( Anguilla rostrata ). Val-Cur significantly improved the growth performance (↑weight gain rate, ↑specific growth rate, and ↓feed conversion rate) and activities of intestinal digestive enzymes (amylase and lipase) and antioxidant enzymes (superoxide dismutase) in American eels. Additionally, Val-Cur significantly improved serum biochemical indices (↑high-density lipoprotein cholesterol, ↓low-density lipoprotein cholesterol, ↓aspartate and alanine aminotransferases). Furthermore, Val-Cur increased intestinal microbial diversity, reduced the abundance of potentially pathogenic bacteria ( Spiroplasma , Clostridium , and Pseudomonas ), and elevated the abundance of beneficial digestion-promoting bacteria ( Romboutsia , Phyllobacterium , Romboutsia sedimentorum , and Clostridium butyricum ) conducive to glucose metabolism ( P < 0.05). To the best of our knowledge, this study is the first to explore water-soluble curcumin in aquaculture, and the findings will lay the groundwork for the potential application of water-soluble curcumin in the field of aquaculture.

To elucidate the effects of different cross-linking methods on the structural and functional properties of ternary complexes, this study systematically engineered β-lactoglobulin-hyaluronic acid-neochlorogenic acid (β-Lg-HA-3CQA) complexes using six distinct strategies: non-covalent cross-linking, ultrasonic-assisted treatment, enzymatic grafting, free radical cross-linking, genipin conjugation, and the Maillard reaction. Structural and functional analyses demonstrated that covalent strategies, significantly outperformed non-covalent and ultrasonic treatment in terms of structural integrity and functional efficacy. Notably, the Maillard-enzymatic approach exhibited the optimal performance, with a solubility of 93.71 %, 3CQA binding content of 73.05 μg/mL, and curcumin entrapment efficiency of 86.55 %. It also significantly enhanced the system stability, as well as its thermal and environmental tolerance. In contrast, ultrasonic treatment offered only transient improvements in dispersion and failed to overcome the intrinsic structural instability of non-covalent complexes. This systematic comparison of multiple cross-linking methods provides both a theoretical framework and practical foundation for optimizing the protein-polysaccharide-polyphenol complexes, thereby enhancing their functional properties and broadening their applicability in food and pharmaceutical fields.

Curcumin, a bioactive compound characterized by its limited solubility in water and low bioaccessibility, which requires an effective delivery system to enhance stability and absorption. This research focused on creating Pickering emulsions stabilized by zein-adzuki bean seed coat polyphenol-sodium alginate composite nanoparticles (ZAS) to improve curcumin delivery. The effects of different concentrations of sodium alginate (SA) on the properties of ZAS were investigated. Fourier transform infrared spectroscopy (FTIR) showed that there were hydrophobic and hydrogen bonds interactions between SA and nanoparticles, and Differential Scanning Calorimetry demonstrated an improvement in thermal stability. Accelerated oxidation experiments showed that ZAS stabilized Pickering emulsions had good anti-oxidant capacity. In comparison to the control group, the addition of SA increased the loading rate of curcumin to 81.65 ± 1.56 % with improved bioaccessibility (27.99 ± 0.26 %) and in vitro digestion stability (55.72 ± 0.22 %). The findings indicate that ZAS nanoparticles modified with SA markedly enhance the stability of Pickering emulsions, while also attaining a high loading efficiency of 81.65 % and bioaccessibility of 27.99 %. This suggests their potential utility in functional food applications.
