BACKGROUND The Maillard reaction involves the interaction of various amino acids and reducing sugars, resulting in food browning. It often produces appealing aromas and flavors. The complexities of the reaction are such that it can be challenging to identify the often numerous and frequently volatile products formed by it. In the present study, we sought to identify and evaluate an unusual product with anti-oxidant activity arising from a fructose-histidine Maillard reaction model. The anti-oxidant profile of this product was assessed by computational means. RESULTS The fructose-histidine Maillard reaction products (FH-MRPs) were generated by heating a 2:1 mixture of the sugar and the amino acid at 140 °C for 2 h. Chromatographically separable fractions, labelled DM-1 to DM-8, were obtained using silica gel as the stationary phase and dichloromethane/methanol (DCM/MeOH) mixtures as the mobile one. Fraction DM-5 exhibited the highest 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity, and further bio-assay guided fractionation led to isolation and identification of 1-(1 H -imidazo[4,5- c ]pyridin-4-yl)ethenone (IMPE) as the active principal, the structure of which was established by nuclear magnetic resonance (NMR) spectroscopic and mass spectral techniques. A mechanism for the formation of IMPE from its precursors is proposed. Density functional theory (DFT) calculations suggest this novel heterocyclic compound exerts its anti-oxidant effects by interacting with DPPH and 2,2'-Azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radicals. Essentially, IMPE was non-toxic below 300 ug mL −1 , showing a concentration-dependent free radical clearance capacity and reducing power within the 100–1000 μg mL −1 range, and moreover, exhibiting significant Fe 2+ chelating abilities wihin the 50-200 μg mL −1 range. CONCLUSION This study identified the unique FH-MRP, IMPE, and found that it acts as food antioxidant through the chelation of metal ions. © 2024 Society of Chemical Industry.

Resveratrol (RES) is a natural non-flavonoid polyphenolic antioxidant. The low solubility of RES in water (26 mg/L) and oil (175.51 mg/kg) limited its antioxidant activity and bioavailability. In order to overcome these issues, in this work, a lipophilic resveratrol laurate was efficiently synthesized using RES as starting material in the presence of enzymes. The catalytic activities of enzymes (Lipase CALA, Lipozyme TL 100L, Lipase CALB, Novozym 435, Lipozyme TLIM, and Lipozyme RMIM) were investigated and screened. Moreover, reaction conditions were optimized using response surface methodology. Meanwhile, thermodynamic and kinetic analyses were also conducted to investigate the enzymatic mechanism. Results showed that, Lipozyme RMIM showed the best performance for resveratrol monolaurates regioselective synthesis. Two products, 3-resveratrol monolaurate (3-RC12) and 4′-RC12, were prepared and purified. The optimal reaction conditions for the preparation of 3-RC12, and 4′-RC12 were 1:65 substrate ratio and 11.7 wt% lipase dosage at 75 °C for 33.2 h. Under these conditions, the RES conversion reached a maximum of 98.04 ± 1.76%. The regioselective products were analyzed for antioxidant activity. Notably, the esterification site significantly impacts resveratrol monoesters' antioxidant activity. And 3-RC12 demonstrates superior ABTS and DPPH radical cation scavenging activity, surpassing RES and 4′-RC12. Notably, in yield and antioxidant activity, 3-RC12 is better than 4′-RC12, which make it a potential antioxidant for industrial application.

Abdominal wall defects caused by trauma, congenital rupture, and intra-abdominal infection remain challenging due to the large wound area and complex complications. Herein, an assembled mesenchymal stem cell (MSCs)-laden granular hydrogel (termed assembled GSD@FPs), loaded with basic fibroblast growth factor (bFGF) and platelet-derived growth factor (PDGF), is developed. This hydrogel is constructed through dynamic covalent cross-linking ( via borate ester bonds) among dopamine-grafted gelatin methacrylamide (GelMA-DA), phenylborate-modified hyaluronic acid (HA-PBA), and epigallocatechin-3-gallate (EGCG), serving as multifunctional bulk building blocks for cell delivery and abdominal wall repair. The designed assembled granular hydrogels possess good rheological properties, self-healing, injectability, and tissue-adhesion properties. Detailed in vitro cell experiments are conducted, revealing that the GSD@FPs granular hydrogels can effectively promote cell proliferation, cell migration and angiogenesis. Furthermore, in abdominal wall defects, assembled GSD@FPs significantly accelerates the tissue healing process by simultaneously inhibiting the inflammatory response, promoting collagen deposition, and promoting cell proliferation and angiogenesis. Importantly, the assembled GSD@FPs granular hydrogels can also provide mechanical support and increase the thickness of regenerated tissue (1727.8 ± 169.6 μm for the control group, 3204.2 ± 278.5 μm for the assembled GSD@FPs group at 14 d). Eventually, the GSD granular hydrogels biodegraded, facilitating tissue remodeling and generating new muscle tissues. Therefore, this study provides a promising strategy with great potential for application in abdominal wall repair.

Cellulose nanocrystals (CNCs) were obtained from the extraction and bleaching of jute cellulose as the enhancer, 2-hydroxypropyl-β-cyclodextrin (HP-β-CD) as the carrier, the flavonoids-anthocyanidins and cinnamaldehyde as the bioactive agent , and finally a novel kind of polylactic acid (PLA)-based composite membrane was derived by electrostatic spun method. With the increasing concentration, HP-β-CDs cooperated with CNCs to regulate or control the release rate of bioactive compounds , which had a synergistic effect on the performance of the PLA matrix. The mechanical strength of PLA-3.2 composite with tannic acid (TA) surface cross-linking was 29.6 % higher than neat PLA, and could also continuously protect cells from oxidative stress and free radicals . In addition, excellent cell biocompatibility was found, and attributed to the interaction between bioactive compounds and cell membrane . In addition, we also found two excellent properties from our experimental results: obvious intelligent color reaction and good antibacterial ability. Finally, PLA-3.2 composites could be degraded by soil and are conducive to plant root growth . Hence, this work could solve many of the current problems of biodegradability and functionality of biopolymers for potential applications in areas such as intelligent bioactive food packaging.

Surface modification is an important approach to improve osseointegration of the endosseous implants, however it is still desirable to develop a facile yet efficient coating strategy. Herein, a metal–phenolic network (MPN) is proposed as a multifunctional nanocoating on titanium (Ti) implants for enhanced osseointegration through early immunomodulation. With tannic acid (TA) and Sr 2+ self-assembled on Ti substrates, the MPN coatings provided a bioactive interface, which can facilitate the initial adhesion and recruitment of bone marrow mesenchymal stem cells (BMSCs) and polarize macrophage toward M2 phenotype. Furthermore, the TA-Sr coatings accelerated the osteogenic differentiation of BMSCs. In vivo evaluations further confirmed the enhanced osseointegration of TA-Sr modified implants via generating a favorable osteoimmune microenvironment. In general, these results suggest that TA-Sr MPN nanocoating is a promising strategy for achieving better and faster osseointegration of bone implants, which can be easily utilized in future clinical applications.

The treatment of diabetic chronic wounds is still faced with great challenges, mainly due to wound infection, excessive inflammation, and peripheral vascular disease in the wound area. Therefore, it is of great importance to develop a novel multifunctional hydrogel with high efficiency to accelerate diabetic wound healing. Curcumin (Cur), a Chinese herbal, has shown great potential in enhancing the healing of diabetic chronic wounds because of its immunomodulatory and pro-angiogenic properties. However, its low aqueous solubility, poor bioavailability, and chemical instability have limited its clinical applications. To address these current bottlenecks, novel poly(vinyl alcohol) (PVA)–chitosan (CS)/sodium alginate (SA)–Cur (PCSA) hydrogels were prepared for the first time, and they demonstrated all of the above intriguing performances by the Michael addition reaction of CS and Cur. PCSA hydrogels show multiple dynamic bonds, which possess strong mechanical properties (tensile stress: ∼0.980 MPa; toughness: ∼258.45 kJ/m3; and compressive strength: ∼7.38 MPa at strain of 80%). These intriguing performances provided an optimal microenvironment for cell migration and proliferation and also promoted the growth of blood vessels, leading to early angiogenesis. Importantly, the experimental results demonstrated that PCSA hydrogels can effectively transform pro-inflammatory M1 macrophages into anti-inflammatory M2 macrophages without the need for additional ingredients in vitro. Benefiting from these characteristics, a full-thickness diabetic wound in a rat model demonstrated that PCSA hydrogels can effectively accelerate wound healing via ROS-scavenging, downregulation of IL-1β, and upregulation of CD31 expression, resulting in angiogenesis and collagen deposition. This strategy not only provides a simple and safe Cur-based hydrogel for diabetic wound healing but also highlights the significant potential for the development of high-performance biomaterials for promoting diabetic wound healing using traditional Chinese medicine.

On-demand responsive hydrogels are a promising solution for effective wound management as they can adjust their properties in response to changes in the wound environment, allowing them to provide tailored support for the healing process. However, the conventional hydrogels may not fully meet the diverse demands of the intricate healing process. Herein, a novel glycyrrhizic acid (GA) based self-assembly hydrogel coordinated with copper and polyphenol (GCP hydrogel) was developed to exhibit triggered release behavior in response to the microenvironment. The GCP hydrogel coordinated with copper and protocatechuic acid (PA) and self-assembled with GA, also exhibits enzyme-like properties by mimicking the cascade process of superoxide dismutase (SOD) and catalase (CAT), effectively scavenging reactive oxygen species (ROS). Furthermore, the on-demand release of Cu 2+ at different stages of the wound healing process can not only enhance the antibacterial ability of methicillin-resistant Staphylococcus aureus (MRSA) but also intelligently promote angiogenesis with outstanding biocompatibility. In addition, the GCP hydrogel effectively modulated the activity of macrophages in response to inflammatory stimuli, exhibiting remarkable anti-inflammatory abilities and promoting tissue regeneration. The multifunctional GCP hydrogel platform has the potential to create a dynamic microenvironment that is conducive to tissue regeneration, making it an ideal candidate for smart wound management.

Kidney stones, which have high prevalence and recurrence rates, often cause severe oxidative damage and inflammation. The ultrasmall hydrodynamic diameter of nanoparticles is crucial for their enrichment in the kidneys to exert biological activity. However, integrating crystallization inhibition and therapeutic functions into a single ultrasmall nanoparticle is challenging. A novel ultrasmall iron (Fe)–gallic acid (Ga) metal–phenolic networks (Fe–Ga MPNs) is developed for treating calcium oxalate (CaOx) nephropathies. These MPNs can specifically adsorb on the high-energy ( 1 ¯ 01 $\bar{1}01$ ) crystal face to inhibit the growth of CaOx monohydrate (COM), promoting the phase transition from highly toxic COM to low-risk CaOx dihydrate. Fe–Ga MPNs have broad-spectrum free radical scavenging abilities, reducing oxidative damage and inhibiting cell apoptosis. They exhibit sensitivity toward kidney damage, accumulating in injured renal tissue, reducing tubule injury and inflammation, improving tubule function, and inhibiting crystal formation. Fe–Ga MPNs also inhibit pro-inflammatory macrophage polarization and upregulate anti-inflammatory and highly phagocytic macrophage polarization. RNA sequencing analysis shows that Fe–Ga MPNs induce transcriptomic changes mainly involving immune regulation and citrate homeostasis pathways. In conclusion, the multifunctional nanonetwork Fe–Ga MPNs, with crystallization inhibition, antioxidant, and immune regulation properties, show great potential in treating CaOx nephropathies.

The consumption of wampee has traditionally been utilized to alleviate gastrointestinal inflammation and associated disorders; however, its exact mechanism has remained unknown. The aim of this study was to elucidate the therapeutic efficacy and underlying mechanism of wampee polyphenol extract (WPE) in dextran sulfate sodium (DSS)-induced ulcerative colitis (UC). The findings revealed that WPE alleviated diverse symptoms of UC, regulated various inflammatory cytokines, and effectively protected the colon tissue structure and barrier integrity, thereby inhibiting LPS translocation. Moreover, WPE restored the richness and diversity of gut microbiota and optimized its structure at the phylum and genus levels, causing a notable improvement in short- chain fatty acid (SCFA) metabolism, particularly acetic acid, propionic acid, and butyric acid. Consequently, WPE was demonstrated to effectively suppress the LPS-induced TLR4-p38 MAPK/NF-κB signaling pathway by modulating gut microbiota and SCFA metabolism. These findings provided a theoretical basis for the use of wampee as a potential functional natural food for UC.

To better treat bacteria-infected wounds and promote healing, new wound dressings must be developed. In this study, we obtained PA@Fe by chelating iron trivalent ions (Fe 3+ ) with protocatechualdehyde (PA), which has a catechol structure. Subsequently, we reacted it with ethylene glycol chitosan (GC) via a Schiff base reaction and loaded vancomycin to obtain an antibacterial Gel@Van hydrogel with a photothermal response. The as-prepared Gel@Van hydrogel exhibited good injectability, self-healing, hemostasis, photothermal stability, biocompatibility, and antioxidant and antibacterial properties. Moreover, Gel@Van hydrogel achieved highly synergistic antibacterial efficacy through photothermal and antibiotic sterilization. In a mouse skin-damaged infection model, Gel@Van hydrogel had a strong ability to promote the healing of methicillin-resistant Staphylococcus aureus ( MRSA )-infected wounds, indicating the great potential application value of Gel@Van hydrogel in the field of treating and promoting the healing of infected wounds.

Chitosans were prepared by H 2 O 2 oxidative depolymerization from squid pens with low molecular weights (LMW) of 13,025, 7011, 4169, 2242 and 963 Da. The bile acid binding capacities and antioxidant properties of LMW chitosans were studied in vitro . LMW chitosans exhibited stronger bile acid binding capacities than that of chitosan. The scavenging ability of LMW chitosans against DPPH radicals improved with increasing concentration, and EC 50 values were below 1.3 mg/mL. The EC 50 values of LMW chitosans against hydroxyl radicals ranged from 0.93 to 3.66 mg/mL. All LMW chitosans exhibited a strong ferrous ion chelating effect and reducing power. At 1 mg/mL, the scavenging ability of chitosan-963 towards superoxide radicals was 67.76%. These results indicated that LMW chitosans which have stronger bile acid binding capacity and antioxidant activities may act as potential antioxidants in vitro .

A miscellaneous polysaccharide, PAP55–3-1, with a molecular weight of 23.03 kDa, was isolated from Periplaneta americana through extraction with dilute alkali solution, ethanol precipitation, and column chromatography purification. Structural analysis shows that PAP55–3-1 is mainly composed of five monosaccharides: galactosamine hydrochloride, glucosamine hydrochloride, galactose, glucose and mannose. Its main glycosidic bonds are: Manp-(1→, Galp-(1→, →3)-Galp-(1→, →3,6)-Manp-(1→, →2,6)-Manp-(1→, →6)-Manp-(1→, →4)-Galp-(1→, →6-Glcp-(1→, →6)-Galp-(1→, →2)-Manp-(1 →, →3,4)-Glcp-(1→, →3,6)-Galp-(1→. In vitro experiments demonstrated that PAP55–3-1 can effectively inhibit reactive oxygen species (ROS) and O 2 − production following H 2 O 2− induction. After H 2 O 2− induction, HIF-1α (hypoxia-inducible factor) was translocated in mitochondria PAP55–3-1 increased localization of HIF-1α was located on mitochondria to maintain the stability of mitochondrial function stability, thereby effectively inhibiting H 2 O 2 -induced mitochondrial oxidative damage. Additionally, PAP55–3-1 inhibited the M1 polarization of macrophages stimulated by H 2 O 2 and promoted the phenotype polarization of macrophages from M1 to M2, displaying anti-inflammatory and pro-repair properties. In vivo experimental results indicated that PAP55–3-1 promoted wound healing in mice. Immunohistochemical experiments revealed a reduction in CD68 expression and increase in CD206 expression in both positive and the high-dose polysaccharide group control group. This further demonstrated that PAP55–3-1 promotes the phenotype polarization of macrophages from M1 to M2, exerting anti-inflammatory and wound-healing activities.
