Electrochemical reduction of carbon dioxide (CO2ER) has become an effective solution to relieve the energy crisis and tackle climate change. In this study, a series of tin-based organic frameworks modified by In (Sn-MOF/Inx) were successfully synthesized via a simple hydrothermal method and explored for high formate-selective CO2ER. The pure Sn-MOF exhibits maximum formate selectivity with a faradaic efficiency (FEformate) of approximately 85.0% and a current density of 15 mA cm−2 at −1.16 VRHE, while the In (6%)-modified Sn-MOF (Sn-MOF/In6) delivers a much higher maximum FEformate (around 97.5%) and a current density of 16 mA cm−2 at −0.96 VRHE. Remarkably, the Sn-MOF/In6 exhibits a significantly larger specific surface area (183.3 m2 g−1) compared to the Sn-MOF (65.2 m2 g−1). These findings indicate that introducing In, an alien element with a slightly different outer orbital electron number from that of Sn, can significantly boost the selectivity and activity for CO2ER to formate. This study presents an efficient way to modify MOF catalysts through a well-designed introducing process.

The growth of solid tumors is inevitably accompanied by hypoxia and angiogenesis. Understanding the true state of tissue hypoxia and angiogenesis during tumor progression will help further comprehend the tumor microenvironment and cancer therapy. Here, we provided a novel strategy for the long-term tracking of tumor hypoxia and angiogenesis in cervical cancer. The hypoxia response elements (HRE)-driven luciferase (LUC) reporter gene transfected cervical cancer cell lines were constructed for hypoxia visualization. The hypoxia response activity of HRE-driven LUC was verified in HeLa cells and then monitored via the HeLa-HRE-LUC tumor-bearing mouse models by bioluminescence imaging (BLI). An oxygen-sensitive nanoparticle was constructed for glycerol monooleate cubic liquid crystal material loaded with Ir(piq) 3 /IR780 and could be used for phosphorescence-fluorescence imaging. The imaging result showed that the nanoparticles labeled the low oxygen region overlapped with the BLI signal region detected by HRE-driven LUC in vivo , which confirmed that the hypoxia visualization platform could genuinely reflect the hypoxia level in vivo . The angiogenesis in vivo was reflected by changes in deoxygenated/oxygenated hemoglobin detected by photoacoustic imaging. Besides these, anticancer drug apigenin down-regulated HIF-1α and VEGF under hypoxia conditions and inhibited the tube formation in vitro . Then, the inhibition effects were verified in the hypoxia and angiogenesis visualization model in vivo . This study confirmed a novel in vivo real-time monitoring of tumor hypoxia and angiogenesis strategy, which can provide useful information for further research on cancer progression and anticancer therapy.

Neuroinflammation is considered as an important pathological mechanism in neurodegenerative diseases. The natural isoquercitrin (IQ) was reported to have potential anti-neuroinflammatory activity. The acylation of glycoside in IQ enhanced its hydrophobicity, which was expected to enhance the protective effect against inflammation. In this study, three carboxylic acids with anti-neuroinflammatory effects including cinnamic acid, ibuprofen (IBU) and acetylsalicylic acid were introduced into the 6''-OH of IQ through the corresponding vinyl esters intermediates ( 8a-8c ). Ultimately, the acylated IQ derivatives (Compound 9a-9c ) were obtained with 35–42% yields using immobilized lipase Novozym 435 as catalyst. Subsequently, their anti-neuroinflammatory activities were evaluated in lipopolysaccharide (LPS)-induced BV2 cells. Compound 9b improved cell viability in the range of ≤50 μM and significantly decreased NO, PGE 2 production and TNF-α, IL-1β release and oxidative stress level with a concentration-dependent manner. Also, it could downregulate iNOS, COX-2, TNF-α and IL-1β expression levels, approximately 40% reduction were achieved when 15 μM compound 9b was employed. In addition, compound 9b resisted phosphorylation and degradation of IkBαs, suppressing the activation of NF-κB signaling pathway, exhibiting excellent neuroinflammatory inhibition. Moreover, the administration of compound 9b (30, 60 mg/kg) alleviated behavioral disorders and neuronal damages in LPS-induced neuroinflammatory mice. Meanwhile, the decreased TNF-α, IL-1β release, expression and the inhibited glial cells activation were obtained in compound 9b -treated group, which was superior to that of IQ or IBU. Overall, these findings demonstrated that compound 9b , formed by the introduction of ibuprofen into IQ, can serve as a novel promising therapeutic agent for anti-neuroinflammation.

Singlet oxygen (1O2) plays a pivotal role in numerous catalytic oxidation processes utilized in water purification and chemical synthesis. The spin-trapping method based on electron paramagnetic resonance (EPR) analysis is commonly employed for 1O2 detection. However, it is often limited to time-independent acquisition. Recent studies have raised questions about the reliability of the 1O2 trapper, 2,2,6,6-tetramethylpiperidine (TEMP), in various systems. In this study, we introduce a comprehensive, kinetic examination to monitor the spin-trapping process in EPR analysis. The EPR intensity of the trapping product was used as a quantitative measurement to evaluate the concentration of 1O2 in aqueous systems. This in situ kinetic study was successfully applied to a classical photocatalytic system with exceptional accuracy. Furthermore, we demonstrated the feasibility of our approach in more intricate 1O2-driven catalytic oxidation processes for water decontamination and elucidated the molecular mechanism of direct TEMP oxidation. This method can avoid the false-positive results associated with the conventional 2D 1O2 detection techniques, and provide insights into the reaction mechanisms in 1O2-dominated catalytic oxidation processes. This work underscores the necessity of kinetic studies for spin-trapping EPR analysis, presenting an avenue for a comprehensive exploration of the mechanisms governing catalytic oxidation processes.

The voltage-gated sodium channel subtype Nav1.6 is involved in the electrophysiological changes of primary sensory neurons that occur in oxaliplatin-induced neuropathic pain , but its regulatory mechanism remains unclear. In this study, Western blot , RT-qPCR, immunofluorescence staining, chromatin immunoprecipitation were used to prove the mechanism of MAPK-ERK-CREB signaling pathway participating in oxaliplatin-induced neuropathic pain by regulating Nav1.6. The results showed that p-Raf1 and p-ERK, key molecules in MAPK/ERK pathway, and Nav1.6 were significantly increased in DRGs of oxaliplatin-induced neuropathic pain rats. Inhibition of p-Raf1 and p-ERK respectively not only reduced the expression of Nav1.6 protein in DRGs of OXA rats, but also caused a decrease in Nav1.6 mRNA, which led us to further explore the transcription factor CREB regulated by MAPK/ERK pathway. Results showed that CREB was co-distributed with Nav1.6. Inhibition of CREB resulted in decreased mRNA and protein expression of Nav1.6, and alleviated oxaliplatin-induced neuropathic pain. A chromatin immunoprecipitation experiment proved that OXA caused p-CREB to directly bind to the promoter region of Scn8A, which is the encoding gene for Nav1.6, and promote the transcription of Scn8A. In summary, in this study, we found that oxaliplatin can activate the MAPK/ERK pathway, which promotes the expression and activation of CREB and leads to an increase in Scn8A transcription, and then leads to an increase in Nav1.6 protein expression to enhance neuronal excitability and cause pain. This study provides an experimental basis for the molecular mechanism of sodium channel regulation in oxaliplatin-induced neuropathic pain.

Given their substantial neutron capture cross-section, extreme hardness, and high chemical and thermal stability, boron-based materials are widely used as building blocks to protect against highly ionizing radiations such as gamma rays and neutrons. Indeed, uncontrolled nuclear radiation exposure can be highly hazardous to radiation workers and the public. In this sense, this work presents an extensive study and experimental evaluation of the nuclear shielding features of hexagonal-boron nitride (h-BN) based nanocomposite, where bisphenol-A based polybenzoxazine (BA-PBz) was used as matrix. The neutron shielding studies were carried out at the nuclear research reactor of Algeria NUR. The surface treatment of h-BN nanoparticles was confirmed by FTIR and XPS techniques. The curing behavior and the degradation phenomena of the nanocomposites were evaluated by DSC-TGA analyses. The distribution of h-BN nanoparticles within the polymer matrix was assessed by TEM and SEM. The results showed that the developed boron nitride-based nanocomposite exhibits intriguing shielding performances and good thermal stability. The DSC-TGA tests exhibit high degradation temperature that reach 279°C. The highest performances were obtained at an h-BN concentration of 7 wt%, where the macroscopic cross was found to be (Σ = 3.844 cm –1 ) with a screening ratio of (S = 96.12%), equivalent to a mean free path (λ) of 0.138 cm.

This study assessed the use of magnetic biosorbents in dispersive solid-phase extraction with a gas chromatograph-electron capture detector for the determination of trifluralin, chlorothalonil, transfluthrin, bromopropylate, and bifenthrin in water samples. To our knowledge, this is the first time that magnetic cork composites are used as an adsorbent in dispersive solid-phase extraction. The advantages of magnetic cork composites include their density regulation and high surface areas. The magnetic composites can be recovered using a magnetic field for desorption purposes, which can improve the operation process and reduce the extraction time. Moreover, the parameters affecting the extraction performances were optimized. The method has a limit of detection between 0.30 and 2.02 μg L−1. Good linearities (R2 > 0.99) were obtained in the linear range of 1.00 to 2000 μg L−1. The relative recoveries of the analytes in tap water, river, and lake water samples spiked at different concentrations ranged between 90% and 104%, and the relative standard deviations were less than 7.1%. Therefore, this study showed that Fe3O4/cork magnetic composites can be used as efficient and eco-friendly biosorbents in dispersive solid-phase extraction for the determination of pesticides in water samples. The use of these composites contributes to the current trend of green chemistry.

Anaerobic digestion (AD) of wastewater is the most promising bioprocess for organic conversion, however, phenol is toxic and resistant to anaerobic degradation. The current study compared the effect of hydrochar and granular activated carbon (GAC) on AD of phenol at four concentrations (100 mg/L, 250 mg/L, 500 mg/L and 1000 mg/L). Results demonstrated that hydrochar significantly improved the methane production rate and reduced the lag phase at all concentrations of phenol. The methane production rate was improved by about 50% at both 100 mg/L and 250 mg/L phenol, while it was raised by >160% at 500 mg/L and 1000 mg/L phenol by hydrochar. The GAC only increased the methane production rate at 500 mg/L and 1000 mg/L due to high adsorption capacity. Further, the adsorption of phenol by hydrochar had no apparent impact on the methane production rate, even though certain amounts of phenol were adsorbed. At 500 mg/L, the amount of methane produced significantly increased, so 16S rRNA transcripts sequencing and metabolomic analysis were conducted. 16S rRNA transcripts sequencing analysis indicated that hydrochar resulted in the enrichment of syntrophic bacteria (e.g., Syntrophorhabdus & Syntrophobacter ) and Methanosaeta , which might be related with direct interspecies electron transfer . Further, it was noticed that the growth of Methanobacterium was repressed at 500 mg/L phenol, while hydrochar promoted its growth. Phenol was degraded into L-tyrosine and then followed the benzoate degradation pathway for methane production as revealed by metabolomic analysis. In addition, metabolomic analysis also revealed that hydrochar promoted the degradation of all metabolites and enhanced the phenol degradation into methane.

Sulfonated polyether ether ketone (SPEEK) has been widely investigated in proton exchange membrane fuel cells (PEMFCs) due to its excellent thermal stability, chemical stability, and low cost compared with Nafion. However, excessive degree of sulfonation will easily lead to the decrease in thermal stabilities and mechanical properties of SPEEK membranes, which limits the enhancement of proton conductivity. In this work, a series of Schiff-base networks (SNWs) with different contents are in situ synthesized in the SPEEK membrane by a Schiff-base co-condensation reaction, and then, the composite membranes are soaked in sulfonic acid for further improvement of proton conductivity. The highest doping amount of the SNW filler in SPEEK can reach 20 wt %. High loading and low leaching rate of H2SO4 are easily achieved owing to the similar size between sulfuric acid molecules and micropores in SNW. Moreover, abundant amino and imine groups in SNW networks contribute to the anchoring of H2SO4 into the pores by acid–base interactions. The proton conductivity of the SPEEK/S-SNW-15 composite membrane can reach 115.53 mS cm–1 at 80 °C and 100% RH. Meanwhile, the composite membrane also exhibits satisfied stability and mechanical property.

Background Notch signaling is critical for regulating the function of vascular endothelial cells (ECs). However, the effect of the intracellular domain of Notch1 (NICD) on EC injury in sepsis remains unclear. Methods We established a cell model of vascular endothelial dysfunction and induced sepsis in a mouse model via lipopolysaccharide (LPS) injection and cecal ligation and puncture (CLP). Endothelial barrier function and expression of endothelial-related proteins were determined using CCK-8, permeability, flow cytometry, immunoblot, and immunoprecipitation assays. The effect of NICD inhibition or activation on endothelial barrier function was evaluated in vitro . Melatonin was used for NICD activation in sepsis mice. The survival rate, Evans blue dye of organs, vessel relaxation assay, immunohistochemistry, ELISA, immunoblot were used to explore the specific role of melatonin for sepsis induced vascular dysfunction in vivo . Results We found that LPS, interleukin 6, and serum collected from septic children could inhibit the expression of NICD and its downstream regulator Hes1, which impaired endothelial barrier function and led to EC apoptosis through the AKT pathway. Mechanistically, LPS decreased the stability of NICD by inhibiting the expression of a deubiquitylating enzyme, ubiquitin-specific proteases 8 (USP8). Melatonin, however, upregulated USP8 expression, thus maintaining the stability of NICD and Notch signaling, which ultimately reduced EC injury in our sepsis model and elevated the survival rate of septic mice. Conclusions We found a previously uncharacterized role of Notch1 in mediating vascular permeability during sepsis, and we showed that inhibition of NICD resulted in vascular EC dysfunction in sepsis, which was reversed by melatonin. Thus, the Notch1 signaling pathway is a potential target for the treatment of sepsis.

Background and Objectives Targeting the problems, like various residual pesticides, the high interference of sample matrix and the slow detection speed, this work studied the rapid screening method of multipesticide residues in rice by taking bensulfuron-methyl, propanil and cypermethrin as examples. Findings Rice matrix solution was obtained by using the improved QuEChERS method, matrix standard solution, and solvent standard solution were prepared to obtain the linear range and matrix effect of each pesticide. QuEChERS method could rapidly pretreat samples, and the matrix effect was soft, with matrix effect values lower than 16%. Mixed solutions of the three pesticides in four combinations were prepared within the linear range. The type of pesticides in rice was determined by support vector machine (SVM), and the pesticide concentration was predicted by partial least square regression (PLSR). SVM provided a supervised identification method for pesticide residues, making the recognition accuracy higher than 90%. The error of PLSR in predicting pesticide concentration was lower than 5.36%. Conclusions UV spectroscopy had fast test speed, so the combination of QuEChERS and UV spectroscopy could improve the efficiency of analysis. SVM combined with PLSR could preliminarily determine the type of pesticides in rice sample, and then accurately predict the concentration of pesticides. Significance and Novelty The QuEChERS sample pretreatment technology and UV spectroscopy in combination with chemometrics was expected to be a new technology for rapid detection of pesticide residues in samples.
