Rapid drug clearance and off-target effects of therapeutic drugs can induce low bioavailability and systemic side effects and gravely restrict the therapeutic effects of inflammatory bowel diseases (IBDs). Here, we propose an amplifying targeting strategy based on orally administered gallium (Ga)-based liquid metal (LM) nano-agents to efficiently eliminate reactive oxygen and nitrogen species (RONS) and modulate the dysregulated microbiome for remission of IBDs. Taking advantage of the favorable adhesive activity and coordination ability of polyphenol structure, epigallocatechin gallate (EGCG) is applied to encapsulate LM to construct the formulations (LM-EGCG). After adhering to the inflamed tissue, EGCG not only eliminates RONS but also captures the dissociated Ga to form EGCG-Ga complexes for enhancive accumulation. The detained composites protect the intestinal barrier and modulate gut microbiota for restoring the disordered enteral microenvironment, thereby relieving IBDs. Unexpectedly, LM-EGCG markedly decreases the Escherichia_Shigella populations while augmenting the abundance of Akkermansia and Bifidobacterium, resulting in favorable therapeutic effects against the dextran sulfate sodium-induced colitis.

This study optimized the process of extracting protein from black garlic using an alkaline dissolution and acid precipitation method through response surface methodology. The optimal extraction conditions were determined as a solid-to-liquid ratio of 1:50, an extraction time of 100 min, an extraction temperature of 30 °C, and an alkaline extraction pH of 9.0. Under these optimized conditions, the actual black garlic protein (BGP) extraction yield was 12.10% ± 0.21%, and the isoelectric point of the obtained BGP was 3.1. Subsequently, this study extracted black garlic protein under optimal conditions and subjected it to enzymatic hydrolysis using different enzymes (trypsin, pepsin, and their mixed enzymes). The functional characteristics, antioxidant activity, and hypoglycemic activity of black garlic protein before and after enzymatic hydrolysis were compared. Among the hydrolysates, the pepsin hydrolysate (BGPH-P) had the smallest particle size (188.57 ± 1.93 nm) and the highest Zeta potential (−29.93 ± 0.42 mV). Scanning electron microscopy showed that BGPH-P had the smallest and most dispersed particles. Fourier-transform infrared (FTIR) spectroscopy revealed that the dual enzymatic hydrolysis hydrolysate (BGPH-PT) exhibited the most stable structure. Compared to BGP, the hydrolysates demonstrated significantly improved solubility, water-holding capacity, and foaming ability ( p < 0.05), while their emulsifying activity, emulsion stability, DPPH radical scavenging capacity, and hypoglycemic activity decreased. In summary, the BGP extracted using the optimized process demonstrated good antioxidant and hypoglycemic activities, while its enzymatic hydrolysate BGPH-P exhibited excellent solubility, water-holding capacity, and emulsifying properties, providing valuable insights for the further development of black garlic protein and its hydrolysates.

The objective of this present study is to evaluate the effect of different drying methods (spray drying, freeze drying and hot-air drying) on physicochemical properties and antioxidant ability of G. lucidum extracts (GLEs) using various techniques. Meanwhile, combined with free amino acids (FAA) analysis, taste activity values (TAV) analysis and electronic tongue to investigate the relationship between the drying method and the sensory quality of GLEs. The study demonstrates that spray drying GLEs (SDE) has a high water solubility of 96.04% ± 0.10% due to its low crystallinity and hollow spherical microstructure. SDE and freeze drying GLEs (FDE) contain high levels of polysaccharides and proteins, which facilitates the formation of polysaccharide-protein chelates, thereby improving antioxidant capacity. However, SDE has higher antioxidant activity when its polysaccharide content is the same. SDE has higher thermal stability to maintain the molecular structure of GLEs. Meanwhile, FDE has a bitter FAAs content up to 65.84 ± 1.12 mg/g and TAV of Histidine is 178.70. Overall, the results suggest that spray drying is an appropriate and preferred drying method to maintain the bioactivity and bioavailability of GLEs. These findings may enhance the understanding of the GLEs flavor property and bioavailability associated with drying methods and provide a theoretical basis for G. lucidum processing.

In this study, pectin extracted from pomelo peel was investigated using three different combination methods of pulsed electric field (PEF) and cellulase. Three action sequences were performed, including PEF treatment followed by enzymatic hydrolysis, enzymatic hydrolysis followed by PEF treatment, and enzymatic hydrolysis simultaneously treated by PEF. The three corresponding pectins were namely PEP, EPP and SP. The physiochemical, molecular structural and functional properties of the three pectins were determined. The results showed that PEP had excellent physiochemical properties, with the highest yield (12.08 %), total sugar (80.17 %) and total phenol content (38.20 %). The monosaccharide composition and FT-IR analysis indicated that the three pectins were similar. The molecular weights of PEP, EPP and SP were 51.13, 88.51 and 40.00 kDa, respectively. PEP showed the best gel properties, emulsification stability and antioxidant capacity among the three products, due to its high galacturonic acid and total phenol content, appropriate protein and low molecular weight. The mechanism of PEF-assisted cellulase hydrolysis of pomelo peel was also revealed by SEM analysis. These results suggested that PEF pretreatment was the best method, which not only improved the efficiency of enzymatic extraction, but also reduced resource waste and increased financial benefits.

This study systematically investigated the antioxidant activities and phytochemical profiles of petroleum ether (PE), ethyl acetate (EtOAc), n-butanol (n-BuOH) and aqueous fractions of edible (tubers) and non-edible portions (peels) ofPachyrhizus erosus. The results showed that both the tubers and peels fromP. erosuswere rich in polyphenols and flavonoids, whereas the EtOAc fraction of peels had the highest polyphenol content, and the PE fraction of peels had the highest total flavonoid content. ABTS, DPPH, and FRAP assays revealed that both the EtOAc fraction of tubers and peels fromP. erosusshowed significant antioxidant activity, whereas the EtOAc fraction of peels possessed better antioxidant activity than that of tubers. UPLC-Orbitrap-MS/MS analysis indicated that thirty compounds were identified from the EtOAc fractions of peels and tubers, including twenty-one flavonoids, six phenolics, two coumarins, and one lignan, some of which have previously been revealed to display significant antioxidant and anti-inflammatory effects via the Nrf2-Keap1 and NF-κB signaling pathways. These findings provide robust scientific evidence for the health-promoting properties and pharmaceutical potential ofP. erosus, and its non-edible portion (peels) has great potential for use as a natural antioxidant in the food, cosmetic, and pharmaceutical industries.

Dark tea has long been recognized for its health-promoting benefits, attributed to its complex phytochemical composition. However, the specific bioactive compounds responsible for these beneficial effects remain inadequately characterized. This study aimed to explore the impact of dark tea water extract (DTE) on digestive enzyme activity, antioxidant capacity, and glucose-lipid balance. DTE was fractioned into four fractions using gel separation, followed by analysis with high-performance liquid chromatography and quasi-targeted metabolomics. The 30 % ethanol elution (EEA) and 50 % ethanol elution (EEB) fractions showed stronger antioxidant and enzyme inhibition effects compared to the whole DTE. The EEA fraction was rich in 5 catechins and 26 additional phytochemicals, while the EEB fraction contained high levels of caffeine, ECG, and 29 other phytochemicals. Notably, significant correlations were observed between quercetin-3,4′-O-di-β-glucopyranoside and 3-(2-Naphthyl)-L-alanine with digestive enzyme inhibition. Cellular studies revealed the ability of EEA and EEB to reduce lipid accumulation, improve glycolipid metabolism, and alleviate oxidative stress by increasing SOD, CAT, and GSH levels while decreasing MDA and ROS in HepG2 cells. Furthermore, 34 flavonoids, 2 alkaloids, 2 terpenes, 2 alcohols and polyols, 2 phenylpropanoids and polyketides, 1 organoheterocyclic compound were directly linked to the antioxidant activity and the modulation of glucose and lipid levels. These findings offer valuable insights into the phytochemical profiles of dark tea and its potential health benefits.

In this study, four different types of essential oil monomer and tannic acid were used to prepare different kinds of essential oil-tannin particles. As well as encapsulating the chemically labile essential oils, these particles could also be used as Pickering emulsifiers to prepare oil-in-water emulsions. These emulsions were co-stabilized using the essential oil-tannin particles in combination with chitosan. The resulting Pickering emulsions were shown to have good storage stability, viscoelastic properties, antioxidant activity , and antibacterial activity. The emulsions co-stabilized by cinnamaldehyde-tannin and chitosan had the smallest particle size and largest shear viscosity, shear modulus, and physical stability. An in vitro digestion study showed that the cinnamaldehyde encapsulated within these emulsions also had a relatively high bioaccessibility. Considerable differences were observed in the stabilities of emulsions prepared with different oil phase volume fractions. The more concentrated emulsions did not exhibit phase separation after 30 days storage, which was attributed to the fact that the close packing of the oil droplets prevented their movement. This study shows that essential oil-polyphenol particles can be used as multifunctional ingredients, such as antioxidants, antimicrobial agents, and emulsifiers.

Introduction: The root of Paeonia suffruticosa Andrews (P. suffruticosa Andr.), is a traditional Chinese medicine. Numerous studies have shown that it possesses anti-inflammatory, antioxidant, and anti-aging effects due to its rich content of bioactive compounds such as polyphenols and paeonol. Thus, it finds extensively applied in the fields of medicine and cosmetics. However, there are few reports on the photoprotective effects of P. suffruticosa Andr. root bark, this study aims to investigate its research in this area. Methods: This study utilized P. suffruticosa Andr. root bark sourced from Kunming, Yunnan Province, China. The P. suffruticosa Andr. root extract (PSAE) was obtained using AB-8 resin. The photoprotective effect of PSAE was assessed using HaCaT cells, HFF cells, and a 3D Reconstructed Human full T-Skin™ model. Mechanistic investigations were performed using RT-qPCR, WB, IF, H&E staining, Masson’s trichrome staining and IHC staining. Finally, an assessment of the effects on humans was conducted. Results: The total phenolic content in the obtained PSAE was 48.9%. Antioxidant activity studies demonstrated that PSAE effectively inhibits DPPH radicals, superoxide anions, hydroxyl radicals, and ABTS radicals, while also enhancing the inhibition rates of collagenase and hyaluronidase. In vitro studies on photoaging resistance revealed that PSAE significantly reduced the UV-induced increases in reactive oxygen species (ROS) levels and senescence-associated β-galactosidase (SA-β-gal) activity. Mechanistic studies indicated that PSAE suppressed the overexpression of IRS1 and its downstream effectors, including PI3K, AKT, and mTOR induced by UV irradiation. A human efficacy assessment was conducted by evaluating parameters such as transepidermal water loss (TEWL), epidermal moisture content, roughness and elasticity, confirming the efficacy of PSAE in humans.Discussion: In summary, PSAE attenuates UV-induced oxidative damage, genetic damage, and collagen degradation associated with photoaging by modulating the IRS/PI3K/FOXO signaling pathway. This study elucidated the mechanism through which PSAE, thereby providing strong support for its application in cosmetic anti-aging formulations.

Sustainable and intelligent packaging systems, utilizing biomass-derived matrix and bioactive indicators represent promising approaches for real-time detection and monitoring of freshness of food, as well as prolonging the shelf-life and improving the quality and safety of food products. Herein, we reported pH-responsive collagen nanocomposite films reinforced by curcumin-loaded Laponite nanoplatelets which could act as smart indicators for monitoring the freshness of shrimps. Caffeic acid, as a bio-based modifier, was covalently grafted onto the surfaces of the nanoplatelets loaded with curcumin . The collagen nanocomposite films were fabricated by the incorporation of functionalized nanoplatelets through all-aqueous casting-evaporation approach. The triple-helical structure of collagen molecules was stabilized by the functionalized nanoplatelets mainly through hydrogen bonding and electrostatic interactions with uniform distribution into the film matrix. Moreover, the resultant nanocomposite films not only demonstrated favorable hydrophobicity and water vapor barrier properties, but also exhibited effective antioxidant activity and anticipated color responsiveness to pH changes and NH 3 presence. The consistent changes in color of the nanocomposite film with the shrimps following days of storage supported its feasibility and potential for monitoring the freshness of shrimps. Therefore, we envision that the developed collagen-based nanocomposite films showcase great potential in the application of sustainable and intelligent packaging system for dynamic visualization of seafood freshness.

To address the disadvantages of traditional rubber antioxidants with low molecular weight such as easy migration, poor extraction resistance, low thermal stability and antioxidant efficiency, in this work, we first prepared an antioxidant containing primary and secondary antioxidant groups (MC-MPA) through thiol-ene click reaction between N-(4-aminophenyl)maleimide (MC) and 3-mercaptopropionic acid (MPA). Then, we synthesized a chitosan-based macromolecular antioxidant with long-chain alkene, aromatic secondary amine, and thioether groups (COS-UC-MC-MPA) by sequentially grafting functional compounds 10-undecanonyl chloride (UC) and antioxidant MC-MPA onto natural macromolecular polymer chitosan (COS). The free radical scavenging ability of COS-UC-MC-MPA was assessed by using 2,2-Diphenyl-1-picrylhydrazyl (DPPH), and the results demonstrated that COS-OC-MC-MPA has high scavenging rate of 76.5 % for DPPH free radicals after 1 min of contact with DPPH dimethyl sulfoxide solution, showing excellent free radical scavenging ability. In addition, the results of accelerated thermo-oxidative aging test, thermogravimetric (TG) test, kinetic analysis of thermal-oxidative degradation, extraction resistance test, and volatilization resistance test indicated that COS-UC-MC-MPA with superior migration resistance and volatilization resistance could effectively enhance the antioxidative properties, thermal stability and thermal-oxidative stability of rubber composites. Moreover, the results of rubber processibility analysis (RPA) test and scanning electron microscopy (SEM) implied that the addition of COS-UC-MC-MPA is also conductive to improving the dispersibility of silica in rubber matrix. This study supplies a new perspective for the preparation of environmentally friendly and highly efficient macromolecular antioxidants for rubber composites.

Curdlan demonstrates significant biological activities and exhibits functional properties such as thickening, water retention, and gelling capabilities. However, the inherent structural simplicity and constrained functionality of pure curdlan-based hydrogels present significant limitations for their utilization in specialized applications requiring multifunctional properties. In this study, curdlan were used as the matrix to prepare multifunctional hydrogels with photothermal conversion, antibacterial and antioxidant capabilities through the addition of tannic acid and iron ions and by employing heating-annealing approach. The molecular interactions between tannic acid and curdlan were elucidated systematically, and the detailed characterization of the hydrogel's functional properties, encompassing its rheological characteristics, thermal stability, swelling kinetics, photothermal conversion capability, and biological activities including antioxidant capacity and antimicrobial efficacy were investigated. The results demonstrated that tannic acid formed stable hydrogen-bonding interactions with curdlan molecules, which significantly affected the structural reorganization of curdlan's triple-helix architecture during renaturation. After introducing the Fe 3 + , the iron-tannic acid chelation complex was served as an effective cross-linking agent for curdlan polymer chains, resulting in enhanced mechanical strength of the composite hydrogel while simultaneously imparting remarkable photothermal conversion efficiency to the material system. And the composite hydrogel demonstrated significant free radical scavenging capacity, potent antibacterial activity, and good cell compatibility. The developed composite hydrogel including superior antioxidant activity, effective antibacterial performance, and efficient photothermal conversion capability, shows the potential as a promising candidate for wound dressing, thereby significantly expanding the potential utilization of curdlan-based biomaterials.

Facial masks have become omnipresent daily, providing the skin with sufficient moisture and activated nutrients. However, conventional wet facial masks use large amounts of preservatives and antimicrobials, which are not friendly to human health and ecological environment. Herein, emulsion electrospinning was innovatively used to prepare eco-friendly resveratrol (RES) and phenyl ethyl resorcinol (PR)-loaded polyvinyl alcohol (PVA) dry nanofiber facial mask, avoiding the usage of preservatives and antimicrobials. The SEM results showed that the dry PVA/RES/PR facial masks have a mean diameter of about 205 nm. When used, the active substances (RES and PR) inside the nanofiber facial mask are released by moisture spray, activating the active substances released in response to water, providing an in-situ moisturizing effect (improved 13 %), antioxidant performance (free radical scavenging rate, 86 %), and antibacterial performance (antimicrobial efficiency of 99.98 % and 99.99 % against E. coli and S. aureus ). Moreover, the dry PVA/RES/PR facial mask can be sealed with biodegradable paper packaging instead of plastic packaging. The novel environmental dry facial mask prepared with emulsion electrospinning is a suitable material to study and apply in cosmetics.
