Application of a CYP1B1-Targeted NIR Probe for Breast Cancer Diagnosis, Surgical Navigation, and CYP1B1-Associated Chemotherapy Resistance Monitoring

Zhihao Wu,Tao Shi,Qi Shao,Dongmei Chen,Peisheng Gao,Jie Wang,Ting Xu,Qingqing Meng,Shaoshun Li

Early detection and precise treatment for breast cancer are crucial, given its high global incidence rate. Hence, the development of novel imaging targets is essential for diagnosing and monitoring resistance to chemotherapy, which is pivotal for achieving precise and personalized treatment for breast cancer patients. In our previous work, we successfully developed a near-infrared (NIR) probe 1 for CYP1B1-targeted imaging. In this study, we aimed to investigate the utility of the probe as a NIR fluorescence and photoacoustic dual-mode imaging probe for the detection and surveillance of breast cancer. Western blotting of cancer cell lines has confirmed that CYP1B1 is widely expressed in breast cancer and gynecological cancer. In vitro NIR fluorescence imaging capability of the probe for tracking CYP1B1-positive tumor cells was validated by using confocal microscopy. Further studies, including in vivo fluorescence and photoacoustic dual-model imaging and ex vivo biological distribution analysis on a triple-negative breast cancer xenograft mouse model, demonstrated that the probe selectively accumulated in tumor tissue within as early as 0.5 h postinjection. The results of the surgical resection experiment revealed that the tumor could be entirely removed under the guidance of NIR imaging, thereby indicating the probe’s efficacy in surgical navigation. CYP1B1 expression was found to be upregulated in adriamycin (ADR)-resistant breast cancer cells, MCF-7/ADR. Consequently, the sensitivity of CYP1B1 overexpressed cells, MCF-7/1B1, to ADR was significantly reduced, with an IC50 value of 0.586 ± 0.0934 μM, compared to the parental MCF-7 cells with an IC50 value of 0.183 ± 0.0444 μM. In vivo and ex vivo imaging assays conducted on MCF-7/ADR tumor-bearing mice demonstrated that the probe was specifically enriched in tumor sites, suggesting its potential for monitoring chemotherapy resistance in breast cancer. This study expands the scope of application for NIR probe 1, establishing its utility in breast cancer diagnosis through fluorescence-photoacoustic dual-model imaging, monitoring of chemotherapy resistance, and guidance for surgical resection. This strategy paves the way for novel approaches to precise and personalized treatment for breast cancer patients.

Comparative analysis of spectral variable selection methods for NIR-based multi-component detection of Xanthoceras sorbifolium Bunge seed kernels

Shengxin Li,Ziyan Zhang,Zhiran Zhang,Sen Zhou,Mengkai Liu,Xichao Li,Zheng Zheng,Jie Sun

The moisture, oil, protein, and nervonic acid contents are crucial criteria for assessing the nutritional value and processing quality of Xanthoceras sorbifolia Bunge seed kernels (XSKs). Near-infrared spectroscopy (NIRS) offers a viable alternative to conventional chemical methods for rapid and non-destructive sample analysis. This study employed a highly adaptive method known as competitive adaptive reweighted sampling (CARS), which facilitates variable selection for different components and streamlines the feature-variable screening process. Using this method, various algorithms were applied to establish NIRS detection models for the four components of XSKs, and accuracy evaluations were conducted using the prediction set determination coefficient ( R P 2 ), root-mean-square error (RMSEP), and other parameters. The results indicated that CARS effectively extracted key information on various components in NIRS. Based on the feature variables selected by CARS, the partial least squares regression (PLSR) model exhibited optimal performance regarding moisture ( R P 2  = 0.978, RMSEP = 0.117 %) and nervonic acid content ( R P 2  = 0.988, RMSEP = 0.072 %). Meanwhile, the particle swarm optimization-enhanced support vector regression (PSO-SVR) model and back-propagation neural networks (BPNN) achieved superior performance for protein ( R P 2  = 0.989, RMSEP = 0.243 %) and oil content ( R P 2  = 0.978, RMSEP = 0.467 %), respectively. These models enable rapid and non-destructive detection of multi-component content, thereby facilitating the expeditious identification and selection of XSKs with high nutritional value and superior quality.

Regulation of Monomer Allocation and Surface Passivation via Dual Ligands for the Synthesis of NIR-IIc Ag2Se Quantum Dots

Zhen-Ya Liu,Wei Zhao,Li-Ming Chen,Fei Ding,An-An Liu,Dai-Wen Pang

Narrow bandgap silver selenide (Ag2Se) is a promising candidate for noninvasive in vivo imaging in the long-wavelength region of the second near-infrared window (NIR-IIc, 1700–2000 nm), yet precise control over its size remains challenging due to ineffective regulation of nucleation and growth, largely constrained by its extremely low solubility product. Here, we reveal a growth bottleneck in the Ag2Se quantum dots (QDs) synthesis arising from rapid monomer depletion during nucleation. The sole use of strongly binding thiol ligands further confines the system to a thermodynamically metastable size regime, hindering the continuous growth required for precise spectral tuning. By employing a dual-ligand strategy combining strongly binding thiol ligands with labile oleate ligands, we modulate monomer supersaturation and the dynamic adsorption–desorption equilibrium on the nanocrystal surface, thereby suppressing nucleation to preserve sufficient monomer for subsequent monomer diffusion-limited growth. This approach yields size-tunable Ag2Se QDs with emission peaks extending into the NIR-IIc window. Furthermore, the synergistic passivation by zinc oleate ligands significantly improves the size dispersity and colloidal stability of large-sized Ag2Se QDs. Our in-depth investigation not only addresses challenges in size control of QDs exhibiting nonclassical growth but also highlights the transformative potential of low-toxicity, NIR-IIc-emissive Ag2Se QDs for optoelectronics and noninvasive deep-tissue imaging.

Senolytic Therapy Enabled by Senescent Cell-Sensitive Biomimetic Melanin Nano-senolytics

Hairui Zhang,Xiaoling Xu,Xin Shou,Wucan Liao,Chengkang Jin,Changjiang Chen,Chen Zhang,Wenhua Gao,Junfeng Zhang,Weihong Ge,Liyun Shi

Cellular senescence is a significant risk factor for aging and age-related diseases (ARD). The canonical senolytics Dasatinib and Quercetin (DQ) have shown promise in clearing senescent cells (SnCs); however, the lack of selectivity poses a challenge in achieving optimal outcomes. Despite the recent occurrence of the nanomaterial-based approaches targeting SnCs, limited therapeutic effects and potential toxicity still remain a major concern. Herein, we developed a “double locks-like” nanoplatform that integrated Galactan coating and mesoporous polydopamine to encase the senolytic drug DQ. By this way, DQ was only released in SnCs that were featured with higher levels of β-galactosidase (β-gal) and low PH. Additionally, the nanoparticles were equipped with 2,2,6,6-Tetramethylpiperidine-1-oxyl (Tempo) to gain enhanced photothermal converting potential. Consequently, the synthesized nanosenolytics demonstrated remarkable specificity and efficacy in eradicating SnCs, and accordingly reversed pulmonary fibrosis in mice without affecting normal tissues. Upon exposure of near-infrared (NIR) light, the nanoparticles demonstrated to efficiently remove senescent tumor cells inducted by chemotherapy, thereby hindering the outgrowth and metastasis or breast cancer. Collectively, the present study develops an “On/Off” switchable nanoplatform in response to SnCs, and produces a more safe, efficient and feasible way to delay aging or alleviate age-associated diseases. Self-Calibrated Identification of Metastatic Lymph Nodes Using Full-NIR-II Tunable Ag2Se Quantum Dots Engineered by Short-Chain Phosphines

Wei Ge,Beibei Gao,Shuxian Li,Guangbo Yu,Lu Gao,Zhemin Shen,Fu Wang,Junle Qu

Ratiometric fluorescence imaging in the second near-infrared (NIR-II) region holds great promise for the accurate identification of metastatic lymph nodes (MLNs), but faces challenges in biocompatible probe design with simplified composition and broad spectral tunability. Here, a minimalist, self-calibrating ratiometric probe based on homologous Ag 2 Se quantum dots (QDs) is developed. Through rational redesign of tributylphosphine-mediated coordination kinetics, continuous size-dependent emission tuning across 935–1710 nm, with the synthetic mechanism and structure–property relationships systematically elucidated, is achieved. The inherently low-toxicity Ag 2 Se core, combined with poly(amino acid) coating, ensures excellent biosafety, as validated by in vitro cytotoxicity and in vivo biochemical assays. Two spectrally distinct QDs (emitting at 1040 and 1305 nm, with full width at half maximum of 176 and 119 nm) enable multiplexed anatomical imaging, allowing both real-time intraoperative detection and postoperative assessment of MLNs, with a diagnostic threshold ratio of ≈0.9. This work presents a scalable strategy for full-NIR-II monochromatic QD synthesis and establishes a minimal-component biocompatible platform for precision ratiometric diagnostics.

Oleic Acid Copolymer as A Novel Upconversion Nanomaterial to Make Doxorubicin-Loaded Nanomicelles with Dual Responsiveness to pH and NIR

Jin Zhang,Xiaoyue Tang,Chuanqing Huang,Zeyu Liu,Yong Ye

Oleic acid (OA) as main component of plant oil is an important solvent but seldom used in the nanocarrier of anticancer drugs because of strong hydrophobicity and little drug release. In order to develop a new type of OA nanomaterial with dual responses to pH and near infrared light (NIR) to achieve the intelligent delivery of anticancer drugs. The novel OA copolymer (mPEG-PEI-(NBS, OA)) was synthesized by grafting OA ando-nitrobenzyl succinate (NBS) onto mPEGylated polyethyleneimine (mPEG-PEI) by amidation reaction. It was further conjugated with NaYF4:Yb3+/Er3+nanoparticles, and encapsulated doxorubicin (DOX) through self-assembly to make upconversion nanomicelles with dual response to pH and NIR. Drug release behavior of DOX, physicochemical characteristics of the nanomicelles were evaluated, along with its cytotoxic profile, as well as the degree of cellular uptake in A549 cells. The encapsulation efficiency and drug loading capacity of DOX in the nanomicelles were 73.84% ± 0.58% and 4.62% ± 0.28%, respectively, and the encapsulated DOX was quickly released in an acidic environment exposed to irradiation at 980 nm. The blank nanomicelles exhibited low cytotoxicity and excellent biocompatibility by MTT assay against A549 cells. The DOX-loaded nanomicelles showed remarkable cytotoxicity to A549 cells under NIR, and promoted the cellular uptake of DOX into the cytoplasm and nucleus of cancer cells. OA copolymer can effectively deliver DOX to cancer cells and achieve tumor targeting through a dual response to pH and NIR.

808 nm NIR Laser-Excited Upconversion Nanoplatform for Combinatory Photodynamic and Chemotherapy with Deep Penetration and Acid Bursting Release Performance

Yingtao Zhong,Yun Dong,Tie Chen,Lingzhi Yang,Min Yao,Yunshi Zhi,Haoyi Yang,Jian Zhang,Wenchuan Bi

Combinatory photodynamic and chemotherapy have demonstrated superior performance in cancer ablation over singular therapeutics. However, photodynamic therapy (PDT) often exhibits suboptimal efficacy for deep-seated tumors, owing to the limited penetration depth of illumination light, while chemotherapy is generally accompanied by severe side effects. Therefore, it is imperative to develop a functional nanoplatform for combinatory PDT and chemotherapy, which could, for PDT, achieve enhanced light penetration and, for chemotherapy, realize reduced therapeutic threshold dosage and a controllable drug release profile (e.g., minimized release in blood circulation but bursting release in the tumor microenvironment). Herein, we demonstrate a therapeutic nanoplatform composed of poly(acrylic acid) (PAA)-modified silica-coated Nd3+-doped upconversion nanoparticles decorated with methylene blue (MB) and doxorubicin (DOX) in silica and a PAA layer, respectively. Notably, 808 nm light is used to excite upconversion nanoparticles and further trigger the photosensitization behavior of MB in PDT, while the quick acid response of the PAA layer in the tumor acid environment introduces DOX bursting for optimized chemotherapy with significantly decreased therapeutic threshold dosage and minimized side effects. Importantly, the anticancer efficiency of the nanoplatform in vitro and in vivo shows an IC50 and a tumor inhibition rate of 12.55 μg mL–1 and 89.81%, respectively. This study provides a strategy for combinatory cancer therapy.

Reduction-sensitive polypeptide nanogel conjugated BODIPY-Br for NIR imaging-guided chem/photodynamic therapy at low light and drug dose

Le Liu,Tuanwei Li,Zheng Ruan,Pan Yuan,Lifeng Yan

A heavy atoms modified reaction-active photosensitizer NHS-BODIPY-Br possessing efficient near infrared (NIR) fluorescence emission and singlet oxygen generation properties has been synthesized, which can be used for synchronous NIR imaging and photodynamic therapy (PDT). A reduction-sensitive PEGylated polypeptide nanogel was prepared by ring-opening polymerization (ROP) of l -cystine- N -carboxy anhydride. Poly (ethylene glycol) methyl ether was used as initiator and hydrophilic segment, the as-prepared nanogel was conjugated with the NHS-BODIPY-Br molecule by chemical linkage, and it can be directly used as a macrophotosentizer for NIR imaging-guided PDT. In addition, the nanogel also showed good encapsulating ability for doxorubicin (DOX). In the presence of glutathione (10 mM), the obtained NIR nanogel showed obvious reduction-induced drug release behavior. In vitro tests on internalization of the NIR nanogel by HepG2 cells indicated its efficiency in detecting cancer cells. Meanwhile, MTT assays performed on HepG2 cells confirmed that the cancer cells growth could be obviously suppressed (almost all cells) when exposed to an extremely low energy light (25 mW/cm 2 , 10–15 J/cm 2 ) and low dose of DOX (3–5 μg/ml), indicates an efficient NIR image-guided chem/photodynamic therapy.

Mitochondrial-targeting and NIR-responsive Mn3O4@PDA@Pd-SS31 nanozymes reduce oxidative stress and reverse mitochondrial dysfunction to alleviate osteoarthritis

Yuquan Li,Junxu Yang,Xiaoming Chen,Hao Hu,Nihan Lan,Jinmin Zhao,Li Zheng

Mitochondrial reactive oxygen species (mROS) play a crucial role in the process of osteoarthritis (OA), which may be a promising target for therapy of OA. In this study, novel mitochondrial-targeting and SOD-mimic Mn 3 O 4 @PDA@Pd-SS31 nanozymes with near-infrared (NIR) responsiveness and synergistic cascade to scavenge mROS were designed for the therapy of OA. Results showed that the nanozymes accelerated the release of Pd and Mn 3 O 4 under NIR irradiation, exhibiting enhanced activities of SOD and CAT mimic enzymes with reversed mitochondrial dysfunction and promoted mitophagy to effectively scavenge mROS from chondrocytes, modulate the microenvironment of oxidative stress, and eventually inhibit the inflammatory response. Nanozymes were excreted in vivo through intestinal metabolic pathway and had good biocompatibility, effectively reducing the inflammatory response and relieving articular cartilage degeneration in OA joints, with a reduction of 93.7 % and 93.8 % in OARSCI scores for 4 and 8 weeks respectively. Thus, this study demonstrated that the mitochondria targeting and NIR responsive Mn 3 O 4 @PDA@Pd-SS31 nanozymes could efficiently scavenge mROS, repair damaged mitochondrial function and promote cartilage regeneration, which are promising for the treatment of OA in clinical applications.

A molecular rotor-based NIR probe with large stokes shift for highly sensitive tracking of mitochondrial viscosity in live tumors

Fan Wang,Zhiyu Wang,Kun Yu,Yuqing Wang,Qi Su,Chunxv Han,Wenxuan Hu,Lei Hu,Ping Li,Hui Wang

Abnormal mitochondrial viscosity is closely associated with tumor initiation and progression. Therefore, the development of fluorescent probes capable of real-time, precise monitoring of mitochondrial viscosity changes in tumor cells holds significant potential for the early diagnosis and therapeutic evaluation of tumors. This study designed and synthesized a series of near-infrared (NIR) fluorescent probes ( W1–W3 ) with D-π-A structural framework based on a phenothiazine-bithiophene-pyridinium moiety. All three probes ( W1 – W3 ) featured emission at approximately 650 nm, accompanied by a substantial Stokes shift of about 150 nm. Among them, W2 and W3 exhibited excellent responsiveness to viscosity. Their molecular rotor character results in quenched fluorescence in low-viscosity environments and intensified emission in high-viscosity media, enabling highly sensitive detection of viscosity changes. Cellular experiments revealed that W2 possesses outstanding biocompatibility and precise mitochondria-targeting ability. Remarkably, it achieved effective bioimaging at a concentration as low as 500 nM. The probe also succeeded in dynamically monitoring viscosity changes during drug-induced (e.g., DXM, rotenone) mitochondrial dysfunction. Notably, W2 detected increased mitochondrial viscosity in tumor cells triggered by apoptosis inducers (e.g., DOX, cisplatin), thereby confirming its capability to track dynamic changes in mitochondrial viscosity. In vivo and in vitro imaging results established the potential of probe W2 for tumor imaging. With its capability for dynamic viscosity monitoring from cellular to in vivo levels, probe W2 positions itself as a promising tool to advance applications in early tumor diagnosis, drug efficacy assessment, and precision therapy.

Visualization of mitochondrial monoamine oxidase dynamics in live cells and DILI via a ratiometric NIR probe with “ICT+ESIPT” mechanism

Ting Cao,Mengjin Li,Hong Ma,Zhefeng Fan

Monoamine oxidase (MAOs) is a key enzyme that maintains amine homeostasis and participates in liver pathological processes. Real time in situ monitoring of its activity is crucial for elucidating the mechanism of drug-induced liver injury (DILI). This study successfully constructed a ratio type fluorescent probe Cy-1 with mitochondrial targeting ability and near-infrared emission characteristics. This probe is based on MAOs catalyzed oxidative deamination reaction, and achieves fluorescence response through a clever “ICT (Intramolecular Charge Transfer) + ESIPT (Excited-State Intramolecular Proton Transfer)” dual mechanism, producing significant ratiometric signal changes in the near-infrared region. Experiments have shown that Cy-1 has a wide linear range (10–115 μg/mL), high sensitivity (0.86 μg/mL), and high selectivity in vitro, and can specifically localize to mitochondria. The probe was successfully used for in situ differential monitoring of endogenous MAOs activity in living cells, and visualized semi quantitative monitoring of MAOs activity changes in vivo was achieved in an acetaminophen (APAP) induced drug-induced liver injury (DILI) model. Cy-1 is expected to become a powerful chemical tool for understanding the mechanisms of MAOs in physiological and pathological processes, especially in the occurrence and development of diseases such as liver injury, as well as evaluating the efficacy of related drugs.

Se/Ti3C2 nanoparticles with antimicrobial and antioxidant properties: Rapid phototherapy of periodontitis by modulating GPX1 expression in macrophages under NIR

Zhen Wan,Qian Wu,Tian Deng

Periodontitis is a chronic inflammatory disease initiated by bacterial pathogens, resulting from complex interactions between microbial infection and host immune responses. In current clinical practice, antibiotics represent the most frequently utilized adjunctive therapy for periodontitis. However, the overuse of antibiotics not only contributes to the emergence of bacterial resistance but also demonstrates limited efficacy in inflammation resolution. In this study, we synthesized selenium/Ti 3 C 2 (Se/Ti 3 C 2 ) through a one-step redox method using Ti 3 C 2 as a template. The Se/Ti 3 C 2 demonstrated significant antibacterial activity against Escherichia coli ( E.coli ), Staphylococcus aureus ( S.aureus ), and Porphyromonas gingivalis ( P.gingivalis ) under near-infrared (NIR) light irradiation, as evidenced by comprehensive evaluations including colony-forming unit (CFU) counting assays, live-dead fluorescence staining, scanning electronic microscopy (SEM) observations, growth curves, and membrane potential staining assays. Furthermore, the anti-inflammatory and antioxidant properties of Se/Ti 3 C 2 were investigated using various methodologies, including 1,1-diphenyl-2-pyridine-hydratedacyl (DPPH) scavenging assay, 2,2′-azino-bis(3-ethylbenzothiazoline)-6-sulphonic acid (ABTS) scavenging assay, glutathione peroxidase (GPX) activity measurement, intracellular reactive oxygen species (ROS) fluorescence staining, flow cytometry, and quantitative real-time polymerase chain reaction (qPCR) assays. Through GPX1 gene knockdown experiments, we demonstrated that the anti-inflammatory effects of Se/Ti 3 C 2 were closely associated with GPX1 expression. RNA sequencing analysis was subsequently performed to elucidate the underlying mechanisms of its anti-inflammatory and antioxidant activities. Finally, in vivo experiments using a mouse periodontitis model demonstrated the remarkable therapeutic potential of Se/Ti 3 C 2 in periodontitis treatment. Therefore, Se/Ti 3 C 2 holds great promise as a potential alternative to antibiotics, emerging as a novel therapeutic material for periodontitis treatment.
