Alzheimer’s disease (AD) is a neurodegenerative disease characterized by the deposition of amyloid-β peptides (Aβ). Aβ accumulation leads to the formation of neurofibrillary tangles, inflammation, axonal injury, synapse loss, and neuronal apoptosis. Thus, reducing Aβ levels should exert a neuroprotective effect against AD. Ginsenoside Rf, an extract from Panax notoginseng, has potent anti-fatigue, anti-nociception, anti-oxidation, and anti-inflammation properties. However, it is unclear whether ginsenoside Rf is effective in the treatment of AD. Here, we reported that ginsenoside Rf could significantly attenuate Aβ-induced apoptosis in N2A cells, as reflected by a dramatic increase in mitochondrial membrane potential and decrease in Ca2 + concentration, reactive oxygen species, and active caspase-3 expression. Meanwhile, ginsenoside Rf could alleviate the Aβ-induced inflammation reaction, such as the decrease of interferon-gamma (IFN-γ) and active caspase-1 expression and the increase of interleukin-13. Furthermore, we also found that Rf is able to accelerate Aβ clearance and subsequently reduces Aβ level in N2A cells stably transfected with human Swedish mutant APP695 (N2A-APP). More importantly, daily Rf treatment (20 mg/kg, i.p.) throughout the experiment dramatically improved spatial learning and memory in Aβ42-induced mouse model of AD. Taken together, these results indicate that ginsenoside Rf may decrease Aβ-induced neurotoxicity and memory decline via anti-inflammatory response during AD development, suggesting that Rf may be a potential therapeutic agent for treating AD.

The polycystic ovary syndrome (PCOS) model was established in rats and correlation between the expression of macrophage migration inhibitory factor (MIF) and cytokinesis with the MAPK signalling pathway in the rat ovary was measured. The PCOS model in rats was established by dehydroepiandrosterone (DHEA). Thirty sexually immature female Sprague-Dawley rats were randomly and equally assigned to three groups: control group, PCOS group, and PCOS with high-fat diet (HFD) group. Serum hormones were assayed by radioimmunoassay (RIA). The ovaries were immunohistochemically stained with MIF, and the expression of MIF, p-JNK and p-p38 was detected by Western blotting in ovaries. The serum testosterone level, LH concentration, LH/FSH ratio, fasting insulin level and HOMA IR index in the PCOS group (6.077±0.478, 13.809±1.701, 1.820±0.404, 10.83±1.123 and 1.8692±0.1096) and PCOS with HFD group (6.075±0.439, 14.075±1.927, 1.779±0.277, 10.20±1.377 and 1.7736±0.6851) were significantly higher than those in the control group (4.949±0.337, 2.458±0.509, 1.239±0.038, 9.53±0.548 and 1.5329±0.7363), but there was no significant difference between the PCOS group and PCOS with HFD group. The expression levels of MIF, p-JNK, and p-p38 in the PCOS group (0.4048±0.013, 0.6233±0.093 and 0.7987±0.061) and PCOS with HFD group (0.1929±0.012, 0.3346±0.103 and 0.3468±0.031) were obviously higher than those in control group (0.2492±0.013, 0.3271±0.093 and 0.3393±0.061), but no significant difference was observed between PCOS group and PCOS with HFD group. It was suggested that MIF may participate in the pathogenesis of PCOS through the MAPK signalling pathway in PCOS rats induced by DHEA.

In this study, we have provided a highly efficient, convenient, and universal protocol for preparing polyvinylidene fluoride (PVDF) membranes with low blood contact activation via in situ cross-linking copolymerization of 2-hydroxyethl methacrylate (HEMA) and acrylic acid (AA) in a solution of PVDF. The modified membranes were prepared from PVDF solution by phase inversion technology. The composition and morphology of the modified membranes were confirmed by attenuated total reflectance-Fourier transform infrared (ATR-FTIR) spectroscopy, thermogravimetric (TG) analysis, and scanning electron microscopy (SEM). Protein adsorption, clotting time, and contact activation on the modified PVDF membranes were systematically studied, the results indicating that after the incorporation of AA and HEMA, the modified PVDF membranes possessed anticoagulant properties in addition to low contact activation of blood components when in contact with blood. Therefore, fluorinated PVDF membranes with surfaces enriched with carboxyl and hydroxyl groups possessed the potential for use in long-term blood-contacting devices.

A total of 328 agricultural product samples highly suspected to be contaminated, from flour companies, feed companies, and livestock farms throughout China, were surveyed for deoxynivalenol (DON) contamination using a self-assembly enzyme-linked immunosorbent assay (ELISA) kit. An ELISA kit for DON was developed with a 4.9 ng mL−1limit of detection (LOD) in working buffer and a 200 ng g−1LOD in authentic samples. The DON contamination detection rate was 88.7%, concentrations ranged from 200.9 to 6480.6 ng g−1, and the highest DON contamination was found in distillers’ dried grains with solubles with an average of 3204.5 ng g−1. Wheat bran and wheat were found to be the most commonly contaminated samples, and the corn meal samples had the lowest average DON level. This ELISA kit is a powerful alternative method for the rapid, sensitive, specific, accurate, and high-throughput determination of DON and can meet the maximum requirement levels. This survey suggests that DON contamination in the Chinese market is serious, and the contamination risk deserves attention. Essential preventive measures should be implemented to ensure food safety and human health. Keywords:deoxynivalenol;contamination;survey;ELISA kit;agricultural productKey Contribution:A total of 328 agricultural product samples highly suspected to be contaminated were surveyed for deoxynivalenol using a self-assembly ELISA kit.

Herein, we established an ionic liquid (IL)-based liquid-solid transformation microextraction (IL-LTME) combined with HPLC-UV detection for the simultaneous determination of triclosan (TCS) and its methylated product, methyltriclosan (MTCS), in human fluids. The IL-LTME method was based on an in situ metathesis between hydrophilic IL and ion-exchange salt to form a solid hydrophobic IL. According to the above principle, a hydrophilic IL, [C 12 MIM]Br, was selected as the extractant, and NH 4 PF 6 as ion-exchange salt. The prominent advantages of the newly developed method are: (1) the in-situ reaction between the extractant [C 12 MIM]Br and ion-exchange salt NH 4 PF 6 changed the IL from hydrophilic to hydrophobic that avoiding the stick of ionic liquid on the tube wall; (2) bubbling with NH 3 greatly increased the contact area between IL-extractant and analytes resulting in improved extraction recovery; and (3) solidification of the [C 12 MIM] PF 6 provided a good separation and avoided the use of specialized equipment. A series of main parameters were optimized by single-factor screening and central composite design as follows: 0.9 mL of NaOH, 2.0 min of second ultrasonically time, 10 min of centrifugation time, 21 mg of extractant [C 12 MIM]PF 6 , 2.4 min of ultrasonic time, 65 mg of NH 4 PF 6 and 13.8 min of cooling time. Under the optimized conditions, the limits of detection for TCS and MTCS were 0.126–0.161 μg L −1 in plasma samples, and 0.211–0.254 μg L −1 in urine samples, respectively. The extraction recoveries for TCS and MTCS were in the range of 94.1–103.8%. The intra-day and inter-day precisions were 1.00–4.74% and 1.02–5.21%, respectively. In general, the IL-LTME method is environment-friendly, time-saving, economical, high efficient and robust with low detection limits and high recoveries. Thus, the newly developed method has excellent prospects for sample pretreatment and analysis of trace TCS and MTCS in blood and urine samples.

Microplastics have a great potential to sorb organic pollutants from the adjacent environment. In this study, the sorption of tetracycline , a polar and ionizable antibiotic, on three types of microplastics (polyethylene (PE), polypropylene (PP) and polystyrene (PS)) were investigated in batch sorption experiments. The sorption isotherms were well fitted by the Langmuir model, indicating that not only hydrophobic interactions but also other interactions (e.g. electrostatic interactions) played important roles in the sorption process. PS had the maximum sorption capacity, following the order PS > PP > PE, which can be attributed to polar interactions and π-π interactions. The sorption of tetracycline on microplastics was significantly influenced by pH, with sorption capacity increasing gradually, peaking at pH 6.0 and then decreasing, likely due to the influence of tetracycline speciation with the change of pH. Fulvic acid was selected as representative dissolved organic matter (DOM) to examine the effect on sorption. The increasing concentration of fulvic acid inhibited the sorption of tetracycline on three microplastics, decreasing them by more than 90% at the fulvic acid concentration of 20 mg/L, which implied a greater affinity of tetracycline to fulvic acid than to microplastics. Increasing salinity from 0.05 to 3.5% had negligible effects on the sorption of tetracycline on the three microplastics. Our results highlight the importance of pH and DOM on the sorption of tetracycline on microplastics, and suggest the relatively minor role of microplastics in the fate and transport of tetracycline in the aquatic environment in the presence of DOM.

We used surface-modified titanium (Ti) substrates with a multilayered structure composed of chitosan-catechol (Chi-C), gelatin (Gel) and hydroxyapatite (HA) nanofibers , which were previously shown to improve osteogenesis, as a platform to investigate the interaction of osteogenesis and angiogenesis during bone healing . Combined techniques of Transwell co-culture, wound healing assay , enzyme linked immunosorbent assay (ELISA), quantitative real-time polymerase chain reaction (qRT-PCR), western blotting and immunohistochemical staining were used to evaluate adhesion, morphology and migration of adipose-derived mesenchymal stem cells (Ad-MSCs) and human umbilical vein endothelial cells (HUVECs) grown on different Ti substrates. We investigated the effect of substrates on the osteogenic differentiation of Ad-MSCs and reciprocal paracrine effects of Ad-MSCs on HUVECs or vice versa . The multilayered Ti substrates directly regulated the cellular functions of Ad-MSCs and angiogenic HUVECs and mediated communication between them by enhancing paracrine effects via cell–matrix interactions in vitro . The in vivo results showed that the change of microenvironment induced by surface-modified Ti implants promoted the adhesion, recruitment and proliferation of MSCs and facilitated coupled osteogenesis and angiogenesis in bone healing. The study proved that multilayer-film-coated Ti substrates positively mediated cellular biological function in vitro and improved bone healing in vivo . Statement of Significance Recent studies have revealed that osteogenesis and angiogenesis are coupled, and that communication between osteoblasts and endothelial cells is essential for bone healing and remodeling processes; however, these conclusions only result from in vitro studies or in vivo studies using transgenic murine models. Relatively little is known about the communication between osteoblasts and endothelial cells in peri-implants during bone healing processes. Our results revealed the cellular/molecular mechanism of how multilayered Ti substrates mediate reciprocal paracrine effects between adipose-derived mesenchymal stem cells and human umbilical vein endothelial cells; moreover, the interactions between the cell-matrix and peri-implant was proven in vivo with enhanced bone healing. This study contributes to our understanding of the fundamental mechanisms of angiogenesis and osteogenesis that affect peri-implantation, and thus, provides new insights into the design of future high-quality orthopedic implants.

B lymphocyte-induced maturation protein 1 (Blimp-1), a transcription factor containing zinc finger , is required and sufficient to trigger terminal differentiation of B cells in mammals. The Blimp-1 ( OnBlimp-1 ) from Nile tilapia ( Oreochromis niloticus ) was identified and characterized its expression pattern during B cell activation and maturation. The cDNA of OnBlimp-1 open reading frame is 2547 bp encoding a protein of 848 amino acids and the predicted molecular weight is 93.36 kDa. OnBlimp-1 contains a SET domain and five ZnF_C2H2 domains, which shares high homology with that of other species. OnBlimp-1 transcription was detected in all examined tissues with high expression in the spleen (SPL). Analysis of sorted lymphocyte populations , including IgM + and IgM − cells from peripheral blood (PBL), SPL and anterior kidney (AK), indicated that the OnBlimp-1 transcription was highly expressed in the IgM + B cells. Upon LPS stimulation, OnBlimp-1 expression was up-regulated in tissues of PBL, SPL and AK significantly. The expression of OnBlimp-1 , as well as the secreted IgM , was significantly up-regulated in the SPL and AK leukocytes stimulated with anti-OnIgM monoclonal antibody and LPS in vitro , respectively. Above results suggest that OnBlimp-1, a cytokine regulating the terminal differentiation of activated B cells to antibody-secreting cells, is likely to play important roles in B cell activation and maturation in Nile tilapia.

The cotton bollworm Helicoverpa armigera , is one of the world’s major pest of agriculture, feeding on over 300 hosts in 68 plant families. Resistance cases to most insecticide classes have been reported for this insect. Management of this pest in agroecosystems relies on a better understanding of how it copes with phytochemical or synthetic toxins. We have used genome editing to knock out a cluster of nine P450 genes and show that this significantly reduces the survival rate of the insect when exposed to two classes of host plant chemicals and two classes of insecticides. Functional expression of all members of this gene cluster identified the P450 enzymes capable of metabolism of these xenobiotics. The CRISPR-Cas9-based reverse genetics approach in conjunction with in vitro metabolism can rapidly identify the contributions of insect P450s in xenobiotic detoxification and serve to identify candidate genes for insecticide resistance.

Selective propylene epoxidation to propylene oxide (PO) with hydrogen peroxide (H 2 O 2 ) was carried out in a catalytic semi-batch reactor. High propylene epoxidation activity (44 h − 1 ) was observed over Nb based mesoporous silicate materials Nb-TUD-1 under mild operating conditions. The physical and chemical properties of the Nb based silicates characterized using BET, FTIR, TPD , TEM and UV–Vis revealed that the site isolation and surface acidity are crucial for PO production. Catalyst synthesis methods were investigated for their effects on PO productivity, PO selectivity and H 2 O 2 utilization efficiency. It is found that Nb-TUD-1 material synthesized by the sol–gel method is more active and selective than impregnated materials for liquid phase propylene epoxidation. Surface characterization confirms that thus synthesized Nb-TUD-1 catalysts have more Lewis acidity and less Bronsted acidity compared to the catalysts by impregnation.

Well-defined polyurethane (PU)-based grafted copolymers with poly(2-(dimethylamino)ethyl methacrylate) (PDEM) side chains were prepared by a combination of radical polymerization and condensation reaction. They can self-assemble in water to form nanoparticles with PU as the core and hydrophilic PDEM chains as the shell. Tuning the pH value of the solutions allowed these nanoparticles to stabilize either oil-in-water (O/W) or water-in-oil (W/O) Pickering emulsions. Stable O/W emulsions were obtained with PU-based nanoparticles in the pH 3–5 or pH 11–12 range due to their better affinity to the aqueous phase. In contrast, changing the pH value to the range of 8–9 significantly changed the stabilizing behavior of the nanoparticles, leading to the formation of stable W/O emulsions. Additionally, the effect of oil polarity on the emulsification behavior of the PU-based nanoparticles was investigated. For highly non-polar oil like n-hexane, only O/W emulsions were generated and the resulting emulsions showed no response to pH change.

Polyethyleneglycol-modified surfaces have become popular for anti-biofouling applications. In this paper, a novel glucose biosensor was fabricated by immobilizing glucose oxidase (GOx) onto the carboxymethyl-PEG-carboxymethyl (CM-PEG-CM) biomaterial film on the surface of glassy carbon electrode (GCE). The biosensor was applied in whole blood directly, which was based on the low values of polymer–water interfacial energy, resistance to protein adsorption and cell adhesion. The entrapped GOx could preserve its bioactivity and exhibited an excellent electrochemical behavior with a formal potential of −290 mV in phosphate buffer solution (PBS) (pH = 7.4). Response studies to glucose were carried out using differential pulsed voltammetry (DPV). The results indicated that the modified electrode can be used to determine glucose without interference from l -ascorbic acid (AA) and uric acid (UA) with the low detection limit of 1.24 × 10 −5 M. The data obtained from the biosensor showed good agreement with those from a biochemical analyzer in hospital. The GOx biosensor modified with CM-PEG-CM will have essential meaning and practical application in future that attributed to the effect of anti-biofouling and good performance.
