Untargeted metabolomics combined with vitro antioxidant to comprehensively evaluate the effect of sodium sulfite immersion on the holistic quality of mung bean sprouts

Yaxin Li,Zheng Zhou,Qi Wu,Baoguo Chen,Shuhong Ye,Yuna Cui,Yan Ding

Mung bean sprouts are widely consumed as a seasonal fresh vegetable, renowned for their affordability and richness in antioxidants and bioactive compounds. This study employed ultra-high-performance liquid chromatogram-Q-Exactive HF mass spectrometry (UHPLC-QE-MS) and multivariate statistical analysis to comprehensively evaluate the chemical profile of mung bean sprouts following sulfite immersion. The findings revealed a significant alteration in the overall chemical composition of mung bean sprouts following sodium sulfite immersion. Eleven components, including four sulfur-containing compounds, were identified as characteristic markers distinguishing between non-immersed and sodium sulfite-immersed mung bean sprouts. Esterification and addition reactions were inferred to occur during sodium sulfite immersion, leading to the transformation of flavonoid and saponin sulfates. Commercial samples analysis indicated that sulfur-containing compounds were detectable in 9 of 11 commercial mung bean sprouts. Meanwhile, when sodium sulfite concentration exceeded 3.00 mg/mL and immersion time exceeded 360 min, the contents of total polyphenol and flavonoid were significantly reduced and the antioxidant activity was adversely influenced.

Undenatured type II collagen nanofibrils with sodium alginate coating: Structural characterization, physicochemical properties and capability to load curcumin

Yuan Wang,Jinfang Liu,Like Mao,Fang Yuan,Yanxiang Gao

In this study, the structural design and physicochemical property enhancement of undenatured type II collagen (UC-II) nanofibrils with sodium alginate (SA) coating induced by calcium ions (Ca 2+ ) were investigated. The research aimed to elucidate the impact of Ca 2+ concentration on the morphology, thermal stability , and digestive resistance, as well as to assess the potential of UC-II/SA nanofibrils as a delivery system for curcumin (Cur). A series of Ca 2+ concentrations (1–9 mM) were methodically applied to optimize the condition that maintains the triple-helical structure of UC-II, thereby enhancing its functional properties. It was found that the Ca 2+ level up to 5 mM effectively preserved the structural integrity and improved thermal stability of UC-II, with the added benefit of ensuring the substantial delivery of active fragment to small intestine (70.7 %), which was 3.43 times greater than that of uncoated UC-II. Moreover, incorporating Cur into the UC-II/SA nanofibrils resulted in a 300 times increase in Cur solubility and showcased the superior dispersion stability, antioxidant activity , and sustained release profile during simulated digestion. These findings underscored the dual functionality of the UC-II/SA system as both a stabilizing agent for UC-II nanofibrils and an efficient carrier for Cur delivery.

UV-B Radiation Exhibited Tissue-Specific Regulation of Isoflavone Biosynthesis in Soybean Cell Suspension Cultures

Mian Wang,Yiting Wang,Muhammad Bilal,Chong Xie,Pei Wang,Xin Rui,Runqiang Yang

Isoflavones, a class of substances with high biological activity, are abundant in soybeans. This study investigated isoflavone biosynthesis in soybean cell suspension cultures under UV-B radiation. UV-B radiation enhanced the transcription level and activity of key enzymes involved in isoflavone synthesis in cell suspension cultures. As a result, the isoflavone contents significantly increased by 19.80% and 91.21% in hypocotyl and cotyledon suspension cultures compared with the control, respectively. Meanwhile, a significant difference was observed in the composition of isoflavones between soybean hypocotyl and cotyledon suspension cultures. Genistin was only detected in hypocotyl suspension cultures, whereas glycitin, daidzein, and genistein accumulated in cotyledon suspension cultures. Therefore, UV-B radiation exhibited tissue-specific regulation of isoflavone biosynthesis in soybean cell suspension cultures. The combination of suspension cultures and abiotic stress provides a novel technological approach to isoflavone accumulation.

Tough polyacrylic acid hydrogels with stable swelling and active functionalities enabled by quaternized cellulose nanofibrils and iron ions for absorbent pad interlayers

Wenjiao Ge,Shan Cao,Han Yu,Xiaohui Wang

Hydrogels are highly sought-after absorbent materials for absorbent pads; however, it is still challenging to achieve a satisfactory balance between mechanical performance, water absorption capacity, and active functionalities. In this work, we presented double-network hydrogels synthesized through acrylic acid (AA) polymerization in the presence of quaternized cellulose nanofibrils (QCNF) and Fe 3+ . Spectroscopic and microscopic analyses revealed that the combined QCNF and Fe 3+ facilitated the formation of double-network hydrogels with combined chemical and physical crosslinking. The synergistic effect of QCNF and Fe 3+ resulted in impressive mechanical properties, including tensile strength of 1.98 MPa, fracture elongation of 838.8 %, toughness of 7.47 MJ m −3 , and elastic modulus of 0.35 MPa. In comparison to the single-network PAA hydrogel, the PAA/QCNF/Fe 3+ (PQFe) hydrogels showed higher and relatively stable swelling ratios under varying pH levels and saline conditions. The PQFe hydrogels exhibited notable antioxidant activity, as evidenced by the 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay, and demonstrated effective antibacterial activity against both Escherichia coli ( E. coli ) and Staphylococcus aureus ( S. aureus ). These hydrogels show promising potential as an absorbent interlayer in absorbent pads for active food packaging.

Treg-enhancing and immunomodulating microgel scaffold promotes cell ingrowth and heart function recovery post-acute myocardial infarction in vivo

Shuqin Wang,Kai Wang,Wangbei Cao,Liang Song,Shifen Li,Zihe Zhai,Liyin Shen,Yang Zhu,Wenxing Liu,Changyou Gao

The ingrowth of endogenous cells into tissue scaffolds and modulation of Treg population take a pivotal function in restoring the structure and function of heart post-acute myocardial infarction (MI). However, the traditional hydrogels are failed to achieve such functions due to their critically small mesh size and inherent bioinertness. In this study, a Treg-regulated microgel scaffold (GTK-TK-drug) was prepared by inter-microgels assembly of microgels having cyclodextrin (CD) or adamantane (Ad) with a particle size of about 220 μm. The raw materials to obtain the microgels included gelatin methacryloyl (GelMA), reactive oxygen species (ROS)-responsive crosslinking agent poly(ethylene glycol)-thioketal-poly(ethylene glycol) (PEG-TK-PEG), ROS-responsive release prodrug aminooxyacetic acid (PEG-TK-AOA), PEG-CD or PEG-Ad, and photoinitiator lithium phenyl-2,4,6-trimethylbenzoylphosphinate (LAP). Quantitative real-time PCR (qPCR), cell counting kit-8, and immunofluorescence staining revealed the effectiveness of AOA for the differentiation of T cells into Treg and the ingrowth of cells in vitro and in vivo . During the short treatment of MI in vivo , the levels of plasma inflammatory factors, cardiomyocyte apoptosis and inflammatory cells were reduced significantly. In the long-term animal experiments, the recovery of cardiac function was significantly enhanced as characterized by echocardiography and the degree of myocardial fibrosis compared to the unassembled microgels control.

Time-space regulating prodrug hydrogels for prevention of peritendinous adhesion

Zhipeng Ni,Haiying Zhou,Haojie Yu,Li Wang,Chenguang Ouyang,Jian Yang,Yanzhao Dong,Ahmad Alhaskawi,Tian Tu,Hui Lu

The bidirectional regulations of antiadhesion and regeneration are crucial for Achilles tendon injuries. However, the precise prevention and treatments of peritendinous adhesions both in time and space are still great challenges. Here, a series of time–space regulating prodrug (tannic acid (TA), epigallocatechin (EGC) and epigallocatechin gallate (EGCG)) hydrogels are designed for the prevention of peritendinous adhesion in Achilles tendon contusion and rupture injuries. In the term of space, the self-healing and deformable hydrogels prepared by crosslinking phenylboric acid (PBA) functionalized polyphosphazene (PZBA) with guar gum (GG) through ROS-responsive phenylborate ester (PBAE) can specifically aggregate to the high ROS sites via the ROS-responsive degradation and recombination of PBAE. Thus, providing support and acting as barriers to inhibit fibroblast proliferation. In the term of time, the prodrug strategy using ROS-responsive PBAE as bridging linker can achieve long-term anti-inflammatory effect over 2 weeks. Furthermore, the degradation time of hydrogels matched the course of peritendinous adhesion, thus achieved bidirectional regulations (decrease the gene expression of TGF-β1, COL-1 and COL-3 to antiadhesion in the early stage but increase them to promote regeneration in the late stage). This study presents a promising approach employing smart prodrug hydrogels with time–space regulating properties for prevention of peritendinous adhesion.

Vitamin C derived carbon dots: inhibiting amyloid aggregation and scavenging reactive oxygen species

Tiange Fan,Xiuyun Cao,Chao Wang,Xu Shao,Xin Wang,Ping Guan,Xiaoling Hu

The pathogenesis of Alzheimer's disease is very complex, so its multifunctional treatment is of great significance, in which the synergistic therapy of the amyloid cascade hypothesis and oxidative stress hypothesis shows good results. In this study, bifunctional carbon dots (CACDs) that can inhibit amyloid aggregation and relieve oxidative stress were synthesized. Studies have shown that CACDs have a good antioxidant effect. When the concentration of CACDs is 100 μg mL−1, the elimination efficiency of DPPH (2,2-diphenyl-1-picrylhydrazyl) reaches 65.57%, and the scavenging efficiency of ˙O2− and ˙OH reaches 79.6% and 73.9%, respectively. Moreover, CACDs possess outstanding scavenging abilities against the ROS in cells, thus resulting in the mitigation of cellular oxidative damage. Meanwhile, CACDs can also bind to lysozyme proteins through hydrophobic interactions to further interfere with the amyloid self-assembly process. According to thioflavin T (ThT) analysis, the inhibition efficiency of CACDs on amyloid proteins gradually increases with increasing concentration. Circular dichroism spectroscopy (CD) further indicates that the CACDs can inhibit the transition of the protein structure to the β-sheet structure. CACDs and Aβ42 also have strong binding ability. CACDs have excellent biocompatibility and can alleviate the cytotoxicity caused by Aβ oligomers. The results demonstrate that CACDs have promising applications in multifunctional materials and important applications in multi-target therapy of Alzheimer's disease.

Water-dispersible macromolecular antioxidants for toughening and strengthening cellulose membranes

Jie Wang,Wei Gao,Yu Jin,Wangmao Tian,Yutao Zhang,Chengcheng Hu,Baoxia Wang,Shuqi Dong,Liang Yuan

Biodegradable packaging materials from cellulose are eco-friendly alternatives to traditional petroleum-based plastics. Balancing its mechanical properties as well as protective values (antioxidation, oxygen barrier, etc.) is critical. However, most studies to improve its antioxidation performance were accompanied by sacrificed mechanical properties. In the current work, a series of linear -COOH functionalized phenolic polymers were prepared from phenolic compounds (vanillin, 3,4-dihydroxy benzaldehyde) through a facile tri-component thiol-aldehyde polycondensation . While circumventing the cumbersome protection-deprotection of phenol groups, the one-pot strategy also affords water dispersible polymers for fabricating composites with cellulose nanofibers in an aqueous medium. After introducing 5–10 wt% of the copolymers , a minor soft phase was formed inside the composites, contributing to enhanced mechanical strength , toughness, antioxidation capability, and ultra-violet blocking performance, while its oxygen barrier property was well maintained.

Water-in-water emulsions stabilized by konjac glucomannan/tragacanth gum hydrogels for riboflavin delivery

Nitong Bu,Huanglong Lin,Di Zhang,Xianrui Chen,Pingping Tan,Yuanbo Zhong,Liying Huang,Jie Pang,Lin Wang,Ruojun Mu

Water-in-water (W/W) emulsions as innovative transport for hydrophilic functional ingredients inherently lack stability due to the ultralow interfacial tension and thick interfacial layer. In this work, we first proposed a simple strategy to construct stable dextran (DEX)-in-poly (ethylene glycol) (PEG) W/W emulsions by utilizing the unique phase preference of konjac glucomannan/tragacanth gum (KT) hydrogels formed based on the cooling-induced intermolecular hydrogen bonds. KT hydrogels were demonstrated to present selective partition to PEG phase and could form physically crosslinking network, thus significantly improving the rheological behavior and stability of DEX-in-PEG (D/P) emulsions with no flocculation after 31 d of storage at 25 °C. Meanwhile, the average droplet size of emulsions decreased from 15.37 to 8.48 μm with TG concentration tuning from 0.2 % to 0.8 %. When encapsulating riboflavin (RIB) in D/P emulsions stabilized by KT-4 hydrogel, the storage and lighting stability of RIB were enhanced with retention rate of 60.94 % and 21.89 %, respectively. In addition, the release of RIB in the simulate gastrointestinal environment substantially reduced and the antioxidant activity of RIB was maximumly protected due to the unique pH-responsiveness of KT hydrogel network. This work provides a feasible strategy for designing stable W/W emulsions stabilized by natural polysaccharide hydrogels, and biocompatible carriers for hydrophilic functional ingredients.

Whey protein nanoparticles: Enhancing solubility, environmental resistance, and bio-accessibility of pterostilbene

Xiuping Liang,David Julian McClements,Zhengyu Jin,Long Chen

Pterostilbene (Pte) is a dietary phenolic compound derived from plants that offers numerous physiological advantages. Nonetheless, its low water solubility and limited oral bioavailability restrict its bioactivity. To address these limitations, nanoparticles were prepared using anti-solvent precipitation method. Optimal nanoparticles were achieved at a 10:1 mass ratio of whey protein isolate (WPI) to Pte, resulting in a particle size of 104.54 nm, a polydispersity index of 0.42, and an impressive encapsulation efficiency of Pte at 85.02%. The water solubility of Pte in the nanoparticles was found to be 53.14 times higher than that of free Pte and remained stable under various environmental conditions. Furthermore, the DPPH and ABTS + free radical scavenging activities of the nanoparticles were maximized at 55.58% and 92.97%, respectively. Pte release from WPI-Pte nanoparticles was well-fitted to Korsmeyer-Peppas model and followed a non-Fickian mechanism controlled by both diffusion and erosion. This environmentally friendly, straightforward, and efficient approach shows promise for large-scale production of Pte products with amplified health benefits.

Study of Zn2+ complexation driven zein-based W/O/W Pickering emulsions for enhancing curcumin bioaccessibility and fluorouracil bioactivity

Long Deng,Junqiu Liao,Weiqi Liu,Shiyuan Liu,Xiaoxiao Liang,Rujin Zhou,Yanbin Jiang

Overcoming gastrointestinal tract barriers to enhance bioavailability is a major challenge for oral delivery. The innovative design of multi-compartment functional Pickering emulsions would provide an ideal option for targeted co-delivery of bioactive components. In this study, pH-responsive water-in-oil-in-water (W/O/W) Pickering double emulsions co-loaded with curcumin and fluorouracil were fabricated by zein/anthocyanin (ACN)/Zn 2+ nanoparticles (ZAZn NPs) via a one-step emulsification process. The unique emulsifier was designed by depositing ACN-Zn 2+ complexes on zein nanoparticles (zein NPs). The binding of ACN to the nanoparticle surface through hydrogen bonding interaction enhanced the hydrophilization and stimuli-response of zein NPs. Furthermore, the co-ordination of Zn 2+ and ACN facilitated the formation of nanoparticles with near-neutral wettability, which served as the dominant driving force for the one-step generation of W/O/W Pickering emulsions. The droplet size, distribution and internal structure of Pickering double emulsions were influenced by nanoparticle concentration and oil fraction. Due to Zn 2+ complexation with curcumin at the oil/water interface, the W/O/W Pickering emulsions obtained exhibited high curcumin encapsulation efficiency (91.2%) and free fatty acid release degree (65.9%), resulting in increased bio-accessibility (71.0%). The simulated gastrointestinal release demonstrated that curcumin and fluorouracil achieved controlled intestinal release, which was driven by a pH-mediated competitive adsorption-desorption mechanism. Moreover, in vitro cell experiments showed that co-encapsulated Pickering double emulsions exhibited 2.5 times greater bio-activity than those containing fluorouracil only. This facile strategy for preparing Pickering double emulsions is promising for the co-delivery of different physico-chemical bio-active compounds, which would improve oral bio-accessibility and bio-activity.

Study on the interaction mechanism and physicochemical properties of luteolin and its glucoside compounds with β-lactoglobulin

Zihao Chen,Min Fu,Jun Chen,Guowen Zhang,Ti Li,Qin Geng,Taotao Dai

Molecular docking simulations and multispectral approaches were used to study the non-covalent interactions between β-lactoglobulin (β-LG) and chosen luteolin (Lu), isoorientin (Iso), and cynaroside (Cyn). The fluorescence quenching results demonstrated that Lu, Iso, and Cyn spontaneously formed stable complexes with β-LG through static quenching. Thermodynamic parameters and molecular docking simulations demonstrated that hydrogen bonds were the primary forces driving the interactions between the protein and flavonoids. The binding constants followed the order: β-LG-Lu (2.87 × 10⁴ L/mol) > β-LG-Iso (1.83 × 10⁴ L/mol) > β-LG-Cyn (1.52 × 10⁴ L/mol). Additionally, the C-glycosylated flavonoid (Iso) showed a higher propensity to form hydrogen bonds with β-LG compared to the O-glycosylated flavonoid (Cyn). The results of circular dichroism indicated that the β-LG structure became looser with the introduction of Lu, Iso, and Cyn. Furthermore, the assembly of β-LG-flavonoid complexes decreased the surface hydrophobicity and oil-water interfacial tension of β-LG, improved its foaming and emulsifying capabilities, and protected the antioxidant properties of the flavonoids to a certain extent. This study investigated the interaction mechanisms between flavonoids with different glycosyl types and β-LG, providing insights for the incorporation of suitable flavonoids into dairy products, and offering theoretical perspectives on preserving the antioxidant properties of flavonoids.
