Development of pH-Sensitive Intelligent Packaging Films Based on Polyvinyl Alcohol Incorporated With Lycium ruthenicum Anthocyanin and Curcumin for Real-Time Monitoring of Mutton Freshness

Yonghua Bao,Weiyuan Chen,Xiaohong Zhou,Sudan Ye,Chunhui Fu

A multifunctional intelligent packaging film was fabricated by co-incorporating Lycium ruthenicum anthocyanins (LRAs) and curcumin (CUR) into a polyvinyl alcohol (PVA) matrix. Microstructural and spectroscopic analyses supported the successful incorporation of both pigments and their intermolecular interactions with the PVA matrix. Notably, the physicochemical and functional properties of the composite films varied with the LRA/CUR ratio. Specifically, the CUR-dominant formulation (1:2) exhibited the highest elongation at break (86.13%) and antioxidant activity (DPPH scavenging rate of 63%) among the binary formulations, together with stronger antibacterial activity. Conversely, the LRA-dominant film (2:1) exhibited greater color stability and the strongest ammonia response (maximum Δ E = 35.02). When applied to chilled mutton stored at 4°C for 8 days, the intelligent films displayed progressive color transitions from yellow-orange to reddish-orange as spoilage progressed. These chromatic variations were accompanied by increases in pH (from 6.42 to 7.41) and total volatile basic nitrogen (TVB-N) content (from 9.29 to 39.41 mg/100 g). Overall, the LRA/CUR-PVA films combined freshness indication with active preservation, demonstrating their potential for intelligent chilled-meat packaging.

The solid dispersion of a novel curcumin analogue (CACN136) prepared using PVP/Mannitol composite carrier significantly improved oral bioavailability and depression-like behavior in mice

Yumeng Wei,Ke Gong,Rongzhu Li,Qiong Zhang,Yueting Zhong,Wenmei Zhao,Ying Zuo,Shifeng Chu,Ling Zhao

CACN136-SD increased CACN136 solubility and rapid dissolution. • Solid dispersion enhanced CACN136 oral bioavailability in rats. • CACN136-SD improved depression-like behaviors in CUMS mice. • CACN136-SD regulated brain 5-HT and MAO without obvious toxicity. Objective CACN136 is a novel curcumin analogue with antidepressant potential, but its poor aqueous solubility may restrict oral absorption. This study aimed to develop a rapidly dissolving solid dispersion of CACN136 (CACN136-SD) to further enhance its oral bioavailability and antidepressant efficacy. Methods CACN136-SD was prepared using a solvent method with PVP and mannitol as composite carriers. Physicochemical characterization was performed through in vitro experiments, followed by pharmacokinetic and antidepressant behavioral evaluations. Results CACN136-SD increased the apparent aqueous solubility of CACN136 approximately 20-fold. Cumulative dissolution at 10 min reached 95.69%, versus 6.64% for the active pharmaceutical ingredient (API). Differential scanning calorimetry and X-ray powder diffraction showed markedly reduced drug crystallinity, with CACN136 present predominantly in an amorphous or highly disordered state within the carrier matrix. Fourier-transform infrared spectroscopy showed changes in the local vibrational environment of CACN136 after incorporation into the matrix. The apparent terminal half-life of CACN136-SD was approximately 23 times that of the API, and its systemic exposure (AUC-t) was 8.95 times higher. In the chronic unpredictable mild stress (CUMS) model, CACN136-SD significantly alleviated depression-like behaviors and produced greater changes in serum and hippocampal 5-hydroxytryptamine (5-HT) and monoamine oxidase (MAO) levels than unformulated CACN136. Preliminary safety assessments detected no obvious cardiac, hepatic, or renal toxicity under the tested conditions. Conclusion CACN136-SD improved the solubility, dissolution, and oral exposure of CACN136 and showed enhanced antidepressant-like activity associated with changes in monoamine-related markers. This solid dispersion represents a promising antidepressant candidate with potential for clinical translation. Download: Download high-res image (159KB) Download: Download full-size image

Intelligent indicator films based on cold plasma-modified chitosan loaded with curcumin-resveratrol nanoemulsions: pH-responsive mechanism, colorimetric performance and application for tilapia freshness monitoring

Meng Qiu,Tingting Yang,Guanghua Xia,Jiamei Wang,Zhongyuan Liu,E Liao,Wei Xu,Liming Zhang,Wenmin Wu

Zein nanoemulsions encapsulated curcumin and resveratrol for pH-sensitive smart films. • Cold plasma (CP) modification regulated bioactive release in composite films. • CP at 30 kV endowed films with excellent mechanical, barrier and antioxidant properties. • The films showed pH-responsive color change to volatile amines from tilapia spoilage. Food safety risks associated with spoilage and plastic packaging have driven the development of sustainable intelligent packaging systems. In this study, zein-curcumin-resveratrol‑sodium alginate nanoemulsions were incorporated into chitosan films to construct pH-responsive active indicator films. Cold plasma (CP) modification was applied to regulate surface properties and enhance sensing performance. CP modification increased surface roughness and active sites, improving the interaction between films and volatile amines. The 30 kV CP-modified film exhibited optimal comprehensive properties, with improved mechanical strength (tensile strength of 5.36 ± 0.18 MPa and elongation at break of 46.95 ± 3.14%), antioxidant activities (73.81 ± 1.50% for DPPH and 70.26 ± 1.41% for ABTS), pH responsiveness, barrier properties and UV protection. Moreover, colorimetric responses (ΔE and a* values) showed a significant positive correlation with tilapia TVB-N accumulation (R 2  > 0.95), verifying the quantitative relationship between film discoloration and fish freshness deterioration. This study provides a practical intelligent packaging strategy for real-time aquatic product quality monitoring. Download: Download high-res image (158KB) Download: Download full-size image

The effect of the molecular weight of hyaluronic acid on the physicochemical characterization of hyaluronic acid-curcumin conjugates and in vitro evaluation in glioma cells

Cihui Tian,Sajid Asghar,Yurui Xu,Zhipeng Chen,Mei Zhang,Lin Huang,Junxiu Ye,Qineng Ping,Yanyu Xiao

In this study, a redox-sensitive glioma-targeting micelle system was designed to deliver curcumin (CUR) by conjugating it to hyaluronic acid (HA-s-s-CUR, HSC) via disulfide linkage. The effect of the molecular weight of HA on the physicochemical characteristics of HSC conjugates and their in vitro effects in glioma cells were also explored. These conjugates formed nano-scale micelles (209–926 nm) independently in aqueous solution. The micelles greatly increased the solubility of CUR and improved its stability, which is crucial for harnessing the therapeutic potential of this active molecule. The redox sensitivities of different HSC micelles were measured by using a dynamic light scattering method and in vitro release assay, which showed that the low (50 kDa) and medium molecular weight (200 kDa and 500 kD) HA-based conjugates were sensitive to GSH, whereas higher molecular weights (1000 kDa and 2000 kDa) did not show redox-sensitivity. Increased cytotoxicity and uptake of low and medium molecular weight-modified HSC conjugates by the glioma cells further confirmed that the sensitive micelles are more effective for intracellular drug delivery compared to the high molecular weight-modified HSC conjugates or the plain CUR . In summary, the molecular weight of HA affects the physicochemical attributes of HSC conjugates. Only HSC micelles made with HA molecules less than 500 kDa exhibit redox sensitivity.

Combination of Astragali Polysaccharide and Curcumin Improves the Morphological Structure of Tumor Vessels and Induces Tumor Vascular Normalization to Inhibit the Growth of Hepatocellular Carcinoma.

Decai Tang,Shuo Zhang,Xiaoxia Shi,Jiafei Wu,Gang Yin,Xiying Tan,Fuyan Liu,Xingdong Wu,Xiangyu Du

Normalizing the disordered tumor vasculature, rather than blocking it, is a novel method for anticancer therapy. Astragali polysaccharide (APS) and curcumin were reported to be active against carcinomas. However, the effect and mechanism of the combination of APS and curcumin on vascular normalization in hepatocellular carcinoma (HCC) was not clear. In the present study, effects of combined APS and curcumin on tumor vascular normalization were evaluated in HepG2 tumor-bearing mice. Photoacoustic tomography (PAT) was performed to observe the morphological structure of tumor vessels in vivo. The microstructure of the tumor vessels was also analyzed through scanning electron microscopy. Additionally, the expression of CD31 and NG2 was analyzed by immunohistochemical staining. Tumor vessels of HepG2 tumor-bearing mice treated with the combination were sparse with uniform growth, morphology rules, and complete vascular walls, which had fewer branches and sprouts. ECs of tumor vessels were arranged regularly and were tightly connected, tending toward normalization. The expression of CD31 was reduced while NG2 was increased significantly by the combination of APS and curcumin. The results indicated that APS and curcumin in combination showed a better effect on inhibiting tumor growth in an orthotopic nude-mouse model of HCC. More important, the combination induced normalization of tumor vascular better than APS or curcumin administration alone, improving the morphological structure of tumor vessels and promoting maturation of tumor vessels. The results of the present study provided a reasonable possibility for combination therapy of APS and curcumin in the treatment of HCC via tumor vascular normalization.

Highly sensitive Curcumin-conjugated nanotheranostic platform for detecting amyloid-beta plaques by magnetic resonance imaging and reversing cognitive deficits of Alzheimer's disease via NLRP3-inhibition

Ruan Yuting,Xiong Ying,Fang Wenli,Yu Qun,Mai Yingren,Cao Zhiyu,Wang Kexi,Lei Ming,Xu Jiaxin,Liu Yan,Zhang Xingcai,Liao Wang,Liu Jun

Background Alzheimer's disease (AD) is the most common neurodegenerative disorder without effective therapy and lack diagnosis strategy for preclinical AD patients. There is an urgent need for development of both early diagnosis and therapeutic intervention of AD. Results Herein, we developed a nanotheranostics platform consisting of Curcumin (Cur), an anti-inflammatory molecule, and superparamagnetic iron oxide (SPIO) nanoparticles encapsulated by diblock 1,2-dio-leoyl- sn -glycero-3-phosphoethanolamine- n -[poly(ethylene glycol)] (DSPE-PEG) that are modified with CRT and QSH peptides on its surface. Furthermore, we demonstrated that this multifunctional nanomaterial efficiently reduced β-amyloid plaque burden specifically in APP/PS1 transgenic mice, with the process noninvasively detected by magnetic resonance imaging (MRI) and the two-dimensional MRI images were computed into three-dimension (3D) plot. Our data demonstrated highly sensitive in vivo detection of β-amyloid plaques which more closely revealed real deposition of Aβ than previously reported and we quantified the volumes of plaques for the first time based on 3D plot. In addition, memory deficits of the mice were significantly rescued, probably related to inhibition of NLR Family Pyrin Domain Containing 3 (NLRP3) inflammasomes. Conclusions Gathered data demonstrated that this theranostic platform may have both early diagnostic and therapeutic potential in AD. Graphical Abstract

Curcumin attenuates renal interstitial fibrosis of obstructive nephropathy by suppressing epithelial-mesenchymal transition through inhibition of the TLR4/NF-кB and PI3K/AKT signalling pathways

Zhaohui Wang,Zhi Chen,Bingsheng Li,Bo Zhang,Yongchao Du,Yuhang Liu,Yao He,Xiang Chen

Context Renal interstitial fibrosis (RIF) is characterized by the accumulation of inflammatory cytokines and epithelial-mesenchymal transition (EMT). Curcumin exerts antifibrogenic, anti-inflammatory and antiproliferative effects. Objective To explore the mechanisms underlying the effects of curcumin on RIF.Materials and methods Eight-week-old male C57BL/6 mice were intragastrically administered curcumin (50 mg/kg/day) for 14 days after undergoing unilateral ureteral obstruction (UUO) operations. Renal function (blood urea nitrogen [BUN] and serum creatinine [Scr]) and inflammatory cytokine levels were tested using colorimetric assays and ELISA, respectively. EMT markers were evaluated through immunohistochemistry, western blotting and qPCR. Transforming growth factor beta 1 (TGF-β1; 10 ng/mL) and lipopolysaccharides (LPS; 100 ng/mL) were used to stimulate EMT and an inflammatory response in human renal proximal tubular epithelial (HK-2) cells, respectively, for further investigation. Results In vivo, curcumin significantly improved the levels of BUN and Scr by 28.7% and 21.3%, respectively. Moreover, curcumin reduced the levels of IL-6, IL-1β and TNF-α by 22.5%, 30.3% and 26.7%, respectively, and suppressed vimentin expression in UUO mice. In vitro, curcumin reduced the expression of vimentin and α-smooth muscle actin in TGF-β1-induced HK-2 cells. In LPS-induced HK-2 cells, curcumin decreased the release of IL-6, IL-1β and TNF-α by 43.4%, 38.1% and 28.3%, respectively. In addition, curcumin reduced the expression of TLR4, p-PI3K, p-AKT, p-NF- κB and p-IκBα in both LPS- and TGF-β1-induced HK-2 cells.Discussion and conclusions Curcumin repressed EMT and the inflammatory response by inhibiting the TLR4/NF-κB and PI3K/AKT pathways, demonstrating its potential utility in RIF treatment.

A novel fast-releasing formulation containing curcumin and anti-microbial peptide-encapsulated transdermal hydrogels into microneedles to reduce inflammation in the therapy of neonatal sepsis

Qian Min,Ma Di,Zhao Xiaoqi,Guo Wanxu,Zhang Yunfeng

Neonatal sepsis is a leading cause of neonatal mortality worldwide. Typically, intramuscular injections of gentamycin and vancomycin have been the preferred treatment for sepsis. The use of these drug molecules has many complications because of some side effects, proper dosage calculations, and harmless injection supply. In the present study, we developed new combinations of therapeutic drug molecules containing anti-inflammatory curcumin with bioactive antimicrobial peptides (AMP). We designed and developed a biocompatible and thermally responsive chitosan/polyacrylamide polymeric microneedle array to deliver AMP/curcumin nanoparticles (Nano-Cur) transdermally. The prepared drug-loaded microneedles were investigated for their chemical and morphological structure, in vitro insertion, and transdermal drug release. This investigation established that AMP/Nano-Cur-loaded microneedles have potential effects in sepsis treatment.

Curcumin protects against hepatic stellate cells activation and migration by inhibiting the CXCL12/CXCR4 biological axis in liver fibrosis:A study in vitro and in vivo

Lifeng Qin,Jinmei Qin,Xiumei Zhen,Qian Yang,Liyi Huang

The C-X-C motif chemokine 12/C-X-C chemokine receptor type 4 (CXCL12/ CXCR4) biological axis plays an important role in the pathogenesis of liver fibrosis . Curcumin is known to have an anti-fibrosis effect, but the specific mechanism needs to be elucidated. There is currently no evidence illustrating a connection between curcumin and the CXCL12/CXCR4 axis in liver fibrosis . Here, we investigated the contribution of curcumin on CXCL12/ CXCR4 biological axis in liver fibrosis. Our results showed that curcumin remarkably improved hepatic function and liver fibrosis, and the effects are similar as silymarin . The alleviation of liver fibrosis with curcumin treatment was associated with a reduction of CXCL12, CXCR4, α-SMA and RhoA. In addition, curcumin markedly inhibited the proliferation and migration of HSC-T6 cells. This study indicates that curcumin could protect against hepatic stellate cells activation and migration by inhibiting the CXCL12/CXCR4 biological axis in liver fibrosis.

Characterization, adsorption kinetic and in vitro release behavior of curcumin loaded with porous mannitol and porous lactose: Template agent method vs. Pore-forming agent method

Zhe Li,Xiaosui Luo,Abid Naeem,Zhengji Jin,Qiong Li,Yongmei Guan,Lihua Chen,Weifeng Zhu,Liangshan Ming

Polyvinylpyrrolidone K30 was used as the templating agent, and ammonium bicarbonate was used as the pore-forming agent to make porous mannitol and porous lactose by the template and pore-forming agent method, respectively. Compared with the template method, the porous particles prepared by the pore-forming agent method have larger pore diameter (320.276 nm and 250.528 nm) and specific surface area (1.018 m 2 /g and 0.913 m 2 /g). The molecular docking results showed that mannitol/lactose interacted with curcumin and adhered to each other through hydrogen bonding. The adsorption kinetics process of porous mannitol and porous lactose prepared by template agent, pore-forming agent and curcumin were different. Among the curcumin-loaded porous particles prepared by the two methods, the curcumin-loaded porous lactose prepared by the pore-forming agent method had the fastest release rate and the highest cumulative release rate (95 %). Curcumin releases consistent with the Peppas release kinetics model and the diffusion mechanism.

Fabrication and characterization of curcumin-loaded complex acid-induced cold gels of soybean and red bean protein isolates incorporated with common and waxy corn starches

Qi Qi,Xiaopian Yan,Yuxuan Mao,Fang Wang,Hao Zhang,Xiuying Xu,Yuzhu Wu,Chengbin Zhao,Jingsheng Liu

Soybean protein isolate (SPI) and red bean protein isolate (RBPI) were mixed with common starch (CS) and waxy starch (WS) from corn respectively, and then subjected to glucono-δ-lactone (GDL)-induced crosslinking to form the protein–starch complex cold gels, which was used as a delivery carrier for curcumin. The complex gel with addition of CS had denser network structure and higher gel strength, resistance to deformation and viscoelasticity, while the complex gel with WS had the opposite effect. The protein interacted more strongly with CS than WS, enhancing the structural and thermal stability of the complex gel, which resulted in a higher curcumin encapsulation ratio in the gel and its chemical stability during light and heat treatment. The intensity and location of many characteristic absorption peaks in the infrared spectra changed during simulated gastrointestinal digestion, which indicated the destruction of gel structure, leading to the transformation of α-helix and β-sheet into β-turn and random coil in the secondary structure. The addition of CS and WS reduced the release of curcumin in gastric phase, but promoted its directional release in intestinal phase, due to the changes in the digestion pattern of the complex gel, especially the complex gel with addition of WS had a better controlled release effect, which was also related to the more disordered and flexible secondary structure of its gastric digests. The protection of curcumin by gel network and the targeted release behavior of curcumin during intestinal digestion improved its bioaccessibility. In addition, the curcumin in RBPI-based gels had higher encapsulation ratio, chemical stability and bioaccessibility than that in SPI-based gels. Therefore, protein–starch complex cold gels can be used as controlled release carriers for the protection and delivery of sensitive bioactive compounds or nutrients.

Curcumin Prevents Neuroinflammation by Inducing Microglia to Transform into the M2-phenotype via CaMKKβ-dependent Activation of the AMP-Activated Protein Kinase Signal Pathway

Qiao Peifeng,Ma Jingxi,Wang Yangyang,Huang Zhenting,Zou Qian,Cai Zhiyou,Tang Yong

Background: Neuroinflammation plays an important role in the pathophysiological process of various neurodegenerative diseases. It is well known that curcumin has obvious anti-inflammatory effects in various neuroinflammation models. However, its effect on the modulation of microglial polarization is largely unknown. Objective: This study aimed to investigate whether curcumin changed microglia to an anti-inflammatory M2-phenotype by activating the AMP-activated protein kinase (AMPK) signaling pathway. Methods: LPS treatment was used to establish BV2 cells and primary microglia neuroinflammation models. The neuroinflammation mouse model was established by an intracerebroventricular (ICV) injection of lipopolysaccharide (LPS) in the lateral septal complex region of the brain. TNF-α was measured by ELISA, and cell viability was measured by Cell Counting Kit-8 (CCK-8). The expression of proinflammatory and anti-inflammatory cytokines was examined by Q-PCR and Western blot analysis. Phenotypic polarization of BV2 microglia was detected by immunofluorescence. Results: Curcumin enhanced AMPK activation in BV2 microglial cells in the presence and absence of LPS. Upon LPS stimulation, the addition of curcumin promoted M2 polarization of BV2 cells, as evidenced by suppressed M1 and the elevated M2 signature protein and gene expression. The effects of curcumin were inhibited by an AMPK inhibitor or AMPK knockdown. Calmodulin-dependent protein kinase kinase β (CaMKKβ) and liver kinase B1 (LKB1) are upstream kinases that activate AMPK. Curcumin can activate AMPK in Hela cells, which do not express LKB1. However, both the CaMKKβ inhibitor and siRNA blocked curcumin activation of AMPK in LPS-stimulated BV2 cells. Moreover, the CaMKKβ inhibitor and siRNA weaken the effect of curcumin suppression on M1 and enhancement of M2 protein and gene expression in LPS-stimulated BV2 cells. Finally, curcumin enhanced AMPK activation in the brain area where microglia were over-activated upon LPS stimulation in an in vivo neuroinflammation model. Moreover, curcumin also suppressed M1 and promoted M2 signature protein and gene expression in this in vivo model. Conclusion: Curcumin enhances microglia M2 polarization via the CaMKKβ-dependent AMPK signaling pathway. Additionally, curcumin treatment was found to be neuroprotective and thus might be considered as a novel therapeutic agent to treat the neurodegenerative disease such as Alzheimer's disease, Parkinson's disease, etc.
