Synergistic Antioxidant Effects of Cysteine Derivative and Sm-Cluster for Food Applications

Lingxia Chen,Lijun Wang,Lifu Ma,Chao Wang,Xinshu Qin,Minlong Wang,Xiaohe Zhang,Ruoyan Yang,Bing Fang,Jie An

The incorporation of antioxidants in food products is essential to prevent or delay deterioration, thereby addressing food spoilage. Thiol compounds, recognized for their natural antioxidant properties, are widely used in various foods; however, their antioxidant capacity is often limited. This study investigates the potential enhancement of thiol antioxidant capacity through the addition of a soluble, low-toxic inorganic Sm-cluster. Our findings demonstrate that the Sm-cluster significantly bolsters the antioxidant efficacy of thiol compounds. We explored, for the first time, the in vitro antioxidant activities of an Sm-oxo/hydroxy cluster combined with a cysteine derivative for potential food applications. The composition exhibited a robust inhibition of aromatic aldehyde flavor compound oxidation and displayed strong, dose-dependent DPPH (2,2-diphenyl-1-picrylhydrazine) radical scavenging activity. Notably, the antioxidant activity of the Sm-cluster/cysteine derivative was further enhanced under strong visible light conditions, which typically increased the likelihood of oxidation. These results suggest that the combination of inorganic cluster and thiol compounds presents a promising natural alternative to traditional antioxidants in the food industry.

Study of Zn2+ complexation driven zein-based W/O/W Pickering emulsions for enhancing curcumin bioaccessibility and fluorouracil bioactivity

Long Deng,Junqiu Liao,Weiqi Liu,Shiyuan Liu,Xiaoxiao Liang,Rujin Zhou,Yanbin Jiang

Overcoming gastrointestinal tract barriers to enhance bioavailability is a major challenge for oral delivery. The innovative design of multi-compartment functional Pickering emulsions would provide an ideal option for targeted co-delivery of bioactive components. In this study, pH-responsive water-in-oil-in-water (W/O/W) Pickering double emulsions co-loaded with curcumin and fluorouracil were fabricated by zein/anthocyanin (ACN)/Zn 2+ nanoparticles (ZAZn NPs) via a one-step emulsification process. The unique emulsifier was designed by depositing ACN-Zn 2+ complexes on zein nanoparticles (zein NPs). The binding of ACN to the nanoparticle surface through hydrogen bonding interaction enhanced the hydrophilization and stimuli-response of zein NPs. Furthermore, the co-ordination of Zn 2+ and ACN facilitated the formation of nanoparticles with near-neutral wettability, which served as the dominant driving force for the one-step generation of W/O/W Pickering emulsions. The droplet size, distribution and internal structure of Pickering double emulsions were influenced by nanoparticle concentration and oil fraction. Due to Zn 2+ complexation with curcumin at the oil/water interface, the W/O/W Pickering emulsions obtained exhibited high curcumin encapsulation efficiency (91.2%) and free fatty acid release degree (65.9%), resulting in increased bio-accessibility (71.0%). The simulated gastrointestinal release demonstrated that curcumin and fluorouracil achieved controlled intestinal release, which was driven by a pH-mediated competitive adsorption-desorption mechanism. Moreover, in vitro cell experiments showed that co-encapsulated Pickering double emulsions exhibited 2.5 times greater bio-activity than those containing fluorouracil only. This facile strategy for preparing Pickering double emulsions is promising for the co-delivery of different physico-chemical bio-active compounds, which would improve oral bio-accessibility and bio-activity.

Synthesis and structure characterization of boron complexes containing C2-symmetric tertiary amine ligands: Assessment catalytic degradation of dyes and antioxidant properties

Minggang Hu,Xiaochen Wang,Can Wang,Hongtao Chu,Xinfeng Song,Guohua Dong,Jiyuan Zhang,Jin Xing

In this study, a new and simple method for synthesizing boron complexes was presented. The boron complexes with C 2 -symmetric ligands were prepared from in situ generated tertiary amine by the reaction of salicylaldehyde with primary amine in the presence of sodium borohydride. The boron complexes were characterized by 1 H and 13 C NMR spectroscopy, elemental analysis, and mass spectrometry. The crystal structures of complexes 1 and 2 were determined by single crystal X-ray diffraction method. The complex is a mononuclear boron complex with a tertiary amine ligand. The boron center is tetracoordinated with a distorted tetrahedral geometry. Catalytic activities of the boron complexes in the reduction of dyes, was investigated using sodium borohydride as a reducing agent. Complexes 1 and 2 were effective catalysts for degradation of dyes. Kinetic studies reveal that the reduction of methylene blue is of the first order. Free radical scavenging activities of the boron complexes have been tested using DPPH, ABTS and hydroxyl radicals, respectively. The result shows that the boron complexes have effective antioxidant activity.

TA-V Nanozymes with Acid Resistance Capabilities Effectively Target and Alleviate Ulcerative Colitis Lesions via Oral Delivery

Zhenzhen Jin,Xiangjing Cao,Qinglong Guo,Cong Zhang,Qingrong Li,Wenqi Wang,Yong Lv,Yan Ma,Xianwen Wang

As one of the most common inflammatory bowel diseases (IBDs), ulcerative colitis (UC) has become a rising global health issue that affects people’s quality of life. Conventional therapeutic drugs have low bioavailability and may cause serious side effects due to their lack of acidic resistance and lesion-targeting capabilities. The development of novel nanomedicines to overcome these problems is urgently needed. Nanozymes have attracted attention because of their excellent catalytic efficiency in various harsh environments. In this study, tannic acid-vanadium (TA-V) nanozymes with multienzymatic and excellent antioxidant abilities, which exhibit acidic resistance and a negative surface charge, were successfully developed. All these characteristics make it possible that these nanozymes are not easily decomposed by gastric acid and can effectively accumulate in colitis lesions with a positive charge through oral delivery. In vitro and in vivo experiments further demonstrated the excellent prophylactic and therapeutic value of these compounds in the treatment of UC by scavenging reactive oxygen/nitrogen species (ROS/RNS) and mitigating the oxidative stress environment, thus downregulating the levels of the proinflammatory cytokines IL-1β, IL-6, and TNF-α. Furthermore, TA-V also showed excellent biosafety and biocompatibility without causing obvious damage to the main organs. This work provides novel preventative and therapeutic TA-V nanozymes that might have potential clinical applications in UC treatment.

Qualitative and Quantitative Analysis of Chemical Components in Yinhua Pinggan Granule with High-Performance Liquid Chromatography Coupled with Q-Exactive Mass Spectrometry

Imranjan Yalkun,Haofang Wan,Lulu Ye,Li Yu,Yu He,Chang Li,Haitong Wan

Yinhua Pinggan Granule (YPG) is an approved compounded traditional Chinese medicine (TCM) prescription for the treatment of cold, cough, viral pneumonia, and related diseases. Due to its complicated chemical composition, the material basis of YPG has not been systematically investigated. In this study, an analytical method based on high-performance liquid chromatography (HPLC) coupled with Q-Exactive mass spectrometry was established. Together with the help of a self-built compound database and Compound Discoverer software 3.1, the chemical components in YPG were tentatively identified. Subsequently, six main components in YPG were quantitatively characterized with a high-performance liquid chromatography–diode array detector (HPLC-DAD) method. As a result, 380 components were annotated, including 19 alkaloids, 8 organic acids, 36 phenolic acids, 27 other phenols, 114 flavonoids, 75 flavonoid glycoside, 72 terpenes, 11 anthraquinones, and 18 other compounds. Six main components, namely, chlorogenic acid, puerarin, 3′-methoxypuerarin, polydatin, glycyrrhizic acid, and emodin, were quantified simultaneously. The calibration curves of all six analytes showed good linearity (R2> 0.9990) within the test ranges. The precision, repeatability, stability, and recovery values were all in acceptable ranges. In addition, the total phenol content and DPPH scavenging activity of YPG were also determined. The systematic elucidation of the chemical components in YPG in this study may provide clear chemical information for the quality control and pharmacological research of YPG and related TCM compounded prescriptions.

Selenium-containing polyurethane nanofibers with MnO2 nanoparticles and gelsevirine promote diabetic wound healing by modulation of ROS and inflammation

Wajiha Ahmed,Shifen Li,Min Liang,Pai Peng,Wali Muhammad,Qiaoxuan Wang,Changyou Gao

Reactive oxygen species (ROS) and subsequent inflammatory cascades hinder the healing of diabetic wounds, which should be tackled simultaneously when designing wound dressings. In this study, ROS-responsive di‑selenium-containing polyurethane nanofibers (PUF) loaded with manganese dioxide nanoparticles (MnO 2 NPs) and gelsevirine (GSV) with an average diameter of 0.6 ± 10 μm, were prepared to specifically target ROS and inflammation control, thereby enhancing healing in diabetic wounds. The resulting nanofibers exhibited a porous architecture and enhanced drug delivery capability. The MnO 2 /GSV/PUF dressing showed negligible cytotoxicity with cell viability over 80 %, effective ROS-scavenging ability of 87 %, and inhibition of pro-inflammatory cytokine expression. This dual mechanism of ROS-scavenging and inflammation modulation accelerated the healing of diabetic wounds up to 92 % in 14 d in a full-thickness diabetic wound model, as evidenced by reduced cytokine levels, enhanced epithelialization, and uniform collagen deposition with the highest percentage of 60 %.

Remarkable Enhancement of Antioxidant Activity of the Ovalbumin–EGCG Conjugate through a Novel Preceding Selective Protection Grafting Strategy

Yumin Wang,Songbai Liu

Conventional radical grafting of proteins with catechins consumed the most antioxidant-active hydroxyls during grafting, thus failing to effectively retain antioxidant activity in conjugates. In this study, a novel strategy of selective protection of the most reactive hydroxyls before grafting was developed to preserve the most reactive hydroxyls and effectively retain antioxidant activity in conjugates. Selective protection of the most reactive hydroxyls of (−)-epigallocatechin-3-gallate (EGCG) was successfully realized in a yield of 87% applying trimethyl orthopropionate and catalytic calcium triflate at 40 °C. The novel ovalbumin (OVA)–EGCG conjugate with 93% grafting ratio was prepared by radical grafting with the selectively protected EGCG and subsequent deprotection. Substantially enhanced antioxidant performance of the novel OVA–EGCG conjugate in liposomes was unveiled with notably reduced curcumin degradation and leakage. The strategy and approaches developed in this study will be valuable to effectively improve the antioxidant activities of protein–catechin grafting conjugates.

Regulation of tea polyphenols in gluten-glucose Maillard reaction: Evaluation and analysis

Heng-Yu Liang,Hao-Xiang Gao,Zan Jing,Qiang He,Wei-Cai Zeng

Maillard reaction plays an important role in the flavour and color of some food. However, the excessive occurrence of Maillard reaction may affect the quality and safety of food. In this study, a simulated Maillard system composed of gluten and glucose was established to evaluate and analyze the regulatory role of tea polyphenols (TP) in it. The addition of tea polyphenols alleviated excessive browning of the Maillard reaction and significantly inhibited the accumulation of fructosamine and late glycosylation end products in the system. Based on the analysis of laser particle sizer and atomic force microscope, the large particle size distribution and aggregation structure of the Maillard reaction products in the tea polyphenol group disappeared. Further experiments showed that with the addition of tea polyphenols, the free radicals content in the system decreased, while the amount of reaction substrates increased. It was also shown that tea polyphenols effectively inhibited the isomerism of glucose to fructose and affected the content of free sulfhydryl and free amino groups in the system. The results indicated that tea polyphenols can regulate the Maillard reaction between gluten and glucose, which gives it the potential for application in the field of bakery and thermally processed foods.

Selective extraction and analysis of phenolic acids in herbal plants using Fe3O4@MXene@PEI aerogel

Kai Hu,Xiaobing Liu,Huifen Ma,Di Zhao,Junying Song,Huahui Zeng,Zhenqiang Zhang

A magnetic MXene aerogel (Fe 3 O 4 @MXene@PEI) was prepared by crosslinking amino modified MXene with polyethyleneimine using epichlorohydrin as a cross-linker. Adsorption properties of Fe 3 O 4 @MXene@PEI aerogel for phenolic acids were evaluated by adsorption kinetics and isotherms experiments, showing that the high adsorption affinity was governed by multilayer chemisorption process. An efficient MSPE/HPLC method was developed for the determination of phenolic acids with excellent selectivity, good linearity (0.025–5.0 μg mL −1 ), low LODs (0.007–0.017 μg mL −1 ), and satisfactory recoveries (80.0–120.0 %). Moreover, the antioxidant activity of the Fe 3 O 4 @MXene@PEI purified compounds was superior to that of the conventional method as demonstrated by the results of scavenging experiments on 2,2 -diphenyl-1-picrylhydrazyl radical scavenging assay. Finally, 65 organic acids were identified in the Fe 3 O 4 @MXene@PEI treated honeysuckle extracts by UHPLC-Q-Exactive Orbitrap MS/MS analysis. The proposed sorbent exhibits remarkable promise for the selective separation and purification of organic acids from herbal products.

Study on antifungal properties of modified chitosan combined with essential oil and its application in preserving fruits

Lingling Li,Chunhua Wu,Qian Chen,Zhengjun Shi,Kaimeng Xu,Yanfei Niu,Xiaoping Rao

The presence of chemical pesticides has an impact on human health and there is an urgent need for the development of green antifungal alternatives. This study investigated the freshness application of dehydroabietic acid-modified chitosan product (CSDA) mixed with three essential oil (litsea cubeba oil (LCO), wintergreen essential oil (WEO), cinnamon essential oil (CEO)). The antifungal activity of modified chitosan/essential oils was tested by the punching method and the antifungal mechanism was investigated by the transmission electron microscope (TEM). The results showed that fungal inhibition rate of CSDA-WEO on Penicillium digitatum reached 100% at a concentration of 0.05 μL/mL and on Penicillium italicum reached 100% at a concentration of 1 μL/mL. Both molds were not well inhibited by CSDA-LCO and CSDA-CEO. CSDA-WEO was able to affect the cell walls and energy metabolism of the molds, resulting in nutrient loss and death. Fruits were preserved with 0.1 mg/g CSDA and 1 μL/mL WEO. CSDA-WEO maintained the antioxidant activity of citrus , inhibited respiration and water loss, and extended shelf life up to 24 days. CSDA-WEO extended the shelf life of longan to 18 days and kiwi slices to 6 days. CSDA-WEO is expected to serve as an alternative to chemical fungicides and shows promise as a broad-spectrum fruit and vegetable preservative.
