NIR Light-, Temperature-, pH-, and Redox-Responsive Polymer-Modified Reduced Graphene Oxide/Mesoporous Silica Sandwich-Like Nanocomposites for Controlled Release

Panjun Wang,Shuo Chen,Ziquan Cao,Guojie Wang

Here a novel quadruple-responsive nanocarrier based on reduced graphene oxide/mesoporous silica sandwich-like nanocomposites (rGO@MS) modified by pH- and temperature-responsive poly(N,N-dimethylaminoethyl methacrylate) (PDMAEMA) with a linker of disulfide was constructed via surface-initiated atom transfer radical polymerization. The polymer chains would be used as gatekeepers to control the release of the loaded cargo molecules under pH, temperature, NIR light and redox stimuli. The cargo molecules (rhodamine B) were demonstrated to release from the polymer-modified nanocomposites triggered by the quadruple-stimuli. It is noted that the release of the loaded rhodamine B from the nanocarriers could be enhanced greatly under the synergistic effect of multiple stimuli. The prepared quadruple-responsive polymer-modified nanocomposites show a bright prospect in the field of smart nanocarriers for controlled release.

Alternative Copper-Based Single-Atom Nanozyme with Superior Multienzyme Activities and NIR-II Responsiveness to Fight against Deep Tissue Infections

Jiaxiang Bai,Yonghai Feng,Wenming Li,Zerui Cheng,Jessica M. Rosenholm,Huilin Yang,Guoqing Pan,Hongbo Zhang,Dechun Geng

Nanozymes are considered to represent a new era of antibacterial agents, while their antibacterial efficiency is limited by the increasing tissue depth of infection. To address this issue, here, we report a copper and silk fibroin (Cu-SF) complex strategy to synthesize alternative copper single-atom nanozymes (SAzymes) with atomically dispersed copper sites anchored on ultrathin 2D porous N-doped carbon nanosheets (CuNx-CNS) and tunable N coordination numbers in the CuNx sites (x = 2 or 4). The CuNx-CNS SAzymes inherently possess triple peroxidase (POD)-, catalase (CAT)-, and oxidase (OXD)-like activities, facilitating the conversion of H2O2 and O2 into reactive oxygen species (ROS) through parallel POD- and OXD-like or cascaded CAT- and OXD-like reactions. Compared to CuN2-CNS, tailoring the N coordination number from 2 to 4 endows the SAzyme (CuN4-CNS) with higher multienzyme activities due to its superior electron structure and lower energy barrier. Meanwhile, CuNx-CNS display strong absorption in the second near-infrared (NIR-II) biowindow with deeper tissue penetration, offering NIR-II-responsive enhanced ROS generation and photothermal treatment in deep tissues. The in vitro and in vivo results demonstrate that the optimal CuN4-CNS can effectively inhibit multidrug-resistant bacteria and eliminate stubborn biofilms, thus exhibiting high therapeutic efficacy in both superficial skin wound and deep implant-related biofilm infections.

NIR diagnostic imaging of triple-negative breast cancer and its lymph node metastasis for high-efficiency hypoxia-activated multimodal therapy

Pan Yi,Liu Longcai,He Yichen,Ye Luyi,Zhao Xin,Hu Zhiming,Mou Xiaozhou,Cai Yu

Background Triple-negative breast cancer (TNBC) possesses special biological behavior and clinicopathological characteristics, which is highly invasive and propensity to metastasize to lymph nodes, leading to a worse prognosis than other types of breast cancer. Thus, the development of an effective therapeutic method is significant to improve the survival rate of TNBC patients. Results In this work, a liposome-based theranostic nanosystem (ILA@Lip) was successfully prepared by simultaneously encapsulating IR 780 as the photosensitizer and lenvatinib as an anti-angiogenic agent, together with banoxantrone (AQ4N) molecule as the hypoxia-activated prodrug. The ILA@Lip can be applied for the near-infrared (NIR) fluorescence diagnostic imaging of TNBC and its lymph node metastasis for multimodal therapy. Lenvatinib in ILA@Lip can inhibit angiogenesis by cutting oxygen supply, thereby leading to enhanced hypoxia levels. Meanwhile, large amounts of reactive oxygen species (ROS) were produced while IR 780 was irradiated by an 808 nm laser, which also rapidly exhausted oxygen in tumor cells to worsen tumor hypoxia. Through creating an extremely hypoxic in TNBC, the conversion of non-toxic AQ4N to toxic AQ4 was much more efficiency for hypoxia-activated chemotherapy. Cytotoxicity assay of ILA@Lip indicated excellent biocompatibility with normal cells and tissues, but showed high toxicity in hypoxic breast cancer cells. Also, the in vivo tumors treated by the ILA@Lip with laser irradiation were admirably suppressed in both subcutaneous tumor model and orthotopic tumor models. Conclusion Utilizing ILA@Lip is a profound strategy to create an extremely hypoxic tumor microenvironment for higher therapeutic efficacy of hypoxia-activated chemotherapy, which realized collective suppression of tumor growth and has promising potential for clinical translation.

Efficient Ultra-Broadband Ga4GeO8:Cr3+ Phosphors with Tunable Peak Wavelengths from 835 to 980 nm for NIR pc-LED Application

Leqi Yao,Qiyue Shao,Meiling Shi,Tianqi Shang,Yan Dong,Chao Liang,Jinhua He,Jianqing Jiang

Near-infrared (NIR) phosphor-converted light-emitting diodes (pc-LEDs) hold great potential for applications ranging from night vision to non-destructive detection. However, it remains a long-standing challenge to develop NIR phosphors simultaneously with longer-wavelength broadband emissions and higher efficiency. Herein, ultra-broadband Ga 4 GeO 8 :Cr 3+ (GGO:Cr 3+ ) phosphors are developed, with the NIR emission covering 700–1300 nm. Furthermore, tunable emission bands peaking from 835 to 980 nm are achieved simply by varying the Cr 3+ concentration. Particularly, emission maxima (λ max ) of GGO: x Cr 3+ shift from 850 to 900 nm without intensity loss when increasing x values between 0.02 and 0.10. An internal quantum yield of 60% is achieved for GGO:0.02Cr 3+ (λ max ≈ 850 nm, full width at half maximum (FWHM) ≈215 nm). The origin of tunable ultra-broadband emissions of GGO:Cr 3+ is revealed on the basis of structural and time-resolved spectroscopic analysis. The pc-LED fabricated by GGO:0.02Cr 3+ exhibits a maximum NIR output power of ≈56 mW at 400 mA drive current, and its application in high-penetration quality analysis of fruits is also demonstrated. The results indicate that GGO:Cr 3+ phosphors have high promise for practical applications in NIR pc-LED devices.

Bioinspired Jellyfish-like Carbon/Manganese nanomotors with H2O2 and NIR light Dual-propulsion for enhanced tumor penetration and chemodynamic therapy

Yi Xing,Mengyun Zhou,Xiangmei Liu,Minghang Qiao,Liping Zhou,Tailin Xu,Xueji Zhang,Xin Du

Chemodynamic therapy (CDT), which catalyzes the production of highly toxic reactive oxygen species (ROS), has been widely used in tumor therapy in the past five years. However, most of the current nanomaterials for CDT are difficult to penetrate into the tumor effectively due to the lack of motility, resulting in an unsatisfactory CDT effect. Herein, bioinspired jellyfish-like carbon/manganese nanomotors (JCMNs) are designed to achieve hydrogen peroxide (H 2 O 2 ) and NIR light dual-propulsion for the enhanced CDT therapy. Based on the jellyfish-like asymmetric structure, the manganese component can catalyze H 2 O 2 to generate chemical concentration gradient, propelling JCMNs to dynamically generate ROS in cells by self-diffusiophoresis. In addition, the carbon component of JCMNs can absorb near-infrared (NIR) light to produce a temperature gradient, driving JCMNs to enhance tumor penetration by self-thermophoresis. The combination of chemical and NIR light propulsions significantly improves the efficiency of tumor CDT, revealing that it should be a reasonable strategy to promote the development of nanobiomedicine by endowing nanosystems with propulsion capacity.

A reversible CHEF-based NIR fluorescent probe for sensing Hg2+ and its multiple application in environmental media and biological systems

Meiqing Zhu,Long Sun,Xina Liu,Xiaohui Pang,Fugang Fan,Xiaofan Yang,Rimao Hua,Yi Wang

Hg 2+ poses a great threat to human health and the environment due to its bioaccumulation and permanent damage. Herein, a reversible CHEF-based near-infrared fluorescent probe 2-(3-((E)-4-((E)-4-(diethylamino)-2- hydroxybenzylidene)amino)styryl)-5,5-dimethylcyclohex-2-en-1-ylidene)propanedinitrile ( DHEY ) capable of specifically recognizing Hg 2+ was constructed. DHEY exhibits advantages of large Stokes shift (157 nm), excellent selectivity , high sensitivity (LOD = 3.2 μg/L), and fast response efficiency (<3 min). Interestingly, DHEY can also realize rapid and effective detection of Hg 2+ after being recycled 7 times. The successful recovery of trace Hg 2+ in different environmental water samples fully demonstrates the potential of DHEY for actual applications. In particular, DHEY enables real-time observation of the distribution and translocation pattern of exogenous Hg 2+ in HeLa cells and zebrafish. This work provides important theoretical support for investigating the fate of heavy metal ions in the environment using fluorescence techniques.

Entropy-Mediated High-Entropy MXenes Nanotherapeutics: NIR-II-Enhanced Intrinsic Oxidase Mimic Activity to Combat Methicillin-Resistant Staphylococcus Aureus Infection

Xiaojun He,Yuna Qian,Chenglin Wu,Jiayao Feng,Xiaoshuai Sun,Qinxiang Zheng,Xiaokun Li,Jianliang Shen

Bacterial infections, such as bacterial keratitis (BK) and subcutaneous abscess, pose significant challenges to global healthcare. Innovative and new antibacterial agents and antibacterial strategies are in demand to control infections in this era of high drug resistance. Nanotechnology is gradually emerging as an economically feasible and effective anti-infection treatment. High-entropy MXenes (HE MXenes) are used to confer desirable properties with exposed active sites to high-entropy atomic layers, whose potential application in the field of biomedicine remains to be explored. Herein, monolayer HE MXenes are fabricated by implementing transition metals with high entropy and low Gibbs free energy to fill the gap in the biocatalytic performance of non-high-entropy MXenes. HE MXenes are endowed with extremely strong oxidase mimic activity ( K m = 0.227 m m ) and photothermal conversion efficiency (65.8%) in the second near-infrared (NIR-II) biowindow as entropy increases. Subsequently, HE MXenes realize NIR-II-enhanced intrinsic oxidase mimic activity for killing methicillin-resistant Staphylococcus aureus and rapidly removing the biofilm. Furthermore, HE MXenes can effectively treat BK and subcutaneous abscess infection induced by methicillin-resistant Staphylococcus aureus as nanotherapeutic agents with minuscule side effects. Overall, monolayer HE MXenes demonstrate promising clinical application potential in the treatment of drug-resistant bacterial infections and promote the healing of infected tissues.

Super Broadband Near-Infrared Solid Solution Phosphors with Adjustable Peak Wavelengths from 1165 to 875 nm for NIR Spectroscopy Applications

Shuai Huang,Yu Yan,Mengmeng Shang,Yining Wang,Yixin Sun,Peipei Dang,Jun Lin

Cr 3+ -activated near-infrared (NIR) phosphors have attracted wide attention owing to applications in phosphor-converted light-emitting diodes (pc-LED) for spectroscopy. Because a broader spectrum covers more information to detect, the width of the spectrum is an important parameter to consider. Here, the blue LED excitable ultra-broadband Cr 3+ -activated LiInGeO 4 phosphors, which exhibit unprecedented long-wavelength NIR emission with a wide full-width at half maximum (FWHM) of 305 nm, are developed. The overall emission tuning from 1165 to 875 nm with broadened FWHM from 305 to 508 nm is realized through designing continuous solid solution hosts of LiIn 1− z Sc z GeO 4 /(LiIn) 1− y Mg 2 y GeO 4 and finite solid solution hosts of (LiIn) 1− y Zn 2 y GeO 4 :Cr 3+ . The observation of such long-wavelength NIR from Cr 3+ not Cr 4+ is studied and confirmed by electron paramagnetic resonance, X-ray absorption near-edge structure, and steady-/transient-state photoluminescence. The formation of finite solid solutions induces the change in the local structure of Cr 3+ that ultimately leads to the observation of super broadband (FWHM = 508 nm) luminescence. The luminescence mechanism is discussed in this study. Finally, this study demonstrates that the as-synthesized phosphors with tunable NIR luminescence can be well used in NIR pc-LEDs for spectroscopy detection.

Self-generated Schottky barriers in niobium carbide MXene nanocatalysts for theory-oriented sonocatalytic and NIR-II photonic hyperthermia tumor therapy

Jiahuan Xu,Liang Chen,Sujun Ding,Xinyue Dai,Yanfei Dai,Yu Chen,Xuejun Ni

Two-dimensional (2D) transition metal carbides/nitrides (MXenes) have been extensively applied in biomedicine . However, the potential of MXene-based nanomedicine in sonodynamic therapy (SDT) has been rarely explored. Here, under the guidance of theoretical calculation, Nb 2 C MXenes are engineered as a high-performance sonocatalyst/sonosensitizer through mild in-situ self-oxidation. The work function values of Nb 2 C MXenes with different oxidation degrees are calculated to determine the optimized conditions to construct efficient Schottky heterojunction in the self-oxidized Nb 2 C MXenes. Experimentally, the niobium oxide clusters are homogeneously self-generated in-situ in Nb 2 C MXenes to form efficient Schottky barriers after the mild hydrothermal oxidation. The results reveal that the Schottky heterojunction can accelerate the separation of sono-triggered electron-hole pairs and inhibit their recombination, which profoundly augments the reactive oxygen species (ROS) generation efficiency of the in-situ self-oxidized Nb 2 C MXenes (denoted as Nb 2 C-Ox) under ultrasound (US) irradiation. The mild oxidation also allows the Nb 2 C-Ox to inherit the high photothermal performance of Nb 2 C MXenes in the second near-infrared (NIR-II) window. Accordingly, the developed Nb 2 C MXene-originated sonocatalyst/sonosensitizer can achieve high therapeutic efficacy based on the sonodynamic effect and NIR-II photonic hyperthermia. Therefore, this work proposes a theoretical calculation-oriented strategy to design and fabricate new MXene-based sonocatalysts/sonosensitizers for biomedical applications .

Influence of glass matrix SiO2–B2O3–NaF on the formation of NIR-shielding functional units in energy-saving window glasses

Guang Yang,Yongwei Liu,Chuanfan Yang,Fang Xia,Yupeng Wu,Yaroslav Shpotyuk,Viktor Takats,Huidan Zeng,Yinsheng Xu,Hongfei Chen,Yanfeng Gao

This work presents the influence of glass matrix SiO 2 –B 2 O 3 –NaF on the formation of near-infrared (NIR)-shielding functional units in energy-saving glass windows. The structural analyses by XRD , XPS , and Raman confirmed that the NIR-shielding performance is insensitive to SiO 2 or B 2 O 3 glass network formers but is significantly impacted by the concentration of NaF. This is because the presence of a reasonable amount of NaF provides the much-needed sodium for the formation of NIR-shielding functional units Na 5 W 14 O 44 but too much NaF (>22 mol.%) can inhibit the formation of Na 5 W 14 O 44 by transforming the W– O –W chain in the interconnected [WO 6 ] and [WO 4 ] polyhedrons into W–O - bonds. By adjusting NaF to the optimal level (16 mol.%) and adding only a small amount of H 2 WO 4 (1.9 mol.%) as the tungsten source in the raw materials, excellent optical properties and thermal insulation performance have been achieved in the produced bulk glass, with a high transmission of 65.8% at 550 nm in the visible light region and a low transmission of 6.6% at 1550 nm in the NIR region. This performance is much better than soda lime glass and ITO glass and is comparable to cesium tungsten bronze-based membrane coated glass. This study paves a way for glass composition design to achieve high NIR-shielding performance with the minimum amount of tungsten source, leading to cost-effective glass fabrication and enhanced mechanical strength of energy-saving glass windows.

Eco-friendly cellulose-based hydrogel functionalized by NIR-responsive multimodal antibacterial polymeric ionic liquid as platform for promoting wound healing

Wenqi Song,Tiantian Xu,Liwei Qian,Sufeng Zhang,Chaoli Wang,Yuzhen Zhao,Zemin He,Valentin Nica,Zongcheng Miao

With the trend of sustainable development and the complex medical environment, there is a strong demand for multimodal antibacterial cellulose wound dressing (MACD) with photothermal therapy (PTT). Herein, a novel MACD fabrication strategy with PTT was proposed and implemented through graft polymerization of an imidazolium ionic liquid monomer containing iron complex anion structure. The fabricated hydrogels exhibited excellent antibacterial properties because of the efficient photothermal conversion ability (68.67 %) of ionic liquids and the intrinsic structural characteristic of quaternary ammonium salts. The antibacterial ratio of cellulosic hydrogel dressings to S. aureus and E. coli could reach 99.57 % and 99.16 %, respectively. Additionally, the fabricated hydrogels demonstrated extremely low hemolysis rates (<5 %) and excellent cell viability (~>85 %). Furthermore, in vivo antibacterial experimental results proved that the fabricated antibacterial dressings could significantly accelerate wound healing. Therefore, the proposed strategy would provide a new method of designing and preparing high-performance cellulose wound dressings.

In situ assembled titanium carbide-based heterojunctions for the synergistic enhancement of NIR-II photothermal/photodynamic therapy against breast cancer

Hai Zhu,Xuequan Zhang,Qiusheng Wang,Jin Deng,Zhuangzhuang Zhang,Xiaoxian Zhang,Jun Cao,Bin He

The combined use of photothermal therapy (PTT) and photodynamic therapy (PDT) could circumvent the drawbacks of each individual therapeutic strategy, resulting in an enhanced antitumor effect. However, the lack of highly effective photo-agents that are irradiation-safe in the biologically transparent window hinder the advancement of phototherapy clinically. Hence, in this study, a charge separation engineering strategy was adopted to fabricate a nanoplatform with heterojunctions, namely, in situ TiO2-loaded MXene (Ti3C2/TiO2 heterojunctions). This nanoplatform exhibited reduced bandgap (1.68 eV), enhanced NIR-II photothermal conversion efficiency (44.98%), and extended absorption edge compared to pristine TiO2 for enhanced photodynamic effect. More importantly, the proliferation of tumor cells could be efficiently inhibited at a 5 mm chicken breast depth after 1064 nm laser irradiation, and the intracellular ROS production significantly increased under 660 nm or even 1064 nm laser irradiation with heterojunctions (HJs) compared with that of TiO2. Moreover, the in vivo data further confirmed that the as-prepared heterojunctions could efficiently eradicate tumors efficiently via improved photothermal effect with NIR-II laser irradiation and upregulated ROS production. Collectively, the reported HJs strategy provides an opportunity for the success of combinational PTT and PDT therapy in tumor treatment.
