Developed a dual-stage sequential curcumin releasing SIS-based self-adhesive hydrogel lung patch. • Achieved strong wet adhesion, suture-free air-leak sealing, and staged anti-inflammatory therapy. • Validated reduced pneumothorax, fibrosis, and inflammatory pleural adhesions in rat and pig pulmonary injury repair models. Postoperative pulmonary air leakage and uncontrolled inflammation remain major challenges after lung resection, while existing patches primarily provide mechanical sealing without active regulation of the healing microenvironment. Here, we report a self-adhesive, dual-stage sequential drug-releasing lung patch (Hydrogel patch–Cur) that integrates rapid sealing with stage-specific immunomodulation. The patch is constructed by coating a decellularized porcine small intestinal submucosa (SIS) scaffold with a bilayer gelma-based hydrogel system, enabling sequential curcumin (Cur) delivery through structurally distinct networks. The outer low-crosslinked gelma30–CS layer enables rapid drug release and enhanced wet-tissue adhesion, while the inner highly crosslinked gelma80 layer provides sustained release for prolonged immune regulation. This design establishes a time-dependent therapeutic profile that matches the dynamic inflammatory phases of lung repair. The patch exhibits strong adhesion (32-fold higher adhesion than uncoated SIS) on wet tissues and effective sealing performance without the need for sutures or staples. In vitro studies demonstrate promoted macrophage polarization toward a pro-regenerative phenotype and significant suppression of pro-inflammatory cytokines. In vivo, the patch rapidly prevents air leakage and pneumothorax, reduces inflammation and fibrosis, and accelerates lung tissue regeneration in both rat and porcine models, outperforming conventional methods and a clinically used self-adhesive patch. This study presents a clinically translatable strategy that combines self-adhesive sealing with gradient drug delivery for improved postoperative lung repair.

Bisphenol A (BPA), a pervasive endocrine-disrupting chemical, has been increasingly implicated in thyroid carcinogenesis, yet the links between redox imbalance and metabolic alterations remain incompletely defined. Here, we combined LC-MS/MS quantification of BPA and its major conjugates with in vitro and in vivo thyroid cancer models to examine oxidative stress markers and lipid indices assessed by targeted biochemical assays and to evaluate a curcumin-modified, quercetin-loaded chitosan nanomicelle system (Que@CSNPs-Cur). We observed BPA enrichment in tumor-bearing mice, accompanied by increased lipid indices (free fatty acids, triglycerides, and total cholesterol) and elevated oxidative stress markers (ROS, TOS, and MDA), as assessed by targeted enzymatic and colorimetric assays. At the molecular level, BPA exposure was associated with reduced PTEN expression and increased PI3K/AKT phosphorylation, consistent with enhanced proliferative and invasive phenotypes in papillary thyroid carcinoma cells. Que@CSNPs-Cur restored PTEN levels, reduced PI3K/AKT activation, attenuated redox and lipid indices, and promoted caspase-3-dependent apoptosis. In xenograft mice, Que@CSNPs-Cur suppressed tumor growth, decreased oxidative stress markers and lipid indices, and reduced tumor BPA residue. Collectively, these results suggest that BPA exposure is linked to PTEN/PI3K/AKT dysregulation, oxidative stress and lipid disturbances. Que@CSNPs-Cur is a promising redox-oriented nanotherapeutic strategy for BPA-associated thyroid cancer phenotypes. Notably, the present study does not determine whether oxidative stress is upstream or downstream of PTEN suppression.

Persistent excessive inflammation and oxidative stress are main causes for the impaired wound healing in diabetic patient. To reduce the excessive reactive oxygen species (ROS) and inflammatory response of wound in acute inflammatory phase is a key strategy for diabetic wound healing. In this work, a kind of metal organic frameworks (MOFs) with curcumin as ligand and Zn 2+ as center were synthesized by a eutectic method, and a controllable double release system of curcumin and Zn 2+ for diabetic wound repair was constructed by loading the MOFs into the hierarchical micro/nanofibrous poly (L-lactic acid) (PLLA) scaffolds. The results showed that the synthetic MOFs could significantly improve the bioavailability of curcumin, and the fabricated hierarchical micro/nanofibrous scaffolds could not only promote the adhesion and growth of skin tissue cells, but also release curcumin and Zn 2+ on-demand in the early stage of inflammation. In vivo experiments further demonstrated that the scaffolds could effectively inhibit the inflammatory response and promote collagen deposition, angiogenesis as well as re-epithelialization of the skin wound, which ultimately promoted diabetic would healing in mice. The mechanism lying behind is closely related to the finding that ROS generation and inflammatory reaction in the acute inflammation stage of wound healing were significantly reduced by the scaffolds. Our results suggest that the designed hierarchical micro/nanofibrous scaffolds with curcumin and Zn 2+ eutectic MOFs incorporated can effectively promote the skin wound healing in diabetic mice, providing a new strategy for the treatment of chronic wound.

Problem Unhealthy dietary habits and inadequate existing treatment technologies have led to high morbidity and mortality rates in patients with gastric cancer, and the research shows that curcumin has excellent antitumor properties, however, the non-water-soluble characteristic of curcumin limits its applications. To break the limitation, the targeted drug carriers become the urgent need to treat gastric cancer. Method The heterogeneous magnetic α-Fe 2 O 3 /Fe 3 O 4 nanoparticles were obtained by rapid combustion method with citric acid (CA) and ferric nitrate as raw materials and anhydrous ethanol as solvent and fuel, and then modified with citric acid (CA) and chitosan (CTS), and loaded curcumin (Cur) to form α-Fe 2 O 3 /Fe 3 O 4 -CA-CTS-Cur nanosystem. Results The particle sizes of heterogeneous magnetic α-Fe 2 O 3 /Fe 3 O 4 nanoparticles and α-Fe 2 O 3 /Fe 3 O 4 -CA-CTS-Cur nanosystem were 24.9 nm and 35.6 nm, respectively, and their respective saturation magnetization were 70.9 emu/g and 30.3 emu/g. The magnetic α-Fe 2 O 3 /Fe 3 O 4 -CA-CTS-Cur nanosystem had good stability, excellent drug release rate, and enhanced apoptosis to gastric cancer cells (SGC-7901) through the caspase pathway and ferroptosis of cells. Conclusions The magnetic drug delivery nanosystem was successfully constructed, and revealed significant inhibitory and killing effects on SGC-7901 cells, and low toxic and side effects on normal cells.

The objective of this study was to formulate Pickering emulsions stabilized by transglutaminase cross-linked mulberry leaf protein (TG-MLP) nanoparticles as a delivery system for curcumin (Cur) and to assess its bioaccessibility both in vivo and in vitro. The encapsulation efficiency of curcumin in high-internal-phase Pickering emulsions (HIPEs) prepared at pH 10 with a 20 mg/mL concentration of TG-MLP reached 93%. Compared to Oil-Cur, Cur-HIPEs exhibited superior antioxidant activity. Furthermore, Cur-HIPEs demonstrated enhanced stability against ultraviolet irradiation, storage under dark and visible light, and heating, in contrast to Oil-Cur. Among the various conditions tested, HIPEs stabilized by TG-MLP nanoparticles at an ionic strength of 1000 mM offered the most effective protection for curcumin. Moreover, TG-MLP nanoparticles at pH 8 provided better stability for the formulated HIPEs compared to those at pH 6 and 10. During simulated gastrointestinal digestion, the bioaccessibility of curcumin in Cur-HIPEs was significantly increased to 30.1% compared to Oil-Cur. In murine studies, higher levels of curcumin were detected in the stomach, small intestine, rectum, ileum, and feces following administration of Cur-HIPEs, indicating improved protection, absorption, and potential biological activity during digestion. Consequently, HIPEs offer excellent protection and delivery for curcumin during digestion.

Here, we systematically evaluated the therapeutic efficacy and mechanisms of action of in-house synthesized hydroxyethyl starch-curcumin nanoparticles (HES-CUR NPs) in ulcerative colitis (UC). Using a dextran sodium sulfate (DSS)-induced UC mouse model, we analyzed the effect of HES-CUR NPs on colonic tissue and the gut microbiota through the assessment of organ indices and serum biochemical markers, histopathology, immunohistochemistry, Western blot, and 16S rRNA sequencing. We found that HES-CUR NPs significantly suppressed the TLR4/NF-κB inflammatory pathway (downregulated TLR4, p-IKKβ, IL-1β, IL-6, NFKB, MyD88, and p-IKKB/IKKB; p < 0.01), activated the Nrf2/HO-1 antioxidant pathway (enhanced Nrf2 nuclear translocation and HO-1 expression), and upregulated the expression of tight junction proteins (claudin-1, occludin, ZO-1), thereby restoring intestinal barrier integrity. Microbiota analysis revealed that HES-CUR NPs increased gut microbiota diversity (elevated Chao1, Shannon, and ACE indices) and enriched beneficial bacteria abundance (Ligilactobacillus murinus, Lactobacillus johnsonii). Antibiotic intervention partially attenuated the therapeutic effects of the HES-CUR NPs, confirming that their effects involved microbiota-dependent mechanisms. Compared to CUR and SASP, HES-CUR NPs exhibited significantly enhanced bioavailability and efficacy (p < 0.01), attributed to the targeted delivery and sustained-release properties of the hydroxyethyl starch nanocarrier. No toxicity was observed, as indicated by normal spleen, liver, and thymus indices and stable levels of TP, ALT, AST, ALB, GLB, and ALB/GLB. Our findings indicated that HES-CUR alleviates UC through a synergistic, multi-target mechanism involving inflammatory pathway suppression, antioxidant pathway activation, intestinal barrier repair, and microbiota modulation, providing a theoretical foundation for developing efficient and safe natural nanomedicines targeting UC.

Epilepsy is a chronic brain disease with various causes, among which oxidative stress-induced neuronal damage is a significant pathological factor and has been widely studied as one of the key pathogenic mechanisms. Hydrogen peroxide (H₂O₂), a representative reactive oxygen species (ROS), plays an important role in epilepsy. However, real-time imaging capable of revealing the distribution of H₂O₂ in the living brain remains limited. To address this, we developed a feasible fluorescent probe library based on the unique reaction between the α-ketoester moiety and H₂O₂, combined with dicyanoisophorone derivatives possessing excellent optical performance. In consequence, DCI-BT-PY was optimized as a novel fluorescent probe for H₂O₂ through parallel testing of its physicochemical and optical properties. Notably, due to its good blood-brain barrier (BBB) penetration, DCI-BT-PY was used to monitor H₂O₂ upregulation in the brains of epileptic mice, revealing a close relationship between H₂O₂ and epilepsy. Furthermore, using the Sombati epilepsy model of hippocampal neuronal cells and the mouse model of epilepsy, the neuroprotective effect of curcumin on oxidative stress injury of nerve cells was explored, showing that it may alleviate epilepsy by activation of the Nrf2/HO-1 signaling pathway for suppressing the neurotoxic damage. This finding offers new insights into the pathogenesis and treatment of epilepsy. Additionally, this work provides a reliable tool for evaluating H 2 O 2 fluctuations in the brains of epilepsy-induced mice, thereby establishing a solid foundation for antioxidant therapies targeting epilepsy.

To address the ecological pollution caused by synthetic antimicrobial agents in conventional food packaging films, we have successfully developed a fully bio-based and sustainable food packaging film that exhibits exceptional antibacterial and antioxidant properties. In this study, we designed and synthesized two novel natural alcohol esters of amino acid (NAEAAs)—L-Trp-Men and L-Trp-Bor—through esterification reactions involving L -tryptophan ( L -Trp) and L -Menthol or Borneol, respectively. A multifunctional composite film system was constructed using natural corn starch (CS) and sodium carboxymethyl cellulose (CMC) as the matrix materials, with NAEAAs serving as the antimicrobial active components and curcumin (Cur) as the antioxidant. Experimental results demonstrated that the CS/CMC/Men2/Cur2 composite film, containing 2 wt% additives, exhibited nearly complete (≈100 %) broad-spectrum antibacterial efficiency against both Escherichia coli and Staphylococcus aureus , alongside a DPPH radical scavenging rate of 45.69 ± 1.39 %. Cytotoxicity tests confirmed the excellent biocompatibility of NAEAAs, which hydrolyze into amino acids and natural alcohols, thereby minimizing environmental contamination. These fully natural material-based films present a novel approach for developing green, pollution-free, and renewable bio-based food packaging materials.

Skin scarring after injury represents a global challenge, imposing significant impacts on patients' appearance, function, and mental health, thus highlighting the clinical urgency of exploring scar prevention strategies. This study developed a multifunctional nanostructured lipid carrier hydrogel for promoting scarless wound healing based on the innovative concept of “Medicines and Excipients all-in-one”. Using anti-inflammatory and wound-healing Bletilla striata polysaccharide (BSP) and Blumea balsamifera oil (BBO) as functional excipients, we prepared integrated BBO-BSP nanostructured lipid carriers (NLC). Here, BSP served as a natural polymeric emulsifier, while BBO served as the active liquid oil. Furthermore, with BSP as the gel matrix, BBO-BSP NLC hydrogel (NLCG) were constructed, and then curcumin was loaded to form Cur@BBO-BSP NLCG, achieving the synergistic wound-healing function of multiple components. Characterization showed that Cur@BBO-BSP NLC had an average particle size of 172.6 ± 1.0 nm, a polydispersity index of 0.106 ± 0.037, an entrapment efficiency of 89.52 ± 2.28 %, and a drug loading capacity of 4.73 ± 0.08 %. Cur@BBO-BSP NLCG significantly enhanced curcumin stability, exhibiting sustained release over 264 h in vitro . Antioxidant assays revealed that Cur@BBO-BSP NLCG scavenged DPPH and hydroxyl radicals at rates of 94.10 ± 3.24 % and 94.88 ± 4.03 %, respectively, while demonstrating good biosafety and promoting L929 cell migration. In vivo experiments in rabbit ear models confirmed that Cur@BBO-BSP NLCG accelerated wound healing and inhibited scar formation. The mechanism may be related to its ability to regulate the expression of factors such as CD31, VEGF, TNF-α, IL-6, and TGF-β 1 , thereby enabling multidimensional regulation of vascular remodeling, inflammatory microenvironment and fibroblast function. The proposed “functional excipient-active medicine” collaborative delivery strategy offers a promising new paradigm for scarless wound healing therapy.

Objectives. Curcumin (Cur) is a natural polyphenol isolated from turmeric and has potent anti-inflammatory and antioxidant activities. This study aimed to explore the effects and possible mechanisms of curcumin on oxygen-glucose deprivation/reoxygenation (OGD/R)-induced injury in cultured rat astrocyte primary cells. Methods. After screening for effective doses, the cultured rat astrocyte primary cells were divided into three groups: control, OGD/R, and OGD/R + curcumin (10 μM, 20 μM, and 40 μM). Cell viability was detected using CCK8 assays. The level of malondialdehyde and superoxide dismutase activity was determined using commercial kits. The endothelial nitric oxide synthase and adenosine triphosphate concentrations were determined by enzyme-linked immunosorbent assay. The mRNA levels of the inflammatory indexes interleukin (IL)-6, tumor necrosis factor (TNF)-alpha, and interleukin (IL)-1β were evaluated by quantitative reverse-transcription polymerase chain reaction. Annexin V-fluorescein isothiocyanate/propidium iodide was used to detect apoptosis. JC-1 was used to assess the mitochondrial membrane potential. The protein expression of apoptosis-related proteins (B-cell lymphoma-2 (Bcl-2), BCL-2-associated X (Bax), and cleaved caspase 3), mitochondria-related proteins (dynamin-related protein 1 (DRP1), phosphorylated DRP1 (p-DRP1), and mitofusin 2), and essential proteins of the extracellular signal-regulated kinase (ERK) signaling pathway (ERK1/2, p-ERK1/2) were analyzed by western blot. Results. Our data indicated that curcumin reversed OGD/R-induced cell viability loss, oxidative stress, inflammatory cytokine production, and cell apoptosis in a dose-dependent manner. Furthermore, curcumin attenuated OGD/R-induced mitochondrial dysfunction and ERK1/2 phosphorylation in a dose-dependent manner. Conclusions. Curcumin protected against OGD/R-induced injury in rat astrocyte primary cells through improving mitochondrial function and regulating the ERK signaling pathway.

This study investigates the early synergistic effects of curcumin and licorice through vertical cooperation, which can simultaneously target both the upstream and downstream components of neuroinflammation. It evaluates their neuroprotective effects and potential mechanisms in a d -galactose/sodium nitrite-induced Alzheimer’s disease mouse model.Eighty C57BL/6 mice were divided into eight groups ( n = 10): wild-type (WT), AD model, curcumin monotherapy (AD + CL, 100 mg/kg), G. glabra monotherapy (AD + GG, 100 mg/kg), low-dose combination (AD + COM-L, 50 + 50 mg/kg), high-dose combination (AD + COM-H, 100 + 100 mg/kg), donepezil (3 mg/kg), and SN50 (NF-κB inhibitor, 400 µg/kg). Cognitive function was assessed via Morris Water Maze in WT, AD, AD + CL, AD + GG, and AD + COM-H groups, while all groups underwent molecular analyzes. The high-dose combination most effectively restored spatial memory, reducing escape latency by ~ 43% versus monotherapies. Molecularly, it synergistically reduced tau-related proteins (MAPT, GSK-3β) and suppressed the TLR4/MyD88/NF-κB axis, lowering inflammatory mediators (IL-6, TNF-α, CXCL1, PTGS2). IL-6 was further reduced by 28.6% and 40.0% compared to curcumin and G. glabra alone, respectively. The combination also enhanced antioxidant defense (increased SOD) and anti-apoptotic capacity (upregulated BCL-2) while reducing oxidative lipid damage (lower MDA). Network pharmacology identified 40 shared AD targets, with enrichment in NF-κB and IL-17 pathways, validated experimentally. In conclusion, curcumin and G. glabra exert synergistic neuroprotection by concurrently inhibiting the TLR4/NF-κB pathway and modulating IL-17 signaling, with G. glabra potentially targeting the TLR4/HMGB1 axis and curcumin directly inhibiting NF-κB activation, forming a complementary mechanistic interplay. This multi-target action disrupts the interplay between neuroinflammation and tau pathology, underscoring the combination’s therapeutic potential for AD intervention.

Most anticancer drugs cannot kill cancer stem cells (CSCs) effectively, which lead to the failure of anticancer chemotherapy, such as relapse and metastasis. In this study, we prepared a multifunctional oligosaccharides of hyaluronan (oHA) conjugates, oHA-histidine-menthone 1,2-glycerol ketal (oHM). The oHM conjugates possess pH-sensitive menthone 1,2-glycerol ketal (MGK) as hydrophobic moieties and oHA as the target of CD44 receptor. Anticancer drugs, curcumin(Cur) and paclitaxel(PTX), were loaded into oHM micelles via self-assembly. Then, oHM micelles were mineralized through controlled deposition of inorganic calcium and phosphate ions on the nanoparticular shell via a sequential addition method to fabricate the “nano-eggs.” The formed nano-eggs had a smaller size (120.6 ± 4.5 nm) than oHM micelles (158.6 ± 6.4 nm), indicating that mineralization made the appearance of compact nanoparticles. Interestingly, when the nano-eggs were put into the acidic conditions (pH 6.5), their outer shell(inorganic minerals) will be destroyed with the larger size, while the “nano-eggs” were stable under pH 7.4. For both nano-eggs and oHM micelles, the Cur and PTX were released in a sustained manner depending on the pH of the solution. However, the nano-eggs showed much lower released than the oHM micelles due to the dissolution of the inorganic minerals and pH-sensitive ketal at mildly acidic environments (pH 6.5). In vivo study, the nano-eggs could get to the tumor site more effectively than oHM micelles. CSCs were sorted by a side population assay from MDA-MB-231 breast cancer cell lines over-expressing CD44 receptors. Antitumor activity was also evaluated on MDA-MB-231 xenografts in nude mice. The antitumor efficacy indicated that nano-eggs with co-delivery of Cur and PTX produced the strongest antitumor efficacy, and nano-eggs showed strong activity against cancer stem cells. These double pH-sensitive nano-eggs may provide a promising strategy for drug delivery to both cancer cells and cancer stem cells.
