Rechargeable and sunlight-activated Sr3Y2Ge3O12:Bi3+ UV–Visible-NIR persistent luminescence material for night-vision signage and optical information storage

Zhihao Zhou,Xiu Wang,Xiaodong Yi,Hong Ming,Zhijun Ma,Mingying Peng

Persistent luminescence (PersL) materials that can be excited by natural sunlight are in great demand to reduce energy consumption, but the design and development of efficient sunlight-activated PersL phosphor remains a daunting challenge so far. Herein, we report the development of a new kind of sunlight-activated PersL material based on Bi 3+ -doped Sr 3 Y 2 Ge 3 O 12 (SYGO) phosphor, which exhibits an intense UV–Visible-NIR PersL and possesses a super-long PersL longer than 60 h. This newly developed PersL material can be effectively and repeatedly activated by sunlight radiation in all kinds of weather conditions because of its high response to both direct sunlight and diffuse solar radiation. Owing to the existence of stabilized deep traps with suitable depths, SYGO:Bi 3+ also shows excellent photostimulated luminescence (PSL) properties. A series of excitation-time-dependent TL experiments, PL and PersL spectra were systematically studied to reveal the trapping and de-trapping processes of this material. Furthermore, by utilizing the unique PersL and PSL properties, we demonstrate the applications of SYGO:Bi 3+ for night-vision signage and optical information storage. Our achievement in the case of Bi 3+ -doped SYGO phosphor is expected to promote the future development of more excellent sunlight-activated and photostimulated PersL materials towards versatile practical applications.

NIR II-Excited and pH-Responsive Ultrasmall Nanoplatform for Deep Optical Tissue and Drug Delivery Penetration and Effective Cancer Chemophototherapy

Lipeng Zhu,Duyang Gao,Lisi Xie,Yunlu Dai,Qi Zhao

The limited tumor tissue penetration of many nanoparticles remains a formidable challenge to their therapeutic efficacy. Although several photonanomedicines have been applied to improve tumor penetration, the first near-infrared window mediated by the low optical tissue penetration depth severely limits their anticancer effectiveness. To achieve deep optical tissue and drug delivery penetration, a near-infrared second window (NIR-II)-excited and pH-responsive ultrasmall drug delivery nanoplatform was fabricated based on BSA-stabilized CuS nanoparticles (BSA@CuS NPs). The BSA@CuS NPs effectively encapsulated doxorubicin (DOX) via strong electrostatic interactions to form multifunctional nanoparticles (BSA@CuS@DOX NPs). The BSA@CuS@DOX NPs had an ultrasmall size, which allowed them to achieve deeper tumor penetration. They also displayed stronger NIR II absorbance-mediated deep optical tissue penetration than that of the NIR I window. Moreover, the multifunctional nanoplatform preferentially accumulated in tumor sites, induced tumor hyperthermia, and generated remarkably high ROS levels in tumor sites upon NIR-II laser (1064 nm) irradiation. More importantly, our strategy achieved excellent synergistic effects of chemotherapy and phototherapy (chemophototherapy) under the guidance of photothermal imaging. The developed nanoparticles also showed good biocompatibility and bioclearance properties. Therefore, our work demonstrated a facile strategy for fabricating a multifunctional nanoplatform that is a promising candidate for deep tumor penetration as an effective antitumor therapy.

Concurrent photothermal therapy and photodynamic therapy for cutaneous squamous cell carcinoma by gold nanoclusters under a single NIR laser irradiation

Pei Liu,Weitao Yang,Lei Shi,Haiyan Zhang,Yan Xu,Peiru Wang,Guolong Zhang,Wei R. Chen,Bingbo Zhang,Xiuli Wang

Phototherapy, particularly photothermal therapy (PTT) and photodynamic therapy (PDT), has become a promising therapeutic technique for the treatment of skin cancers because of its minial invasiveness, high efficacy, and low side effects. Nevertheless, single modality therapy, either PTT or PDT, has limited clinical effectiveness in treating skin cancers. Thus, combined applications of PTT and PDT have been frequently reported; however, PTT and PDT often require their respective photoagents and excitation light sources, resulting in challenges in clinical transformation. In this study, to address these issues, we report the use of biocompatible gold nanoclusters Au25(Capt)18 for the concurrent PTT and PDT treatment of cutaneous squamous cell carcinoma (cSCC) using an 808 nm near-infrared (NIR) laser. Utilizing their high light-thermal conversion efficiency, potent generation of singlet oxygen, and strong photothermal stability, Au25(Capt)18 nanoclusters potentiated a significant proliferation suppression of cSCC XL50 cells in vitro and the inhibition of cSCC tumors on SKH-1 mice in vivo. In particular, under 808 nm light irradiation, the tumor-cell-killing contributions of PTT and PDT were estimated to be 28.86% and 71.14%, respectively, by using an ROS scavenger to quench the PDT effect. Tumor-infiltrating CD4+ T and CD8+ T cells were observed after one course of concurrent PTT and PDT. Preliminary toxicity studies indicated low adverse effects of the Au25(Capt)18 nanoclusters. Through this study, we report the use of a simple nanostructure for simultaneous PTT and PDT applications to effectively kill cSCC and to induce anti-tumor immune responses. Our study could lead to the development of effective photoagents for current, synergistic applications of different phototherapies with targeted immunological responses for the treatment of cancers.

Effect of substituents on the performance of N-carbazolyl double benzylidene ketones in visible/NIR light one/two-photon polymerization

Jie Xie,Xiaobing Wang,Yonghui Wang,Jiawei Li,Chun Cao,Ming Jin

Developing high-performance photoinitiation systems with high sensitivity is of great significance in one/two-photon polymerization. Especially, photoinitiators with rapid photobleaching effects, deep curing abilities and potential for two-photon lithography have great commercial prospects. However, such photoinitiators often require complex organic synthesis, thus having relatively limited varieties. Double benzylidene ketone is widely used as a high-performance dye and can be constructed through a one-step classical aldol condensation reaction. The optimization of its structure often involves introducing extended conjugated structures or different terminal substituents. However, the synergistic impacts of side substituents with different electron effects are rarely studied. Here, six novel carbazolyl double benzylidene ketones with tert -butyl on the terminal position and F atom or N , N -dimethylamino groups substituted at the side position on conjugated structures are designed and prepared to explore the structure–activity relationships. Notably, results show that introducing electron-donating groups at both positions can effectively induce a redshift in the absorption spectra of molecules, and the introduction of an electron-withdrawing F atom on the side position induces impressive photobleaching during photopolymerization under visible LED irradiation. Moreover, CZs/MDEA with PETA exhibit fast writing speed and excellent nano-patterning capability under 780 nm laser in two-photon polymerization (TPP) and have potential applications in 3D microfabrication technologies.

Enhanced Penetration and Retention of CuS-Based Nanosystem Through NIR Light and In Situ Enzyme Response for Improved Tumor Therapy

Fangli Gao,Liang Zhu,Liting Jiang,Jie Zhang,Shenglu Ji,Weihua Gao,Guanglei Ma,Yi Chang,Xiaoming Ma,Yuming Guo

The efficient way to increase the therapeutic efficacy of nanomedicines is by encouraging the penetration and enhancing the retention of nanoparticles at the tumor site. However, it is a serious dilemma that small nanoparticles can penetrate deep into the tumor tissue but easily be cleared into the surrounding tissues. In order to solve this dilemma, a smart nanosystem is created to address this problem, ensuring both the effective penetration of tiny nanoparticles (NPs) and their appropriate retention at the tumor site. CuS NPs that is modified with peptides are prepared facilely, and can aggregate in situ through the intermolecular crosslinking reaction catalyzed by the transglutaminase (TGase) abundantly expressed at the tumor site, resulting in an outstanding photothermal effect for tumor therapy. Upon NIR irradiation, the photothermal effect of CuS-K and CuS-Q induced the disintegration of liposomes and prompted the release of CuS-K, CuS-Q, and indocyanine green (ICG). Simultaneously, CuS-K and CuS-Q aggregated under the catalysis of TGase after being internalized by tumor cells to enhance photothermal therapy. The current study provides valuable inspiration to design nanomedicines with prolonged circulation time in the blood system, better penetration, and retention at the tumor site, and multimodal tumor therapy to achieve the desired therapeutic efficacy.

Design of ZIF-based hybrid nanoparticles with hyaluronic acid-augmented ROS behavior for dual-modality PA/NIR-II FL imaging

Botao Qu,Yahong Han,Juan Li,Qian Wang,Bingyu Zhao,Xiaoyang Peng,Ruiping Zhang

Photoacoustic (PA) imaging has emerged as a promising bio-imaging technique due to its non-invasive visualization of lesions at great penetration depths. Fluorescence (FL) imaging in the second near-infrared window (NIR-II, 1000–1700 nm) achieves a higher imaging resolution and lower background signals compared to NIR-I. However, the single imaging method possesses its own disadvantages. Thus, we have demonstrated ZIF-8–IR820–MnPc–HA nanoparticles (ZIMH NPs) that can achieve visualization and localization of tumors in mice models with the help of a dual-modality PA/NIR-II FL imaging performance. Meanwhile, these excellent nanoparticles also induce the efficient generation of singlet oxygen (1O2) upon 808 nm laser illumination, and display excellent photodynamic therapy efficacy in cells, further indicating their potential application for in vivo PDT. In ZIMH NPs, hyaluronic acid (HA) impressively acts as a “sponge”, enhancing the generation of 1O2 and facilitating the cellular therapeutic effects. We believe that ZIF-8–IR820–MnPc–HA NPs present a brand-new strategy for the exploration of efficient PDT photosensitizers with dual-modality imaging performance for use in various biomedical applications.

Preparation and characterization of a mussel-inspired and programmable PDA@MXene/PNIPAM hydrogel actuator with ultrafast temperature and NIR responsiveness

Jie Zhang,Jia Liu,An Liu,Shu He,Wei Shao

In recent years, smart hydrogels have been widely applied to various types of hydrogel actuators due to their unique stimulus responsibility, such as temperature responsiveness, near-infrared (NIR) light responsiveness, pH responsiveness, etc. Herein, segmented poly (N-isopropyl acrylamide) (PNIPAM) based hydrogel actuators with both temperature and NIR light actuation behaviors were constructed. Polydopamine (PDA) was firstly used to modify MXene nanosheets to fabricate an efficient photothermal conversion material. Then they were added into PNIPAM matrix to prepare PDA@MXene/PNIPAM hydrogels. The prepared PDA@MXene/PNIPAM hydrogels exhibited three-dimensional network structure with the tensile strength of 26.84 kPa, and they were successfully assembled into segmented hydrogel actuators. The assembled segmented hydrogel actuator exhibited high bending angle of 470° in 1.75 s in 55 °C hot water and bending angle of 203° in 15 s when irradiated by 6 W/cm 2 NIR light. The segmented hydrogel actuators displayed good programmable actuation performance. Finally, they were applied as smart devices, such as hydrogel hook for grasping object, switch for controlling circuit and valve for governing fluid flow. This work provides a novel strategy for fabricating programmable hydrogel actuators and will inspire new ideas and constructions for smart soft hydrogel actuators.

Kinetically and thermodynamically controlled one-pot growth of gold nanoshells with NIR-II absorption for multimodal imaging-guided photothermal therapy

Chen Ming,Chen Xiao-Tong,Zhang Lian-Ying,Meng Wei,Chen Yong-Jian,Zhang Ying-Shan,Chen Zhi-Cong,Wang Hui-Min,Luo Chun-Mei,Shi Xiu-Dong,Zhang Wen-Hua,Wang Mao-Sheng,Chen Jin-Xiang

Since the successful clinical trial of AuroShell for photothermal therapy, there is currently intense interest in developing gold-based core-shell structures with near-infrared (NIR) absorption ranging from NIR-I (650–900 nm) to NIR-II (900–1700 nm). Here, we propose a seed-mediated successive growth approach to produce gold nanoshells on the surface of the nanoscale metal–organic framework (NMOF) of UiO-66-NH2 (UiO = the University of Oslo) in one pot. The key to this strategy is to modulate the proportion of the formaldehyde (reductant) and its regulator / oxidative product of formic acid to harness the particle nucleation and growth rate within the same system. The gold nanoshells propagate through a well-oriented and controllable diffusion growth pattern (points → facets → octahedron), which has not been identified. Most strikingly, the gold nanoshells prepared hereby exhibit an exceedingly broad and strong absorption in NIR-II with a peak beyond 1300 nm and outstanding photothermal conversion efficiency of 74.0%. Owing to such superior performance, these gold nanoshells show promising outcomes in photoacoustic (PA), computed tomography (CT), and photothermal imaging-guided photothermal therapy (PTT) for breast cancer, as demonstrated both in vitro and in vivo.

Eco-friendly cellulose-based hydrogel functionalized by NIR-responsive multimodal antibacterial polymeric ionic liquid as platform for promoting wound healing

Wenqi Song,Tiantian Xu,Liwei Qian,Sufeng Zhang,Chaoli Wang,Yuzhen Zhao,Zemin He,Valentin Nica,Zongcheng Miao

With the trend of sustainable development and the complex medical environment, there is a strong demand for multimodal antibacterial cellulose wound dressing (MACD) with photothermal therapy (PTT). Herein, a novel MACD fabrication strategy with PTT was proposed and implemented through graft polymerization of an imidazolium ionic liquid monomer containing iron complex anion structure. The fabricated hydrogels exhibited excellent antibacterial properties because of the efficient photothermal conversion ability (68.67 %) of ionic liquids and the intrinsic structural characteristic of quaternary ammonium salts. The antibacterial ratio of cellulosic hydrogel dressings to S. aureus and E. coli could reach 99.57 % and 99.16 %, respectively. Additionally, the fabricated hydrogels demonstrated extremely low hemolysis rates (<5 %) and excellent cell viability (~>85 %). Furthermore, in vivo antibacterial experimental results proved that the fabricated antibacterial dressings could significantly accelerate wound healing. Therefore, the proposed strategy would provide a new method of designing and preparing high-performance cellulose wound dressings.

Glutathione-Exhausting Nanoprobes for NIR-II Fluorescence Imaging-Guided Surgery and Boosting Radiation Therapy Efficacy via Ferroptosis in Breast Cancer

Min Wei,Jingwen Bai,Xiao Shen,Kangliang Lou,Yiyang Gao,Ruichan Lv,Peiyuan Wang,Xiaolong Liu,Guojun Zhang

Breast-conserving surgery (BCS) is the standard of care for early breast cancer patients, while the high ratio of reoperation is still a challenge due to inaccurate margin assessments. In patients with locally advanced or advanced breast cancer, radiotherapy is an important treatment for local control or improvement of quality of life. However, enhancing sensitization to radiotherapy is an unmet medical need. To solve the above clinical predicaments, a glutathione (GSH) exhausting virus-like silicon dioxide nanoprobe with Gd coating and folic acid (FA) modification is designed. After loading ICG in the mesopores, the VGd@ICG-FA probe efficiently targets tumor cells with high resolution, due to its virus-like morphology and folate acid anchoring. Especially, the fabricated nanoprobe enables the identification of tiny cancers and navigates precise surgery under NIR-II fluorescence imaging. Moreover, after the nanoprobes enter into the cytoplasm of cancer cells, tetrasulfide linkages in the silica framework are broken under the triggering of high GSH concentrations. In turn, the broken framework exhausts GSH to disrupt intracellular reactive oxygen species (ROS) homeostasis, and Gd produces more ROS under radiotherapy, further activating ferroptosis, and resulting in the enhancement of radiotherapy in breast cancer. Therefore, our nanoprobe exhibits tremendous potential as a NIR-II fluorescence imaging agent with no systematic side effects for precise cancer surgery and nanotherapeutics for boosting radiation sensitivity in future clinical translation of breast cancer.

“Three-in-one” platform based on Fe-CDs nanozyme for dual-mode/dual-target detection and NIR-assisted bacterial killing

Mei Yang,Jun Yao,Borui Su,Nini Xin,Ting Zhou,Mingze Zeng,Chengheng Wu,Dan Wei,Jing Sun,Hongsong Fan

As nanozymes, carbon dots (CDs) have attracted increasing attention due to their remarkable properties. Besides general enzyme activity, their photoluminescence and photothermal properties have been explored rarely, whereas their synergistic effects might produce CDs-based nanozymes of high performance. Here, iron-doped CDs (Fe-CDs) with tunable fluorescence and enhanced peroxidase-like activity were designed to develop a novel “three-in-one” multifunctional platform to provide dual-mode/dual-target detection and near infrared (NIR)-assisted antibacterial ability. This proposed strategy for a H2O2 test exhibited a wide linear relationship with a low limit of detection (LOD) of 0.16 μM (colorimetric) and 0.14 μM (ratiometric fluorescent). Furthermore, due to the nature of cholesterol being oxidized to H2O2 by cholesterol oxidase, sensitive and selective detection of cholesterol was realized, and the LOD was 0.42 μM (colorimetric) and 0.27 μM (ratiometric fluorescent), surpassing that reported previously. This result suggested that Fe-CDs could be used for dual-mode quantification of large family of H2O2-producing metabolites, thereby paving the way for developing multi-mode sensing strategies based on nanozymes. Moreover, this platform showed synergistic effects for antibacterial application, indicating great prospects for bacterial killing as well as wound disinfection and healing. Hence, this platform could contribute to the construction of multifunctional CDs with high performance.

A novel multifunctional mitochondrion-targeting NIR fluorophore probe inhibits tumour proliferation and metastasis through the PPARγ/ROS/β-catenin pathway

Jianv Wang,Jing Jia,Qingqing He,Yang Xu,Hongye Liao,Xia Xiong,Li Liu,Changzhen Sun

Recent advancements in tumour-targeted therapies and immunotherapy offer hope to patients with various malignancies. However, the uncontrolled growth and metastatic infiltration of malignant tumours remain a huge therapeutic challenge. Therefore, this study aimed to develop an integrated multifunctional diagnostic and treatment reagent IR-251 that can not only be used for tumour imaging but also to inhibit tumour growth and metastasis. Besides, our results showed that IR-251 targeted and damaged the mitochondria in cancer cells via organic anion-transporting polypeptides . Mechanistically, IR-251 induced ROS overproduction by inhibiting PPARγ and then inhibiting the β-catenin signalling pathway and downstream protein molecules related to the cell cycle and metastasis. Moreover, the excellent anti-tumour proliferation and metastasis ability of IR-251 were verified in vitro/in vivo . And histochemistry staining revealed that IR-251 inhibited tumour proliferation and metastasis, which showed no significant side effect. In conclusion, this novel, multifunctional, mitochondria-targeting near-infrared fluorophore probe IR-251 has great potential in achieving accurate tumour imaging and inhibiting tumour proliferation and metastasis, and the underlying mechanism of action of IR-251 is mainly via the PPARγ/ROS/β-catenin pathway.
