Preparation and characterization of zein–caseinate–pectin complex nanoparticles for encapsulation of curcumin: pectin extracted by high-speed shearing from passion fruit (Passiflora edulis f. flavicarpa) peel

Xiefei Li,Yupeng Lin,Yumeng Huang,Xin Li,Fengping An,Hongbo Song,Qun Huang

BACKGROUND Pectin extracted by high-speed shearing from passion fruit peel (HSSP) is a potentially excellent wall material for encapsulating curcumin, which has multiple advantages over pectin prepared by heated water extraction. HSSP was used to fabricate complex nanoparticles of zein–sodium caseinate–pectin for encapsulation of curcumin in this study. The influence of heating on the physicochemical properties of the composite nanoparticles was also investigated, as well as the effect of composite nanoparticles on the encapsulation efficiency, antioxidant activity and release characteristics of curcumin. RESULTS The nanoparticles were formed through electrostatic interactions, hydrogen bonds and hydrophobic interactions between the proteins and HSSP. A temperature of 50 °C was more favorable for generating compact and small-sized nanoparticles, which could effectively improve the encapsulation efficiency and functional properties. Moreover, compared to other pectin used in the study, the nanoparticles prepared with HSSP showed the best functionality with a particle size of 234.28 ± 0.85 nm, encapsulation rate of 90.22 ± 0.54%, free radical scavenging rate of 78.97% and strongest protective capacity in simulated gastric fluid and intestinal release effect. CONCLUSION Zein–sodium caseinate–HSSP is effective for encapsulating and delivering hydrophobic bioactive substances such as curcumin, which has potential applications in the functional food and pharmaceutical industries. © 2024 Society of Chemical Industry.

Curcumin Induces Macrophages Polarization to Alleviate Ulcerative Colitis by Regulating Macrophage Nuclear Morphology via Affecting the Interaction Between Enteric Glial Cells Glial-Derived Neurotrophic Factor and Casein Kinase 2

Huixia Zhang,Mengge Ren,Qi An,Ziling Liu,Chenyang Li,Yuchen Bai,Dongxing Shi,Ying Wang,Fubin Qiu,Shangyun Lu

Macrophages exhibit dynamic functional phenotype shifts under diverse influences, releasing either anti-inflammatory or pro-inflammatory cytokines and playing a pivotal role in ulcerative colitis (UC) pathogenesis. In recent years, the role of enteric glial cells (EGCs) in maintaining intestinal homeostasis has been increasingly recognized. In this study, we investigated whether the alleviation of UC by curcumin is associated with the regulation of EGCs-macrophage interactions, thereby affecting macrophage polarization and its specific mechanisms. Through animal experiments and flow cytometry, we evaluated the efficacy of curcumin in alleviating UC and its impact on macrophages. Furthermore, GDNF intervention confirmed its pivotal role in curcumin's ability to alleviate UC. Finally, molecular docking, cell experiments, laser confocal microscopy, and transmission electron microscopy were employed to elucidate the molecular mechanism. It was found that curcumin can alleviate UC by promoting macrophages' polarization into anti-inflammatory subtypes, increasing the level of glial cell neurotrophic factor (GDNF) in UC mice, and GDNF treatment can alleviate DSS-induced colon injury. Importantly, we demonstrated that GDNF regulates macrophages' polarization by interacting with CK2 to affect the morphological characteristics of macrophage nucleus. The results of this study further enrich the existing pathways for curcumin to alleviate UC and provide new ideas for the prevention and treatment of UC.

ZIF-8 Based Active Wound Dressing for pH Modulation and pH-Responsive Delivery of Curcumin and Zn2+: Effect of Acidity on the Drug Release for Antibacterial and Antioxidant Efficacy

Seojin Jung,Seokhee Chang,Na-Eun Kim,Yoon-Jae Song,Jooyoun Kim

A wound evolves with variable pHs depending on the healing stage, and the maintenance of mild acidic condition can help healing by deterring the formation of a biofilm. Herein, a dual-layer wound dressing with an acid-responsive zeolitic imidazolate framework–8 (ZIF-8) layer and an acid-modulating layer is designed to control the acid-dependent release of therapeutic agents for the improved healing process. The outer dressing layer is coated with curcumin-incorporated ZIF-8, where curcumin and zinc ions are released upon dissociation of ZIF-8 in an acidic condition. The inner layer, a pH modulator, is fabricated by electrospinning polyvinylpyrrolidone with addition of acetic acid. In vitro release test shows that the outer layer dressing attains a considerably higher release of curcumin and Zn 2+ under an acidic condition. Combined with the inner acidic layer, the dual layer dressing exhibits facilitated release of effective agents regardless of environmental pHs, attributed to the proper pH modulation to about 6.5. The anti-oxidant activity of curcumin and antibacterial activity of Zn 2+ are confirmed. The study highlights the novel proof-of-concept design of active wound dressing demonstrating the dual functions of pH regulation and the stimulus-responsive drug delivery.

A Co-Encapsulation of Coenzyme Q10 and Curcumin in Liposomes Coated with Chitosan (Q10-Cur-Lip-Chi) with Enhanced Solubility and Stability for Good Release Performance and Antioxidative Activity

Yu Linjin,Chao Chuyan,Li Qinglan,Ye Songling,Lin Jiasen,Zhong Shuling,Xuan Qiancheng,Xu Kailin,Zhao Suqing

Background: Coenzyme Q10 (Q10) is a powerful lipophilic antioxidant with poor solubility in aqueous media. Curcumin (Cur) is a natural polyphenolic phytochemical molecule with poor aqueous solubility. The liposome is an improved administration of drugs because it is biocompatible and permeable for nutraceutical delivery. Chitosan, a hydrophilic polymer, is often used as a polymer coating for its good biocompatible and biodegradable properties, and its relatively low toxicity level. Methods: Q10 and Cur co-loaded liposomes coated with chitosan (Q10-Cur-Lip-Chi) were constructed. The co-encapsulation of Q10 and Cur in liposomes coated with chitosan was verified by TEM, DLS, DSC, FT-IR, and XRPD. The release profile and antioxidant activity of Q10-Cur-Lip-Chi were accessed. Results: The particle size of Q10-Cur-Lip-Chi was about 1440 nm with narrow particle distribution. A satisfactory encapsulation efficiency (EE) of Q10 was about 98%, and 25% for that of Cur. Q10-Cur- Lip-Chi showed higher solubility and better pH resistance with 98.5% of Q10 and Cur retention at pH 7.0 - 9.0. Q10-Cur-Lip also showed great salt stability with a vesicle size change of less than 5%. PSof Q10-Cur-Lip-Chi changed less than 10% at 4°C of storage. Q10-Cur-Lip-Chi also exhibited a good controlled release profile with its accumulative release of less than 34% for Q10 and 30% for curcumin after 24 h. The Q10-Cur-Lip-Chi performed a synergistic effect on antioxidant activity reaching 41.86±1.84%, which was 5.9 times higher than that of Q10, 2.5 times higher than that of Cur, and 1.7 times higher than that of the mixture. Conclusion: The co-encapsulation Q10-Cur-Lip-Chi improves the solubility and stability of Q10 and Cur for good release performance and antioxidative activity.

Fabrication of Bacterial Cellulose Nanofibers/Soy Protein Isolate Colloidal Particles for the Stabilization of High Internal Phase Pickering Emulsions by Anti-solvent Precipitation and Their Application in the Delivery of Curcumin

Shen Rui,Lin Dehui,Liu Zhe,Zhai Honglei,Yang Xingbin

In this study, the anti-solvent precipitation and a simple complex method were applied for the preparation of bacterial cellulose nanofiber/soy protein isolate (BCNs/SPI) colloidal particles. Fourier transform IR (FT-IR) showed that hydrogen bonds generated in BCNs/SPI colloidal particles via the anti-solvent precipitation were stronger than those generated in BCNs/SPI colloidal particles self-assembled by a simple complex method. Meanwhile, the crystallinity, thermal stability, and contact angle of BCNs/SPI colloidal particles via the anti-solvent precipitation show an improvement in comparison with those of BCNs/SPI colloidal particles via a simple complex method. BCNs/SPI colloidal particles via the anti-solvent precipitation showed enhanced gel viscoelasticity, which was confirmed by dynamic oscillatory measurements. Furthermore, high internal phase Pickering emulsions (HIPEs) were additionally stable due to their stabilization by BCNs/SPI colloidal particles via the anti-solvent precipitation. Since then, HIPEs stabilized by BCNs/SPI colloidal particles via the anti-solvent precipitation were used for the delivery of curcumin. The curcumin-loaded HIPEs showed a good encapsulation efficiency and high 2,2-diphenyl-1-picrylhydrazyl (DPPH) removal efficiency. Additionally, the bioaccessibility of curcumin was significantly increased to 30.54% after the encapsulation using the prepared HIPEs. Therefore, it can be concluded that the anti-solvent precipitation is an effective way to assemble the polysaccharide/protein complex particles for the stabilization of HIPEs, and the prepared stable HIPEs showed a potential application in the delivery of curcumin.

Development of curcumin-loaded methoxy poly(ethylene glycol)-block- poly(caprolactone)-block-poly(1, 4, 8-Trioxa [4.6] spiro-9-undecanone) nanoparticles and studies on their in vitro anti-tumor activities

Yongli Shi,Weiwei Ma,Min Gao,Yaxing Yang

The purpose of this paper was to fabricate a novel methoxy poly(ethylene glycol)-block-poly(caprolactone)-block-poly(1, 4, 8-Trioxa [4.6] spiro-9-undecanone) (mPEG- b -PCL- b -PTOSUO, mPECT) triblock copolymer and study on the in vitro anti-tumor activity of curcumin-loaded mPECT nanoparticles (NPs). The mPEG- b -PCL- b -PTOSUO NPs were fabricated with solvent evaporation. Transmission electron microscope (TEM) and laser particle analyzer were applied to investigate the morphology and size distribution of the obtained mPECT NPs. The cytotoxicity of the copolymer (mPECT) was reflected by cell viability. Curcumin (CUR), as a model drug, was encapsulated into mPECT NPs. The in vitro anti-tumor activity of CUR-loaded mPECT NPs were also studied. 1 H nuclear magnetic resonance ( 1 H NMR), Raman, and Fourier transform infrared spectroscopy (FTIR) spectra confirmed the obtaining of mPECT. TEM photograph showed that most of mPECT NPs were in spherical shapes with a uniform size distribution. High cell viability suggested that the cargo-free mPECT NPs had no obvious cytotoxicity. Fluorescent photographs illustrated that CUR-loaded mPECT NPs could be up-taken by SW1990 cells. The medicated NPs could inhibit the proliferation of SW1990 cells. Therefore, the mPECT NPs could be used as a vehicle to improve the bioavailability and anti-tumor effects of CUR.

Effects of atmospheric cold plasma voltage on the interfacial properties of soy protein isolate-sodium alginate nanoemulsions co-loaded with curcumin and resveratrol and their application in surimi gels during chilled storage

Tingting Yang,Zhongyuan Liu,Guanghua Xia,Dawei Yu,Zhidong Liu,Yixiang Wang,Liming Zhang,Jiamei Wang

ACP voltage-modified SPI-SA nanoparticles co-encapsulated curcumin and resveratrol. • 30 kV ACP-treated nanoemulsion had optimal stability, highest EE, and superior emulsifying properties. • 30 kV ACP-nanoemulsion enhanced surimi gel strength, WHC, and network compactness. • ACP-modified nanoemulsion inhibited lipid oxidation and water migration, keeping surimi quality for 15 days. This study applied atmospheric cold plasma (ACP) at 0–40 kV to prepare curcumin- and resveratrol-loaded SPI-SA nanoemulsions and evaluated their performance in surimi gels. Moderate ACP treatment at 30 kV uniquely optimized the interfacial structure, achieving monomodal droplet size, the highest zeta potential (−56.6 + 0.62 mV) and maximum encapsulation efficiency (curcumin 89.34 ± 0.32%, resveratrol 88.33 ± 0.22%), loading capacity (curcumin 11.91 ± 0.04%, resveratrol 17.66 ± 0.04%), along with superior antioxidant activity (DPPH and ABTS radical-scavenging activities 72.66 ± 1.31% and 67.05 ± 2.21%). Fourier transform infrared spectroscopy (FTIR) and X-ray diffraction (XRD) confirmed that ACP-induced SPI unfolding, combined with SA enrichment and polyphenol anchoring, promoted a dense hydrogen-bonded interfacial film with enhanced molecular ordering, leading to excellent storage stability and emulsifying properties. In surimi gels, the 30 kV nanoemulsion acted as an optimal active filler, reinforcing the myofibrillar protein network through physical filling and interfacial cross-linking. This resulted in the highest gel strength (3090.05 ± 23.28 g·mm) and water holding capacity (81.51 ± 0.95%), lowest cooking loss (8.20 ± 0.04%) and TBARS values (0.80 ± 0.03 mg MDA/Kg), shortest T₂ relaxation times, and densest microstructure during 15-day refrigerated storage. Unmodified nanoemulsion only provided moderate improvements. Thus, 30 kV is the optimal ACP voltage for preparing stable, high-efficiency nanoemulsions and enhancing surimi gel quality, offering a theoretical basis for ACP-modified protein-polysaccharide delivery systems. Involvement of the inhibition of mitochondrial apoptotic, p53, NF-κB pathways and the activation of Nrf2/HO-1 pathway in the protective effects of curcumin against copper sulfate-induced nephrotoxicity in mice

Chongshan Dai,Meng Li,Yue Liu,Diem Hong Tran,Haiyang Jiang,Shusheng Tang,Jianzhong Shen

Chronic copper exposure could cause potential nephrotoxicity and effective therapy strategies are limited. This study investigated the protective effects of curcumin on copper sulfate (CuSO 4 )-induced renal damage in a mouse model and the underlying molecular mechanisms. Mice were administrated orally with CuSO 4 (100 mg/kg per day) in combination with or without curcumin (50, 100 or 200 mg/kg per day, orally) for 28 days. Results showed that curcumin supplementation significantly reduce the Cu accumulation in the kidney tissues of mice and improved CuSO 4 -induced renal dysfunction. Furthermore, curcumin supplantation also significantly ameliorated Cu exposure-induced oxidative stress and tubular necrosis in the kidneys of mice. Moreover, compared to the CuSO 4 alone group, curcumin supplementation at 200 mg/kg per day significantly decreased CuSO 4 -induced the expression of p53, Bax, IL-1β, IL-6, and TNF-α proteins, levels of NF-κB mRNA, levels of caspases-9 and − 3 activities, and cell apoptosis, and significantly increased the levels of Nrf2 and HO-1 mRNAs in the kidney tissues. In conclusion, for the first time, our results reveal that curcumin could trigger the inhibition of oxidative stress, mitochondrial apoptotic, p53, and NF-κB pathways and the activation of Nrf2/HO-1 pathway to ameliorate Cu overload-induced nephrotoxicity in a mouse model. Our study highlights that curcumin supplementation may be a promising treatment strategy for treating copper overload-caused nephrotoxicity.

Involvement of the activation of Nrf2/HO-1, p38 MAPK signaling pathways and endoplasmic reticulum stress in furazolidone induced cytotoxicity and S phase arrest in human hepatocyte L02 cells: modulation of curcumin

Chongshan Dai,Lei Lei,Bin Li,Yang Lin,Xilong Xiao,Shusheng Tang

Furazolidone (FZD) is extensively used as the antiprotozoal and antibacterial drug in clinic. The previous study has shown that curcumin pretreatment could improve FZD induced cytotoxicity by inhibiting oxidative stress and mitochondrial apoptotic pathway. The current study aimed to investigate the potential roles of endoplasmic reticulum (ER) stress, p38 mitogen-activated protein kinases (p38 MAPK) signaling pathway in curcumin against FZD cytotoxicity by using human hepatocyte L02 cells. The results showed that curcumin could markedly attenuate FZD induced cytotoxicity. Compared with FZD alone group, curcumin pretreatment significantly reduced the expression of phospho (p)-p38, cyclin D1, p-checkpoint kinase 1 (ChK1) and breast cancer associated gene 1 (BRCA1) protein, followed to attenuate S phase arrest. Meanwhile, curcumin pretreatment prevented FZD induced ER stress, evidenced by the inhibition of glucose-regulated protein 78 and DNA damage inducible gene 153/C/EBP-homologous protein (GADD153/CHOP) protein expression. Moreover, compared with the control, FZD exposure activated the protein and mRNA expression levels of nuclear factor erythroid 2-related factor 2 (Nrf2) and heme oxygenase 1 (HO-1), which were further activated by curcumin treatment. These results reveal that curcumin could prevent FZD induced cytotoxicity and S phase arrest, which may involve the activation of Nrf2/HO-1 pathway and the inhibition of p38 MAPK pathway and ER stress.

Salvianolic acid B protects against myocardial damage caused by nanocarrier TiO2; and synergistic anti-breast carcinoma effect with curcumin via codelivery system of folic acid-targeted and polyethylene glycol-modified TiO2 nanoparticles

Lingling Ding,Jiawei Li,Rui Huang,Zhidong Liu,Chunhua Li,Shaozi Yao,Jinyan Wang,Dongli Qi,Nan Li,Jiaxin Pi

Targeted delivery by the folate ligand is an effective way to enhance an anti-breast carcinoma effect, due to its high affinity for the folate receptor, which is overexpressed in many tumor cells. In this study, we firstly synthesized a folic acid (FA)-targeted and polyethylene glycol (PEG)-modified TiO 2 nanocarrier. Then, an FA-PEG-TiO 2 nanoparticle (NP) codelivery system loaded with curcumin and salvianolic acid B were prepared by emulsion evaporation–solidification at low temperature. The obtained folate-targeted NPs (FA-NPs) showed more cytotoxicity on MCF7 cells and MDA-MB-231 cells than a nontargeted NP group. Apart from a synergistic anti-breast cancer effect with curcumin, salvianolic acid B protects the cardiovascular system from oxidative injury by the TiO 2 nanocarrier. With coumarin 6 as a fluorescent probe to observe cellular uptake of NPs, the results of in vitro cellular uptake demonstrated FA-NPs exhibited higher cellular uptake and accumulation in MCF7 cells and MDA-MB-231 cells than nontargeted NPs. Then, in vivo biodistribution of NPs was further qualitatively and quantitatively confirmed by in vivo imaging. More importantly, the animal study further suggested that FA-NPs had significantly stronger antitumor effects via receptor-mediated targeted delivery. Consequently, FA-PEG-TiO 2 NPs loaded with curcumin and salvianolic acid B could be a promising drug-delivery system to treat breast cancer.

Precise ratiometric co-loading, co-delivery and intracellular co-release of paclitaxel and curcumin by aid of their conjugation to the same gold nanorods to exert synergistic effects on MCF-7/ADR cells

Shaohui Xu,Zhu Jin,Zhipeng Zhang,Wei Huang,Yuanyuan Shen,Zhongmin Wang,Shengrong Guo

It is necessary for effective combinational effects that multiple chemotherapeutic agents enter and act on the same tumor cells simultaneously at a suitable ratio. Due to their individual physicochemical or biopharmaceutical properties, they cannot reach the same cells at its predetermined ratio after co-administration. Herein, we report a novel system with precise ratiometric co-loading, co-delivery and intracellular co-release of paclitaxel (PTX) and curcumin (CUR), to investigate their synergistic effects. We prepared the system by conjugating PTX and CUR at different ratios onto the same gold nanorods (GNRs). We demonstrated that PTX and CUR could be precisely ratiometric co-loaded to form dual-drug conjugated biotin-PEG modified GNRs (abbreviated as PTX/ [email protected] ), which could co-deliver dual drugs into tumor cells with a predetermined ratio and co-release them intracellularly. The PTX/ [email protected] with the mass ratio of PTX and CUR at 1:1 exhibited the best synergistic effect on multidrug resistant MCF-7/ADR cells while the free PTX and CUR mixture with the mass ratio of at 1:0.75 displayed the strongest synergism. Cytotoxicity studies showed that PTX/ [email protected] are superior to free drug mixture. Furthermore, PTX/ [email protected] could remarkably induce apoptosis of MCF-7/ADR cells under near-infrared irradiation. Moreover, we found that PTX/ [email protected] significantly inhibited P-glycoprotein (P-gp) expression in MCF-7/ADR cells.
