trans -Porphyrin containing phenyl group at both ends was synthesized via corresponding dipyrromethanes (DPM) in the presence of TFA and DDQ. The polyethyleneglycole (PEG) chains were attached at the trans positions of ABA type porphyrinic moiety. The prepared PEGylated porphyrin (with PEG arms at the periphery) P-(PEO) 2 was self-assembled into spheres of uniform diameter while their DMF solution was dialyzed into water. Furthermore, the addition of fullerene (C 60 ) to P-(PEO) 2 caused complexation between porphyrin and fullerene which lead to interesting and trenchant morphology having worm like lateral aggregates. The assembly process and the as-prepared edifices were characterized using UV–vis absorption spectroscopy and the morphology of these assemblies was investigated using transmission electron microscope (TEM). Furthermore, the DNA binding with porphyrin aggregates has been demonstrated via electrochemical (cyclic voltammetric) response of screen printed electrodes modified . The obvious increase in redox current disclosed the intermolecular interactions of the adducts with the DNA. These studies of unprecedented adducts can further be explored for potential applications in biosensors .

Herein we introduce the first successful assay of biomolecule by in situ selective crystallization based quartz crystal microbalance (QCM). Selective crystallization of CaCO3 on QCM sensor surface was utilized as an efficient mass amplification strategy and enhanced the sensitivity of QCM significantly. High specificity is guaranteed by the cooperation of two functional groups: −N(CH3)3 and −COOH. Passivation of sensor surface is realized by self-assembly of −N(CH3)3, which effectively inhibited the nonspecific crystallization. The DNA target is detected through hybridization of probe DNA labeled with −COOH, which can effectively promot the in situ surface crystallization of CaCO3. The concentration of target DNA is reflected by the frequency shift of QCM which is directly induced by the surface crystallization. The selective crystallization based QCM platform is simple, straightforward, high sensitive, and high specific. We demonstrate the excellent LOD (2 aM DNA) and a linear range of 10aM to 1 nM for DNA. Detection of Ramos cells are also realized with a LOD of five cells and a linear range of 5–6000 cells.

Three oligomeric linear poly(beta-amino esters) (PAEs) were synthesized and used to improve the gene transfection efficiency and reduce the cytotoxicity of poly(L-lysine) (PLL). The synthesized PAEs can co-assemble with PLL and plasmid DNA to form PLL/PAE/DNA ternary complexes of nanoscale sizes and moderate surface charges. The existence of multiple tertiary amines on PAE backbones imparts the ternary complexes with strong proton buffering capacity and therefore enhances the endosomal escape ability of the ternary complexes. In HeLa cells, the PLL/PAE/DNA ternary complexes show up to 476-fold enhancement in gene transfection efficiency when compared with the PLL/DNA binary complexes. Meanwhile, the incorporation of PAE oligomers significantly reduced the cytotoxicity of PLL. The strategy proposed here may provide insights into the development of PLL based gene delivery systems.

The microbubble is a kind of clinically applied ultrasound contrast agent in disease diagnosis that can also rupture under sonication to increase membrane permeability and promote gene entry into targeted cells. However, the development of ultrasound-mediated gene delivery might be restricted because genes and microbubbles were separated and would not reach the targeted cells simultaneously. Herein, a kind of crosslinked positive microbubbles (CPMBs) were prepared to load DNA as gene vectors to promote gene delivery efficiency. The BSA shell of the CPMBs was crosslinked with disulfide bonds, which obviously enhanced the stability of the CPMBs. Furthermore, the CPMBs revealed sonoporation effects comparable to those of clinically applied SonoVue microbubbles. As DNA and CPMBs were electrostatically linked as an entirety, they would reach cells simultaneously. Thus, with the aid of ultrasound, these DNA-loaded microbubbles promoted DNA entry into cytoplasm more effectively and obtained higher cellular uptake efficiency and better transfection efficiency than DNA-mixed microbubbles. Confocal microscopy results showed that rupturing of the CPMBs/DNA entire microbubbles under sonication could carry DNA directly into the cytoplasm or nucleus. All results indicated that the cytocompatible DNA-loaded microbubbles have promising prospects in ultrasound-mediated gene delivery.

Driven by the urgent need to detect trace heavy metal ions in various real water samples, this article demonstrates for the first time an electrochemical biosensor based on DNA modified Fe3O4@Au magnetic nanoparticles (NPs). Three DNA probes are designed to contain certain mismatched base pairs. One is thiolated and modified on the surface of Fe3O4@Au NPs (DNA 1). The other two probes (DNA 2 and 3) are labeled with two independent electrochemical species. Stable structures of cytosine–Ag+–cytosine and thymine–Hg2+–thymine formed in the presence of Ag+ and Hg2+ can assist the hybridization of DNA 1/DNA 2 and DNA 1/DNA 3, which locate corresponding electrochemical species onto the surface of the magnetic NPs. The achieved nanocomposites are then used as selective electrochemical probes for the detection of heavy metal ions by recording the square wave voltammetry signals. Simultaneous detection of Ag+ and Hg2+ is demonstrated without significant interference, and their individual high sensitivities are fundamentally preserved, which meet the requirements of U.S. Environmental Protection Agency (USEPA). Furthermore, the proposed method has been challenged by various real water samples. The results confirm the DNA modified magnetic NPs based sensing method may have potential applications for the monitoring of heavy metal ions in real sample analysis.

This paper reported that the peroxidase-like activity of DNA–Ag/Pt nanoclusters (NCs) can be inhibited selectively by Hg2+. The interaction between Hg2+ and the DNA–Ag/Pt NCs was discussed through characterization by TEM, DLS and XPS, etc. It was found that the integration of both Hg0 and Hg2+ to the DNA–Ag/Pt NCs resulted in the DNA–Ag/Pt NCs aggregation and the decrease of Pt2+ content on the DNA–Ag/Pt NCs surface, which finally impaired their catalytic activity. Based on the above mechanism, a colorimetric detection for Hg2+ was developed with a detection limit (LOD) of 5.0 nM and a linear range from 10 nM to 200 nM. The method was highly selective toward Hg2+ over other common metal ions, low-cost and simple, which facilitated the application of detecting Hg2+ in tap water.

Trichloroethylene (TCE) had been shown to induce aberrant DNA methylation changes in mouse liver, which could be critical in the development of hepatocellular carcinoma. To understand the effect of methionine, a methyl donor, on TCE-induced mouse liver cancer, we exposed B6C3F1 mice to TCE in the presence or absence of methionine. We found that supplementation with methionine attenuated the TCE-induced hypo- and hyper-methylation status of Cdkn1a and Ihh in mouse liver, respectively. The TCE-induced mRNA expression changes of Cdkn1a, Ihh and Jun were also alleviated by methionine. However, methionine supplementation accelerated the TCE-induced mRNA over-expression of Mki67, and enhanced the TCE-induced mRNA down-regulation of Dnmt3a and Tet2. In summary, our results showed that methionine could attenuate at least part of the TCE-induced DNA methylation changes, but the additional gene expression changes induced by supplementary of methionine does not support the assumption that methionine could prevent TCE-induced liver carcinogenesis.

In order to improve the sensitivity of ion imprinted electrochemical (IIE) sensor to detect copper ions, a novel dual signal amplification (DSA) sensor was proposed using deoxyribonucleic acid (DNA) modified IIE sensor. Chitosan (CS) was used as a functional monomer for IIE sensor. The IIE sensor was prepared by mixing copper ion templates with CS and removing the templates. Gold nanoparticles (AuNPs) were electrodeposited onto the surface of IIE sensor to improve their conductivity and provide a connecting platform for DNA probes. And then the DNA probe was fixed on the surface of AuNPs to prepare a DSA sensor. The signal of DSA sensor was significantly amplified by the synergistic effect of IIE sensor and DNA probe. The feasibility, parameter optimization and electrochemical performance were analyzed by experimental comparison, surface micro scanning and kinetic study. The results show that compared with IIE sensor, the sensitivity of DSA sensor is significantly improved, with a value of 1.6712 × 10 -6 A·(µM) -1 . The exchange current density is 5.9765 × 10 -8 A·cm −2 . The detection range is 10 nM to 100 nM (R 2 = 0.99651) with the detection limit of 4.17 nM (3σ/slope). The recommended method has high sensitivity and selectivity, as well as very high recovery rates in tap water and milk.

Clinical diagnosis of ovarian cancer lacks high accuracy due to the weak selection of specific biomarkers along with the circumstance biomarkers localization. Clustering analysis of proteins transported on exosomes enables a more precise screening of effective biomarkers. Herein, through bioinformatics analysis of ovarian cancer and exosome proteomes, two coexpressed proteins, EpCAM and CD24, specifically enriched, were identified, together with the development of an as-derived dual-aptamer targeted exosome-based strategy for ovarian cancer screening. In brief, a DNA ternary polymer with aptamers targeting EpCAM and CD24 was designed to present a logic gate reaction upon recognizing ovarian cancer exosomes, triggering a rolling circle amplification chemiluminescent signal. A dynamic detection range of 6 orders of magnitude was achieved by quantifying exosomes. Moreover, for clinical samples, this strategy could accurately differentiate exosomes from healthy persons, other cancer patients, and ovarian cancer patients, enabling promising in situ detection. By accurately selecting biomarkers and constructing a dual-targeted exosomal protein detection strategy, the limitation of insufficient specificity of traditional protein markers was circumvented. This work contributed to the development of exosome-based prognosis monitoring in ovarian cancer through the identification of disease-specific exosome protein markers.

The development of new methods for fabricating artificial multienzyme systems has attracted much interest because of the potential applications and the urgent need for multienzyme catalysts. Controlling the enzyme ratio is critical for improving the cooperative enzymatic activity in multienzyme systems. Herein, we introduce a versatile strategy for fabricating a multienzyme system by coimmobilizing horseradish peroxidase (HRP) and glucose oxidase (GOx) on magnetic nanoparticles multifunctionalized with dopamine derivatives through DNA-directed immobilization. This multienzyme system exhibited precise enzyme ratio control, high catalytic efficiency, magnetic retrievability, and enhanced stability. The enzyme ratio was conveniently adjusted, as required, by regulating the quantity of functional groups on the multifunctionalized nanoparticles. The optimal mole ratio of GOx/HRP was 2:1. The Michaelis constant Km and specificity constant (kcat/Km, where kcat is the catalytic rate constant) of the multienzyme system were 1.41 mM and 5.02 s–1 mM–1, respectively, which were approximately twice the corresponding values of free GOx&HRP. The increased bioactivity of the multienzyme system was ascribed to the colocalization of the involved enzymes and the promotion of DNA-directed immobilization. Given the wide variety of possible enzyme associations and the high efficiency of this strategy, we believe that this work provides a new route for the fabrication of artificial multienzyme systems and can be extended for a wide range of applications in diagnosis, biomedical devices, and biotechnology.

The Phi29 DNA polymerase is renowned for its processivity in synthesizing single-stranded DNA amplicons by rolling around a circularized DNA template. However, DNA synthesis rolling circle amplification (RCA) is significantly hindered by the secondary structure in the circular template. To overcome this limitation, an engineered circular template without secondary structure could be utilized to improve the sensitivity of RCA-based assays without increasing its complexity. We herein proposed an entropy-driven special RCA technology for the detection of HPV16 E7 gene at room temperature. The strategy is composed of a molecular beacon containing a loop region for nucleic acid target recognition and a stem region to initiate RCA. With the target analyte, the stem region of the molecular beacon will be exposed and then hybridized with a special circular template to initiate the DNA amplification. We tested different designs of the molecular beacon sequence and optimized the assay’s working conditions. The assay achieved a sensitivity of 1 pM in 40 min at room temperature. The sensitivity of this assay, at 1 pm, is about a hundred-fold greater than that of conventional linear RCA performed in solution. Our proposed sensor can be easily reprogrammed for detecting various nucleic acid markers by altering the molecular beacon’s loop. Its simplicity, rapid assay time, and low cost make it superior to RCA sensors that utilize similar strategies.

Ochratoxin A (OTA) poses a great threat to food and environmental safety owing to its high toxicity. Herein, we developed a sensitive electrochemical aptasensor that integrates aptamer, enzyme-free DNA circuits, and electrochemical technology for OTA detection. In the presence of target OTA, the aptamer recognition first initiated the EDC circuit to releasing DNA (T2) from a ternary DNA complex (S-O-T2). The released T2 as the trigger to activate the CHA circuit and expose amounts of G-quadruplex sequences on modified electrode surface. Eventually, hemin was embedded in the G-quadruplex structure and generated an amplified electrochemical signal. Under optimal conditions, this developed biosensor is capable of detecting OTA ranging from 1 pg/mL to 10 ng/mL with a low detection limit of 0.135 pg/mL, which could be contributed to the design of target-triggered EDC and CHA cascade amplification. Additionally, the proposed biosensor was applied to analyze OTA in corn samples with satisfactory results, demonstrating that the proposed method has great potential in food safety and environmental monitoring.
