Oxygen vacancies promote electron transfer in PRT@MnO2-x with boosting oxidase-mimicking activity for dual-mode detection of β-galactosidase

Wenli Lai,Qian Lin,Cunshan Zhang,Bin Chen,Jinyu Li,Lingting Wu,Maoru Lin,Hengfen Dai,Liqing Lin

A novel oxygen vacancy-rich PRT@MnO 2-x nanozyme was fabricated via a facile one-pot ultrasonic method. • The as-prepared material possesses prominent oxidase-like activity, relying on in-situ water activation rather than exogenous oxygen. • A sensitive colorimetric/fluorescent dual-mode sensing platform was constructed for β-Gal activity with low detection limits. • The developed strategy shows satisfactory performance in human serum, possessing promising clinical application potential. Vacancy engineering has been widely applied to modulate the enzyme-like activity of nanomaterials by adjusting their surface/electronic structures and creating additional active sites. Regulating oxygen vacancies in nanomaterials and elucidating their oxygen activation mechanisms are crucial for designing high-performance nanozymes. Herein, we employed KMnO 4 as the manganese source and protamine as the stabilizer to prepare transition metal manganese oxide (PRT@MnO 2-x ) rich in oxygen vacancies. The as-prepared PRT@MnO 2-x exhibited superior oxidase-like activity, which is proposed to originate from oxygen vacancy-mediated electron transfer and valence band hole-driven water oxidation to ·OH. Steady state kinetic constant (K m ) suggested that PRT@MnO 2-x had a stronger affinity toward 3,3′,5,5′-tetramethylbenzidine and possessed prominent catalytic performance (K m and V max were 0.074 mM and 0.031 mM s −1  mg −1 ). An innovative method is proposed to employ PRT@MnO 2-x nanosheets with abundant oxygen vacancies for colorimetric/fluorescent dual-mode detection of β-Galactosidase (β-Gal) activity. Notably, the OXD-like activity of PRT@MnO 2-x is largely independent of dissolved oxygen or molecular oxygen. Instead, it facilitates electron transition from the valence band to the conduction band through the form of oxygen vacancies, enabling the oxidation of water to ·OH on the valence band of PRT@MnO 2-x . The detection limits (LOD) determined by colorimetric and fluorescence analysis for β-Gal activity are 0.015 U/L and 0.054 U/L respectively, and satisfactory spiked recovery results were achieved in human serum samples. This method has the advantages of low cost, simple operation and rapid response. The dual-mode signal sensor is expected to provide more accurate and robust detection of β-Gal than the single-mode probe, and has a wider application prospect. Synergistic photo-electro-piezo effects in a polypyrrole/MnO2-PVDF nanofiber membrane for higher energy efficiency of hybrid solar evaporation systems

Muhammad Sultan Irshad,Wancheng Qin,Naila Arshad,Naveed Mushtaq,Muhammad Zeeshan,Xiaochao Fan,Tianxiang Zhou,Yi E,Irshad Hussain,Tao Mei,Yajun Qi,Van-Duong Dao,Nang Xuan Ho,Xianbao Wang

Energy harvesting technologies are gaining attention because of their potential to generate power from ambient sources like light and mechanical motion. The development of advanced electrospun polyvinylidene fluoride (PVDF) flexible films with dual functionality (photothermal-piezoelectric effect) has become a prominent area of research, focusing on optimizing their hydrophilicity. Herein, electrospun MnO 2 nanosheets/PVDF flexible films are sequentially fabricated to transform the dielectric and ferroelectric properties, which result in an open-circuit voltage of 20 V and a short-circuit current of 0.10 μA under repeated mechanical force of 10 N at 2 Hz. Wearable films demonstrated the ability to harvest the biomechanical energy of human movements, such as finger and joint tapping, as well as wearable shoe soles that harvest energy from walking and running. More importantly, the impregnation of polypyrrole over MnO 2 /PVDF films (MnPVDF@PPy) and post-hydrophilic modification achieved an enhanced evaporation rate of 1.85 kg m −2 h −1 and waste heat recovery into open-circuit voltage of 135 mV and a short-circuit current of 33 mA under one sun irradiation. The unique interplay between photo-piezo-electro mechanisms ensures efficient energy flow, robustness, and adaptability in dynamic environments by addressing critical challenges such as water scarcity and intelligent sensing with a single, multifunctional platform.

Chondroitin Sulfate-Based MPDA@MnO2 Nanocomposite Hydrogels: A Smart Drug Delivery System with pH/ROS Responsiveness and Photothermal-Enhanced Therapeutic Effects

Xu Wang,Qin Ding,Rui Ran,Qiangguo Chen,Xian Li,Xu Ye

Chronic wounds, particularly those complicated by infection, present significant challenges in clinical management. The microenvironment of these wounds is typically characterized by the accumulation of reactive oxygen species (ROS) and abnormal local pH levels, both of which impede the healing process. Baicalin (BA), a natural flavonoid, exhibits anti-inflammatory activity, ROS-scavenging capability, and pro-healing effects. In this study, hydrogels were synthesized through photoinitiated radical polymerization of methacrylic anhydride (MAA) and dopamine (DA)-modified chondroitin sulfate (ChSMA-DA), grafting degrees of MA and DA were 58%, 23%, MPDA@MnO2nanoparticles (NPs), and methacrylated gelatin (GelMA). The gelation time, microtopography, swelling behavior, and water retention of the hydrogels were investigated, along with their degradation, rheological properties, and photothermal effects. The results indicate that swelling ratio (SR) and water retention (WR) of optimal HG-MPDA@MnO2-M sample were 5.7, 82.42%, exhibited responsive behavior upon weakly acidic environment with pH 6.5 and elevated ROS levels, and exhibited a stable photothermal effect (photothermal conversion efficiency was 22.7%) under 808 nm near-infrared (NIR) light. Following the incorporation of the drug model BA, the cumulative release percentage over 24 h under the combined stimulation of pH 6.5, 1 mmol·L−1H2O2, and 808 nm NIR was 81.1%, significantly higher than either factor alone. These hydrogels show promise as an injectable dressing for chronic wounds, effectively integrating the internal microenvironment of the wound tissue with external NIR to modulate drug release.

A tumor microenvironment-responsive and mannose-mediated MnO2-based nano-delivery system for synergistic targeted chemotherapy in triple-negative breast cancer

Guanghui Mei,Hanwen Wang,Xinhua Lin,Lingyi Huang

Triple-negative breast cancer (TNBC) remains a formidable challenge due to its aggressive progression and the absence of established therapeutic targets. This study engineered a multifunctional, tumor microenvironment (TME)-responsive nanoplatform MnO 2 @Man/DOX, which was designed for synergistic targeted chemotherapy and TME modulation. The platform comprises a manganese dioxide (MnO 2 ) core for redox regulation and a mannose (Man) shell for active targeting and metabolic sensitization, stabilized with bovine serum albumin and sodium dodecyl sulfate. MnO 2 @Man/DOX nanoparticles are nearly spherical (289 nm) and exhibit dual-responsiveness by efficiently depleting intracellular glutathione and catalyzing endogenous hydrogen peroxide into cytotoxic hydroxyl radicals via Fenton-like reactions. In vitro, the nanoplatform demonstrated a remarkable 8-fold reduction in IC 50 (0.52 μg/mL) compared with free doxorubicin (4.2 μg/mL) in 4T1 cells. Transcriptomic analysis suggested that MnO 2 @Man/DOX is associated with TNF signaling and apoptosis-related pathways, including extrinsic, intrinsic, and endoplasmic reticulum stress-mediated programs. In vivo evaluations in 4T1 tumor-bearing mice confirmed preferential tumor accumulation and superior growth inhibition with a high biosafety profile, including a hemolysis rate below 5% and minimal systemic toxicity. By integrating receptor-mediated targeting, redox homeostasis disruption, and chemosensitization, MnO 2 @Man/DOX offers a promising metabolism-oriented strategy for treating refractory TNBC.

Phytic acid-induced amorphous and porous transformation of MnO2@GO with enhanced capacitive performance for efficient uranium removal via capacitive deionization

Dingge Guo,Chunpei Yan,Yaxi Zhu,Bin Huang,Yong Qian,Tianxiang Jin

Addressing uranium-contaminated wastewater is crucial for environmental protection and public health. This study introduces an innovative and efficient capacitive deionization (CDI) method for extracting U(VI) from wastewater, utilizing a phytic acid (PA)-modified MnO 2 @GO aerogel as the electrode material. The incorporation of PA effectively disrupts the dense crystalline structure of MnO 2 , transforming it into an amorphous and porous framework. This structural change significantly enhances the specific surface area by approximately 450 % and creates more active sites for ion adsorption. Electrochemical characterization reveals that the PA modification improves capacitive performance by enhancing capacitance and reducing charge transfer resistance. Consequently, the PA-MnO 2 @GO aerogel exhibits excellent electrochemical properties and enhanced capacitive deionization performance, enabling rapid U(VI) removal under low voltage in a CDI cell configuration. This CDI cell achieves a maximum U(VI) removal capacity of 636 mg g −1 . Furthermore, the material demonstrates exceptional cycling stability and selectivity, maintaining approximately 83 % of its initial removal capacity after 20 cycles and exhibiting high adsorption efficiency even in the presence of interfering ions. This research presents an economical and effective novel approach for uranium extraction from radioactive wastewater using CDI technology, significantly contributing to the safe utilization of nuclear energy and environmental conservation.

BCNO Quantum Dots and MnO2 Nanosheets-Based Fluorescence and Digital Image Colorimetry Sensing for the Detection of 2, 4-Dichlorophenoxyacetic Acid

Junxia Wang,Ying Qin,Na Li,Yu He,Gongwu Song

2, 4-dichlorophenoxyacetic acid is a widely used pesticide, necessitating the development of a low-cost, reliable, and rapid detection method. In this work, BCNO quantum dots were synthesized by a one-step microwave method. A fluorescence “turn-off-on” probe for 2, 4-dichlorophenoxyacetic acid detection was developed using MnO 2 nanosheets due to their strong oxidation and broad absorption spectrum. MnO 2 nanosheets can quench the fluorescence of BCNO quantum dots through internal filter effect. Alkaline phosphatase hydrolyzes trisodium L-ascorbate-2-phosphate to produce ascorbic acid, which decomposes MnO 2 nanosheets, restoring the fluorescence of BCNO quantum dots. With the increase of alkaline phosphatase concentration, the ascorbic acid produced gradually increased; thus, the fluorescence intensity of BCNO quantum dots increased. 2, 4-dichlorophenoxyacetic acid inhibits alkaline phosphatase activity, preventing the decomposition of MnO 2 nanosheets and hindering fluorescence recovery. The probe exhibits a linear detection range of 1.0–40.0 mg/L and a detection limit of 0.067 mg/L. Color changes corresponding to 2, 4-dichlorophenoxyacetic acid concentration were analyzed using a smartphone application, establishing a digital image colorimetry method. The results demonstrated a good linear relationship within the range of 0–50 mg/L. The method was validated with real samples, demonstrating its utility in environmental and food safety monitoring.

Constructing hollow urchin-like Ag-K/Co3O4-MnO2 spheres with low Ag loading as a highly efficient catalyst for formaldehyde oxidation

Suhong Lu,Nan Sun,Liwei Gong,Chengyu Xu,Rongrong Guo,Yiyao Li,Shuo Wang,Jurong Liu,Yuzhen Fang,Guilong Liu

Hollow urchin-like Co 3 O 4 -MnO 2 spheres were synthesized via hydrothermal method, followed by co-impregnation with 3 wt% Ag and K (3, 5, 7 wt%). The optimized Ag-5 K/Co 3 O 4 -MnO 2 catalyst achieved complete HCHO conversion at 50 °C under 150 ppm HCHO, 21 vol% O 2 /N 2 , RH = 30 %, and WHSV = 36,000 mL g cat -1 h −1 . Systematic parameter studies (0 ∼ 80 % RH, 50 ∼ 350 ppm HCHO, 5 ∼ 21 vol% O 2 , 36,000 ∼ 144,000 mL g cat -1 h −1 WHSV) demonstrated the robust catalytic performance of Ag-5 K/Co 3 O 4 -MnO 2 across wide operational ranges. Remarkably, Ag-5 K/Co 3 O 4 -MnO 2 demonstrated excellent stability and nearly 100 % HCHO conversion over a continuous period of 100 h with the WHSV of 36,000 mL g cat -1 h −1 . The hollow architecture enhanced reactant diffusion, while interconnected nanorod arrays provided abundant Ag/K anchoring sites. Potassium doping increased Ag/Ag + , (Mn 2+ + Mn 3+ )/Mn 4+ and Co 3+ /Co 2+ ratios via strengthened Ag-support interactions, promoting oxygen activation. Mechanistic analysis indicated HCHO oxidation proceeds via HCHO → DOM → HCOO – + OH – →CO 2  + H 2 O route.

Design of a P–O–M (M = Mn, Zn) d-pπ Backbonding Electrolyte Additive for 40 Ah Electrolytic Zn–MnO2 Batteries

Mingyan Chuai,Hao Tong,Zimin Yang,Siting Deng,Mingqiang Wu,Jidan Xing,Guoliang Chai

An electrolytic Zn–MnO2 battery is highly valued due to its cost-effectiveness, environmental friendliness, and abundant resource availability. However, the battery’s performance is hindered by the slow kinetics at the poorly conductive MnO2 cathode and hydrogen evolution at the Zn anode. Here, a strategy of P–O–M (M = Mn, Zn) d-pπ backbonding design is proposed for phosphorus–oxygen electrolyte additives, which can be realized by tuning the atomic dipole moment-corrected Hirshfeld (ADCH) population charge of the P/O atom. The reversibility of d-pπ backbonding not only leads to the fast kinetics of Mn2+/Zn2+ at electrodes during both charge and discharge processes to suppress the competitive hydrogen evolution reaction but also enhances the electronic conductivity at the electrode–electrolyte interfaces to sustain the high areal capacity of batteries. Hydroxymethyl dimethyl phosphite (HPD) with d-pπ backbonding is a preferred additive with a suitable ADCH charge. The assembled electrolytic Zn–MnO2 (HPD) battery exhibits a high discharge capacity of 14.05 mAh cm–2 at an areal capacity of 15 mAh cm–2 and superior cycling stability over 1500 cycles. The Zn–MnO2 (HPD) soft-pack battery exhibits a discharge capacity of over 1.60 Ah at a discharge rate of 0.5 C and maintains a Coulombic efficiency of ∼80% over 100 cycles. Furthermore, the assembled 50 V 40 Ah commercial Zn–MnO2 (HPD) battery can drive an electric vehicle for 10 km. The ADCH charge regulation provides a feasible and effective method for developing high-performance aqueous batteries by achieving d-pπ backbonding.

Au/MnO2 Featuring Efficient Oxidase-mimic Activity at Neutral pH: Toward Portable Dual-modal Sensor for sulfide detection integrated with smartphone

Mi Li,Haorong Cheng,Yang Song,Xinxin Shi,Tongxiang Li,Yan Chen,Zhao Li,Lin Tian

The rapid detection of sulfide compounds including glutathione (GSH) and Na2S2O8 is of great interest because they play an essential role in the process of food processing. Here, we design a colorimetric/smartphone dual-mode sensing platform to detect GSH and Na2S2O8 simultaneously, depending on gold nanoparticle-decorated MnO2 nanosheets (Au/MnO2 NSs). Because of the distinct structure, Au/MnO2 possesses superior catalytic activity similar to native oxidase capable of catalysing the directly oxidation of 3,3’,5,5’-tetramethylbenzidine (TMB) under near-neutral pH conditions. Meanwhile, this catalytic reaction results in an obvious color change or the solution, which senses GSH and Na2S2O8 through redox and diazotization reactions. The designed sensor enables value-assisted and visual determination of GSH and Na2S2O8 with limits of detection of 2.3 nM. Based on the dual-modal sensing system, a smartphone-mediated “all-in-one” sensing platform is designed to realize the real-time and intelligent monitoring of GSH and Na2S2O8. This work provides a new avenue to construct multifunctional sensors to achieve the visualization and portable determination of sulfide compounds.

A B,N co-doped carbon nanotube array with anchored MnO2 nanosheets as a flexible cathode for aqueous zinc-ion batteries

Yan-bing YUAN,Zong-bin ZHAO,Hong-hui BI,Run-meng ZHANG,Xu-zhen WANG,Jie-shan QIU

For rechargeable aqueous zinc-ion batteries (ZIBs), the design of nanocomposites comprised of electrochemically active materials and carbon materials with novel structures has great promise in addressing the issue of electrical conductivity and structural stability in the electrode materials during electrochemical cycling. We report the production of a novel flexible electrode material, by anchoring MnO 2 nanosheets on a B,N co-doped carbon nanotube array (BNCNTs) grown on carbon cloth (BNCNTs@MnO 2 ), which was fabricated by in-situ pyrolysis and hydrothermal growth. The generated BNCNTs were strongly bonded to the surface of the carbon fibers in the carbon cloth which provides both excellent electron transport and ion diffusion, and improves the stability and durability of the cathode. Importantly, the BNCNTs offer more active sites for the hydrothermal growth of MnO 2 , ensuring a uniform distribution. Electrochemical tests show that BNCNTs@MnO 2 delivers a high specific capacity of 310.7 mAh g −1 at 0.1 A g −1 , along with excellent rate capability and outstanding cycling stability, with a 79.7% capacity retention after 8000 cycles at 3 A g −1 . Triple-signal lateral flow immunoassay for the detection of bovine casein using an M13 phage-displayed nanobody and MnO2 nanozyme conjugate

Wenyi Dong,Ajuan You,Yulou Qiu,Wanwan Liu,Jia Qiu,Yutao Zhu,Biao Zhang,Xianshu Fu,Zihong Ye,Xiaoping Yu

Cow's milk allergy is a significant global health concern, with bovine casein recognized as its predominant allergen. Rapid, sensitive, and accurate detection of bovine casein in food products is therefore essential. Here, a triple-signal lateral flow immunoassay (LFIA) for casein detection was developed by integrating an M13 phage-displayed nanobody (M13-Nb) with multifunctional MnO 2 nanosheets (MnO 2 NSs) as the signal probe. Anti-casein M13-Nb was selected from a naïve phage-displayed nanobody library through four rounds of biopanning. MnO 2 NSs exhibiting intrinsic colorimetric properties, oxidase-like catalytic activity, and photothermal capability were synthesized and conjugated with M13-Nb, enabling the MnO 2 NSs@M13-Nb-based LFIA to generate colorimetric, amplified colorimetric, and photothermal signals simultaneously. The visual limits of detection (vLOD) for these modes were 125, 15.63, and 31.25 ng/mL, respectively, with corresponding quantitative LODs of 60.97, 6.72, and 23.88 ng/mL. The assay showed high specificity and accuracy, with recovery rates of 87.92%–114.25% in spiked food samples. This triple-signal LFIA provides a rapid and reliable platform for casein detection in foods and highlights the potential of combining M13-Nb and MnO 2 NSs for on-site monitoring of food allergens.

One-step electrochemical exfoliation-deposition of MnO2 anchoring on graphite nanosheets as an effective host material for high-performance sulfur cathode

Chen Fang,Zhan Linqian,Zhang Songqing,Liang Zhongxin,Fan Xuliang,Ma Lin,Zhou Xiaosong

Lithium-sulfur (Li-S) battery with high energy density is a promising substitute for the conventional lithium-ion battery. However, the application of Li-S battery is limited by the low conductivity of sulfur and the diffusion of soluble intermediate polysulfide. To solve this issue, we have successfully prepared a graphite nanosheet/manganese dioxide (GN/MnO 2 ) composite as the sulfur host material by a one-step electrochemical exfoliation-deposition method. Combined with the high electrical conductivity of GN and the strong interaction between MnO 2 and polysulfide, the derived GN/MnO 2 /S cathode could exhibit superior electrochemical performance and high stability during the charge-discharge process. Even after 250 cycles, the GN/MnO 2 /S composite remained a stable discharge capacity of 493.5 mAh/g with the capacity retention of 68.5%.
