Antimicrobial peptides (AMPs), acknowledged as host defense peptides, constitute a category of predominant cationic peptides prevalent in diverse life forms. This study explored the antibacterial activity of α-conotoxin RgIA, and to enhance its stability and efficacy, D-amino acid substitution was employed, resulting in the synthesis of nine RgIA mutant analogs. Results revealed that several modified RgIA mutants displayed inhibitory efficacy against various pathogenic bacteria and fungi, includingCandida tropicalisandEscherichia coli. Mechanistic investigations elucidated that these polypeptides achieved antibacterial effects through the disruption of bacterial cell membranes. The study further assessed the designed peptides’ hemolytic activity, cytotoxicity, and safety. Mutants with antibacterial activity exhibited lower hemolytic activity and cytotoxicity, with Pep 8 demonstrating favorable safety in mice. RgIA mutants incorporating D-amino acids exhibited notable stability and adaptability, sustaining antibacterial properties across diverse environmental conditions. This research underscores the potential of the peptide to advance innovative oral antibiotics, offering a novel approach to address bacterial infections.

Bitter acids (BA) are main component of Humulus lupulus L. (hops). They are known for beer brewing and have various biological and pharmacological properties, especially the bone-protective effect confirmed by our previous in vivo study. Here we aimed to elucidate the anti-senior osteoporosis (SOP) effect of BA on osteoblasts and explore its underlying mechanism. In vitro SOP model was established by D-galactose (D-gal) injured osteoblasts, and the bone formation markers and apoptosis level were measured. mCherry-EGFP-LC3 adenovirus infection and autophagic markers including beclin1 and LC3 proteins were detected to investigate the autophagy level in osteoblasts. To further verify whether BA play the bone-protective role through regulating autophagy, the autophagy inhibitor 3-MA was used, and the cell proliferation, ALP activity, bone mineralization, apoptosis rate and SA-β-gal staining areas were measured. Finally, the protein expressions of AKT/mTOR signaling pathway were detected by Western blotting, and AKT agonist SC79 and mTOR agonist MHY1485 were used to further study the mechanism of BA on AKT/mTOR-mediated autophagy. The results showed that BA stimulated osteoblastic differentiation and inhibited apoptosis proteins Bcl-2/Bax in D-gal-treated osteoblasts. BA also increased the expression of autophagic markers beclin1 and LC3-II/LC3-I in D-gal-treated osteoblasts. mCherry-EGFP-LC3 autophagic double fluorescent adenovirus showed BA promoted the generation of autolysosomes and autophagosomes in D-gal-injured osteoblasts, indicating that BA might prevent osteoblastic bone loss through activating autophagy. Autophagy inhibitor 3-MA was used to further verify whether BA played the bone-protective role via regulating autophagy. The results revealed the promotion effects of BA on proliferation, ALP activity, and mineralized nodule formation in D-gal-injured osteoblasts were eliminated after autophagy blocking with 3-MA, and the inhibitory effects of BA on apoptosis rate and SA-β-gal staining areas were also eliminated. Moreover, BA reduced the phosphorylation levels of AKT, mTOR, p70S6K, and 4EBP in AKT/mTOR pathway, and the promotion of BA on the autophagic markers was blocked after the activation of AKT and mTOR by SC79 and MHY1485. In conclusion, it was the first time to demonstrate that BA improved cell activities and bone formation in aging osteoblasts, and revealed the mechanism of BA against SOP in osteoblasts was activating AKT/mTOR-mediated autophagy.

Organonitrogen chemicals are widely used and in great demand in the food, pharmaceutical and chemical industries. Hence, it is important to develop green and efficient synthesis paths to organonitrogen chemicals for their use as new resources. Herein, we propose the preparation of organonitrogen chemicals from chitin biomass monomer N -acetyl- d -glucosamine (GlcNAc) promoted by natural deep eutectic solvents (NADESs). The selective generation of 3-acetamido-5-(1′,2′-dihydroxyethyl) furan (Chromogen III) and 3-acetamido-5-acetylfuran (3A5AF) from GlcNAc was achieved by the design of NADESs components. GlcNAc reacts in binary NADES proline-glycerol (Pro-Gly) to generate 26 % of Chromogen III and 39 % of 3A5AF in ternary NADES proline-glycerol-lactic acid (Pro-Gly-LA). In addition, the intermolecular hydrogen bonds in NADESs were confirmed through 1 H diffusion-ordered spectroscopy (DOSY) NMR technology. And the results of 13 C NMR chemical shift titration indicated interaction between NADESs and the C3 and C6 position of GlcNAc. Based on the NMR experiments a mechanism is proposed.

A Raman-active boronate modified surface-enhanced Raman scattering (SERS) microporous array chip based on the enzymatic reaction was constructed for reliable, sensitive, and quantitative monitoring of D-Proline (D-Pro) and D-Alanine (D-Ala) in saliva. Initially, 3-mercaptophenylboronic acid (3-MPBA) was bonded to Au-coated Si nanocrown arrays (Au/SiNCA) via Au–S bonding. Following this, H 2 O 2 obtained from D-amino acid oxidase (DAAO)-specific catalyzed D-amino acids (D-AAs) further reduced 3-MPBA to 3-hydroxythiophenol (3-HTP) with a new Raman peak at 882 cm −1 . Meanwhile, the original characteristic peak at 998 cm −1 remained unchanged. Therefore, the I 882 /I 998 ratio increased with increasing content of D-AAs in the sample to be tested, allowing D-AAs to be quantitatively detected. The Au/SiNCA with large-area periodic crown structure prepared provided numerous, uniform “hot spots,” and the microporous array chip with 16 detection units was employed as the platform for SERS analysis, realizing high-throughput, high sensitivity, high specificity and high-reliability quantitative detection of D-AAs (D-Pro and D-Ala). The limits of detection (LOD) were down to 10.1 µM and 13.7 µM throughout the linear range of 20–500 µM. The good results of the saliva detection suggested that this SERS sensor could rapidly differentiate between early-stage gastric cancer patients and healthy individuals. Graphical Abstract

Volatile organic compounds (VOCs) emitted by plants provide essential insights into plant health, quality, and environmental stressors. Among these, D -limonene emissions fluctuate with agricultural stress and fruit quality and have emerged as a potential biomarker for monitoring plant condition. However, developing a portable, low-power, and user-friendly sensor for D -limonene detection in field conditions remains challenging. In this study, we report a novel colorimetric biosensor that employs a mild one-pot synthesis method to immobilize acetylcholinesterase (AChE) on a zinc-based metal–organic framework (AChE@Zn-MOF) and utilizes phenolphthalein as a pH indicator for highly sensitive D -limonene detection. The biosensor retains enzymatic activity comparable to free AChE while exhibiting enhanced stability across a range of environmental conditions. D -limonene inhibits AChE activity, disrupting acetylcholine hydrolysis and inducing a detectable pH shift, resulting in a visual color change. To enhance field practicality, the biosensor was integrated into commercial pH test strips coated with an agarose gel membrane to improve moisture retention and gas absorption. Coupled with smartphone-assisted image processing, the system enables detection of D -limonene at concentrations as low as 1 ppm and allows visualization of its release during mango post-harvest storage and citrus pest infestations. This approach establishes a foundation for portable, colorimetric smart sensing technologies in agriculture, facilitating real-time monitoring of VOCs like D -limonene in the field.

The healing of wounds in aging skin is a challenging issue that has not been thoroughly studied. Composite hydrogels made from natural polysaccharides have shown potential as dressings for various types of wounds. In this study, we prepared a polysaccharide-based composite hydrogel to provide a new strategy for treating aging skin wounds. First, chitosan (CS) was modified and Gastrodin (GAS) was grafted onto its main chain structure to obtain GAS/CS. Then GAS/CS was mixed with oxidized Gastrodia elata polysaccharides (GEP) to form a hydrogel using the principle of adsorbent reaction, and Puerarin (PUE), a natural plant ingredient, was embedded by gelatin microspheres and then loaded into the hydrogel. As a result, the composite hydrogel effectively reduced oxidative stress in tissue cells at the wound site and inhibited bacterial growth. It also reduced inflammation, promoted angiogenesis, and enhanced collagen deposition, which facilitated the repair of all aspects of the wound healing process in aging mouse skin. In short, this study explored the anti-aging effects of polysaccharides from Gastrodia elata , which might serve as both a treatment and an auxiliary for aging skin wounds. Additionally, the Gastrodia elata polysaccharide hydrogel was expected to be a promising drug carrier for skin repair.

Pyrolysis, a promising thermochemical conversion technique, can convert lignocellulosic biomass into various value-added products. Herein, the selective production of 1,4:3,6-dianhydro-α- d -glucopyranose (DGP) was achieved through fast pyrolysis of cellulose based on oxalic acid-assisted torrefaction (OAT) pretreatment. The mixture of cellulose and oxalic acid first underwent torrefaction, followed by fast pyrolysis to yield DGP. Simultaneously, the air atmosphere was considered to facilitate the formation of esterification intermediates, thereby promoting the formation of DGP. The effects of torrefaction temperature, oxalic acid amount, as well as pyrolysis temperature were carefully explored for DGP production. A yield of 9.6 wt% and a selectivity of 57.5 % for DGP were achieved from the micro-scale pyrolysis of cellulose under the optimized conditions. In lab-scale tests, the DGP yield reached 12.5 wt% from cellulose. The OAT-based pyrolysis was confirmed to be applicable to the waste cellulose-rich materials, with a DGP yield of 8.2 wt% from waste cotton and 6.0 wt% from spent paper in lab-scale experiments. Additionally, the OAT-based pyrolysis also showed the potential for poly-generation of DGP and formic acid at different stages.

The glucagon-like peptide-1 (GLP-1) analogue used to treat diabetes is an increasingly popular polypeptide protein therapeutic, commonly marketed as an injection. However, the effective oral administration of peptide drugs remains challenging because of their extremely low bioavailability. In recent years, a number of delivery systems that have been shown to be effective in improving the therapeutic efficacy of oral drugs. Herein, liraglutide was employed as a model drug and amphoteric sulfobetaine (SB12) was selected for the surface modification of poly (D, L-lactide-co-glycolic acid) (PLGA) nanoparticles (NPs) to obtain hydrophilic and electroneutral SB12-NPs. The functional SB12-NPs were first screened to identify the optimal prescription process and obtained from the final prescription were evaluated. The particle size, zeta potential , encapsulation efficiency (EE%) and drug-loading (DL%) of SB12-NPs were 87.25 ± 0.77 nm, −3.91 ± 1.88 mV, 77.45% ± 1.62%, and 10.46% ± 0.21%, respectively. The cellular uptake of Lira-SB12 NPs was significantly better than that of free liraglutide, and verified that it was transported mainly through endocytosis mediated by clathrin- and lipid raft–mediated. The trans-mucous permeability (2.86-fold) and intestinal permeability (1.79-fold) of SB12-NPs were significantly higher than those of free liraglutide. Single and multiple doses of SB12-NPs showed that the blood sugar level of diabetic mice could be lower to about 70% of the initial value. The SB12-NPs demonstrated a higher relative bioavailability of 9.59% compared with that of oral pure liraglutide (5.13%). Thus, SB12-modified PLGA NPs with hydrophilic and electroneutral surface properties can significantly improve mucus permeability and oral bioavailability, and have the potential to be applied for oral delivery of peptides and proteins .

Rare earth oxides (REOs) hold a pivotal position in enhancing electrocatalytic performance, yet the comprehension of their underlying mechanisms poses significant challenges. In this study, REOs loaded with a ternary alloy, abundant in oxygen vacancies, were synthesized through a one-pot reduction method and exhibited remarkable oxygen evolution reaction (OER) activity. The interaction at the interface between Fe 0.3 NiCo and CeO 2 induced substantial lattice distortion and modulated the oxygen vacancy concentration within the REOs. A unique electron transport channel, incorporating transition metal (3d) -oxygen vacancy (Vo) -rare earth (4 f) elements, was constructed at the interface, significantly boosting the OER activity of NiCo-CeO 2 . Notably, a current density of 10 mA cm −2 was achieved at a low overpotential of 151 mV. Comprehensive characterizations revealed that the interplay between the alloy and REOs regulated the oxygen vacancy concentration of the latter. In-situ attenuated total reflection infrared spectroscopy (ATR-IR) further confirmed that REOs facilitated the adsorption of O 2 /-OH, thereby enhancing OER activity. Theoretical calculations also indicated that oxygen vacancies effectively shifted the d and f band centers near the Fermi level, promoting d-f electron exchange and ensuring efficient electron transfer. This study offers a novel perspective for the development of efficient RE-based catalysts.

An electrolytic Zn–MnO2 battery is highly valued due to its cost-effectiveness, environmental friendliness, and abundant resource availability. However, the battery’s performance is hindered by the slow kinetics at the poorly conductive MnO2 cathode and hydrogen evolution at the Zn anode. Here, a strategy of P–O–M (M = Mn, Zn) d-pπ backbonding design is proposed for phosphorus–oxygen electrolyte additives, which can be realized by tuning the atomic dipole moment-corrected Hirshfeld (ADCH) population charge of the P/O atom. The reversibility of d-pπ backbonding not only leads to the fast kinetics of Mn2+/Zn2+ at electrodes during both charge and discharge processes to suppress the competitive hydrogen evolution reaction but also enhances the electronic conductivity at the electrode–electrolyte interfaces to sustain the high areal capacity of batteries. Hydroxymethyl dimethyl phosphite (HPD) with d-pπ backbonding is a preferred additive with a suitable ADCH charge. The assembled electrolytic Zn–MnO2 (HPD) battery exhibits a high discharge capacity of 14.05 mAh cm–2 at an areal capacity of 15 mAh cm–2 and superior cycling stability over 1500 cycles. The Zn–MnO2 (HPD) soft-pack battery exhibits a discharge capacity of over 1.60 Ah at a discharge rate of 0.5 C and maintains a Coulombic efficiency of ∼80% over 100 cycles. Furthermore, the assembled 50 V 40 Ah commercial Zn–MnO2 (HPD) battery can drive an electric vehicle for 10 km. The ADCH charge regulation provides a feasible and effective method for developing high-performance aqueous batteries by achieving d-pπ backbonding.

Bimetallic single-atom catalysts have garnered considerable interest in the field of the oxygen reduction reaction due to their unique electronic configurations and synergistic catalytic effects. However, precise modulation of d-orbital electron distribution at single-atom sites and comprehensive elucidation of the underlying catalytic mechanisms continue to present significant challenges. Herein, the FeCo(mlm)–N–C catalyst, integrating atomically dispersed Fe–Co dual-metal sites and Fe3C nanoparticles, was synthesized by using an encapsulation and ligand exchange strategy. Comprehensive analyses and theoretical simulations reveal that the incorporation of Fe3C nanoparticles induces significant d-orbital electron delocalization at the Fe active sites. This tailored electronic configuration effectively modulates Fe d–O p hybridization between the Fe active sites and adsorbed OH*. Consequently, it optimizes the occupancies of bonding and antibonding orbitals, thereby accelerating OH* desorption. This mechanism enables FeCo(mlm)–N–C to exhibit excellent catalytic performance and remarkable stability in both acidic and alkaline environments, while demonstrating superior activity in zinc-air batteries and proton exchange membrane fuel cells. This work not only presents a highly efficient non-noble metal electrocatalyst but also provides valuable insights into the rational development of advanced transition metal–nitrogen–carbon catalysts for energy-related applications.

Elevating the lattice oxygen p -band center (ε p ) is critical to trigger the lattice oxygen mechanism (LOM) for boosting oxygen evolution reaction (OER) activity in advanced electrocatalysts, but it also causes a reduced d -band center (ε d ) due to the coupled metal-oxygen d - p orbital, resulting in a sluggish regeneration of lattice oxygen. Herein, we surmount this challenge by heterojunction and defect-engineering to raise both ε d and ε p of CeO 2 NPs/D-Ni(OH) 2 . The formative built-in electric field and defect reduce the electronic density of Ni(OH) 2 , effectually decreasing the deprotonation energy and promoting its surface reconstruction into γ-NiOOH, the formative high valence Ni δ+ raises ε p to trigger LOM. Meanwhile, the ε d is upshifted by the high f -band of CeO 2 , accelerating the adsorption of oxygenous species to replenish the lattice oxygen vacancies. Consequently, the CeO 2 NPs/D-Ni(OH) 2 demonstrates exceptional OER activity, requiring only 169 mV to reach 10 mA cm −2 , and maintains impressive stability over 300 h at 1000 mA cm −2 in 1 M KOH. This work presents a novel strategy to balance lattice oxygen participation and regeneration by rising d - p orbital for inducing and maintaining LOM activity.
