Constructing J-aggregates of Cyanine Dye for NIR-II in vivo Dynamic Vascular Imaging and Long-Term Targeting of Tumors

Jiaqi Zhou,Hao Li,Hui Li,Jiayi Ding,Zhong Du,Jiabao Xiong,Hongyang Yao,Xueliang Zhang,Nuernisha Alifu,Biao Dong

Cyanine molecules with the second near-infrared (NIR-II) emission hold great potential for bioimaging owing to their great biocompatibility, but the scissor-like structure of these molecules poses a major bottleneck in obtaining efficient NIR-II fluorescence probes. Constructing J-aggregates represents a promising strategy for obtaining biomedical NIR-II emissive materials. However, achieving J-aggregates in cyanine dyes with large torsion angles between the heterocyclic rings poses a challenge. In this study, we introduced the guanidine of tumor molecular targeted peptide 1 (TMTP1) to increase steric hindrance of IR-783 and reduce the angle of IR-783 scissors. The near-coplanar structure of IR-783@peptide TMTP1 composite facilitates the formation of a novel J-aggregates (IR-783-LP-TMTP1) with super-stable effect for NIR-II in vivo dynamic vascular imaging and remarkable tumor targeting capbility. The stable emision wavelength and high spatial resolution of J-aggregates was demostrated for brain and ear vasculature bioimaging under 808 nm laser excitation. Additionally, J-aggregates exhibits robust tumor-targeting capability towards cervical tumors, indicating their potential in cervical cancer diagnosis. This work develops a molecular design strategy to construct bright NIR-II J-aggregates with super-stable and robust tumor-targeting properties and paving the way for improving bioimaging performance of similar molecules.

Hyaluronic Acid-Functionalized Gold Nanorods with pH/NIR Dual-Responsive Drug Release for Synergetic Targeted Photothermal Chemotherapy of Breast Cancer

Weijun Xu,Junmin Qian,Guanghui Hou,Aili Suo,Yaping Wang,Jinlei Wang,Tiantian Sun,Ming Yang,Xueli Wan,Yu Yao

Tumor-targeted delivery of photothermal agent and controlled release of concomitant chemotherapeutic drug are two key factors for combined photothermal chemotherapy. Herein, we developed a pH/near-infrared (NIR) dual-triggered drug release nanoplatform based on hyaluronic acid (HA)-functionalized gold nanorods (GNRs) for actively targeted synergetic photothermal chemotherapy of breast cancer. Targeting folate (FA), dopamine, and adipic acid dihydrazide triconjugated HA was first synthesized and used to decorate GNRs via Au–catechol bonds, and then an anticarcinogen doxorubicin (DOX) was conjugated onto HA moieties via an acid-labile hydrazone linkage, resulting in multifunctional nanoparticles GNRs-HA-FA-DOX. The nanoparticles exhibited excellent stability and had a pH and NIR dual-responsive drug release behavior. In vitro studies showed that the nanoparticles could be efficiently internalized into breast cancer MCF-7 cells and kill them under NIR irradiation in a synergistic fashion via inducing cell apoptosis. Pharmacokinetics and biodistribution studies in tumor-bearing mice indicated that the nanoparticles had a long blood circulation with a half-life of 2.4 h and exhibited a high accumulation of 11.3% in tumor site. The tumors of mice treated with combined chemotherapy and photothermal therapy were completely suppressed without obvious systemic toxicity after 20 d of treatment. These results demonstrated a great potential of GNRs-HA-FA-DOX nanoparticles for targeted synergistic therapy of breast cancer.

Glucose-triggered NIR-responsive photothermal antibacterial gelatin/dextran hydrogel simultaneously targeting the high glucose and infection microenvironment in diabetic wound

Yachao Yu,Hua Zhao,Jingmei Liu,Chenghao Li,Pengyuan Liu,Pei Cheng,Yongli Liu,Wenna Guo,Fangxia Guan,Minghao Yao

The treatment of diabetic wounds with bacterial infection is a major challenge in the medical field. Microenvironment-responsive hydrogel dressings have shown great advantages, and photothermal antibacterial therapy is a potential antimicrobial strategy to avoid the generation of resistant bacteria. In this work, a glucose-triggered near-infrared (NIR)-responsive photothermal antibacterial hydrogel was designed and named GOGD based on a cascade reaction of glucose oxidation and polyphenol polymerization. The GOGD hydrogel was composed of gelatin and oxidized dextran (Odex), and loaded with a natural plant polyphenol gallic acid (GA) and the dual-biological enzymes (glucose oxidase GOx and horseradish peroxidase HRP). In response to the high glucose environment, GOx in the hydrogel decomposed glucose to produce hydrogen peroxide, which further catalyzed GA polymerization with HRP to produce poly-GA possessing NIR photothermal capability, thus endow GOGD hydrogel with glucose-triggered NIR responsive photothermal antibacterial property simultaneously targeting the high glucose and infection microenvironment in diabetic wounds. Furthermore, the GOGD hydrogel demonstrated good biocompatibility and a strong ability to scavenge reactive oxygen species (ROS), thereby protecting cells from oxidative damage. In a mouse model, this hydrogel not only displayed excellent hemostatic properties but also significantly enhanced the healing of Staphylococcus aureus -infected diabetic wounds by regulating inflammation and promoting angiogenesis. Therefore, the proposed GOGD hydrogel provides a novel approach to diabetic wound treatment by utilizing its unique glucose-responsive mechanism combined with integrated NIR-photothermal bacterial inhibition. This well-designed material holds great promise for significantly improving the healing of infected diabetic wounds and offers new prospects for future advancements in wound therapy.

Lipoic Acid Capped Ag2S Quantum Dots for Mitochondria-Targeted NIR-II Fluorescence/Photoacoustic Imaging and Chemotherapy/Photothermal Treatment of Tumors

Yang Xuan,Zhou Su,Meng Guan,Yinan Zhao,Wenliang Zhang,Yating Gao,Xinyu Bian,Mingfan Zhang,Donghui Zhao,Shubiao Zhang

Subcellular targeted therapy has recently emerged as a significant approach in cancer treatment. Mitochondria, as crucial organelles that not only provide cellular energy but also play roles in cell differentiation, signal transmission, reactive oxygen species production, apoptosis, and calcium homeostasis, are considered potential targets for cancer therapy. Silver sulfide quantum dots (Ag2S QDs) have demonstrated exceptional capabilities in near-infrared fluorescence and photoacoustic imaging as well as photothermal treatment. Therefore, the development of Ag2S QDs with mitochondrial-targeting ability is of particular significance. Herein, we developed a simple method for synthesizing aqueous-phase Ag2S QDs using lipoic acid, an enzymatic substrate involved in cellular respiration, as a ligand and mitochondrial-targeting molecule. These Ag2S QDs, which exhibit excellent water solubility and biocompatibility, can be produced in a single step and act as a mitochondrial-targeted theranostics. By adsorbing doxorubicin (DOX), we successfully created multifunctional nanoprobes called Ag2S-DOX. In vitro and in vivo experiments demonstrated that Ag2S-DOX was suitable for fluorescence and photoacoustic dual-mode imaging as well as synergistic mitochondrial-targeted chemotherapy and photothermal treatment, offering a promising strategy for tumor therapy.

Nanocarrier-based drug delivery system with dual targeting and NIR/pH response for synergistic treatment of oral squamous cell carcinoma

Ran Li,Chaoqiong Wan,Yanwei Li,Xiaofeng Jiao,Tiantian Liu,Yixuan Gu,Ruifang Gao,Jun Liu,Bing Li

Oral squamous cell carcinoma (OSCC) is highly heterogeneous and aggressive, but therapies based on single-targeted nanoparticles frequently address these tumors as a single illness. To achieve more efficient drug transport, it is crucial to develop nanodrug-carrying systems that simultaneously target two or more cancer biomarkers. In addition, combining chemotherapy with near-infrared (NIR) light-mediated thermotherapy allows the thermal ablation of local malignancies via photothermal therapy (PTT), and triggers drug release to improve chemosensitivity. Thus, a novel dual-targeted nano-loading system, DOX@GO-HA-HN-1 (GHHD), was created for synergistic chemotherapy and PTT by the co-modification of carboxylated graphene oxide (GO) with hyaluronic acid (HA) and HN-1 peptide and loading with the anticancer drug doxorubicin (DOX). Targeted delivery using GHHD was shown to be superior to single-targeted nanoparticle delivery. NIR radiation will encourage the absorption of GHHD by tumor cells and cause the site-specific release of DOX in conjunction with the acidic microenvironment of the tumor. In addition, chemo-photothermal combination therapy for cancer treatment was realized by causing cell apoptosis under the irradiation of 808-nm laser. In summary, the application of GHHD to chemotherapy combined with photothermal therapy for OSCC is shown to have important potential as a means of combatting the low accumulation of single chemotherapeutic agents in tumors and drug resistance generated by single therapeutic means, enhancing therapeutic efficacy.

NIR-I Light-Activated Antibiotic Delivery & PDT via TiO2/Graphene Metastructure for Enhanced Antibacterial Activity and Osseointegration of Ti Implants

Minggang Yang,Yufan Zhang,Zhenhao Hou,Jianbo Wu,Fuwei Liu,Jun Wu,Kelvin W.K. Yeung,Wenhao Qian,Xuanyong Liu,Liang Kong,Yunpeng Li,Jiajun Qiu,Guocheng Wang

Antibiotic therapy is a key strategy for treating infections associated with orthopedic implants, yet its limited effectiveness and potential to disrupt bone healing highlight the need for innovative approaches. Herein, a TiO 2 -Graphene (TiO 2 -G) metastructure is developed on Ti implant surface using a hydrothermal method coupled with plasma-enhanced chemical vapor deposition (PECVD), showing strong near-infrared light (NIR) absorption. The antibiotic doxycycline (DOX) is successfully loaded onto TiO 2 -G and exhibited enhanced NIR release. The promising antibacterial efficacy is proven by both in vitro and in vivo tests with NIR irradiation for 5 min, which is ascribed to the synergistic photocatalytic activity of TiO 2 -G and NIR-responsive release of DOX. Interestingly, after 5 min of NIR irradiation, the TiO 2 -G metasurface neutralized the immediate negative effects of photodynamics and even upregulated the expression of osteogenic genes (osteocalcin (OPN), osteopontin (BSP), and bone sialoprotein (OCN)), with particularly enhanced effects observed on day 14. Moreover, the sustained release function of TiO 2 -G significantly mitigated the cytotoxicity of free antibiotics with antibacterial capabilities comparable to those of TiO 2 -G/DOX under NIR irradiation for 5 min. Consequently, the in vivo studies proved that the TiO 2 -G metastructure enhanced the osteointegration of the implant even in the absence of infection when loaded with DOX.

Block cationic copolymer/quaternary ammonium chitosan-based composite antibacterial hydrogel dressings with NIR photothermal effects for bacteria-infected wound healing

Qian Wang,Ze Zhang,Jiang Yin,Lingyi Shen,Lingli Zhu,Carl Redshaw,Qilong Zhang

Wound infections caused by pathogenic bacteria can cause delayed wound healing and even threaten the life of patients. Drug resistance and effectiveness with regard to traditional wound dressings have become urgent issues which need to be addressed. The antibacterial effect of hydrogels, as novel wound dressings, is through contact-active antibacterial was limited by the contact area and time. In this paper, MXene (a 2D carbonitride) has been employed as the substrate and via the in-situ reduction of silver nanoparticles and the polymerization of dopamine on its surface the photothermal agent MXene-Ag-PDA was obtained. This was then introduced into the hydrogel matrix via the block cationic polymer (AM-VBIMBr-AAPBA), oxidized sodium alginate (OSA) and quaternary ammonium chitosan (HACC). Multifunctional composite hydrogels (PNVAO/M-A-P) possessing photo-thermal effects and inherent antibacterial activity were obtained after crosslinking, and were employed as medical materials to promote the healing of infected wounds. The cationic groups contained in the PNVAO/M-A-P were capable of capturing and killing bacteria by electrostatic interactions, and produced local heat killing adsorption and residual bacteria in the surrounding solution under NIR irradiation, thereby shortening the antibacterial cycle and improving the sterilization efficiency. Meanwhile, the introduction of glycerin in the hydrogel improved its anti-freezing and anti-drying properties, enabling the hydrogel to function stably under extreme environments. PNVAO/M-A-P composite hydrogels based on chemical and dynamic crosslinking exhibited the desired mechanical strength, and via their self-healing ability can adapt to dynamic wounds. The hydrogel dressing showed excellent anti-freezing and anti-drying properties, high bovine serum albumin (BSA) adsorption capacity (308.17 mg·g −1 ), good biocompatibility (the cell viability exceeded 100 % after 48 h of incubation), antioxidant properties and excellent antibacterial activity (the sterilization rate can reach 99.9 %). Furthermore, the hydrogel was applied to infected wound treatment and was able to control the wound infection, reduce pro-inflammatory factor production, promote collagen deposition and neovascularization formation, and thus promote wound healing. Therefore, multifunctional composite hydrogels with synergistic antibacterial effects can be used as an novel dressing for the treatment of bacterial infected wounds.

Broadband photoluminescence toward the NIR II region and stable green ceramic pigments based on a novel NaBaScSi2O7:xCr silicate phosphor

Xuejiao Wang,Sihan Yang,Feng Jiang,Jiantong Wang,Changshuai Gong,Ji-Guang Li

Cr3+-activated near-infrared (NIR) phosphors are the most promising materials for commercialization. However, the emission of most Cr3+-activated phosphors is typically located in the NIR-I region and achieving luminescence towards the NIR-II region (>900 nm) via Cr3+ doping remains challenging. This study reports a novel Cr3+-activated NaBaScSi2O7 phosphor that exhibits broadband NIR emission centred at 974 nm with a full width at half maximum of 140 nm under 493 nm blue light excitation. The emission wavelength exceeds that of most Cr3+-activated NIR phosphors. The underlying mechanism for the encouraging phenomenon is attributed to the strong charge polarisation arising from the intrinsic local coordination environment of the Cr3+ ions, as confirmed by charge differential density (CDD) and the electron localization function (ELF) analyses. The photoluminescence properties of phosphors were systematically investigated, and the performance of the fabricated NIR pc-LED device was evaluated for night vision and non-invasive imaging. Notably, the NaBaScSi2O7 phosphor displayed a bright green colour with excellent colour properties and its potential as a stable green ceramic pigment was explored.

3D-printed CoSi/PCL composite scaffold with NIR-II photothermal ability and enhanced adipogenic activity for breast reconstruction after mastectomy

Jupei Zhang,Hangbin Xia,Xuerui Zhou,Zhaoxu Meng,Qishu Jin,Dongmin Chen,Xiaojuan Xia,Yiren Jiao,Jiang Chang,Zhihong Dong,Zhen Zeng,Hongshi Ma,Chen Yang

Breast reconstruction after mastectomy aims to restore breast appearance and function. Current scaffolds often fail to meet the complex clinical demands of post-mastectomy breast reconstruction, which include personalized shaping, prevention of tumor recurrence and metastasis, and promotion of adipose tissue regeneration. This study aims to address these challenges by combining a readily processable biopolymer, polycaprolactone (PCL), with a multifunctional bioactive ceramic, cobalt orthosilicate (Co 2 SiO 4 , CoSi) to form a multi-functional CoSi/PCL composite scaffold through 3D printing technology. The scaffold exhibits a controllable shape with designed macroporous architecture. Its photothermal performances are well presented in both near-infrared (NIR) I and NIR-II regions, enabling effective tumor ablation with minimal side effects. Additionally, the release of bioactive silicate and cobalt ions promotes adipogenesis and angiogenesis, thereby enhancing the integration of the scaffold with surrounding tissues. This approach offers a promising strategy for combining tumor therapy with breast reconstruction, providing a clinically relevant solution for post-mastectomy patients.

3D printed gelatin/PTMC core/shell scaffolds with NIR laser-tuned drug/biomolecule release for cancer therapy and uterine regeneration

Shangsi Chen,Jiahui Lai,Jizhuo Chen,Liwu Zheng,Min Wang

Surgical resection is an efficient treatment for cancerous tissues and uterine fibroids in the women uterus. However, the insufficiency of clinical interventions could result in tumor recurrence, and the defective tissues remained would cause intrauterine adhesions (IUAs) and further affect reproduction capacity. In this study, 3D printed hydrogel/poly( l -lactide- co -trimethylene carbonate) (PLLA-co-TMC, “PTMC” in short) core/shell scaffolds with NIR-tuned doxorubicin hydrochloride (DOX) and estradiol (E2) dual release were designed and fabricated for cancer therapy and uterine regeneration. Gelatin (Gel) and DOX were homogeneously mixed and then 3D printed to form Gel-DOX scaffolds. Gel-DOX scaffolds were then immersed in PTMC-PDA@E2 solution to fabricate Gel-DOX/PTMC-PDA@E2 core/shell scaffolds. Consequently, Gel-DOX/PTMC-PDA@E2 scaffolds could release DOX and E2 in a chronological manner, firstly delivering DOX assisted by phototherapy (PTT) to effectively kill Hela cells and then sustainably releasing E2 to promote uterine tissue regeneration. In vitro experiments showed that core/shell scaffolds exhibited excellent anticancer efficiency through the synergy of DOX release and hyperthermia ablation. Moreover, E2 could be sustainably released for over 28 days in vitro to promote the proliferation of bone marrow-derived mesenchymal stem cells (BMSCs). The novel Gel-DOX/PTMC-PDA@E2 core/shell scaffolds have therefore exhibited potential promise for the treatment of cancer therapy and uterine regeneration.

Xinxin Liu,Zhengfei Hong,Rongrong Li,Shuxian Hou,Xuechao Shi,Guoqiang Yang,Yuxuan Wang,Yimei Wang,Zhiru Tao,Jun Ge,Fei Ge,Yiyan Song,Jun Wang

Wound infections caused by surgery, chronic ulcers, or trauma have been a challenge for clinicians, especially multiple resistant bacteria induced by the misuse of antibiotics, which has increased the risk of bacterial infection. It is very urgent to design and develop antimicrobial materials with potential for clinical application against drug-resistant bacterial infections. In this study, a photothermal montmorillonite-chitosan hydrogel system (MNT-CS Gel) was fabricated as a photothermal antimicrobial dressing. This study found for the first time that montmorillonite (MNT) has good photothermal properties and photothermal conversion efficiency under near-infrared (NIR) light, and on this basis, its application research was carried out. The MNT possesses broad-spectrum antibacterial activity with significant inhibitory effects against E. coli and S. aureus, excellent photothermal stability under NIR light. And all the performances were improved after combining with CS-Gel, especially in bacterial adsorption capacity and photothermal performance. Notably, MNT-CS Gel was used as a photothermal antibacterial agent for the first time, and the antibacterial effects in vitro and their mechanisms have been systematically investigated. It was found that the MNT-CS Gel had significant inhibitory and destructive effects on bacteria in an in vitro pigskin model. The MNT-CS Gel could adsorb the bacteria strongly, inhibit bacterial biofilm formation, induced the generation of ROS, destroy the structure of bacteria, and cause the leakage of nucleic acids. The MNT-CS Gel demonstrated good photothermal antimicrobial properties, suggesting that the MNT-CS Gel has potential for clinical hygiene applications.
