Metal–organic frameworks (MOFs) hold great promise as drug carriers and therapeutic agents in biomedical applications. However, the limited stability of MOF-based carriers in biological environments has garnered increasing attention. Achieving sustained drug release and stability remains a challenge due to the intrinsic instability of MOFs in biological environments. Herein, we report a general postsynthetic modification (PSM) strategy utilizing polyvalent DNA (polyDNA) nanostructures as a universal binder on three representative MOFs. Our findings reveal that the high local density of DNA patches on the MOF surface effectively inhibits competitive ligand-induced linker release and structural collapse. Mechanistic investigations indicate that the cooperative adsorption of adjacent DNA oligos with flexible conformations contributes to enhanced biointerface stability. Importantly, our study demonstrates that polyDNA-modified MOFs exhibit superior stability and enhanced cellular uptake, leading to sustained drug release and improved cytotoxicity against cancer cells. This facile and versatile postsynthesis modification strategy has the potential to advance the application of MOFs in nanomedicine and other fields requiring high structural stability.

Circulating tumor cells (CTCs), though rare and highly heterogeneous, are closely associated with tumor initiation, progression, and prognosis. Their low abundance, structural fragility, phenotypic diversity, and size variability make efficient enrichment particularly challenging. To address these limitations, we developed a novel CTC isolation platform that combines tetrahedral DNA nanostructure-based trivalent aptamer magnetic nanospheres (TDF-Cocktail-MNP) with a magnetic microfluidic separation device. This system enhances the capture of viable, phenotypically diverse CTCs and supports reliable downstream analysis. The TDF structure presents three aptamers: EpCAM targets epithelial CTCs, Vimentin captures mesenchymal types, and EGFR binds to cells overexpressing EGFR—a key marker in triple-negative breast cancer. This multi-target approach enables efficient isolation of heterogeneous CTC populations from whole blood samples. Captured CTCs are enriched using the IsoFlux System, which reduces mechanical stress and preserves cell viability by avoiding repeated magnetic manipulation. After nuclease-mediated removal of the DNA scaffold, cells are classified and quantified through immunocytochemistry. Validated with both spiked and clinical breast cancer samples, this platform shows strong potential for liquid biopsy applications, offering robust CTC detection and valuable insights for personalized cancer diagnostics and treatment.

Flexible mechanical sensors offer extensive application prospects in the field of smart wearables. However, developing highly sensitive, flexible mechanical sensors that can simultaneously detect strain and pressure remains a significant challenge. Herein, we present a flexible mechanical sensor based on AgNPs/MWCNTsCOOH/PDA/PU/PVB nanofiber-covered yarn (AMPPPNY) featuring a DNA-like double-helix wrinkled structure. The sensor is fabricated by electrospraying polyvinyl butyral (PVB) onto a pre-stretched double-helix elastic yarn, followed by electrospinning a polyurethane (PU) nanofiber membrane and inducing the self-polymerization of dopamine (DA) to create an adhesive layer. Then, one-dimensional carboxylated multi-walled carbon nanotubes (MWCNTs-COOH) and zero-dimensional silver nanoparticles (AgNPs) are dispersed onto the structure, synergistically forming a stable conductive network for efficient signal transmission. The integration of conductive fillers with different dimensionalities and DNA-like double-helix wrinkled structure endows the sensor with high strain sensitivity (gauge factor of 11,977) in the strain range of 0–310% and high pressure sensitivity (0.475 kPa −1 ) in the pressure range of 0–2 kPa. Moreover, the fabricated sensor exhibits rapid response and recovery times (130 ms/135 ms) and outstanding cyclic stability (over 10,000 cycles of both strain and pressure). Next, the fibrous sensor is weaved into a large-area fabric, and the developed smart textiles demonstrate impressive performance in detecting both subtle and large human movements. The proposed sensor is a promising candidate for flexible wearable applications. Graphical

Breast tumor–derived exosomes promise to guide diagnosis and therapy. But the small size and low mass of exosomes have higher requirements for target binding and signal amplification. Although hybridization chain reaction (HCR) has been extensively utilized for the sensitive detection of an enormous range of biomolecules, including the exosomes, achieving sufficient amplification efficiency remains challenging. To address this limitation, more and more researchers integrate the reaction system with functional materials of DNA. Inspired by this, we herein report for the first time the cubic DNA nanostructure enhanced hybridization chain reaction (CDN-HCR) to detect breast tumor–derived exosomes. Unlike other nanostructure-assisted HCR approaches, the four cholesterol anchors on the CDN enhance the binding affinity of signal probes for exosomes, while the four triggers combined with the supporting CDN further improve the reaction efficiency of HCR. Furthermore, incorporating an electrochemical detection system, the assay platform enables enzyme-free and label-free detection of exosomes. The quantitative detection of exosomes is easily achieved with a linear range of 5.5 × 10 2 to 5.5 × 10 7 particles/μL and a minimal detectable concentration of 51 particles/μL. Finally, the human serum study proves the feasibility of this technique in clinic applications. Graphical

Background DNA and RNA virus detections commonly rely on qPCR (quantitative polymerase chain reaction) -based methods that need dyes or Taqman probes as signaling probes and expertized instruments. Isothermal amplification such as linear rolling-circle amplification (LRCA) has attracted large interest in its role to act as a supplement to the traditional qPCR. However, the current LRCA-based signaling probes suffered from long time-consumption and complex procedures for nucleic acid detections. Novel systems are urgently needed to integrate the simplicity of LRCA with rapid signal amplification of label-free probes to spread the advantages of isothermal amplification for virus detections. Results A primer-independent LRCA system utilizing AgNCs/DNA as signaling probes was developed for rapid virus detection. LRCA was optimized into a one-step reaction by integrating target-triggered ligation reaction of padlock probes (PlPs) with DNA amplification. After an easy H 2 O 2 treatment to minimize dithiothreitol's interference, LRCA product with tandem repeats of G-rich sequence was hybridized with AgNCs/DNA, promptly amplifying fluorescence signals through the G-proximity enhancement effect. The system precisely discriminated single-base mismatch at ligation junctions of PlPs. The formation of 3′-hairpin structure in PlPs was found to be prioritized over 5′-exposed linear sequences to maximize the fluorescence signals. It requires only 1 μL sample and costs 40 min at 25–30 °C, facilitating a sensitive and broad linear range of detection for E (10 -9 -10 μM) gene in RNA virus (SARS-CoV-2) and VP2/3 gene (10 −9 - 1 μM) in DNA virus (BK human polyomavirus, BKV), and successful application in urine samples. Significance AgNCs/DNA lighting up LRCA products with rapid signal amplifications alongside the extension of single strand DNA enables the direct use of LRCA for nucleic acids detections. The modular design of PlPs, universal signal probes, near-room temperature reaction conditions, small sample requirements and rapid reaction make it adapted for various DNA detections, and give it great promise for future point-of-care tests.

With the continuous in-depth study of the interaction mechanism between viruses and hosts, the virus has become a promising tool in cancer treatment. In fact, many oncolytic viruses with selectivity and effectiveness have been used in cancer therapy. Human enterovirus is one of the most convenient sources to generate oncolytic viruses, however, the high seroprevalence of some enteroviruses limits its application which urges to exploit more oncolytic enteroviruses. In this study, coxsackievirus B5/Faulkner (CV-B5/F) was screened for its potential oncolytic effect against non-small cell lung cancers (NSCLCs) through inducing apoptosis and autophagy. For refractory NSCLCs, DNA-dependent protein kinase (DNA-PK) or ataxia telangiectasia mutated protein (ATM) inhibitors can synergize with CV-B5/F to promote refractory cell death. Here, we showed that viral infection triggered endoplasmic reticulum (ER) stress-related pro-apoptosis and autophagy signals, whereas repair for double-stranded DNA breaks (DSBs) contributed to cell survival which can be antagonized by inhibitor-induced cell death, manifesting exacerbated DSBs, apoptosis, and autophagy. Mechanistically, PERK pathway was activated by the combination of CV-B5/F and inhibitor, and the irreversible ER stress-induced exacerbated cell death. Furthermore, the degradation of activated STING by ERphagy promoted viral replication. Meanwhile, no treatment-related deaths due to CV-B5/F and/or inhibitors occurred. Conclusively, our study identifies an oncolytic CV-B5/F and the synergistic effects of inhibitors of DNA-PK or ATM, which is a potential therapy for NSCLCs.

Herein, we propose an all-DNA self-assembled G-quadruplex/hemin DNAzyme nanosphere (GHN) and apply it to construct an electrochemical biosensor for sensitive detection of kanamycin (KAN). Unlike conventional metal-core-dependent biosensors, the GHN eliminates inorganic components, offering superior biocompatibility, simplified synthesis, and reduced environmental hazards. High loading of G-rich sequences was achieved by performing multiple branches of the hybridization chain reaction on self-assembled DNA spherical core. In addition to exhibiting excellent catalytic performance, the GHN inherently provides a strong current response, facilitating highly sensitive detection of target analytes. Through a one-step displacement reaction triggered by the target, the GHN is captured on the electrode surface, generating a measurable current signal. Under optimal experimental conditions, the electrochemical signal of the proposed sensor showed a good linear relationship with the logarithm of the KAN concentration over the range of 0.1 pM to 1 μM, with a detection limit of 71 fM (S/N = 3). For practical applications, the sensor achieved 93.98% to 102.0% recovery in spiked milk samples, demonstrating its reliability in complex matrices. The target-independent synthesis of GHN allows facile reconfiguration by altering aptamer sequences, enabling universal detection of environmental pollutants and biomarkers. This platform offers a new approach for early monitoring of antibiotic residues, which is expected to provide valuable references for the formulation of relevant environmental protection policies and the safeguarding of human health.

Spatially resolved imaging of intracellular pH during ischemic stroke (IS) is critical for demarcating salvageable penumbral tissue and tracking lesion progression; however, engineering IS-specific pH sensors remains challenging. Herein, we engineered an apurinic/apyrimidinic endonuclease 1 (APE1)-activatable ratiometric DNA nanosensor (LPF) by encapsulating a dual-strand DNA probe (PF) within liposomes. The PF probe operates through a dual-strand molecular switch: the i-motif in the cytosine-rich F-strand undergoes pH-dependent folding, activating Cy3/Cy5 FRET for ratiometric pH sensing; simultaneously, the AP site on the P-strand blocks pH-sensing activity until cleaved by cytoplasmic APE1 upregulated in IS lesions, thereby enabling IS-specific activation. Liposomes enhance delivery to ischemic cells. In vitro studies identified 30-nt and 18-nt P-strand as optimal for protection and activation, respectively, with PF exhibiting high APE1 selectivity under acidic conditions. Confocal imaging demonstrated LPF’s capability to specifically monitor pH changes in oxygen-glucose-deprived (OGD) cells, requiring cytoplasmic APE1 upregulation for activation. In MCAO mice, LPF achieved enzyme-mediated IS-specific activation and dynamically tracked pH gradients. This work establishes the first APE1-activated ratiometric nanosensor for spatially precise pH imaging in IS, providing a platform for stroke diagnosis and personalized intervention.

The lack of selective tumor targeting and the high toxicity of conventional chemotherapy treatments remain major challenges in cancer therapy. Here, we develop a self-controlled DNA nanostructure-CRISPR-12a system, a triple-locked cascade tumor therapy nanocapsule (Tatna), for efficient and targeted tumor treatment. Tatna integrates structural DNA tetrahedrons (DTs) with high drug-loading capacity, Cas12a/crRNA ribonucleoprotein (Cas12a RNP), and doxorubicin (DOX) to enable multisite response for precise drug delivery and augmented tumor treatment. By incorporation of a nucleolin-targeting aptamer, Tatna achieves selective targeting and efficient tumor cell internalization. Encapsulation in pH-responsive poly l-lactic-co-glycolic acid (PLGA) nanocapsule ensures stable circulation and controlled release of both DOX and Cas12a until tumor-specific activation in the acidic microenvironment. The Cas12a RNP, triggered by APE1 mRNA overexpression in tumor cells, induces trans-cleavage of DTs, releasing DOX and Cas12a to transport into the nucleus and induce enhanced cell apoptosis. This self-regulating and multifunctional approach enhances the efficacy of chemotherapy while reducing off-target effects. Tatna’s programmable, tumor-specific delivery system represents a powerful strategy for advancing precision medicine and personalized cancer treatment.

Low-mass soluble β-amyloid oligomers (LSAβO) are critical Alzheimer’s disease (AD) biomarkers with significant diagnostic and therapeutic potential. However, their application in early screening and detection is limited by the reliance on complex analytical instruments and procedures. To address this, we developed a visual sensing platform for LSAβO detection using a functionalized pNIPAM-DNA hydrogel. Exploiting the temperature-responsive nature of pNIPAM, the hydrogel selectively incorporates and enriches LSAβO from solution via temperature-induced expansion and contraction. LSAβO binding to aptamers on the hydrogel triggers the formation of G-quadruplex DNAzymes, which catalyze the oxidation of 3,3′,5,5′-tetramethylbenzidine to generate a visible colorimetric signal. The hydrogel’s small pore size further enhances selectivity by excluding larger oligomers during real sample analysis. This sensor exhibits a linear detection range of 0.1–7.5 nM for LSAβO and a detection limit of 50 pM. Combining LSAβO enrichment and size exclusion, this functionalized pNIPAM-DNA hydrogel platform provides a cost-effective, highly sensitive, selective, and high-throughput approach for preliminary LSAβO screening and detection.

Rapid and precise DNA barcode-based identification of biological species holds significant potential for pharmaceutical authentication and biomedical diagnostics. Herein, we present a polymerase chain reaction (PCR)–surface-enhanced Raman scattering (SERS) platform that integrates SERS tags for ultrasensitive and fast authentication of Hippocampus trimaculatus , a high-value traditional Chinese medicine (TCM). The SERS tags are composed of gold nanostars, near-infrared cyanine7 Raman reporters and carboxylated polystyrene shells, which achieve single-particle detection sensitivity under 780 nm irradiation. The tags also show excellent colloidal and SERS stability under physiologically relevant conditions (e.g., phosphate buffer saline, serum, 1 mM NaCl, and pH 1–12), with signal variations less than 5 %. The carboxylated polystyrene shells enable efficient DNA functionalization. Leveraging these advancements, the PCR-SERS assay detects genomic DNA (gDNA) at concentrations as low as 10 copies/μL within 20 thermal cycles, with remarkable specificity for Hippocampus trimaculatus over four common adulterant species. Notably, the method reduces amplification requirements to 5 thermal cycles (detection limit of 10 6 copies/μL) while completing the entire workflow in less than 30 min (conventional qPCR, 20–30 cycles, 1–2 h). Beyond TCM verification, this PCR-SERS platform holds broad applicability for rapid nucleic acid detection in fields ranging from environmental eDNA monitoring to point-of-care diagnostics.

MicroRNAs (miRNAs) are closely associated with the development and progression of cancer, making the development of highly sensitive and rapid miRNA detection strategies a significant challenge. Herein, we developed an electrochemical sensor based on DNA triangular prism (TP) and hybridization chain reaction (HCR) for the ultra-sensitive, rapid, and specific analysis of miRNA. The TP was modified on the electrode surface to obtain high density of recognition probes. After hybridization of the target miRNA with the recognition probes on the TP, HCR was further activated. Numerous biotin-carrying HCR products were generated on the electrode surface. Then the biotin on the HCR products efficiently bound to streptavidin-horseradish peroxidase (SA-HRP), and under the catalysis of a large amount of HRP, TMB and H 2 O 2 rapidly underwent redox reactions, producing the enhanced electrical signal, thus achieving high-sensitivity analysis of miRNA. The developed strategy has a detection limit of 2.21 aM and demonstrates high specificity with the ability to distinguish single-base-pair differences in miRNA. The strategy also exhibits good reproducibility, stability, and practical application potential. We believe that this developed strategy holds promise as an effective tool for miRNA analysis and is expected to play a crucial role in disease diagnosis and new drug development.
