Introduction Quality evaluation of Huang-qin is significant to ensure its clinical efficacy. Objective This study aims to establish an accurate, rapid and comprehensive Huang-qin quality evaluation method to overcome the time-consuming and laborious shortcomings of traditional herbal medicine quality assessment methods. Methods The contents of baicalin, baicalein and scutellarin in Huang-qin from five different origins were analyzed by FT-IR and NIR spectra combined with multivariate data technology. The quality of Huang-qin from different origins was evaluated by TOPSIS and consistency analysis based on the content of three active ingredients. The correlation between ecological factors and the accumulation of active ingredients was explored. Results Satisfactory prediction results of PLS models were obtained. Relatively, the model based on FT-IR combined with the PLS regression method has higher R 2 and smaller RMSE than the NIR combined with the PLS method. TOPSIS and consistency analysis results showed that the quality of Huang-qin from different geographical origins was significantly different. The results showed that the quality of Huang-qin produced in Shanxi Province was the best among the five origins studied. The results also found that the quality of Huang-qin in different growing areas of the same origin was not completely consistent. The correlation study showed that altitude, sunshine duration and rainfall were the main factors that caused the quality difference of medicinal materials in different geographical origins. Conclusion This study provides a reference for the rapid quantitative analysis of the active components of herbal medicine and the quality evaluation of them.

Near-infrared II (NIR-II) photoacoustic (PA) imaging offers enhanced performance through reduced photon scattering, minimized background interference, and improved resolution at greater depths, thereby driving urgent demand for NIR-II PA nanoprobes in translational biomedicine. In this study, we developed a smart stannous sulfide@cuprous oxide (SnS@Cu 2 O) nanoprobe for colorectal cancer PA imaging within NIR-II window. Under the existence of HS − , the SnS@Cu 2 O nanoprobe exhibits a broad absorption peak within the 1000–1300 nm range, enabling concentration-dependent NIR-II PA signal amplification in colorectal cancer tissues with overexpression of endogenous H 2 S. The in vitro PA intensity was linearly correlated to HS − concentration in the range of 0–1.2 mM. Intratumoral injection of SnS@Cu 2 O nanoprobe (10 mg/kg) induced time-dependent PA signal amplification at the tumor site, exhibited rapid intensification, peaking at 4-h post-administration. Additionally, the SnS@Cu 2 O can catalyze the Cu + with H 2 O 2 in the tumor tissue yielding ·OH via Fenton-like reactions for photothermal-enhanced chemodynamic therapy, enabling NIR-II PA imaging-guided therapy against deep-tissue colorectal tumor. This paper reports on the synthesis and application of a smart SnS@Cu 2 O nanoprobe that was activated by colorectal tumor microenvironment.

Autologous tumor cells demonstrate considerable promise as individualized therapeutic vaccines owing to their endogenous neoantigen repertoire. Nevertheless, their suboptimal immunogenicity substantially impedes clinical translation. Herein, we engineered an innovative whole tumor cell vaccine platform utilizing photothermal Cu 2- x Se nanoparticles, which encapsulate comprehensive tumor-associated antigens and intrinsic immunostimulatory molecules to elicit a polyvalent antitumor immune response. In this work, Cu 2-x Se was applied to prepare the whole tumor cell vaccine for the first time. As a pyroptosis inducer, Cu 2-x Se triggered the expression of damage-associated molecular patterns (DAMPs) as endogenous adjuvants in 4T1 cells (mouse breast cancer cells) under near-infrared (NIR) irradiation. Repeated freeze-thaw cycles inactivated and lysed Cu 2-x Se-loaded 4T1 cells, releasing DAMPs for laser-activated Cu 2-x Se-based whole tumor cell vaccine (LC-TCV) preparation. Mild photothermal therapy (PTT) at the vaccination site by NIR irradiation effectively suppressed tumor growth. This efficacy was primarily attributed to the synergistic effects of multiple natural antigens and adjuvants in LC-TCV, which promoted dendritic cells (DCs) maturation and subsequent immune activation. Consequently, it potentiates the recruitment of cytotoxic T lymphocytes, elicits immunogenic cell death (ICD), and consequently initiates a potent antigen-specific immune cascade. Collectively, this study presents an innovative approach to unleash the potential of personalized whole tumor cell vaccines in cancer therapy.

Triple-negative breast cancer (TNBC) is a highly aggressive and uncommon subtype of breast cancer with a poor prognosis. It is crucial to prioritise the creation of a nanotherapeutic method that is highly selective and actively targeting TNBC. This study explores a new nanosystem, Cu 9 S 8 -SNAP@PM (C-S@P), composed of Cu 9 S 8 -SNAP coated with a platelet membrane (PM). The purpose of this nanosystem is to cure TNBC using multimodal therapy. The utilisation of PM-coated nanoparticles (NPs) enables active targeting, leading to the efficient accumulation of C-S@P within the tumour. The Cu 9 S 8 component within these NPs serves the potential to exert photothermal therapy (PTT) and chemodynamic therapy (CDT). Simultaneously, the S-Nitroso-N-Acetylvanicillamine (SNAP) component enables nitric oxide (NO) gas therapy (GT). Furthermore, when exposed to NIR-II laser light, Cu 9 S 8 not only increases the temperature of the tumour area for PTT, but also boosts CDT and stimulates the release of NO through thermal reactions to improve the effectiveness of GT. Both in vitro and in vivo experimental results validate that C-S@P exhibits minimal side effects and represents a multifunctional nano-drug targeted at tumors for efficient treatment. This approach promises significant potential for TNBC therapy and broader applications in oncology.

Background Osteosarcoma (OS) is a malignant tumor originating from primitive mesenchymal cells, characterized by rapid metastasis, high invasiveness, and significant mortality. The primary challenges in OS management include the effective elimination of residual tumor cells to prevent recurrence and the repair of extensive bone defects caused by surgical intervention. Objective This study aims to develop an innovative biomimetic 3D-printed bioactive glass ceramic (BGC) scaffold modified with two-dimensional nanosheets to address both tumor ablation and bone tissue repair. Materials and methods The nanosheets were constructed via ellagic acid (EA) and ruthenium (Ru) coordination, leveraging the non-topological adhesion properties of catechol in EA to deposit the nanosheets onto the BGC scaffold (EARu-BGC). The therapeutic effects of EARu-BGC were evaluated in vitro and in vivo . Results EARu-BGC sequentially responds to the local microenvironment during OS treatment. During the tumor ablation phase, EARu-BGC induced ferroptosis through the synergistic effects of photothermal and chemodynamic therapy, achieving over 90 % tumor cell ablation and significantly inhibiting tumor volume and weight. In the bone tissue repair phase, EARu-BGC exhibited adaptive ROS scavenging and facilitated a pro-healing microenvironment, promoting osteogenic differentiation. The gradual degradation of the BGC scaffold provided essential minerals and space for new bone formation. In vivo experiments demonstrated that EARu-BGC significantly enhanced osteogenesis, increasing the trabecular number to 1.51 ± 0.15/mm and reducing trabecular separation to 1.50 ± 0.04 mm. Conclusion The EARu-BGC scaffold presents a promising multifunctional platform for OS treatment by effectively balancing antitumor efficacy with bone repair capabilities.

Bacterial infections pose a significant threat to the healing of human skin wounds, as the reactive oxygen species (ROS) they produce can significantly delay the wound healing process, ultimately leading to the formation of chronic wounds and even endangering human life. Therefore, this study successfully constructed a dynamically reversible hydrogel network through dual crosslinking of Schiff base bonds and hydrogen bonds using oxidized hyaluronic acid (OHA), carboxymethyl chitosan (CMCS), and tannic acid (TA). By loading Cu@PDA, the OCTC2 hydrogel wound dressing was prepared, exhibiting synergistic antibacterial and antioxidant properties through TA/Cu 2+ release and NIR-II photothermal effects. We used FTIR, XRD, SEM, and TEM to thoroughly analyse the specific composition and morphology of the Cu@PDA and hydrogel. We measured the injectability, adhesion, NIR-II photothermal, and antibacterial properties of each group of hydrogels. The results showed that TA and Cu@PDA endowed the hydrogel with excellent antioxidant properties, enabling effective scavenging of various free radicals. Additionally, OCTC2, which possesses sustained-release TA/Cu 2+ capability, achieves antibacterial rates of over 94 % against S. aureus and E. coli . Under the combined effect of NIR-II photothermal therapy, it can achieve highly efficient bacterial killing (99.8 %), effectively promoting the healing of bacterial infection wounds. OCTC2 holds promise as a potential ideal hydrogel dressing for treating bacterial-infected wounds.

Chemodynamic therapy (CDT) and photodynamic therapy (PDT) have emerged as promising strategies for treating skin abscesses. However, their therapeutic efficacy is fundamentally constrained by the hypoxic and hydrogen peroxide-deficient microenvironment characteristic of abscesses. Herein, we developed a near-infrared (NIR) light-responsive nanoplatform (ICG/CuO2@PCM) by encapsulating copper peroxide (CuO2) and indocyanine green (ICG) within a phase-change material (PCM), which enables control over self-supplied H2O2/O2 release via an NIR-triggered cascade. Upon irradiation, ICG-mediated photothermal conversion elevates the temperature beyond the PCM melting threshold, triggering a burst release of CuO2 and ICG. CuO2 generates hydroxyl radicals (•OH) through a Fenton-like reaction in the acidic wound microenvironment. Additionally, CuO2 can thermally decompose under photothermal conditions to produce a sustained oxygen supply, which ICG immediately converts into singlet oxygen (1O2), thereby enhancing PDT efficiency. Furthermore, photothermal therapy not only enhances the efficiency of CDT but also provides additional antibacterial support. Remarkably, ICG/CuO2@PCM (NPs), coupled with irradiation, accelerate abscess wound recovery by dispersing biofilm, killing bacteria, and promoting macrophage polarization toward an anti-inflammatory phenotype. Overall, this NIR-responsive nanoplatform highlights the promise in controlled modulation of microenvironment-adaptive antimicrobial strategies to overcome current limitations of hypoxic infections.

Antibacterial photodynamic therapy (aPDT) exhibits promise in bacterial infection models. However, its effectiveness is often limited in scenarios of bacterial infection within hypoxic environments such as deep periodontal pockets. The present study aims to develop a method to increase the efficacy of aPDT by combining rare earth upconversion nanoparticles (UCNPs) with cyanobacteria, forming cyanobacterial oxygen photosynthesis nanoplatform. In addition, due to the non-photochemical quenching (NPQ) mechanism in algae, cyanobacteria convert excess light energy into thermal energy to achieve synergistic aPDT/photothermal therapy (PTT) effects. Two types of UCNPs were loaded with cyanobacterial cells, including NaYF 4 :Yb 3+ ,0.2 mol%Tm 3+ with blue emission for triggering photosynthesis, and NaErF 4 :0.5 mol%Tm 3+ @NaYF 4 :20 mol%Yb 3+ with red emission for exciting the photosensitizer Chlorin e6 (Ce6). The resulting nanoplatform is referred to as Cyan@UCNPs/Ce6. Both the blue light required for oxygen photosynthesis and red light for aPDT are generated via absorption and upconversion of near-infrared (NIR) light, which can deeply penetrate the tissue. Notably, both in vitro and in vivo experiments demonstrate the remarkable biocompatibility and potent synergistic aPDT/PTT effects. Compared with non-NIR irradiation, the metabolic activities of two common periodontitis bacteria ( F.nucleatum , P.gingivalis ) decrease from 0.80±0.04 and 0.71±0.03 to 0.30±0.01 and 0.17±0.01, respectively ( p < 0.05). The findings of this study demonstrate that the developed Cyan@UCNPs/Ce6 nanoplatform is a promising strategy for photoresponsive synergistic aPDT/PTT in the treatment of periodontitis.

Bacterial infection reduces quality of wound healing and even cause severe complications, but most current dressings adopt single antibacterial mode showed limited effectiveness. Herein, a plant extract quercetin (Que) composite chitosan/hyaluronic acid hydrogel (named CBCQ) with near-infrared (NIR) triggered controllable NO releasing was developed for efficient photothermal therapy/nitric oxide (PTT/NO) synergistic antibacterial and wound healing. Specifically, CBCQ hydrogel with chemical/physical crosslinked network was fabricated via host-guest interaction, metal coordination and Schiff base reaction. It exhibited porous structure, excellent injectability, self-healing, adhesion, antioxidant, and biocompatible properties suitable for wound dressings. Notably, superior PTT effect generated from Que-Fe 3+ has been demonstrated, which served as a photothermal switch for the controllable NO release, thereby enable to utilize the physiological functions of NO at different concentrations, improving antibacterial efficiency by integrating PTT and NO benefits, and further allowing sustained antibacterial through Que when NIR was off. Importantly, CBCQ hydrogel achieved better healing of skin incision than commercial 3M medical glue and Film, and significantly improved the healing rate and quality of Methicillin-resistant Staphylococcus aureus (MRSA) infected wound by efficient and sustained antibacterial activity, inhibiting inflammation, and promoting collagen deposition and angiogenesis, highlighting its promising application in skin incisions and infected full-thickness skin wounds.

Photothermal therapy based on conjugated polymers represents a promising antibacterial strategy but still possesses notable limitations. Herein, degradable pseudo conjugated polymers (PCPs) containing photothermal molecular backbones and reactive oxygen species (ROS)-sensitive thioketal bonds are designed. Triphenylphosphine (PPh 3 ) is introduced into PCPs to generate phosphonium-based PCPs (pPCPs), which further assembled with hyaluronic acid into pPCP nanoparticles (pPCP-NPs). pPCP-NPs with quaternary phosphonium cations selectively anchor on and destroy bacterial cell membranes through electrostatic action. Under 1064 nm laser irradiation, pPCP-NPs (pPCP-NPs/+L) produce near-infrared-II (NIR-II) photothermal antibacterial effect, thereby killing bacteria in a sustained manner. pPCP-NPs are readily degraded upon ROS abundant at infection sites, therefore exhibiting enough biosafety. pPCP-NPs/+L display an almost 100% bacterial inhibition rate in vitro and resultin a nearly complete recovery of bacteria-induced mouse wounds. A further metabolomics analysis denotes that pPCP-NPs/+L work in a concerted way to induce bacterial DNA damage, inhibit bacterial carbon/nitrogen utilization and amino acid/nucleotide synthesis. Taken together, degradable pPCP-NPs with both NIR-II photothermal effect and cationic phosphonium structural bacteriostasis provide a new avenue for antibiotics-alternative anti-infection therapy.

The coating optimized with high near-infrared reflectance pigments can effectively reduce the energy consumption for heating and cooling in buildings, thereby alleviating the pressure on global energy consumption. Using a high-temperature solid-state method, blue-violet pigments with high near-infrared reflectance, Ca 2 Zn 1- x M x Si 2 O 7 (M = Mn, 0 ≤ x ≤ 0.4 and Ni, 0 ≤ x ≤ 0.2) solid solutions, were synthesized for the first time, with a maximum solar reflectance of 82.67%. Using XPS analysis, it was determined that the oxidation state of Ni in the pigment is +2, while Mn exists in mixed oxidation states of +2 and +3. UV-VIS-NIR spectroscopy analysis indicates that the blue-violet color of the pigment originates from the d-d transitions of the transition metal ions. As the doping concentration increases, the near-infrared reflectance of the pigment decreases. The DFT calculations have also confirmed that the color of pigments originates from transition metal ions. The as-synthesized pigments were incorporated into acrylic to create colored coatings. Improving the near-infrared solar reflectance of the acrylic coating. The excellent high near-infrared solar reflectance and coating property optimization make the synthesized pigment a potential energy-saving coating.

Intelligent control of the immune response is essential for obtaining percutaneous implants with good sterilization and tissue repair abilities. In this study, polypyrrole (Ppy) nanoparticles enveloping a 3D frame of sulfonated polyether ether ketone (SP) surface are constructed, which enhance the surface modulus and hardness of the sulfonated layer by forming a cooperative structure of simulated reinforced concrete and exhibit a superior photothermal effect. Ppy-coated SP could quickly accumulate heat on the surface by responding to 808 nm near-infrared (NIR) light, thereby killing bacteria, and destroying biofilms. Under NIR stimulation, the phagocytosis and M1 activation of macrophages cultured on Ppy-coated SP are enhanced by activating complement 3 and its receptor, CD11b. Phagocytosis and M1 activation are impaired along with abolishment of NIR stimulation in the Ppy-coated SP group, which is favorable for tissue repair. Ppy-coated SP promotes Collagen-I, vascular endothelial growth factor, connective tissue growth factor, and α -actin (Acta2) expression by inducing M2 polarization owing to its higher surface modulus. Overall, Ppy-coated SP with enhanced mechanical properties could be a good candidate for clinical percutaneous implants through on-off phagocytosis and switchable macrophage activation stimulated with NIR.
