Alicyclobacillus acidoterrestris has received much attention due to its unique thermo-acidophilic property and implication in the spoilage of pasteurized juices. The aim of this study was to investigate the antibacterial characteristics and mechanisms of thymol against A. acidoterrestris vegetative cells and spores. Minimum inhibition concentration (MIC) and minimum bactericidal concentration (MBC) of thymol against vegetative cells were both 0.25 mg/mL. MIC of thymol against spores was 0.5 mg/mL, and the MBC was greater than 1.0 mg/mL. Thymol could effectively inhibit A. acidoterrestris growth, spore germination and guaiacol production. When treated by thymol, the leakage of nucleic acids and proteins from A. acidoterrestris cells and spores increased significantly. SDS-PAGE investigation of bacterial proteins showed that the loss of soluble proteins was obvious as well. Scanning electron microscopy observation exhibited that A. acidoterrestris cells or spores treated with thymol had a severe morphological damage. These results demonstrated that thymol exerted its good antibacterial activities by destroying the integrity of the cell and spore. This study provides a new method for the inactivation of A. acidoterrestris in fruit juice.

The presence of Heyndrickxia sporothermodurans is a widespread concern in dairy industry due to its highly thermal-resistant spores. The "Cleaning-in-place" (CIP) system is the primary method used by dairy industry to control them. But the food safety-related properties of H. sporothermodurans remain poorly understood. Thus, this study proposes to determine the growth characteristics and sporulation of H. sporothermodurans in different matrices, as well as the efficacy of various disinfectants against its biofilm and spore. Therefore, the growth of vegetative cell and spore germination in rich media (brain heart infusion, BHI) and ultra-high temperature (UHT) milk for at least 360 h was investigated in this research. In addition, the biofilms of different maturity induced on stainless steel surfaces were sanitized simulating a CIP procedure, using disinfectants (200 mg/mL peracetic acid, 1.5% sodium hydroxide and 1% nitric acid). Scanning electronic microscopy (SEM) was used as a complementary technique to observe the conditioning of the stainless steel surface after 10 d. Subsequently, the effects of these sanitation methods against the spore were performed in 96-well plates. The observation of growth behavior revealed that the vegetative cells and spore cells may be interconverted in a nutrient-rich environment, resulting in a balanced mode in which spores account for approximately 0.1% of the total cells. This could be used to anticipate the number of contaminated bacteria in a UHT plant environment. The sterilization efficiency tests showed that these sanitation methods were efficient against biofilms but not against spores. Therefore, new inactivation strategies against spores should be proposed and should focus on spores adhesion and the biofilms induced by spores.

Pathogenic bacterial spores pose a considerable threat to human health due to their ability to survive extreme environmental conditions. Thus, sensitive detection and efficient inactivation of these spores are essential for preventing disease transmission. However, most existing studies have predominantly concentrated on detection alone. Herein, we developed a versatile nanoplatform that integrates sensitive bacterial spore sensing and in situ spore inactivation capabilities by assembling terbium ions (Tb 3+ ) with photosensitizing carbon dots (CDs), referred to as Tb-CDs nanoassemblies (NAs). By leveraging the unique optical properties of CDs and Tb 3+ ions, along with the antenna effect of lanthanides, Tb-CDs NAs demonstrated highly sensitive visual detection of the key bacterial spore biomarker, pyridinedicarboxylic acid (DPA), achieving a low detection limit of 0.68 μM and excellent selectivity. Additionally, Tb-CDs NAs exhibited practical sensing capabilities in various environments, including spore suspensions and real samples, while monitoring bacterial spore germination. Further studies confirmed that the DPA-triggered disassembly of Tb-CDs NAs led to the release of CDs, enabling higher affinity for spores and in situ inactivation through singlet oxygen generation. This work underscores the potential of CDs-based assemblies for simultaneous pathogen detection and inactivation, providing promising tools for preventing bacterial spread and contamination.

Oxidative stress from mechanical injury can trigger the development of Wound-Induced Spores (WIS) in Pyropia yezoensis , yet the regulatory mechanisms underlying this process require further elucidation. In this study, comparative transcriptomic analysis between injured and uninjured P. yezoensis thalli revealed significant differential gene expression within the glutathione (GSH) metabolism pathway during WIS development. There was a down-regulation in the expression of genes encoding for γ-glutamyl-cystein synthetase (GCL) and glutathione synthetase (GSS), which were pivotal for GSH biosynthesis, during the development of WIS. Moreover, a diminished expression level was also observed in genes associated with oxidative metabolism of GSH during this process, including those encoding glutathione S-transferase (GST), glutathione peroxidase (GPX), glutathione reductase (GR), ascorbate peroxidase (APX), monodehydroascorbate reductase (MDHAR), and dihydroascorbate reductase (DHAR). However, this transcriptional regulation does not correspond to a significant decrease in intracellular GSH levels, which may be associated with a reduction in the oxidative metabolism of GSH, leading to decreased cellular utilization of GSH. Treatment injured P. yezoensis with Buthionine sulfoximine (BSO) promoted the release probability of WIS, the release efficiency of WIS and germination rate of asexual spores, while supplementation of GSH produced opposite results, suggesting that modulating GSH levels can regulate the release of WIS. Correlation analysis suggested that there was a significant positive correlation between the release of WIS and the germination rate of asexual spores, suggesting that promoting the release of WIS could lead to the acquisition of a greater number of asexual spore seedlings. Our findings underscore the key role of GSH metabolism in WIS regulation and provide a theoretical foundation for the subsequent development of strategies to obtain asexual spore seedlings.

Bacterial spores in food exhibit strong resistance to conventional processing methods, posing a significant challenge to food safety. This scientific aim of the study was to compare the antimicrobial effects of organic acids and plasma-activated lactic acid (PALA) on the specific spoilage bacteria (SSB) spores in cooked crayfish ( Procambarus clarkii ), and further explored the antimicrobial mechanism of PALA against Bacillus cereus spores. Plasma activation enhanced the antimicrobial efficacy of lactic acid (LA), as 1 % PALA showed equivalent antimicrobial efficacy to 1.5 % LA against spores. Microstructural changes and leakage of core contents indicated that PALA treatment caused damage to the inner membrane (IM) of B. cereus spores. Then, the spore germination experiments further revealed that PALA treatment disrupted the function of SpoVA proteins in the IM, leading to a significant decrease in spore germination capacity. Furthermore, inactivated spores did not regain viability after treatment with lysozyme or Ca-DPA, and no mutants were detected among the surviving spores. These results demonstrated that the inactivation effect of PALA on B. cereus spores is closely related to the structural disruption and dysfunction of the spore IM. Additionally, PALA treatment extended the shelf life and maintained the product quality of cooked crayfish, supporting PALA as a promising measure for controlling microbial safety.

Transmission of bacterial endospores between the environment and people and the following germination in vivo play critical roles in both the deadly infections of some bacterial pathogens and the stabilization of the commensal microbiotas in humans. Our knowledge about the germination process of different bacteria in the mammalian gut, however, is still very limited due to the lack of suitable tools to visually monitor this process. We proposed a two-step labeling strategy that can image and quantify the endospores’ germination in the recipient’s intestines. Endospores collected from donor’s gut microbiota were first labeled with fluorescein isothiocyanate and transplanted to mice via gavage. The recipient mice were then administered with Cyanine5-tagged D-amino acid to label all the viable bacteria, including the germinated endospores, in their intestines in situ. The germinated donor endospores could be distinguished by presenting two types of fluorescent signals simultaneously. The integrative use of cell-sorting, 16S rDNA sequencing, and fluorescence in situ hybridization (FISH) staining of the two-colored bacteria unveiled the taxonomic information of the donor endospores that germinated in the recipient’s gut. Using this strategy, we investigated effects of different germinants and pre-treatment interventions on their germination, and found that germination of different commensal bacterial genera was distinctly affected by various types of germinants. This two-color labeling strategy shows its potential as a versatile tool for visually monitoring endospore germination in the hosts and screening for new interventions to improve endospore-based therapeutics.

Salmonella Typhimurium ( S . Tm), a Gram-negative pathogenic bacterium, is one of the most common causes of invasive bacterial diseases. Antibiotic therapy remains the principal therapeutic modality for treating S . Tm infection. However, due to the difficulty in precisely targeted pathogenic bacteria after oral administration, the therapeutic effect remains unsatisfactory. Here, we developed an oral probiotic spores-based biohybrid delivery system (BCs@PME-Au) to treat S. Tm-induced colitis. By employing a one-pot metal deposition method, Polymyxin E (PME) acted as a reducing agent to promote the Au 3+ rapid nucleation and growth into PME-capped Au NPs (PME-Au NPs). By forming Au-S and Au-N bonds with the active sites (-SH, -NH 2 ) of Bacillus coagulans spores (BCs), PME-Au NPs were anchored onto the surface of BCs to construct the biohybrid system BCs@PME-Au. Following oral administration, BCs@PME-Au successfully passed through the gastric acid barrier. After absorbed water and nutrients, BCs germinated into Bacillus coagulans (BC) in the gut and PME-Au NPs were released. Based on the BC’s targeting pathogen infection site and PME-Au NPs’ targeting Gram-negative bacteria, the biohybrid system achieved significantly antibacterial effect of S . Tm. Mechanistically, by blocked the LPS-induced inflammatory pathway TLR4/MyD88/NF-κB, BCs@PME-Au exerted a powerful anti-inflammatory effect. With its robust antibacterial efficacy, targeted delivery, and excellent safety profile demonstrated both in vitro and in vivo, the biohybrid system BCs@PME-Au offers significant promise in treating bacterial colitis.

Hepatopancreatic microsporidiosis (HPM), caused by Enterocytozoon hepatopenaei (EHP), has emerged as a major threat to the shrimp aquaculture industry. However, no effective strategies have yet been developed to control the spread of this disease. In this study, we assessed the anti-EHP activity of ten monomeric compounds in Penaeus vannamei . The results showed that tetrahydropalmatine reduced the EHP copy number by 73.8 % at a concentration of 20 mg/L. In a therapeutic experiment, feeding EHP-infected shrimp a diet containing 0.1 % tetrahydropalmatine significantly lowered the EHP load. Additionally, the growth-related gene juvenile hormone esterase-like carboxylesterase 1 was significantly upregulated, while ecdysteroid-regulated protein and farnesoic acid O-methyltransferase were significantly downregulated, suggesting that tetrahydropalmatine may enhance the growth potential of EHP-infected shrimp. After 30 days of feeding, the EHP-infected shrimp's body weight in the groups fed with tetrahydropalmatine increased by 19.9 % compared to those fed with the normal diet. In a prevention experiment, healthy shrimp fed a diet containing tetrahydropalmatine and subsequently infected with EHP showed an increase in body weight by day 30 post-infection, compared to the control group, although this difference was not statistically significant. Tetrahydropalmatine can inhibit spore germination and electron dense disc formation, which may be an important reason for its suppression of EHP infection. Moreover, tetrahydropalmatine effectively inhibited the horizontal transmission of EHP, suggesting its potential to limit HPM outbreaks. These findings indicate that tetrahydropalmatine may serve as a feed additive for the treatment of HPM caused by EHP.

In this work, a new and simple method was developed for the synthesis of uncovered and high-selectivity spiramycin-based molecularly imprinted microspheres (SP-MIMs) by Pickering emulsion polymerization using spiramycin as templates. And surprisingly the solid particles were absent on the surface of imprinted microspheres, which can be ascribed to the Ganoderma lucidum spores (GLS): they were firstly selected to be the stabilizers for the Pickering emulsion in this new strategy. Through a series of adsorption experiments, the uncovered SP-MIMs were proven to possess more excellent selective recognition and removal ability for template SP in aqueous solution. And SP-MIMs could be reused for many times without significant loss of adsorption capacity, indicating the satisfactory regeneration performance. Therefore, SP-MIMs can be employed as a promising adsorbent for the selective removal of SP from aqueous media, and this strategy will contribute to overcoming the shortcomings of stabilizer particles on the surface of the as-prepared imprinting microspheres, because these solid particles lack the special selective recognition activity of template molecules, and may disturb the adsorption effect of imprinting microspheres.

Ganoderma lucidum spores (GLS) constitute a healthy food for humans, but the hard spore shells are difficult to digest. In this study, digestive properties of GLS in different forms (sporoderm-unbroken, sporoderm-broken [SB-GLS] and sporoderm-removed [SR-GLS]) were investigated in vitro digestion and fermentation. After simulated digestion, morphologies of all three spores changed slightly. After gastric and intestinal digestion, the amounts of polysaccharides in each digestive residue of GLS, SB-GLS and SR-GLS were greatly reduced, by approximately 90.3%, 96.5% and 99.2%, respectively. Triterpenoid quantities in the digestive residues were higher than those before digestion because the absolute weight of residue decreased. Total triterpenoid contents of GLS, SB-GLS and SR-GLS after digestion were 9.2 ± 0.8, 24.7 ± 1.3, and 32.5 ± 1.1 mg/g, respectively. The amounts of CO 2 , CH 4 and H 2 found in the water and alcohol extracts of SR-GLS were similar, but the amount of H 2 in the alcohol extract was higher. In terms of short-chain fatty acids (SCFAs) production, propionic, butyric and valeric acids found in the water extracts were more abundant than those in alcohol extracts, which suggests that the polysaccharides were particularly important for SCFA production. This finding indicates that SR-GLS for the water extract has stronger metabolic promotion effects and better anti-inflammatory properties.

The inactivation effects of high pressure CO2 + nisin (simultaneous treatment of HPCD and nisin, HPCD + nisin), HPCD→nisin (HPCD was followed by nisin), and nisin→HPCD (nisin was followed by HPCD) treatments on Bacillus subtilis spores in aqueous solutions were compared. The spores were treated by HPCD at 6.5 or 20 MPa, 84–86°C and 0–30 min, and the concentration of nisin was 0.02%. Treated spores were examined for the viability, the permeability of inner membrane (IM) using flow cytometry method and pyridine-2, 6-dicarboxylic acid (DPA) release, and structural damage by transmission electron microscopy. A synergistic effect of HPCD + nisin treatment on inactivation of the spores was found, and the inactivation efficiency of the spores was HPCD + nisin > HPCD→nisin or nisin→HPCD. Moreover, HPCD + nisin caused higher IM permeability and DPA release of the spores than HPCD. A possible action mode of nisin-enhanced inactivation of the spores was suggested as that HPCD firstly damaged the coat and cortex of spores, and nisin penetrated into and acted on the IM of spores, which increased the damage to the IM of spores, and resulted in higher inactivation of the spores.

Introduction Pythiosis, caused by Pythium insidiosum ( P. insidiosum ), is an emerging disease with high mortality and morbidity. Despite its clinical severity and geographical strain variations, diagnostic and treatment challenges persist. Materials and methods We queried PubMed and Google Scholar for “ P. insidiosum ” and “pythiosis,” 1084 human pythiosis cases to generate a corresponding distribution map. Molecular biology, morphology, modified sporulation technique, and microscopic observation were employed to understand the biological properties of P. insidiosum with a strain of P. insidiosum isolated from a patient in Guangzhou. Drug susceptibility studies on the isolate were conducted both in vitro and in vivo . Results Molecular biology, morphology, and biological processes confirmed that this strain was P. insidiosum . Epidemiological investigations have revealed that India and Thailand are hotspots for human pythiosis, and sporadic cases are increasing in China, the Americas, and Europe. The modified method of zoospore induction achieved 250-fold greater than the traditional method within 24 h. In vitro drug testing demonstrated superior antibiotic sensitivity (doxycycline (DOX) MIC 4 μg/mL; azithromycin (AZM) MIC 8 μg/mL) versus antifungals (> 128 μg/mL). Subcutaneous infection models in immunocompromised mice showed 80 and 90% survival with oral AZM/DOX monotherapy versus 20% in controls ( P < 0.05), correlating with reduced hepatic fungal burdens and attenuated neutrophilic periarteritis. Conclusion The distribution map underscores pythiosis as a growing climate-sensitive disease that requires enhanced surveillance in non-endemic regions. The modified method of zoospore induction and microstructure observation with Transmission Electron Microscopy (TEM) may be helpful for the rapid and sensitive detection of P. insidiosum . We also provided theoretical and technical support for the effective treatment of pythiosis in humans and animals with DOX and AZM in China, which offers novel insights into the potential of certain antibiotics as effective treatments.
