A series of benzimidazole phenylhydrazone derivatives (6a–6ai) were synthesized and characterized by1H-NMR, ESI-MS, and elemental analysis. The structure of6bwas further confirmed by single crystal X-ray diffraction as (E)-configuration. All the compounds were screened for antifungal activity againstRhizoctonia solaniandMagnaporthe oryzaeemploying a mycelium growth rate method. Compound6fexhibited significant inhibitory activity againstR. solaniandM. oryzaewith the EC50values of 1.20 and 1.85 μg/mL, respectively. In vivo testing demonstrated that6fcould effectively control the development of rice sheath blight (RSB) and rice blast (RB) caused by the above two phytopathogens. This work indicated that the compound6fwith a benzimidazole phenylhydrazone scaffold could be considered as a leading structure for the development of novel fungicides. Keywords:benzimidazole;phenylhydrazone;antifungal activity

BACKGROUND Sclerotinia stem rot (SSR) caused by Sclerotinia sclerotiorum threatens oilseed rape cultivation, and the emergence of fungicide-resistant strains has led to control failures worldwide. Identifying novel chemical alternatives with different modes of action and high antifungal activities is thus crucial. Herein we evaluated the antifungal effects of 3-(2-pyridyl)methyl-2-(4-chlorphenyl)imino- thiazolidine (PMAS) on S. sclerotiorum to determine its efficacy for SSR management. RESULTS PMAS had an inhibitory effect on mycelial growth; the EC 50 values were 17.83 and 21.15 μg mL −1 for the carbendazim-susceptible strain Ss01 and carbendazim-resistant strain Hm25, respectively. PMAS treatment changed the color of inhibited mycelia to green, and the hyphae were sustained in the undifferentiated stage. Cysteine supplementation made this green color disappear, whereas methionine enhanced the color. Moreover, PMAS treatment markedly inhibited oxalic acid biogenesis, increased free thiol content in mycelia, and weakened the activities of oxaloacetase and malate dehydrogenase, but had little effect on the activity of glyoxylate dehydrogenase. Cysteine could reverse the inhibitory effects of PMAS on mycelial morphogenesis and biochemical constituents, except thiol production. In the pot-culture experiment, PMAS showed a good protective effect, with the control efficacy being >91% on SSR. CONCLUSION PMAS appears to be an effective fungicide for SSR management. © 2020 Society of Chemical Industry

Over the past several decades, invasive fungal infections, especially candidiasis, have caused dramatic morbidity and mortality due to ineffective antifungal drugs and severe drug resistance. Herein, new BRD4–histone deacetylase (HDAC) inhibitors were designed to restore the susceptibility of Candida albicans (C. albicans) to fluconazole (FLC). Interestingly, several compounds showed excellent selectivity against fungal HDACs. In particular, compound B2 showed excellent synergistic effect with FLC against resistant C. albicans (FICI = 0.063) with high selectivity against fungal HDACs (SI = 1653) and low cytotoxicity. Compound B2 effectively synergized with FLC and prevented biofilm formation and morphological transition in resistant C. albicans, potentiating the antifungal activity of FLC in vivo and significantly reducing kidney fungal loads. Thus, this drug combination is promising in the treatment of resistant C. albicans infections.

Histatin5 (Hst-5) is a member of the histatin family of antimicrobial peptides secreted by human parotid and submandibular glands. With the natural antibacterial activity, it plays an important role in the first-line barrier of oral cave against pathogens, especially for the fungal intrusion. In this study, we explored the utility of Hst-5 in the treatment of vulvovaginal candidiasis, a common condition of women of the childbearing age. We used a synthesized Hst-5 over five consecutive days as the topical treatment in a murine model of vulvovaginal candidiasis. According to the fungal colony counts, fungal burden in the vagina lavage dropped remarkably after treatment with Hst-5. Furthermore, cytological analysis of the lavage fluid indicated that the number of cast-off cells including cornified epithelial cells and inflammatory cells also decreased; histological evaluation of the vagina tissue revealed less fungi adhering to the vaginal wall in treated animals than in controls. Combined, these results suggested for the first time the potential utility of Hst-5 as a topical treatment for vulvovaginal candidiasis, uncovering the possibility of exploiting the natural antibiotic peptides in other aspects.

Fusariumwilt, mainly caused byFusarium oxysporum, affects ginger yield and quality worldwide. To explore a safe and sustainable method of preventing and controllingFusariumwilt, the inhibitory effects of eugenol onF. oxysporum FOX-1were analysed in this study. The results showed that eugenol inhibited the reproductive and vegetative growth ofF. oxysporum FOX-1in vitro. Microscopic observations revealed that eugenol disrupted the hyphal morphology ofF. oxysporum. In addition, eugenol destroyed the cell membrane integrity of the pathogenic fungi, resulting in the leakage ofF. oxysporum FOX-1intercellular contents, including electrolytes, soluble proteins, nucleic acids, and malonydialdehyde. Results of an in vivo inoculation test showed that eugenol exerted a strong inhibitory effect onFusariumwilt in ginger seedlings. In summary, eugenol had an inhibitory effect on the growth ofF. oxysporum FOX-1and controlledFusariumwilt in ginger seedlings. These findings provide a foundation for future development of botanical antifungal agents to manageFusariumwilt. Keywords:Fusariumwilt;eugenol;essential oil;antifungal activity;Zingiber officinale

As antimicrobial agents, essential oils (EOs) have been used by more and more companies as alternatives to traditional chemicals. In the study, seven EOs from Pinus massoniana , cinnamon , Star anise , Litsea cubeba , Cinnamomum camphora , Lemon eucalyptus , and Camphor from Guangxi were researched for their antifungal performances and chemical components. With the help of the diffusion in a disc way as well as the minimum inhibitory concentration (MIC) to three fungi, the antifungal impacts and the bamboo slices anti-mold tests of these EOs were studied. The compositions of these oils were identified. Litsea cubeba EO, Anise EO, and Cinnamon EO exhibited super antifungal activity against Trichoderma viride , Aspergillus niger , and Penicillium citrinum . Treated by Cinnamon EO, Anise EO, or Litsea cubeba EO, few molds were observed on the surface of bamboo chips, which showed that these EOs had a good antifungal effect on molds of bamboo. Starch-based wood adhesives are widely used as environmentally friendly and formaldehyde-free alternatives in the manufacturing of wood products. However, their poor water resistance and inability to inhibit bacterial/fungal growth limit their practicality. To address this issue, a physicochemical double-crosslinked network structure between citric acid, starch, and wood was prepared in this study by physically entangling the wood structure and chemically crosslinking the starch molecules, with citric acid preventing the activity of various enzymes and denatured proteins within microbial cells, thereby inhibiting bacterial and fungal growth. In water resistance cycling tests, the resulting esterified starch plywood demonstrated a 47.6 % and 13.2 % increase in water resistance compared with starch plywood and commercially available phenolic resin plywood, respectively. In addition, the tighter intermolecular bonding in the crosslinked structure results in a 31 % increase in heat resistance. The physical “nailing” and chemical crosslinking between the adhesive and the wood improved considerably the bonding strength to 98.36 %. The dual role of the physicochemical crosslinking of starch adhesives provides a straightforward strategy for the preparation of inexpensive, water-resistant, and fungal/bacterial-resistant starch adhesives, thus contributing to the promotion of starch adhesives.

Essential oils (EOs) have been shown to have a diversity of beneficial human health effects.Clausenais a large and highly diverse genus of plants with medicinal and cosmetic significance. The aim of this study was to analyze the composition ofClausena lansiumEOs and to investigate their potential antifungal effects. The chemical compositions ofClausena lansiumEOs obtained by hydrodistillation were analyzed by gas chromatography-mass spectrometry (GC-MS). A total of 101 compounds were identified among the diverse extracts ofC. lansium. EOs of leaves and pericarps from different cultivars (Hainan local wampee and chicken heart wampee) collected in Hainan (China) were classified into four clusters based on their compositions. These clusters showed different antifungal activities against fiveCandidaspecies (C. albicans,C. tropicalis,C. glabrata,C. kruseiandC. parapsilosis) using the disc diffusion method.Clausena lansiumEOs of pericarps displayed noteworthy antifungal activitives against all the testedCandidastrains with inhibition zone diameters in the range of 11.1–23.1 mm. EOs of leaves showed relatively low antifungal activities with inhibition zone diameters in the range of 6.5–22.2 mm. The rank order of antifungal activities among the four EO clusters was as follows: Cluster IV> Cluster III > Cluster I ≥ Cluster II. These results represent the first report about the correlation between chemical composition ofC. lansiumEOs and antifungal activity. Higher contents of β-phellandrene, β-sesquiphellandrene and β-bisabolene in EOs of pericarps were likely responsible for the high antifungal activity of Cluster IV EOs. Taken together, our results demonstrate the chemical diversity ofClausena lansiumEOs and their potential as novel antifungal agents for candidiasis caused byCandidaspp. Furthermore, the obtained results showing a wide spectrum of antifungal activities provide scientific evidence for the traditional use of these plants. Keywords:correlation;Clausena lansium;essential oil;antifungal activity;Candidaspp.

The antifungal activity of chitosan combined with clove oil against Penicillium digitatum , the causal agent of citrus green mold, was tested in vitro and in vivo . Chitosan combined with clove oil inhibited mycelial growth more than individual treatments, which was related to the greater release of cellular material and the largest alterations in hyphal morphology of P. digitatum . However, compared to chitosan alone, 1% chitosan coatings combined with various amounts of clove oil (0.5, 1 or 2 mL/L) showed no greater ability in controlling decay development on artificially inoculated citrus fruit. 1% chitosan combined with 0.5 mL/L clove oil appeared to slightly reduce lesion diameter and enhanced the activities of defense enzymes, including chitinase and phenylalanine ammonia-lyase at the later stages of incubation. This study indicated that the synergistic antifungal activity of chitosan-clove oil observed in in vitro studies was not found in in vivo tests. Therefore, the data suggest that a coating of 1% chitosan alone, not combined with clove oil, can effectively contribute to the control of green mold on citrus fruit.

The antifungal activity and encapsulation properties of cinnamaldehyde-loaded microcapsules by tannic acid were investigated. The results showed that cinnamaldehyde exhibited a high in vitro antifungal effect against Aspergillus brasiliensis through altering the morphology of the hyphae, and its inhibition zone diameter (IZD) exceeded 15 mm with a minimum inhibitory concentration of 1.0 mg/mL. Cinnamaldehyde-loaded microcapsules crosslinked with tannic acid have a high encapsulation efficiency (92.53 ± 5.20%) and antifungal activity (37.82 ± 0.63 mm), and the retention rate was 2.5 times of that of unencapsulated cinnamaldehyde after 30-d storage at 37 °C. Furthermore, the antifungal stability was enhanced (IZD 25.37 mm) after 60-d storage at 37 °C as compared with that of cinnamaldehyde (IZD 22.07 mm). The results of cell membrane integrity and the extracellular protein content also indicated that the microencapsulation could effectively protect cinnamaldehyde, delay the decrease of antifungal effect and improve its long-term antifungal activity against A. brasiliensis by increasing the retention rate of cinnamaldehyde after storage.

Aspergillus species are emerging causative agents of non-dermatophyte mold onychomycosis. In this study, 48 Aspergillus isolates were obtained from patients with onychomycosis in Mashhad, Iran, during 2015–2018. The aim is to identify the Aspergillus isolates to the species level by using partial calmodulin and beta-tubulin gene sequencing and MALDI-TOF MS, and to evaluate their in vitro susceptibility to ten antifungal drugs: terbinafine, itraconazole, voriconazole, posaconazole, ravuconazole, isavuconazole, caspofungin, micafungin, anidulafungin and amphotericin B according to CLSI M38-A3. Our results indicate that A.flavus (n = 38, 79%) is the most common Aspergillus species causing onychomycosis in Mashhad, Iran. Other detected species were A. terreus (n = 3), A. tubingensis (n = 2), A. niger (n = 1), A. welwitschiae (n = 1), A. minisclerotigenes (n = 1), A. citrinoterreus (n = 1) and A. ochraceus (n = 1). Aspergillus flavus , A. terreus and A. niger isolates were correctly identified at the species level by MALDI-TOF MS, while all cryptic species were misidentified. In conclusion, A. flavus is the predominant Aspergillus species causing onychomycosis due to Aspergillus spp. in Mashhad, Iran. MALDI-TOF MS holds promise as a fast and accurate identification tool, particularly for common Aspergillus species. It is important that the current database of reference spectra, representing different Aspergillus species is expanded to increase the precision of the species-level identification. Terbinafine, posaconazole and echinocandins were i n vitro most active against the studies Aspergillus isolates and terbinafine could be the first choice for treatment of onychomycosis due to Aspergillus .

In the paper, five quaternary ammonium salts derived from rosin were synthesized, characterized and their antifungal activities against different molds were studied. Choline quaternary ammonium salt and tetramethylammonium hydroxide were synthesized from dehydroabietic acid and the rosin ethanolamine ester quaternary ammonium salts which are new compounds were synthesized from propylene pimaric acid. Their structures were confirmed by FTIR, 1 HNMR, 13 CNMR, and high resolution mass spectrometry. The antifungal activities of the synthesized quaternary ammonium salts from rosin acid were tested by Oxford cup method, and their minimum inhibitory concentration was determined. The results show that quaternary ammonium salts derived from rosin acid have inhibitory effects on bamboo slices to some extent. Among them, propylene pimaric acid ethanolamine ester quaternary ammonium salts (A 1 and A 2 ) showed obvious antifungal activity in the Oxford cup test. The Oxford cup activity test and anti-mildew effect of quaternary ammonium salts (B 1 and B 2 ) synthesized from rosin acid are significant, and there are almost no mould growth on bamboo slices. It can be inferred that A 1 , A 2 , B 1 and B 2 are expected to be used as bamboo mildew inhibitors. Graphical abstract Five quaternary ammonium salts derived from rosin were synthesized and characterized; and their antifungal activities against different molds were studied.
