Anammox bacteria performed the reaction of NH4+ and NO with hydrazine synthase to produce N2H4, followed by the decomposition of N2H4 with hydrazine dehydrogenase to generate N2. Ferroheme/ferriheme, which serves as the active center of both hydrazine synthase and hydrazine dehydrogenase, is thought to play a crucial role in the synthesis and decomposition of N2H4 during Anammox due to its high redox activity. However, this has yet to be proven and the exact mechanisms by which ferroheme/ferriheme is involved in the Anammox process remain unclear. In this study, abiotic and biological assays confirmed that ferroheme participated in NH4+ and NO reactions to generate N2H4 and ferriheme, and the produced N2H4 reacted with ferriheme to generate N2 and ferroheme. In other words, the ferroheme/ferriheme cycle drove the continuous reaction between NH4+ and NO. Raman, ultraviolet–visible spectroscopy, and X-ray absorption fine structure spectroscopy confirmed that ferroheme/ferriheme is involved in the synthesis and decomposition of N2H4 via the core FeII/FeIII cycle. The mechanism of ferroheme/ferriheme participation in the synthesis and decomposition of N2H4 was proposed by density functional theory calculations. These findings revealed for the first time the heme electron transfer mechanisms, which are of great significance for deepening the understanding of Anammox.

Biliverdin, a bile pigment hydrolyzed from heme by heme oxygenase (HO), serves multiple functions in the human body, including antioxidant, anti-inflammatory, and immune response inhibitory activities. Biliverdin has great potential as a clinical drug; however, no economic and efficient production method is available currently. Therefore, the production of biliverdin by the biotransformation of exogenous heme using recombinant HO-expressing yeast cells was studied in this research. First, the heme oxygenase-1 gene ( HO1 ) encoding the inducible plastidic isozyme from Arabidopsis thaliana , with the plastid transport peptide sequence removed, was recombined into Pichia pastoris GS115 cells. This resulted in the construction of a recombinant P. pastoris GS115-HO1 strain that expressed active HO1 in the cytoplasm. After that, the concentration of the inducer methanol, the induction culture time, the pH of the medium, and the concentration of sorbitol supplied in the medium were optimized, resulting in a significant improvement in the yield of HO1. Subsequently, the whole cells of GS115-HO1 were employed as catalysts to convert heme chloride (hemin) into biliverdin. The results showed that the yield of biliverdin was 132 mg/L when hemin was added to the culture of GS115-HO1 and incubated for 4 h at 30 °C. The findings of this study have laid a good foundation for future applications of this method for the economical production of biliverdin. Graphical Abstract

Balanced level of hemin in the body is fundamentally important for normal human organ function. Therefore, environmentally benign, stable, and fluorescent metal nanoclusters (NCs) for selective and sensitive detection of hemin have been investigated and reported. Herein, highly orange red emissive gold NCs are successfully synthesized using glutathione as a reducing and stabilizing agent (GSH-Au NCs). The clusters are characterized using various techniques like Fourier transform infrared spectroscopy (FTIR), transmission electron microscopy (TEM), UV-vis spectroscopy, and fluorescence spectrometer. The fluorescence intensity of as-synthesized Au NCs strongly quenched upon addition of different concentrations of hemin. The decrease in fluorescence intensity of GSH-Au NCs has been applied for determination of hemin concentration in the linear range from 1 to 25 nM with a low limit of detection (LOD) of 0.43 nM. The method was also successfully applied for quantification of hemin in human serum sample. In view of this reality, the system can be considered as a possible strategy and excellent platform for determination of hemin in various areas of application. Graphical abstract

Abtract Heme binds to amyloid β -peptide (A β ) in the brain of Alzheimer΄s disease (AD) patients, thus forming heme-A β complexes and leading the characteristic pathological features of AD. The heme-A β complexes reduce O 2 by 2e − to obtain co-reactant H 2 O 2 . In this work, two-dimensional graphite-like carbon nitride (g-C 3 N 4 ) was hybridized with heme via π-π interaction to construct an electrochemiluminescence (ECL) biosensor for detection of A β peptides. The capture aptamer was firstly immobilized on the gold electrode, and then incubated with target A β peptides and g-C 3 N 4 -heme. The g-C 3 N 4 -heme-A β composite was subsequently anchored on the electrode and showed a signal-on, label-free ECL emission based on dual co-reactants enhancement attributed to in-situ generated H 2 O 2 and original K 2 S 2 O 8 in PBS buffer. Under the optimum experimental conditions, the ECL biosensor has a sensitive response to A β peptides with a linear range of 10 fM-0.1 μM and a detection limit of 3.25 fM. Considering prominent specificity, reproducibility, and stability, the proposed biosensor was used to assess recovery of A β peptides in human serum and the result was satisfactory. We believe that the established method would offer a useful analytical means for quantifying A β peptides in a biological matrix.

Hemin, the most common biological iron form, serves many biological functions. However, studies on hemin detection are scarce. As an unusual natural peroxide, artemisinin is quite stable in the presence of common species. While, it can decompose to generate superoxide radicals in the presence of hemin. And the as-obtained superoxide radicals can effectively oxidize thiamine to form fluorescent thiochrome. Inspired by this, a simple and novel fluorescent assay for hemin detection based on artemisin-thiamine is developed. Based on the linear relationship between the fluorescence intensity and concentration of hemin, the developed artemisinin-thiamine assay can be utilized for sensitive detection of hemin in the concentration range of 2.0–300.0 nM with a detection limit of 0.68 nM. Moreover, this assay shows high selectivity over other cations and biomolecules. Besides, hemin can be detected in serum samples with satisfactory results, which demonstrates its great potential for real sample analysis. The designed assay is free of any fluorescent nanomaterials or probes by just mixing these reagents together, and all the detection processes is performed within 20 min. Therefore, it can offer a simple, highly sensitive and selective sensing platform for fluorescence detection of hemin.

Redox-inactive metal ions can modulate the reactivity of redox-active metal ions in a variety of biological and chemical oxidations. Many synthetic models have been developed to help address the elusive roles of these redox-inactive metal ions. Using a non-heme manganese(II) complex as the model, the influence of redox-inactive metal ions as a Lewis acid on its catalytic efficiency in oxygen atom transfer was investigated. In the absence of redox-inactive metal ions, the manganese(II) catalyst is very sluggish, for example, in cyclooctene epoxidation, providing only 9.9% conversion with 4.1% yield of epoxide. However, addition of 2 equiv. of Al3+ to the manganese(II) catalyst sharply improves the epoxidation, providing up to 97.8% conversion with 91.4% yield of epoxide. EPR studies of the manganese(II) catalyst in the presence of an oxidant reveal a 16-line hyperfine structure centered at g = 2.0, clearly indicating the formation of a mixed valent di-μ-oxo-bridged diamond core, MnIII-(μ-O)2-MnIV. The presence of a Lewis acid like Al3+ causes the dissociation of this diamond MnIII-(μ-O)2-MnIV core to form monomeric manganese(IV) species which is responsible for improved epoxidation efficiency. This promotional effect has also been observed in other manganese complexes bearing various non-heme ligands. The findings presented here have provided a promising strategy to explore the catalytic reactivity of some di-μ-oxo-bridged complexes by adding non-redox metal ions to in situ dissociate those dimeric cores and may also provide clues to understand the mechanism of methane monooxygenase which has a similar diiron diamond core as the intermediate.

Artemisinin (ART) is a type of frontline drug to treat drug-resistant falciparum malaria. Simple, accurate and selective determination of ART is significant to monitor its clinical pharmaceutical efficacy. Herein, a new ratiometric fluorescence method has been designed for the determination of ART with Zn-MOF as fluorescence reference and hemin as catalyst, respectively. Zn-MOF possesses intrinsic fluorescence at 443 nm owing to 2-aminoterephthalic acid ligand. When o -phenylenediamine (OPD) is mixed with hemin, a weak fluorescent signal at 570 nm ascribed to oxidized product of OPD (oxOPD) is observed. In the presence of ART, hemin can catalyze ART to break its peroxide bridge and release a large number of reactive oxygen species, which effectively oxidize OPD into luminescent oxOPD. Therefore, the fluorescence at 570 nm is enhanced significantly while the fluorescence of Zn-MOF remains basically unchanged. Thus, a ratiometric fluorescence sensing platform has been constructed for the detection of ART. This method exhibits wider linear range (0.15 μM-150 μM) with detection limit of 50 nM. This novel and selective method has been used to detect ART in compound naphthoquinone phosphate tablets.

Tetracyclines are one class of widely used antibiotics. Meanwhile, due to abuse and improper disposal, they are often detected in wastewater, which causes a series of environmental problems and poses a threat to human health and safety. As an efficient and environmentally friendly method, enzymatic catalysis has attracted much attention. In previous studies, we have designed an efficient peroxidase (F43Y/P88W/F138W Mb, termed YWW Mb) based on the protein scaffold of myoglobin (Mb), an O2carrier, by modifying the heme active center and introducing two Trp residues. In this study, we further applied it to degrade the tetracycline antibiotics. Both UV-Vis and HPLC studies showed that the triple mutant YWW Mb was able to catalyze the degradation of tetracycline, oxytetracycline, doxycycline, and chlortetracycline effectively, with a degradation rate of ~100%, ~98%, ~94%, and ~90%, respectively, within 5 min by using H2O2as an oxidant. These activities are much higher than those of wild-type Mb and other heme enzymes such as manganese peroxidase. As further analyzed by UPLC-ESI-MS, we identified multiple degradation products and thus proposed possible degradation mechanisms. In addition, the toxicity of the products was analyzed by using in vitro antibacterial experiments ofE. coli. Therefore, this study indicates that the engineered heme enzyme has potential applications for environmental remediation by degradation of tetracycline antibiotics. Keywords:heme enzyme;myoglobin;antibiotics;degradation;environmental remediation

Chemiluminescence (CL)-based analytical methods utilize luminophores that need to be activated with an oxidizing agent to trigger CL emission. Despite its susceptibility to decomposition when exposed to external light or trace metals, hydrogen peroxide (H2O2) has been widely used to develop chemiluminescent methods due to the limited number of suitable alternatives for activating chemiluminescent luminophores. Also, analytical methods based on the well-known luminol/H2O2 CL system have low sensitivity. Dissolved oxygen (DO) is a naturally abundant and environmentally benign alternative oxidant for luminol and other CL luminophores. However, DO alone is inactive and needs an efficient catalyst or a coreaction accelerator for its activation. Because of the narrow bandgap of VS4 (ca. 1.12 eV), it can facilitate fast electron-transfer kinetics with an acceptor molecule such as DO. Here, we introduce vanadium tetrasulfide (VS4) to boost CL for the first time. Under the optimized conditions, VS4 nanodendrite catalyzes the generation of reactive oxygen species by activating DO which subsequently reacts with luminol to generate intense CL. It enhances the CL intensity of luminol/DO by about 10,000 times. Surprisingly, hemin remarkably quenches the generated CL of luminol/DO/VS4 nanodendrites, which is completely opposite to its typical enhancement of luminol CL. Based on the remarkable concentration-dependent quenching of the luminol/DO/VS4 nanodendrite CL by hemin, we have developed a sensitive CL method that can selectively detect hemin in the linear concentration range of 1–250 nM and achieved a limit of detection of 0.11 nM. The practical utility of the developed method was demonstrated by the determination of hemin in a pharmaceutical drug for the treatment of acute intermittent porphyria and in human serum. This study demonstrates that VS4 holds great promise in analytical method development.

The detection of bisphenol A (BPA) was carried out using several simplified electrochemical sensors in which electroactive materials Heme, dopamine (DA), β-cyclodextrins (β-CD) or Fc-ECG were deposited on screen-printed electrodes (SPEs) through one-step. The preparation of the developed sensors were verified by SEM, CV and DPV. Results shown that Heme/SPE sensor demonstrated notable electro-catalytic capabilities and a greater current response than other sensors for BPA detection and optimal operating conditions were determined. The current response vs concentration of BPA showed a linear relationship in the range of 0.05–0.50 μmol·l−1, the concentration limit of detection (S/N = 3) as 5.0 nmol·l−1 and 0.0013 μA·μM−1·cm−2 as the sensitivity of detection. There were no interferences arising from common elements that relate to BPA. Heme/SPE was applied for BPA detection in bottle drinking water, tap water and lake water; findings showed a total R.S.D. of less than 3.1% and average recovery from 95.0 to 104.0%. These results showed that the actual application of BPA detection was rapid and precise when placed in environmental water and even useful for safety verification.

Fluorescent carbon dots (CDs) are promising zero-dimensional carbon nanomaterials with semiconductor-like properties, which have attracted tremendous attention owing to their fascinating features such as excellent optical performance, chemical inertness, and low toxicology. In this investigation, Mg-doped CDs (Mg-CDs) with up- and down-conversion fluorescence (FL) were prepared within 2 min through a one-pot, facile and ultrafast microwave method. The obtained Mg-CDs showed excitation-independent blue emission and were potentially applied for the detection of hematin based on the strong inner filter effect (IFE). The FL intensity ratio ( I 0 / I ) of the synthesized Mg-CDs were proportional to the concentration of hematin in the range of 0.1–75 μM with the limit of detection of 25 nM for Mg-CDs-0.1 and 0.01–25 μM with the limit of detection of 1 nM for Mg-CDs-0.5. Overall, this selective and sensitive assay using Mg-CDs obtained through the current microwave method was believed to demonstrate significant application prospects in complex matrices owing to its simplicity and specificity for hematin.

This study established a novel flow injection (FI) methodology for the determination of phenol in aqueous samples based on luminol chemiluminescence (CL) detection. The method was based on the inhibition that phenol caused on the hemin-catalysed chemiluminescence reaction between luminol and hydrogen peroxide in alkaline solution. Optimum conditions and possible mechanisms have been investigated. The linear range was 2.0 × 10 −9 to 4.0 × 10 −7 g mL −1 for phenol. The proposed method is sensitive with a detection limit of 4.0 × 10 −10 g mL −1 . The relative standard deviation for 11 measurements was 2.3% for 1.0 × 10 −7 g mL −1 phenol. The method was applied for the determination of phenol in waste water samples. The results obtained compared well with those by an official method.
