Soil minerals and organic matters affect ARGs transformation by changing the morphology of plasmid and bacterial responses

Hongyu Shi,Xinyi Hu,Jin Zhang,Wenxuan Li,Jiang Xu,Baolan Hu,Liping Ma,Liping Lou

Soil environment is a vital place for the occurrence and spread of antibiotic resistant bacteria (ARB) and antibiotic resistance genes (ARGs). Extracellular DNA-mediated transformation is an important pathway for ARGs horizontal transfer and widely exists in soil environment. However, little information is available on how common soil components affect ARGs transformation. Here, three minerals (quartz, kaolinite , and montmorillonite) and three organic matters (humic acid, biochar, and soot) were selected as typical soil components. A small amount in suspension (0.2 g/L) of most soil components (except for quartz and montmorillonite) promoted transformant production by 1.1–1.6 folds. For a high amount (8 g/L), biochar significantly promoted transformant production to 1.5 times, kaolinite exerted a 30 % inhibitory effect. From the perspective of plasmid, biochar induced a higher proportion of supercoiled plasmid than kaolinite; more dissolved organic matter and metal ions facilitated plasmid aggregation under the near-neutral pH, thus promoted transformation. As for the influence of materials on recipient, although biochar and kaolinite both increased reactive oxygen species (ROS) level and membrane permeability, biochar up-regulated more ROS related genes, resulting in intracellular ROS production and up-regulating the expression of carbohydrate metabolism and transformation related genes. While kaolinite inhibited transformation mainly by causing nutrient deficiency.

Effects of zero-valent iron (ZVI) on nitrogen conversion, transformation of sulfamethoxazole (SMX) and abundance of antibiotic resistance genes (ARGs) in aerobic granular sludge process

Feiyu Zhang,Shuqian Cheng,Jinbo Zhao,Anjie Li

Even after pre-treatment, livestock and poultry wastewater still contain high concentrations of ammonia and residual antibiotics. These could be removed economically using the aerobic granular sludge (AGS) process with zero-valent iron (ZVI). The interaction of antibiotics and nitrogen in this process needs to be clarified and controlled, however, to achieve good removal performance. Otherwise, antibiotics might generate transformation products (TPs) with higher toxicity and lead to the emergence of antibiotic-resistant bacteria carrying antibiotic resistance genes (ARGs), which could cause persistent toxicity and the risk of disease transmission to the ecological environment. This study investigated the impact of ZVI on AGS for nitrogen and sulfamethoxazole (SMX) removal. The results show that AGS could maintain good ammonia removal performance and that the existence of SMX had a negative impact on ammonia oxidation activities. ZVI contributed to an increase in the abundance of nitrite oxidation bacteria, denitrifying bacteria and the functional genes of nitrogen removal. This led to better total nitrogen removal and a decrease in N 2 O emission. Accompanied by biological nitrogen transformation, SMX could be transformed into 14 TPs through five pathways. ZVI has the potential to enhance transformation pathways with TPs of lower ecotoxicity, thereby reducing the acute and chronic toxicity of the effluent. Unfortunately, ZVI might enhance the abundance of sul1 , sul2 , and sul3 in AGS, which increases the risk of sulfonamide antibiotic resistance. In AGS, Opitutaceae , Xanthomonas , Spartobacteria and Mesorhizobium were potential hosts for ARGs. This study provides theoretical references for the interaction of typical antibiotics and nitrogen in the biological treatment process of wastewater and bioremediation of natural water bodies.

Implications for mitigation of antibiotic resistance: Differential response of intracellular and extracellular antibiotic resistance genes to sludge fermentation coupled with thermal hydrolysis

Chen Cai,Xuesong Hui,Wan Yang,Yu Hua,Huiling Liu,Xiaohu Dai

Thermal hydrolysis pretreatment (THP) can effectively remove the antibiotic resistance genes (ARGs) from sewage sludge, but the rebounding effects in the subsequent anaerobic fermentation are often observed. The purpose of this study was to elucidate the distribution and fate of intracellular and extracellular ARGs (iARGs and eARGs) in the sludge acidogenic fermentation process coupled with THP. Our results revealed that THP significantly reduced the absolute abundance of total ARGs in raw sludge but increased eARGs by 0.3–1.4 log units under practical conditions (140 °C for 30 min). There is no significant difference in the removal of total ARGs between the two acidogenic fermenters receiving raw and hydrolyzed sludge, with iARGs prevailing in the produced biosolids. The succession of bacterial community and the co-occurrence relationships among ARG type, mobile genetic elements and bacterial taxa were observed, suggesting a phylogenetic basis for the iARGs patterns in fermented sludge. However, eARGs were susceptible to biodegradation with a half-life of 2.34 h and they contributed limitedly to the ARGs propagation through transformation. These findings suggest an emphasis on the mitigation of iARGs during the acidogenic fermentation of sludge, which would be achieved by lowering the richness and physicochemical destruction of potential hosts.

Selectively enrichment of antibiotics and ARGs by microplastics in river, estuary and marine waters

Shanshan Wang,Nana Xue,Wenfeng Li,Daoyong Zhang,Xiangliang Pan,Yongming Luo

The partition of antibiotics and antibiotic resistant genes (ARGs) between the microplastics (MPs) and the surrounding water with various salinity are still unclear. In this study, we hypothesized that adsorption of antibiotics on MPs might cause a significant change of the structure of microbial communities, diversity and abundance of ARGs on MPs and this might be further affected by change of salinity. In this study, we investigated adsorption of four common antibiotics (sulfamerazine, tetracycline, chloramphenicol and tylosin) to polyethylene (PE) MPs in river, estuary and marine waters, and the differences of antibiotic resistant genes (ARGs) and bacterial communities on MPs and in the three waters. The results showed that MPs can enrich antibiotics, ARGs and microbes from the surrounding water. Elevated salinity could reduce adsorption of antibiotics to MPs and the abundance of ARGs. For example, MPs can concentrate more antibiotics and ARGs in the fresh river water than in the estuary and the marine waters. In addition, ARGs and bacterial communities on MPs at various salinity were significantly different under the pressure of four antibiotics. On MPs, sul1, sulA/folP-01, tetA, tetC, tetX and ermE increased significantly but a few new ARGs such as sulA/folP-01 and tetA appeared. The structure of the bacterial communities on MPs was different from the surrounding water since some bacteria species found on MPs were barely detected in the surrounding water while some genera on MPs vanished after exposure to antibiotics. As the antibiotics adsorbed and the ARGs on MPs decreased with the water salinity, the structure of the communities on MPs thus varied with salinity change. These findings are important to understand the effects of MPs on the transport, fate and ecological risk of antibiotics and ARGs in different aquatic environments.

Effects of voltage and tetracycline on horizontal transfer of ARGs in microbial electrolysis cells

Chenxi Zhang,Zouafi Saad,Shuai Zhang,Bin Chen,Xin He,Shuai Liu

The abuse of antibiotics leads to the production of antibiotic-resistant bacteria (ARB) and antibiotic resistance genes (ARGs). Microbial electrolysis cells (MECs) have been widely applicated in the field of degrading antibiotics. ARGs were increased via horizontal transfer in single and two-chamber MECs. As one of the critical parameters in MECs, voltage has a particular impact on the ARGs transfer via horizontal transfer. However, there have been few studies of ARGs transfer under the exposure of antibiotics and voltage in MECs. In this study, five concentrations of tetracycline (0, 1, 5, 10, 20 mg/L) were selected to explore the conjugative transfer frequency of plasmid-encoded the ARGs from the donor (E. coli RP4) to receptor (E. coli HB101) in MECs, two voltages (1.5 and 2.0 V) were used to explore the conjugative transfer frequency of ARGs in MECs, then, the transfer of ARGs in MECs under the co-effect of tetracycline and voltage was explored. The results showed that the conjugative transfer frequency of ARGs was significantly increased with the increase of tetracycline concentration and voltage, respectively (p < 0.05). Under the pressure of tetracycline and voltage, the conjugative transfer frequency of ARGs is significantly enhanced with the co-effect of tetracycline and voltage (p < 0.05). The oxidative response induced by electrical stimulation promotes the overproduction of reactive oxygen species and the enhancement of cell membrane permeability of donor and recipient bacteria in MECs. These findings provide insights for studying the spread of ARGs in MECs.

Antibiotic sulfadiazine degradation by persulfate oxidation: Intermediates dependence of ecotoxicity and the induction of antibiotic resistance genes

Heshan Zheng,Yunfei Zhang,Shuo Li,Xiaochi Feng,Qinglian Wu,Yoong Kit Leong,Jo-Shu Chang

To preserve the water resources, this study has analyzed the ecotoxicity and antibiotic resistance genes (ARGs) induction capacity of sulfadiazine degradation intermediates resulting from persulfate activation oxidation enhanced by ultraviolet, ultrasound and microwave. The five degradation pathways caused by the contribution discrepancy of electron transfer and singlet oxygen ( 1 O 2 ) and variations in the ecotoxicity of different degradation products were analyzed. Microcosm experiment exhibited that the microbial community in actual water changed significantly with SDZ and degradation intermediates, in which the dominant genera were Aeromonas , Cupriavidus , Elizabethkingia and Achromobacter . Except for the selective pressure on bacteria, the degradation intermediates also exert a certain degree or even stronger induction on sulfonamide ARGs ( sul4 , sul1 and sul2 ) than SDZ. Furthermore, the potential hosts for sulfonamide ARGs were revealed by network analysis. These results provide a better understanding of antibiotics degradation mechanism and ARGs occurrence, which is useful for controlling the spread of ARGs.

Effects of Perfluoroheptanoic Acid (PFHpA) on Bacterial Community, Elemental Cycling and ARGs Transmission in Sediment

Shi Shengnan,Su Xinran,Shi Linna,Li Shuang,Pu Yunhong,Zeng Qianzhi,Yang Bowen,Wu Yaxuan,Sun Yue,Ma Yulian,Yang Xinyu,Wang Xin,Gong Zheng

As a substitute for perfluorooctanoic acid (PFOA), perfluoroheptanoic acid (PFHpA) has been detected in various environments, while its impact on sediment bacteria was unclear. PFHpA exposure altered the whole and the rare bacterial community structure in sediment. Concurrently, PFHpA selectively enriched bacteria involved in C/N/P/S cycling. PFHpA induced intracellular reactive oxygen species (ROS) overproduction, triggering both the antioxidant defense system and the SOS response. PFHpA upregulated functional gene and enzymatic activities involved in C/N/P cycling, while downregulated those involved in S cycling. Based on the co-occurrence patterns between ARGs and MGEs, it might be speculated that HGT potential was increased upon PFHpA exposure. PLS-PM analysis indicated that the rare bacteria had more significant and direct effects on C/N/P/S cycling and transmission of ARGs. These findings will prove useful for understanding the effects of PFHpA on bacterial community, C/N/P/S cycling and transmission of ARGs in sediment.

Mechanisms of metabolic performance enhancement and ARGs attenuation during nZVI-assisted anaerobic chloramphenicol wastewater treatment

Jiahuan Li,Ning Guo,Shan Zhao,Juan Xu,Yunkun Wang

Anaerobic wastewater treatment is a promising technology for refractory pollutant treatment. The nano zero-valent iron (nZVI) assisted anaerobic system could enhance contaminant removal. In this work, we added nZVI into an anaerobic system to investigate the effects on system performances and metabolic mechanism for chloramphenicol (CAP) wastewater treatment. As nZVI concentrations increased from 0 to 1 g/L, the CAP removal efficiency was appreciably improved from 46.5% to 99.2%, while the CH 4 production enhanced more than 20 times. The enhanced CAP removal resulted from the enrichments of dechlorination-related bacteria ( Hyphomicrobium ) and other functional bacteria (e.g., Zoogloea, Syntrophorhabdus ) associated with refractory contaminants degradation. The improved CH 4 production was ascribed to the increases in fermentative-related bacteria ( Smithella and Acetobacteroides ), homoacetogen ( Treponema ), and methanogens . The increased abundances of anaerobic functional genes further verified the mechanism of CH 4 production. Furthermore, the abundances of potential hosts of antibiotic resistance genes (ARGs) were reduced under high nZVI concentration (1 g/L), contributing to ARGs attenuation. This study provides a comprehensive analysis of the mechanism in metabolic performance enhancement and ARGs attenuation during nZVI-assisted anaerobic CAP wastewater treatment.

Phytoremediation of antibiotics and antibiotic resistance genes (ARGs) by submerged macrophytes: Synergistic removal and mechanistic insights

Huimin Wei,Lu Zhang,Yujie Mao,Yiting Cheng,Cong Wang,Zhi Wang

Antibiotic resistance genes (ARGs) pose a major threat to human disease control and have emerged as a critical global public health concern. While aquatic plants are widely recognized for removing antibiotics, their efficacy in co-removing ARGs and underlying mechanisms remain poorly understood. To fill this gap, we systematically evaluated the removal effects and mechanisms of enrofloxacin (ENR), tetracycline (TC), sulfamethoxazole (SMX) and associated ARGs and mobile genetic elements (MGEs) by Hydrilla verticillata , a widespread aquatic macrophyte. Compared with controls without plant, H. verticillata significantly enhanced TC, ENR, and SMX removal efficiencies by 7.8 %, 38.1 %, and 8.5 %, respectively. Concurrently, it reduced the abundance of ARGs and MGEs by 7.21–86.8 %, confirming its novel dual co-removal function. Notably, plant uptake contributed only 0.03–6.3 % to antibiotic removal. Instead, removal relied on H. verticillata -mediated microbial regulation: mitigating antibiotic-induced microbial detriment, maintaining diversity, and enriching degrading bacteria. We further found that aquatic plants alleviated the risk of ARG dissemination by enhancing microbial diversity, narrowing the potential host range, and simplifying ARG transmission networks. Critically, partial least squares path modeling revealed that MGEs were the primary drivers of ARGs dynamics in treatments with plant (path coefficient: 0.840), whereas antibiotic selection pressure dominated without plant (path coefficient: 0.875). These findings highlight the dual role of H. verticillata in antibiotic removal and ARG suppression, providing novel insights into phytoremediation as a sustainable strategy for restoring antibiotic-contaminated aquatic environments and combating antimicrobial resistance.

New insights into the anaerobic digestion of high carbon wastewater with ciprofloxacin: methane production and ARGs inhibition

Qingmuke Su,Yaguang Du,Dongyun Du,Tian C. Zhang

Ciprofloxacin (CIP), as a quinolone antibiotic, has broad-spectrum antibacterial properties and can affect methanogenic performance in anaerobic digestion (AD). While previous studies focused on synthetic wastewater, the fate of CIP in real distillery wastewater (RDW) and its impact on microbial adaptation mechanisms remain unclear (such as biotransformation pathways, population dynamics, and the enzymes involved) in RDW is largely unclear. In this study, we investigated AD performance, metabolic pathways, and antibiotic resistance gene (ARG) dynamics using real wastewater spiked with CIP (0.3–2 mg/L). Results indicate that 0.5 mg/L CIP (631.83 mL CH 4 /g·VS) enhanced the methane yield by 6.67 % (592.34 mL CH 4 /g·VS in control), correlating with upregulated enzyme in glycosis, TCA cycle, and methanogenesis (F 420 increased). With full use of short-chain acids, transient volatile fatty acid (VFA) inhibition (≤ 50 mg/L on Day 3) was overcome by Day 10. Metagenomics revealed CIP promoted the production of stress proteins (e.g., cysteine synthase activity doubled). Furthermore, CIP (0.3–1 mg/L) suppressed mobile genetic elements (MGEs) encoding horizontal gene transfer, including isfinder (15.15 %) and integrases (6.25 %), while ARG and virulence factor abundances remained unchanged versus control. This study firstly shows that low-dose CIP in RDW increases methanogenesis via metabolic adaptation without exacerbating ARG risks. MGE suppression implies that CIP may lessen the possibility of ARG diffusion in AD systems. These results offer vital information for improving AD performance in the treatment of wastewater contaminated by antibiotics and developing methods to strike a balance between antibiotic removal and ARG control.

Changes in tetracycline partitioning and bacteria/phage-comediated ARGs in microplastic-contaminated greenhouse soil facilitated by sophorolipid

Mingming Sun,Mao Ye,Wentao Jiao,Yanfang Feng,Pingfeng Yu,Manqiang Liu,Jiaguo Jiao,Xiaojia He,Kuan Liu,Yuanchao Zhao,Jun Wu,Xin Jiang,Feng Hu

The emerging mixed contamination of antibiotics and microplastics in greenhouse soil has made the control of antibiotic resistant gene (ARG) transmission a novel challenge. In this work, surfactant sophorolipid was applied to enhance the dissipation of tetracycline (TC) and tet genes in the presence of microplastics in greenhouse soil. During 49   days of incubation, soil bacteria and phages were both found to be the crucial reservoirs of ARGs. Meanwhile, microplastic’s presence significantly inhibited the dissipation of TC and ARGs in the soil. However, sophorolipid application was proved to outweigh the negative impact caused by microplastic existence, and lead to the highest dissipation of soil TC and ARGs. Significant positive correlation was detected between the dissipation rate of water-soluble and exchangeable TC content and bacteria/phage co-mediated ARG levels. This also held true between the two fractions of soil TC and the ratio of ARG level in the bacteria to that in the phages (B ARGs /P ARGs ). The opposite impacts of microplastic presence and sophorolipid amendment on the TC/ARG dissipation found in this work provides new information for understanding ARG transmission between bacteria and phages in the mixed contaminated greenhouse soil.

Application of the quorum sensing inhibitor to improve ARGs removal by membrane-based household drinking water treatment process

Jingqing Liu,Hening Cui,Tuqiao Zhang,Xiaowei Liu,Lili Wang

Antibiotic resistance genes (ARGs) have become an increasing threat to human health. Membrane-based technologies have been applied to address multiple concerns regarding the quality of household drinking water (HDW), including ARGs. However, the long-term operation of membrane-based HDW purifiers would inevitably lead to biofouling, making the membrane turn from cleaner to a secondary pollution source. This study proposed a strategy based on quorum sensing inhibitor (QSI) to control ARGs in the finished water of HDW purifiers. Three reported QSIs, vanillin (VL), cranberry extracts (CBETs), and tannic acid (TA), were selected to investigate their effects on the removal of ARGs by typical HDW purifiers, and the corresponding influencing mechanism was elucidated. The results indicated that ARGs leakage occurred in the absence of QSI but disappeared with the addition of VL, CBETs, and TA. The introduction of VL increased the guarantee rate of ≥ 3 log ARGs removal increased from 63.3 to 70% (control group) to 88.9–100%. The influencing mechanism of QSIs on ARGs removal by membrane filtration can be attributed to direct effect (direct inactivation of the ARGs host) and indirect effects (adjusting the physiochemical properties of the membrane surface and bioaccumulation). This study provides data on the regulatory mechanisms of biofouling induced by QSI and proposes a new strategy for ARGs control by membrane-based processes.
