Gut Bifidobacterium pseudocatenulatum protects against fat deposition by enhancing secondary bile acid biosynthesis

Andong Zha,Ming Qi,Yuankun Deng,Hao Li,Nan Wang,Chengming Wang,Simeng Liao,Dan Wan,Xia Xiong,Peng Liao,Jing Wang,Yulong Yin,Bi'e Tan

Gut microbiome is crucial for lipid metabolism in humans and animals. However, how specific gut microbiota and their associated metabolites impact fat deposition remains unclear. In this study, we demonstrated that the colonic microbiome of lean and obese pigs differentially contributes to fat deposition, as evidenced by colonic microbiota transplantation experiments. Notably, the higher abundance of Bifidobacterium pseudocatenulatum was significantly associated with lower backfat thickness in lean pigs. Microbial-derived lithocholic acid (LCA) species were also significantly enriched in lean pigs and positively correlated with the abundance of B. pseudocatenulatum . In a high-fat diet (HFD)-fed mice model, administration of live B. pseudocatenulatum decreased fat deposition and enhances colonic secondary bile acid biosynthesis. Importantly, pharmacological inhibition of the bile salt hydrolase (BSH), which mediates secondary bile acid biosynthesis, impaired the anti-fat deposition effect of B. pseudocatenulatum in antibiotic-pretreated, HFD-fed mice. Furthermore, dietary LCA also decreased fat deposition in HFD-fed rats and obese pig models. These findings provide mechanistic insights into the anti-fat deposition role of B. pseudocatenulatum and identify BSH as a potential target for preventing excessive fat deposition in humans and animals. Graphical This figure highlights how gut microbiota and bile acids regulate fat deposition in lean and obese pigs. Lean pigs showed increased microbial-derived lithocholic acid (LCA) species and muricholic acid species, closely linked to the abundance of Bifidobacterium pseudocatenulatum . In contrast, obese pigs exhibited gut microbiota favoring hyodeoxycholic acid species accumulation. In high-fat diet-fed mice, live B. pseudocatenulatum supplementation enhances secondary bile acid synthesis, especially LCA production, and reduces fat mass, while bile salt hydrolase (BSH) inhibition negates these effects. Dietary LCA similarly lowers fat deposition in obese pigs and rats. These findings provide mechanistic insights into the anti-fat deposition role of B. pseudocatenulatum and identify BSH as a potential target for preventing excessive fat deposition in humans and animals. Highlights The gut microbiota composition and function differ significantly between lean and obese pigs, and breed-specific microbiomes regulate fat deposition. Gut Bifidobacterium pseudocatenulatum protects against fat deposition by enhancing secondary bile acid biosynthesis. Dietary supplementation with lithocholic acid also helps prevent fat deposition.

Extending Viability of Bifidobacterium longum in Chitosan-Coated Alginate Microcapsules Using Emulsification and Internal Gelation Encapsulation Technology

Ji Rui,Wu Jiahui,Zhang Junliang,Wang Tao,Zhang Xudong,Shao Lei,Chen Daijie,Wang Jian

Bifidobacteria are considered one of the most important intestinal probiotics because of their significant health impact. However, this ability is usually limited by gastrointestinal fluid and temperature sensitivity. Emulsification and internal gelation is an encapsulation technique with great potential for probiotic protection during storage and the gastrointestinal transit process. This study prepared microcapsules using an emulsification and internal gelation encapsulation method with sodium alginate, chitosan, and Bifidobacterium longum as wall material, coating material, and experimental strain, respectively. Optical, scanning electron, and focal microscopes were used to observe the microcapsule surface morphology and internal viable cell distribution, and a laser particle size analyzer and zeta potentiometer were used to evaluate the chitosan-coating characteristics. In addition, microcapsule probiotic viability after storage, heat treatment, and simulated gastrointestinal fluid treatment were examined. Alginate microcapsules and chitosan-coated alginate microcapsules both had balling properties and uniform bacterial distribution. The latter kept its balling properties after freeze-drying, verified by scanning electronic microscopy (SEM), and had a clear external coating, observed by an optical microscope. The particle size of chitosan-coated alginate microcapsules was slightly larger than the uncoated microcapsules. The zeta potential of alginate and chitosan-coated alginate microcapsules was negative and positive, respectively. Heat, acid and bile salt tolerance, and stability tests revealed that the decrease of viable cells in the chitosan-coated alginate microcapsule group was significantly lower than that in uncoated microcapsules. These experimental results indicate that the chitosan-coated alginate microcapsules protect B. longum from gastrointestinal fluid and high-temperature conditions.

Akkermansia muciniphila and Bifidobacterium bifidum Prevent NAFLD by Regulating FXR Expression and Gut Microbiota

Fulin Nian,Longyun Wu,Qiaoyun Xia,Peiying Tian,Chunmei Ding,Xiaolan Lu

Background and aims Non-alcoholic fatty liver disease (NAFLD) is closely associated with gut microbiota and has become the most common chronic liver disease worldwide, but the relationship between specific strains and NAFLD has not been fully elucidated. We aimed to investigate whether Akkermansia muciniphila and Bifidobacterium bifidum could prevent NAFLD, the effects of their action alone or in combination, possible mechanisms, and modulation of the gut microbiota. Methods Mice were fed with high-fat diets (HFD) for 20 weeks, in which experimental groups were pretreated with quadruple antibiotics and then given the corresponding bacterial solution or PBS. The expression of the glycolipid metabolism indicators, liver, and intestinal farnesol X receptors (FXR), and intestinal mucosal tight junction proteins were detected. We also analyzed the alterations of inflammatory and immune status and the gut microbiota of mice. Results Both strains were able to attenuate mass gain ( p <0.001), insulin resistance ( p <0.001), and liver lipid deposition ( p <0.001). They also reduced the levels of the pro-inflammatory factors ( p <0.05) and the proportion of Th17 ( p <0.001), while elevating the proportion of Treg ( p <0.01). Both strains activated hepatic FXR while suppressing intestinal FXR ( p <0.05), and elevating tight junction protein expression ( p <0.05). We also perceived changes in the gut microbiota and found both strains were able to synergize beneficial microbiota to function. Conclusions Administration of A. muciniphila or B. bifidum alone or in combination was protective against HFD-induced NAFLD formation and could be used as alternative treatment strategy for NAFLD after further exploration.

Glycyrrhetinic acid ameliorates gastric mucosal injury by modulating gut microbiota and its metabolites via Thbs1/PI3K-Akt/p53 pathway

Zheng-Meng Jiang,Zhi-Yue Fang,Xing Yang,Xing-Xing Ji,Yuan-Yuan Zhao,Bin-Yan Lin,Ze-Bin Weng,E-Hu Liu

Background Dysbiosis of the gut microbiota is pivotal in the development of gastric mucosa injury (GMI). Glycyrrhetinic acid (GA) is a bioactive triterpenoid compound abundantly present in licorice roots. Although GA's potential in mitigating GMI is recognized, its precise mechanism remains elusive, particularly concerning the role of gut microbiota. Purpose This study aimed to explore the protective effects and mechanisms of GA in preventing HCl/ethanol-induced GMI in rats. Results This study demonstrated the protective effects of GA on gastric mucosa, evidenced by enhanced morphology and structure as revealed through H&E staining. Utilizing fecal microbiota transplantation, GA was found to significantly mitigate oxidative damage, inflammation, and expression of apoptosis-related genes in GMI rats by a gut microbiota-dependent mechanism. 16S rRNA sequencing and metabolomics profiling revealed that GA ameliorated HCl/ethanol-triggered intestinal dysbiosis and imbalances in sphingolipid, arginine, and tryptophan metabolism. By promoting the prevalence of Bifidobacterium longum subsp. infantis ( B. infantis ) in the gut microbiota, GA improved metabolic disturbances linked to injury. Furthermore, its action mechanism was related to the inhibition of the Thbs1/PI3K-Akt/p53 signaling pathway. Conclusion The administration of GA improves GMI by mitigating intestinal dysbiosis and fostering colonization of B. infantis . GA offers therapeutic potential for GMI by modulating the Thbs1/PI3K-Akt/p53 pathway, thus alleviating inflammatory responses associated with gut microbiota imbalance.

Production and analysis of exopolysaccharides from Bifidobacterium bifidum and Bifidobacterium animalis subsp. lactis: Genomic basis, structure, and prebiotic properties

Nanyu Tang,Tao Lin,Chang Xu,Xia Fan,Kai Ma,Changliang Zhang,Shuo Geng,Feng Ji,Xin Rui,Wei Li

This study investigates the production and prebiotic properties of exopolysaccharides (EPS) from Bifidobacterium bifidum 3469 (BBF-EPS) and B. animalis subsp. lactis 9749 (BAL-EPS). To eliminate interference from exogenous polysaccharides present in conventional MRS-c broth, EPS were produced using a modified medium (MRS-TY) lacking complex carbohydrate components, enabling accurate structural characterization of the intrinsically synthesized EPS. Whole-genome sequencing revealed fundamentally different eps biosynthetic gene clusters between the strains. The structural analysis indicated that the compact gene cluster of B. bifidum 3469 was associated with a simpler EPS (molecular weight 1.98 ×10 5 Da; consisting of glucose, galactose, rhamnose, and mannose in a 1.00:0.78:0.34:0.25 molar ratio), whereas the large, complex cluster of B. animalis subsp. lactis 9749 was associated with the production of a galactose-rich EPS (molecular weight 2.02 ×10 5 Da; consisting of galactose, rhamnose, glucose, and mannose in a 5.47:1.46:1.00:0.86 molar ratio). Both EPS supported growth of Lactobacillus strains. In vitro fecal fermentation demonstrated that BBF-EPS enriched Megamonas and Faecalibacterium , leading to increased propionic acid production (51.77 mM). In contrast, BAL-EPS enriched Bifidobacterium and Blautia , enhancing butyric acid (28.67 mM) production. These findings highlight the importance of culture medium optimization for reliable characterization of bifidobacterial EPS, supporting their potential application as targeted prebiotics for intestinal health.

Antibacterial Activity of Bifidobacterium breve Against Clostridioides difficile

Yang Jingpeng,Yang Hong

Bifidobacterium breve (YH68) is widely used in the fields of food fermentation and biomedicine. In this study, we explored the antibacterial activity of the cell free culture supernatant (CFCS) of YH68 against Clostridioides difficile ATCC 9689 (CD) by measuring multiple indexes, including the growth, spores production, toxin A/B production, and the expression levels of the tcdA and tcdB genes of CD. In addition, we examined the changes in major cellular functional groups, structures, permeability, integrity, and the proton motive force (PMF) of the cytoplasmic membrane. The results showed that double-dilution ratio of YH68-CFCS (3 × 109 CFU/mL) was the MIC value. The cell density, spores production, and the toxin production of CD treated with YH68-CFCS were lower than that of the control (p < 0.05). In addition, the gene expression levels of tcdA and tcdB in CD treated with YH68-CFCS were significant downregulated (p < 0.05). Marked differences were observed in the cell membrane and cell wall by a FT-IR spectroscopy and SEM. Analysis of the cell membrane permeability and integrity of the CD cells revealed that YH68-CFCS induced the leakage of a large amount of intracellular K+, inorganic phosphate, ATP, nucleic acids and proteinaceous substances. Furthermore, PMF analysis indicated that there was a significant change in Δψ and ΔpH. These findings demonstrated that the antibacterial activity of YH68-CFCS against CD involved the inhibition of growth, spore production, toxin production, and virulence genes expression; a consumption of PMF in the cytoplasmic membrane, the formation of pore in the cell membrane, together with the enhanced cell membrane permeability; and, eventually, cell completely disintegration.

Mechanism analysis of methionine inhibiting growth of Bifidobacterium bifidum

Zheming Gu,Xin Tang,Bingyong Mao,Qiuxiang Zhang,Jianxin Zhao,Wei Chen,Shumao Cui

Bifidobacterium bifidum rarely utilises yeast extract , which causes significant challenges in its industrial production. This study characterised the utilisation of amino acids by B. bifidum to improve the viability of the strain. In this study, the proliferation effect of B. bifidum in different nitrogen sources was determined, and the amino acid contents of different nitrogen sources were analysed, which indicated that hydrophobic amino acids may inhibit B. bifidum growth. Among different hydrophobic amino acids , methionine was identified as the strongest growth inhibitor. Furthermore, the intracellular pH and NH 4 + contents of B. bifidum CCFM16 were higher in methionine-free medium. Among the NH 3 -producing enzymes related to amino acid metabolism , the metC gene was significantly transcribed in methionine-free medium. In addition, tcyP , metB , metC and metE genes encoding cysteine transporter proteins and the conversion pathway of cysteine to methionine were transcribed 2.03-fold, 7.03-fold, 3.05-fold and 2.92-fold, respectively, in methionine-free medium compared to methionine-containing medium. Compared with methionine-containing medium, the activity of tricarboxylic acid cycle rate-limited enzyme isocitrate dehydrogenase in methionine-free medium also increased from 198.89 to 410.56 nmol/min/mg protein, which probably enhanced the carbon flux and promoted growth. Meanwhile, the activity of pyrroline-5-carboxylate reductase , an enzyme that synthesises proline , increased from 14.76 to 35.28 nmol/min/mg protein, which promoted proline synthesis in response to osmotic stress . Cyclopropane fatty acids were also significantly higher in methionine-free medium, which may promote growth by decreasing cell membrane permeability to protons and enhancing cell membrane fluidity . This study provides a theoretical basis for high-density culture of B. bifidum .

Bifidobacterium bifidum-Mediated Specific Delivery of Nanoparticles for Tumor Therapy

Yu Tang,Chun Chen,Binglei Jiang,Lu Wang,Fujie Jiang,Disen Wang,Yaotai Wang,Haiyan Yang,Xia Ou,Yan Du,Qi Wang,Jianzhong Zou

Purpose Hypoxia is considered to be obstructive to tumor treatment, but the reduced oxygen surroundings provide a suitable habitat for Bifidobacterium bifidum (BF) to colonize. The anaerobe BF selectively colonizes into tumors following systemic injection due to its preference for the hypoxia in the tumor cores. Therefore, BF may be a potential targeting agent which could be used effectively in tumor treatment. We aimed to determine whether a novel BF-mediated strategy, that was designed to deliver AP-PFH/PLGA NPs (aptamers CCFM641-5-functionalized Perfluorohexane (PFH) loaded poly(lactic-co-glycolic acid) (PLGA) nanoparticles) by aptamer-directed approach into solid tumor based on the tumor-targeting ability of BF, could improve efficiency of high intensity focused ultrasound (HIFU) treatment of breast cancer. Methods We synthesized AP-PFH/PLGA NPs using double emulsion method and carbodiimide method. Then, we evaluated targeting ability of AP-PFH/PLGA NPs to BF in vivo. Finally, we studied the efficacy of HIFU ablation based on BF plus AP-PFH/PLGA NPs (BF-mediated HIFU ablation) in tumor. Results The elaborately designed AP-PFH/PLGA NPs can target BF colonized in tumor to achieve high tumor accumulation, which can significantly enhance HIFU therapeutic efficiency. We also found that, compared with traditional chemotherapy, this therapy not only inhibits tumor growth, but also significantly prolongs the survival time of mice. More importantly, this treatment strategy has no obvious side effects. Conclusion We successfully established a novel therapy method, BF-mediated HIFU ablation, which provides an excellent platform for highly efficient and non-invasive therapy of tumor.

Bifidobacterium breve B2798 Attenuates Diet-Induced Obesity Through Gut-Adipose Crosstalk

Yue Zhao,Guoqiang Yao,Guangqi Gao,Lai-Yu Kwok,Jie Yu,Zhihong Sun

Obesity is driven by complex interactions between diet, gut microbiota, and host metabolism. Here, we investigate whether the probiotic Bifidobacterium breve B2798 mitigates high-fat diet (HFD)-induced metabolic dysfunction in C57BL/6J mice. Male mice received HFD for 12 weeks, followed by 12 weeks of B. breve B2798 or saline. Probiotic treatment significantly reduced body weight gain, epididymal and inguinal fat mass, and liver weight, and improved insulin sensitivity, without affecting glucose tolerance. Multi-omics integration revealed that B. breve B2798 partially reversed HFD-induced gut dysbiosis, notably decreasing Coriobacteriia and Deferribacteres while enriching Faecalibaculum rodentium . Fecal metabolomics showed elevated hydroxy fatty acids (e.g., 12-hydroxystearic acid) and modulation of lipid metabolism pathways. In epididymal adipose tissue, transcriptomics uncovered restored expression of Slc2a4 and Acsm3 , with gene set enrichment analysis highlighting enhanced fatty acid oxidation, thermogenesis, and oxidative phosphorylation. Integrated miRNA–mRNA analysis identified regulatory networks potentially mediating these effects. These findings demonstrate that B. breve B2798 ameliorates obesity-related metabolic disturbances via coordinated gut microbiota remodeling and adipose tissue reprogramming, supporting its potential as a microbiome-targeted therapeutic strategy.

Characterization of a novel Glutaminase-free L-asparaginase from Bifidobacterium thermophilum

Jiaping Ge,Xiaojun Huang,Zhengyu Zhang

L-asparaginase (ASNase), as a pivotal amidohydrolase enzyme, has been used in removing acrylamide in food processing and treating acute lymphoblastic leukemia in clinic. In this study, a novel ASNase from Bifidobacterium thermophilum (BtASNase) was successfully cloned and heterologously expressed in Escherichia coli. BtASNase was identified to share maximum 40% structural similarity with other ASNases in PDB database. The purified BtASNase with monomeric size about 35 kDa had the highest specific activity (554.82 IU/mg) at 55 °C and pH 8.0. Further investigation indicated that BtASNase showed great stability at wide pH range (6.0–11.0), and retained more than 85% of its activity for 50 min at 37 °C. To be noted, BtASNase exhibited high L-asparaginase specificity and zero glutaminase activity. To our knowledge, this is a novel exploration of ASNase from Bifidobacterium thermophilum, and the explored BtASNase could be a potential candidate with desirable advantages for overcoming limitations such as glutaminase activity, narrow pH range stability, and low thermostability restrict in industry applications of ASNase.

Photoaffinity probe-enabled discovery of sennoside A reductase in Bifidobacterium pseudocatenulatum

Yang Xu,Shujing Lv,Xiang Li,Chuanjia Zhai,Yulian Shi,Xuejiao Li,Zhiyang Feng,Gan Luo,Ying Wang,Xiaoyan Gao

Sennoside A (SA), a typical prodrug, exerts its laxative effect only after its transformation into rheinanthrone catalyzed by gut microbial hydrolases and reductases. Hydrolases have been identified, but reductases remain unknown. By linking a photoreactive group to the SA scaffold, we synthesized a photoaffinity probe to covalently label SA reductases and identified SA reductases using activity-based protein profiling. From lysates of an active strain, Bifidobacterium pseudocatenulatum ( B . pseudocatenulatum ), 397 proteins were enriched and subsequently identified using mass spectrometry. Among these proteins, chromate reductase/nicotinamide adenine dinucleotide phosphate (NADPH)-dependent flavin mononucleotide (FMN) reductase/oxygen-insensitive NADPH nitroreductase (nfrA) was identified as a potent SA reductase through further bioinformatic analysis and Uniprot database screening. We also determined that recombinant nfrA could reduce SA. Our study contributes to further illuminating mechanisms of SA transformation to rheinanthrone and simultaneously offers an effective method to identify gut bacterial reductases.

Bifidobacterium infantis Relieves Allergic Asthma in Mice by Regulating Th1/Th2

Wenlan Wang,Xiaoming Luo,Qin Zhang,Xujun He,Zhifang Zhang,Xinxin Wang

Background Bifidobacteria are among the probiotics used in treating intestinal diseases and are rarely used for allergic asthma treatment. The present study investigated the mechanism of B. infantis in treating allergic asthma in mice. Material/Methods A total of 40 male Balb/c mice were randomized into control, ovalbumin (OVA), montelukast (Mon), and B. infantis (B10) groups, and allergic asthma was induced in the OVA, Mon, and B10 groups. Airway reactivity was measured on day 29 by methacholine at various doses. The numbers of total cells and inflammatory cells in bronchoalveolar lavage fluid (BALF) were counted by blood cell counter and Diff-Quik staining. Hematoxylin-eosin (HE) staining was performed to observe inflammatory cell infiltration in lung tissues. Total IgE and OVA-specific IgE in serum were measured by ELISA. Mucin 5AC expression was detected by Western blot to evaluate airway obstruction. The levels of Th1 (IFN-γ, IL-2) and Th2 (IL-4, IL-5, IL-13) cytokines in BALF and tissues were detected by ELISA and qRT-PCR, respectively. Results The mice in the OVA group had airway hyperreactivity, while the symptoms in the B10 group and Mon group were effectively relieved. B10 reduced the number of inflammatory cells in BALF as well as inflammatory cell infiltration in tissues. Moreover, the levels of total serum IgE, OVA-specific IgE, and Mucin 5AC were increased in the OVA group, but were reduced in the Mon group and B10 group. B. infantis increased the levels of Th1 cytokines and decreased those of Th2 cytokines. Conclusions B. infantis can reduce the infiltration of inflammatory cells induced by OVA-specific antibodies in mice. B. infantis has therapeutic effects on allergic asthma by promoting Th1 and inhibiting Th2 immune responses.
