Single nanoparticle analysis for homogeneous immunoassay of CA19-9 for serological evaluation

Min Jiang,Zili Huang,Xin Zhao,Rui Liu,Yi Lv

Pancreatic cancer is a malignant tumour with high mortality. Surgery after diagnosis at the early stage is the most effective treatment. As the most effective biomarker of pancreatic cancer, CA19-9 has important clinical significance for both early diagnosis and surgical prediction. Herein, a single nanoparticle homogeneous immunoassay based on Au nanoparticle tagging was proposed for CA19-9 determination, with a dynamic range from 6.25 to 400 U mL−1 in intensity mode and 6.25 to 100 U mL−1 in frequency mode. The limits of detection were 0.72 U mL−1 (intensity mode) and 0.84 U mL−1 (frequency mode). Accurate results were obtained without any washing and separation procedure, and the matrix effect was avoided by monitoring both the intensity and frequency signal obtained from single nanoparticles. The method was successfully applied to the serological evaluation of patients. The results were well correlated with those measured by conventional clinical methods, demonstrating a promising foreground in the preoperative assessment of resectability and evaluation of disease recurrence.

Efficient electrochemical detection of pancreatic cancer marker CA19-9 using biosynthesized gold nanoparticles from mint extract for electrode modification

Xiaohui Wang,Yu Liu,Bude Su,Jinhua Tan,Jianxin Shen,Qi Chen,Yu Gou,Ying Zhang

This study presents a novel electrochemical biosensor for the detection of CA19–9, a crucial pancreatic cancer biomarker, utilizing gold nanoparticles (AuNPs) biosynthesized from mint extract. The green synthesis approach yielded well-dispersed, spherical AuNPs with a characteristic surface plasmon resonance peak at 520 nm. Modification of glassy carbon electrodes with these AuNPs resulted in a significant enhancement of electrochemical properties, including a 50 % increase in peak current and a 66.7 % reduction in charge transfer resistance. The biosensor exhibited a wide linear range from 0.1 to 100 U/mL with excellent linearity (R² = 0.998) and achieved a remarkably low limit of detection of 0.05 U/mL. The sensor demonstrated high specificity against common interfering substances and maintained stability over 100 cycles with less than 5 % decrease in performance. Compared to traditional ELISA methods, this biosensor offers rapid detection, minimal sample preparation, and superior sensitivity. The integration of biosynthesized AuNPs not only enhanced the sensor's performance but also aligned with sustainable practices in nanomaterial synthesis. This innovative approach paves the way for the development of sensitive, eco-friendly, and cost-effective diagnostic tools for early pancreatic cancer detection and monitoring.

4-Thiobenzoic acid-functionalized nanocomposite probes for surface-enhanced raman spectroscopy-based detection of CA19-9 in colorectal adenocarcinoma

Yinan Hu,Tao Liu,Bo Guo,Miao Ni,Yanrong Li

This study presents a novel surface-enhanced Raman spectroscopy (SERS) method for detecting CA19–9 in colorectal adenocarcinoma using 4-thiobenzoic acid (4-TBA) functionalized gold nanoparticles (nanospheres). The optimized SERS probes exhibited a maximum enhancement at pH 7 and 50 μM 4-TBA concentration, with stable signal achieved after 2 hours of incubation. The assay demonstrated a wide dynamic range of 1–100 U/mL, with a lower limit of quantification of 1.0 U/mL. Excellent selectivity was observed against common serum interferents, with normalized SERS intensities within 10 % of CA19–9 alone. Recovery studies in spiked serum samples yielded results ranging from 94.5 % to 105.2 %, indicating minimal matrix effects. Clinical validation using 70 serum samples (50 colorectal adenocarcinoma patients, 20 healthy volunteers) showed strong agreement with a commercial ELISA kit (Pearson's r = 0.987, p < 0.0001). Bland-Altman analysis revealed a mean bias of −0.37 U/mL between SERS and ELISA methods. The SERS-based approach offers advantages including rapid analysis (<1 hour), small sample volume requirements (5–10 μL), and potential for multiplexing. This method shows promise for early detection, disease monitoring, and treatment response evaluation in colorectal adenocarcinoma, with potential applications in both clinical and research settings.

A simple SERS sensor based on antibody-modified Fe3O4@Au MNPs for the detection of CA19-9 in CRC patients

Miaowen Xu,Gaoyang Chen,Yong Huang,Hangyu Song,Zheng Wu,Fengjuan Jiang,Lei Fu,Caili Bi,Xiaowei Cao,Wei Wei

This paper presents functionalized magnetic nanoparticles (Fe3O4@Au MNPs) combined with the surface-enhanced Raman spectroscopy (SERS) technique for sensitive detection of colorectal cancer (CRC) protein biomarker carbohydrate antigen 19-9 (CA19-9). Fe3O4@Au MNPs were constructed by the PEI-mediated seed growth method. Then, the signal molecule 5-5′-dithiobis (succinimidyl-2-nitrobenzoic acid) was used as a bridging agent to link CA19-9 antibody, and the SERS sensor was prepared. Using this sensor, detection of CA19-9 can be realized with only a one step sampling reaction. Fe3O4@Au MNPs combine the advantages of magnetic materials and noble metal nanoparticles, effectively amplifying the signal by creating numerous “hot spots” within the Au particle gaps and enhancing magnetic enrichment. Consequently, this approach lowers the detection limit (LOD) and enhances detection sensitivity. The ratio of characteristic peak intensities I1392/I1069 was selected for calculation, and a linear equation was constructed with a LOD as low as 0.27 U mL−1 by quantitative detections of the standard antigen. Finally, the sensor was used to analyze the clinical serum samples from CRC patients and healthy individuals, and the detection results were consistent with the actual results. This method exhibits notable advantages, including simplicity, high sensitivity, specificity, stability and reproducibility. It is expected to provide a new promising analytical method for clinical CA19-9 immunoassay.

Collard-like Bi2S3@Au nanocomposites-based label free electrochemical immunosensor for quantitative detection of CA19-9

Siyu Wu,Hongyun Ma,Xiaoyan Chen,Wen Zhong,Yingying Gu,Yuqing Miao,Yarui An

Here, a label-free immunosensor was conducted using rGO/Collard-like Bi 2 S 3 @Au nanocomposite system to detect carbohydrate antigen 19-9 (CA19-9) as a diagnostic biomarker for Pancreatic cancer. The unique morphology of Collard-like Bi 2 S 3 increases its specific surface area, which allows it to immobilize more materials and biomolecules on its surface. To realize the covalent bonding of bismuth-based materials with biomolecules, bismuth-based materials were functionalized by Au NPs. Besides, the superior electrochemical activity and large specific surface area of Bi 2 S 3 @Au improved the stability of the sensor, which made it an ideal candidate for regulating the sensing interface in this study. Meanwhile, reduced graphene oxide (rGO) with remarkable electrical conductivity to improve the current response of the sensing interface, thus broadening the linear range of the immunosensor. Based on the above materials, a label free electrochemical immunosensor was constructed for detecting CA19-9. Under optimal conditions, it displays good linear relationship within the range of 0.01 U mL −1 to 100 U mL −1 , the detection limit being 0.0012 U mL −1 . Additionally, the immunosensor prepared in this work is characterized by superior interference immunity, reproducibility and storage stability, and has yielded satisfactory recovery rate in testing the serum samples of patients.

Enzyme-amplified split-type photoelectrochemical immunoassay for quantitative monitoring of carbohydrate antigen 19–9 (CA19–9) based on Au-ZnIn2S4 heterojunction

Shuo Tian,Jiaping Lin,Wei Chen,Dianping Tang,Yang Ou

The development of rapid and sensitive CA19–9 (carbohydrate antigen 19–9) analytical methods for early screening of potential patients, especially in clinical situations, is important for controlling the progression of pancreatic cancer. Herein, a new photoelectrochemical (PEC) immunoassay has been devised for the ultrasensitive detection of CA19–9 using gold nanoparticle-modified ultrathin ZnIn 2 S 4 nanosheets (Au-UZNS) as the photoactive substrate. Notably, the alkaline phosphatase (ALP) labeled on the detection antibody (ALP-dAb) serves as the signal amplifier that catalyzes the production of large amounts of ascorbic acid (AA) after the sandwiched immunoreaction. The generated AA is an outstanding electron donor, thereby improving the intensity of the photoactive substrate. Under the optimized parameters, the Au-UZNS-based PEC immunosensor displays an excellent response to CA19–9 ranging from 0.01 to 500 U mL −1 with a limit of detection (LOD) of 4.7 mU mL −1 . Generally, the CA19–9 levels in the serum of high-risk pancreatic cancer patients range from 120 to 400 U mL −1 , while the levels in normal humans range from 0 to 40 U mL −1 . Therefore, the exceptionally low LOD value of the proposed method is sufficient to meet the threshold for clinical detection and holds significant potential for further application.

Electrochemical Immunoassay for Tumor Marker CA19-9 Detection Based on Self-Assembled Monolayer

Zheng Wei,Xiaoping Cai,Weifeng Cui,Junping Zhang

A CA19-9 electrochemical immunosensor was constructed using a hybrid self-assembled membrane modified with a gold electrode and applied to detect real samples. Hybrid self-assembled membranes were selected for electrode modification and used to detect antigens. First, the pretreated working electrodes were placed in a 3-mercaptopropionic acid (MPA)/β-mercaptoethanol (ME) mixture for 24 h for self-assembly. The electrodes were then placed in an EDC/NHS mixture for 1 h. Layer modification was performed by stepwise dropwise addition of CA19-9 antibody, BSA, and antigen. Differential pulse voltammetry was used to characterize this immunosensor preparation process. The assembled electrochemical immunosensor enables linear detection in the concentration range of 0.05–500 U/mL of CA19-9, and the detection limit was calculated as 0.01 U/mL. The results of the specificity measurement test showed that the signal change of the interfering substance was much lower than the response value of the detected antigen, indicating that the sensor has good specificity and strong anti-interference ability. The repeatability test results showed that the relative standard deviations were less than 5%, showing good accuracy and precision. The CA19-9 electrochemical immunosensor was used for the actual sample detection, and the experimental results of the standard serum addition method showed that the RSD values of the test concentrations were all less than 10%. The recoveries were 102.4–115.0%, indicating that the assay has high precision, good accuracy, and high potential application value.Keywords:self-assembled membrane;gold electrode;electrochemical immunosensor;CA19-9;gastric cancer

Mesoporous Magnetic Graphene for Serum Metabolic Profiling in Non-Invasive Early Detection and Diagnosis of Pancreatic Ductal Adenocarcinoma

Jia Qi,Caiyun Fang,Chunhui Deng,Fangying Shi,Qunyan Yao

Pancreatic ductal adenocarcinoma (PDAC) is a highly aggressive and lethal cancer, typically diagnosed at advanced stages due to its asymptomatic onset and challenges in early detection. To address the critical need for the early diagnosis of PDAC, we developed a laser desorption/ionization mass spectrometry (LDI-MS) platform based on mesoporous silica-modified magnetic graphene (MG@mSiO2). MG@mSiO2 exhibited exceptional ultraviolet (UV) absorption, efficient ionization, and minimal background interference, enabling high-resolution profiling of serum metabolic fingerprints (SMFs). Based on the extracted SMFs, we constructed a Random Forest (RF) model to classify PDAC patients, high-risk (HR) individuals, and healthy controls (HC), achieving an accuracy of 97.5% in the independent test set. Additionally, a six-metabolite biomarker panel was identified, showing strong diagnostic potential with sensitivity and accuracy exceeding 89.1% for distinguishing HC from PDAC. When coupled with the serological marker carbohydrate antigen 19–9 (CA19–9), the integrated strategy delivered significantly improved diagnostic performance, achieving high accuracy ranging from 95.3% to 100% in distinguishing HR and PDAC patients from HC. Furthermore, metabolic pathway analysis revealed key pathways associated with PDAC progression, providing mechanistic insights into the disease. This work provides a powerful diagnostic tool for PDAC screening, establishing a foundation for early detection and precision medicine in clinical practice.

Probe-Integrated Label-Free Electrochemical Immunosensor Based on Binary Nanocarbon Composites for Detection of CA19-9

Zhengzheng Yan,Jun Xing,Ruochong He,Qinping Guo,Ji Li

Convenient and sensitive detection of tumor biomarkers is crucial for the early diagnosis and treatment of cancer. Herein, we present a probe-integrated and label-free electrochemical immunosensor based on binary nanocarbon composites and surface-immobilized methylene blue (MB) redox probes for detection of carbohydrate antigen 199 (CA19-9), which is closely associated with gastric malignancies. Nanocarbon composites consisting of electrochemically reduced graphene oxides and carbon nanotubes (ErGO-CNT) are electrodeposited onto an indium tin oxide (ITO) electrode surface to form a 3D nanocomposite film, which could provide high surface area to immobilize abundant MB probes, facilitate the electron transfer of MB, and therefore, improve sensitivity. Polydopamine (PDA) served as a bifunctional linker is able to immobilize anti-CA19-9 antibodies and stabilize the inner probe, conferring the sensing interface with specific recognition capacity. Electrochemical detection of CA19-9 is achieved based on the decrease of the redox signal of MB after specific binding of CA19-9 with a wide linear range of 0.1 mU/mL to 100 U/mL and a limit of detection (LOD) of 0.54 nU/mL (S/N = 3). The constructed electrochemical immunosensor has good selectivity, repeatability, reproducibility, and stability. Furthermore, determination of CA19-9 in human serum samples is also realized. Keywords:electrochemically reduced graphene oxide;carbon nanotubes;methylene blue;CA19-9;immunosensor

Metabolomics to Reveal Diagnostic Biomarkers in CA19-9 Negative Pancreatic Ductal Adenocarcinoma: A Cross-Sectional Study

Shu-Jun Gu,Liu Xu,Hong-Kun Zhou,Jie Yang

Pancreatic ductal adenocarcinoma (PDAC) with negative carbohydrate antigen 19-9 (CA19-9) lacks effective diagnostic and monitoring indicators. We aimed to identify metabolic characteristics in such patients through metabolomics and explored potential plasma biomarkers. In this study, untargeted metabolomics analysis was performed on plasma samples from 30 healthy controls and 30 CA19-9 positive and 30 CA19-9 negative PDAC patients by using liquid chromatograph–mass spectrometer (LC–MS) technology. Compared with healthy controls, 36 differential metabolites were upregulated, and 3 differential metabolites were downregulated in the plasma of CA19-9 negative PDAC patients. Four upregulated differential metabolites, including palmitic amide, dolichol phosphate, cholesteryl acetate, and vitamin A2 aldehyde, were potential biomarkers for diagnosing CA19-9 negative PDAC patients. Enrichment analysis of KEGG pathways showed that glycine and threonine metabolism, tyrosine and tryptophan biosynthesis, thiamine metabolism, etc., were disrupted in CA19-9 negative PDAC patients. In summary, we determined the metabolic heterogeneity in PDAC patients with normal CA19-9 levels. The detected metabolic biomarkers may offer new approaches for the clinical early diagnosis of CA19-9 negative PDAC in the future.

SERS-based immunoassay and degradation of CA19-9 mediated by gold nanowires anchored magnetic–semiconductor nanocomposites

Yiran Tian,Xiuting Li,Fuyan Wang,Chenjie Gu,Ziqi Zhao,Hongjie Si,Tao Jiang

Here, straight upward Au nanowires (NWs) were successfully grown onto Fe 3 O 4 @TiO 2 matrix through a seed-mediated strategy to intensively improve its photocatalysis and SERS performances, facilitating a peculiar recyclable surface-enhanced Raman spectroscopy (SERS)-based immunoassay of CA19–9 in liquid form based on visible light irradiation. Such immunoassay was also supported by a smart heterobifunctional cross-linking agent-mediated probe of anti-CA19–9/4-MBA without metal nanoparticles . A low limit of detection of 5.65 × 10 −4 IU   mL −1 and a wide linear range from 1000 to 0.001 IU   mL −1 were demonstrated through repeated constructing the sandwich immunostructure with only one batch of nanocomposites . Moreover, the actual levels of CA19–9 for colorectal cancer patients were readily measured by the recyclable immunoassay, the results of which are principally consistent with the conventional CLIA detection. Thus, such a green strategy of visible light-induced recyclable immunoassay could be expected to have a potential practicability in the clinical diagnoses of cancer.

A controllable glycosyl molecularly imprinted strategy on MOF nanozyme for high-specificity colorimetric detection of CA19-9

Zhao Zhenghong,Xu Shuzhen,Xu Yue,Liao Guoming,Pan Changbing,Zhang Lianming,Ye Fanggui,Li Jianping

Molecularly imprinted nanozymes (MINs) offer a compelling alternative to antibody-based assays by integrating specific molecular recognition with enzyme-like catalytic activity. Nevertheless, conventional molecular imprinting of glycoproteins remains technically challenging. In this work, employing CA19-9 as a model glycoprotein biomarker, we develop a surface-directed glycan imprinting strategy by integrating molecular imprinting technology with iron-based metal-organic framework (MIL-88B-NH 2 ) nanozymes. The characteristic glycan of CA19-9 (sialyl Lewis A, SLe a ) is pre-adsorbed onto the amino-rich MIL-88B-NH 2 surface via hydrogen bonding and boronic acid affinity, followed by sol-gel polymerization to grow an imprinted shell. After template removal, the recognition cavities are automatically positioned within nanometers of the intrinsic Fe catalytic sites. Upon specific binding of CA19-9, steric hindrance exerted by the protein backbone impedes diffusion of the chromogenic substrate 3,3′,5,5′-tetramethylbenzidine (TMB) to the surface of MOF, resulting in quantifiable suppression of color development. The developed sensor demonstrates sensitive detection of CA19-9 across a wide linear range from 0.001 to 50 U mL −1 , with a detection limit of 1.01 × 10 −4 U mL −1 (3σ/slope), and shows excellent selectivity in serum samples. This work provides a stable, cost-effective, and visual biosensing platform with significant potential for point-of-care biomarker testing. Graphical
