Hydrolysis and transport characteristics of tyrosol-SCFA esters in rat intestine and blood: Two-step release of tyrosol and SCFAs to enhance the beneficial effects

Xinmiao Wang,Qian Wang,Yuanyuan Hu,Fawen Yin,Xiaoyang Liu,Dayong Zhou

The models of rat everted gut sac and hydrolysis by rat plasma were used to clarify the hydrolysis and transport characteristics of tyrosol-SCFA esters (TYr-SEs). HPLC-UV results indicated that TYr-SEs could be hydrolyzed by intestinal lipase, which showed sustained release of SCFAs and TYr. Meanwhile, TYr-SEs and the liberated SCFAs and TYr could cross the membrane and were transported into blood circulation. TYr-SEs were further hydrolyzed by carboxylesterase in plasma. Obviously, the hydrolysis of TYr-SEs in blood also showed sustained release of SCFAs and TYr. Especially, the rates of hydrolysis and transport correlated positively with the acyl chain lengths. Besides, the above rates of the TYr-SE with a straight chain were greater than those of its isomer with a branched chain. Therefore, the above-mentioned two-step release of SCFAs and TYr clearly demonstrated that TYr-SEs would be an effective approach to enhance the beneficial health effects of SCFAs and TYr.

Effect of polysaccharide fractions from Fortunella margarita on the fecal microbiota of mice and SCFA production in vitro

Peilin Chen,Suzhen Lei,Mingyao Tong,Qing Chang,Baodong Zheng,Yi Zhang,Hongliang Zeng

Fortunella margarita polysaccharides (FMPS) are composed of 4 polysaccharide fragments, namely mannogalactoglucan, galactoglucan, glucan and arabinoglucan. This study investigated the in vitro fermentation of FMPS fractions and their effect on the fecal microbiota of mice and short chain fatty acids (SCFAs) production. The results showed that the structure of the fecal microbiota was changed after FMPS fermentation. At the phylum level, the polysaccharide fractions reduced the relative abundance of Bacteroidetes compared with the inulin group. At the genus level, the polysaccharide fractions increased the abundance of Lactobacillus , and decreased the abundance of Granulicatella , Proteus and Rummeliibacillus . Polysaccharide fractions could promote the proliferation of beneficial bacteria ( Lactobacillus ), and decreased harmful bacteria ( Granulicatella , Proteus , Rummeliibacillus and Serratia ). Furthermore, there was a significant distinction in the genus level flora processed by different polysaccharide fractions, especially mannogalactoglucan. Polysaccharide fractions showed potential prebiotic effects, with mannogalactoglucan, in particular, promoting proliferation of SCFA-producing bacteria. SCFAs were regulated by monosaccharide composition, molecular weight of the FMPS fractions, and the composition of the fecal bacteria. Polysaccharide fractions could modulate metabolic function of the fecal microbiota, altering the levels of SCFAs. These results suggested that FMPS are important functional components in gut health, especially mannogalactoglucan.

Beneficial effects of Dendrobium officinale on metabolic hypertensive rats by triggering the enteric-origin SCFA-GPCR43/41 pathway

Bo Li,Xinglishang He,Hai-Ying Jin,Hui-Ying Wang,Fu-Chen Zhou,Ning-Yu Zhang,Dong-Ying Jie,Lin-Zi Li,Jie Su,Xiang Zheng,Rong Luo,Gui-Yuan Lv,Ning-Hua Jiang,Qiao-Xian Yu,Su-Hong Chen

Given the increasing global trend toward unhealthy lifestyles and dietary decisions, such as “over-consumption of alcohol, and high sugar and fat diets” (ACHSFDs), it is not surprising that metabolic hypertension (MH) is now the most common type of hypertension. There is an urgent, global need for effective measures for the prevention and treatment of MH. Improper diet leads to decreased short-chain fatty acid (SCFA) production in the gut, leading to decreased gastrointestinal function, metabolism, and blood pressure as a result of signaling through G-protein-coupled receptors (GPCRs), ultimately causing MH. Previous studies have suggested that Dendrobium officinale (DO) may improve gastrointestinal function, lower blood pressure, and regulate metabolic abnormalities, but it is not clear whether it acts on MH by increasing SCFA and, if so, how. In this research, it was observed that Dendrobium officinale ultrafine powder (DOFP) could lower blood pressure and improve lipid abnormalities in ACHSFD-induced MH model rats. Moreover, DOFP was found to improve the intestinal flora and increased the SCFA level in feces and serum, as well as increased the expressions of GPCR43/41 and eNOS and the nitric oxide (NO) level. An experiment on isolated aorta rings revealed that DOFP improved the vascular endothelial relaxation function in MH rats, and this effect could be blocked by the eNOS inhibitor L-NAME. These experimental results suggest that DOFP improved the intestinal flora and increased the production, transportation, and utilization of SCFA, activated the intestinal-vascular axis SCFA-GPCR43/41 pathway, improved vascular endothelial function, and finally lowered blood pressure in MH model rats. This research provides a new focus for the mechanism of the effect of DOFP against MH by triggering the enteric-origin SCFA-GPCR43/41 pathway.

Polysaccharides from fermented Asparagus officinalis with Lactobacillus plantarum NCU116 alleviated liver injury via modulation of glutathione homeostasis, bile acid metabolism, and SCFA production

Zhihong Zhang,Songtao Fan,Danfei Huang,Tao Xiong,Shaoping Nie,Mingyong Xie

Lactic acid bacteria strain (LAB) NCU116 fermented Asparagus officinalis polysaccharides (FAOP) have been proven to cause substantial changes in physicochemical properties such as monosaccharide composition and molecular weight, accounting for their enhanced immune activity than unprocessed Asparagus officinalis polysaccharides (AOP). In the current study, the hepatoprotective effects of FAOP in mice with cyclophosphamide (CTX)-induced hepatotoxicity were investigated. FAOP were more effective than AOP in alleviating CTX-induced hepatic damage, including inhibition of hepatic biochemical markers (ALT, AST, AKP and LDH) and pro-inflammatory cytokines (TNF-α and IL-1β) as well as reinforcement of antioxidant systems (T-AOC, SOD, CAT, and MDA). In particular, compared with AOP, FAOP showed superior performance by promoting GSH biosynthesis, and normalizing the expression level of bile acid receptors (FXR and SHP) and key enzymes in bile acid synthesis (CYP7A1, CYP8B1 and CYP27A1). Modulation of disordered homeostasis of bile acids by FAOP can be attributed to the upregulation of hepatic short chain fatty acid (SCFA) receptors GPR41 and GPR109A as well as intestinal SCFA production. Furthermore, serum metabolomics study validated the hepatoprotective superiority of FAOP than AOP with evidence from variations in bile acid compositions and the construction of related metabolic pathways. Therefore, LAB NCU116 fermentation of Asparagus officinalis was practical and effective to obtain promising hepatoprotective polysaccharides, which might arise from enhanced SCFA production than unprocessed AOP.

In Vitro Gastrointestinal Digestion and Microbial Hydrolysis of Hydroxytyrosol-SCFA and Tyrosol-SCFA Acyl Esters: Controlled-Release of SCFAs and Polyphenols

Xinmiao Wang,Qian Wang,Dong Cai,Xu Guo,Peiyong Tong,Fawen Yin,Xiaoyang Liu,Dayong Zhou

Phenolipids such as hydroxytyrosol-SCFA acyl esters (HTy-SEs) and tyrosol-SCFA acyl esters (TYr-SEs) with various alkyl chains lengths (C1–C4) and different isomers (branched-chain and straight-chain) were successfully synthesized. All esters were hydrolyzed by pancreatic lipase to produce polyphenols (HTy and TYr) and SCFAs (iso-butyric acid, acetic acid, propionic acid, and n-butyric acid). Moreover, HTy-SEs (and TYr-SEs) could also be hydrolyzed to free HTy (and TYr) and SCFAs by gut microbiota and Lactobacillus from mice feces. Especially, the hydrolysis rates showed positive correlation with the carbon skeleton length, and the hydrolysis degree (DH) of ester with a branched-chain fatty acid was weaker than that of ester with a straight-chain fatty acid. Besides, the DH values of TYr -SEs were significantly higher than those of HTy-SEs. Therefore, through regulating the structures of polyphenols, carbon skeleton lengths, and isomers, controlled-release of polyphenols and SCFAs from phenolipids will be easily achieved.

Mesophilic fermentation upgrades SCFA production from natural/raw henna plant biomass

Huang Jingang,Guo Kangyin,Shi Binfang,Li Jianping

Henna ( Lawsonia inermis ) plant biomass is a popular source of natural dye lawsone used all over the world. In this study, the effect of temperature on fermentative production of short-chain fatty acids (SCFA) from raw henna biomass was investigated. The results indicated that mesophilic fermentation (35 °C) upgraded the SCFA production as compared with those under normal (25 °C) and thermophilic (25 °C) conditions, increasing by up to 2.2-fold and 1.5-fold, respectively. Acetic acid was the main SCFA component, accounting for 50 ~ 70% of the total SCFA, and followed by n -butyric acid and propionic acid. The enzymatic activities of α-glucosidase in thermophilic series were lower than those in mesophilic series, resulting in a less carbohydrate consumption and SCFA production. Soluble lawsone was positively correlated with fermentation temperature; and the average concentration (87.0 mg/L) in mesophilic series was appropriate to serve as a redox mediator to improve the acidification process for SCFA production. Future work to systematically scrutinize the confounding factors affecting the fermentation process of henna biomass was required. Graphical abstract

Dissecting the anti-obesity components of ginseng: How ginseng polysaccharides and ginsenosides target gut microbiota to suppress high-fat diet-induced obesity

Han-Yan Luo,Jing Fang,Wei-Hao Zhang,Kam-Chun Chan,Yui-Man Chan,Cai-Xia Dong,Song-Lin Li,Ai-Ping Lyu,Jun Xu

Introduction Ginseng demonstrates therapeutic potential in treating obesity, with both experimental and clinical studies suggesting its anti-obesity effects are mediated by gut microbiota. Nonetheless, the specific chemical components responsible for this effect remain largely unidentified. Objectives This study aims to investigate the anti-obesity effects and mechanisms of ginseng polysaccharides (GP) and ginsenosides (GS), the primary chemical components of ginseng, with a focus on their impact on gut microbiota. Methods The impact of GP and GS on high-fat diet (HFD)-induced obesity was assessed using a mouse model. Molecular mechanisms were explored through a combination of chemical analysis, metagenomics, RT-qPCR, ELISA, and biochemical assays. Results GP or GS administration effectively prevented adiposity in HFD-fed mice, and both effects were mediated by gut microbiota. Chemical analysis revealed diverse glycosyl groups in GP and GS. Metagenomics data suggested that GP-enriched species, e.g., Bacteroides stercorirosoris and Clostridiales bacterium encoded carbohydrate-active enzymes GH35, GH43 and PL9_1, while GS-enriched Sulfurospirillum halorespirans encoded GH16_5. These enzymes facilitated the utilization of glycosyl groups in GP and GS, selectively stimulating bacterial growth and reshaping the gut microbiota. Furthermore, bacterial species enriched by GP or GS encoded specific functional genes involved in short-chain fatty acid (SCFA) synthesis (K00625 and K00925 for GP; K18118, K00100, and K18122 for GS) and intestinal gluconeogenesis (IGN) (K01678, K00024, and K01596 for GP; K18118 and K00278 for GS). Consequently, the SCFA-GLP-1/PYY signaling and IGN were activated by both GP and GS to ameliorate obesity phenotypes. Conclusion GP and GS, containing diverse glycosyl groups, selectively stimulate specific gut bacteria, triggering mechanisms involved in SCFA-GLP-1/PYY signaling and IGN activation to reduce adiposity in HFD-fed mice. The study enhances understanding of the chemical components crucial for the gut microbiota-mediated anti-obesity effect of ginseng. The mechanistic understanding provides valuable insights for developing ginseng-based drugs or health products to combat obesity.

Gut microbially produced tryptophan metabolite melatonin ameliorates osteoporosis via modulating SCFA and TMAO metabolism

Yueqi Chen,Chuan Yang,Zihan Deng,Tingwen Xiang,Qingrong Ni,Jianzhong Xu,Dong Sun,Fei Luo

Osteoporosis (OP) is a severe global health issue that has significant implications for productivity and human lifespan. Gut microbiota dysbiosis has been demonstrated to be closely associated with OP progression. Melatonin (MLT) is an important endogenous hormone that modulates bone metabolism, maintains bone homeostasis, and improves OP progression. Multiple studies indicated that MLT participates in the regulation of intestinal microbiota and gut barrier function. However, the promising effects of gut microbiota-derived MLT in OP remain unclear. Here, we found that OP resulted in intestinal tryptophan disorder and decreased the production of gut microbiota-derived MLT, while administration with MLT could mitigate OP-related clinical symptoms and reverse gut microbiota dysbiosis, including the diversity of intestinal microbiota, the relative abundance of many probiotics such as Allobaculum and Parasutterella , and metabolic function of intestinal flora such as amino acid metabolism, nucleotide metabolism, and energy metabolism. Notably, MLT significantly increased the production of short-chain fatty acids and decreased trimethylamine N-oxide-related metabolites. Importantly, MLT could modulate the dynamic balance of M1/M2 macrophages, reduce the serum levels of pro-inflammatory cytokines, and restore gut-barrier function. Taken together, our results highlighted the important roles of gut microbially derived MLT in OP progression via the “gut-bone” axis associated with SCFA metabolism, which may provide novel insight into the development of MLT as a promising drug for treating OP.

Salidroside Alleviates Furan-Induced Impaired Gut Barrier and Inflammation via Gut Microbiota–SCFA–TLR4 Signaling

Ziyue Wang,Lu Li,Wenliang Li,Haiyang Yan,Yuan Yuan

As a food contaminant that can be quickly absorbed through the gastrointestinal system, furan has been shown to disrupt the intestinal flora and barrier. Investigation of the intestinal toxicity mechanism of furan is of great significance to health. We previously identified the regulatory impact of salidroside (SAL) against furan-provoked intestinal damage, and the present work further explored whether the alleviating effect of SAL against furan-caused intestinal injury was based on the intestinal flora; three models, normal, pseudo-germ-free, and fecal microbiota transplantation (FMT), were established, and the changes in intestinal morphology, barrier, and inflammation were observed. Moreover, 16S rDNA sequencing observed the variation of the fecal flora associated with inflammation and short-chain fatty acids (SCFAs). Results obtained from the LC–MS/MS suggested that SAL increased furan-inhibited SCFA levels, activated the mRNA expressions of SCFA receptors (GPR41, GPR43, and GPR109A), and inhibited the furan-activated TLR4/MyD88/NF-κB signaling. Analysis of protein–protein interaction further confirmed the aforementioned effects of SAL, which inhibited furan-induced barrier damage and intestinal inflammation.

Kluyveromyces marxianus XZ1 fermentation products alleviate hyperuricemia via the microbiota–SCFA–gut–kidney urate transporter axis

Yunzhe Jia,Zhirong Wang,Wenqian Zheng,Shuyao Sang,Zhenquan Yang,Xiangfeng Zheng

Hyperuricemia (HUA) is a systemic metabolic disorder closely linked to disrupted host-microbiota interplay. Current first-line drugs (e.g., allopurinol) primarily target urate synthesis but lack tissue-repair capabilities and cause long-term adverse effects. Although microbial metabolites circumvent the inflammatory risks of live probiotics, the urate-lowering mechanisms of Kluyveromyces marxianus fermentation metabolites remain largely uncharacterized. Here, we integrated in vitro bioassays, in vivo animal experiments, and multi-omics profiling to elucidate the anti-hyperuricemic efficacy of an ethyl acetate extract from K. marxianus XZ1. In vitro , the XZ1 extract exhibited significant antioxidant and xanthine oxidase (XOD) inhibitory activities. The cell-free extract exerted a dual-action mechanism: directly suppressing urate synthesis by inhibiting XOD and adenosine deaminase (ADA) activities, and enhancing systemic urate clearance through bidirectional regulation of renal and intestinal urate transporters (upregulating excretory ABCG2/OAT1 and downregulating reabsorptive URAT1/GLUT9). Integrated 16S rRNA sequencing and untargeted metabolomics revealed these systemic benefits are driven by gut microbiota remodeling, forming a “microbiota–short-chain fatty acid (SCFA)–gut–kidney urate transporter” regulatory axis. The intervention enriched SCFA-producing taxa, normalized fecal SCFA profiles, strengthened intestinal barrier integrity and reprogrammed host purine and redox metabolic networks. Collectively, K. marxianus XZ1 fermentation products lower uric acid and protect against HUA-induced multi-organ injury, inflammation, and oxidative stress. These findings highlight its potential as a safe, industrially stable, and multi-target functional food ingredient for early dietary intervention in asymptomatic hyperuricemia.

Maillard reaction enhances prebiotic potential of silver carp hydrolysates: Insights from SCFA production, microbiome, and metabolomics

Peipei Dou,Huijuan Zhang,Ning Ding,Elliot Mubango,Hongbing Fan,Ce Shi,Hui Hong,Yuqing Tan,Yongkang Luo

Silver carp ( Hypophthalmichthys molitrix ) is a low-cost, protein-rich freshwater fish with high nutritional value but remains underutilized in functional food applications. Our previous studies have demonstrated that silver carp hydrolysates (SCH) possess potential in the management of chronic diseases and that their sensory properties and antioxidant activities can be further enhanced through Maillard reaction. However, the prebiotic potential of Maillard reaction-modified SCH has been rarely explored. This study investigates the prebiotic potential of SCH-based Maillard reaction products (SCH-MRPs) using in vitro fermentation model combined with 16S rRNA sequencing and metabolomics. During the fermentation process, moderate Maillard reaction significantly promoted SCFA production (approximately 16.29 %) ( p  < 0.05), particularly acetic, propionic, and butyric acids, which are known to play key roles in maintaining intestinal health and immune function. These changes were associated with increased microbial diversity, as indicated by higher Shannon (41.44 %) and Simpson (18.30 %) indices, along with a 264.24 % increase in the relative abundance of Bacteroides . Metabolomics analysis revealed differential metabolites in the SCH-MRPs group suggested potential involvement in regulating tryptophan metabolism and bile secretion pathways, including indole-3-carboxaldehyde, indolepropionate, and glycochenodeoxycholic acid 3-glucuronide. Correlation analysis showed significant positive correlation between Fusobacterium , Bacteroides , and unidentified_Clostridiales abundances with indoles and derivatives, suggesting a regulatory role in tryptophan metabolism. This study systematically explores the prebiotic potential of Maillard reaction-modified SCH conjugated with oligosaccharides. By integrating microbial community analysis and metabolomics, it reveals a dual effect of SCH-MRPs in enhancing gut microbiota composition and metabolic activity, offering a promising strategy for microbiome-targeted functional food development.

Microbiota-Dependent Alleviation of Ulcerative Colitis by Liubao Tea: Integrated Insights into SCFA and Arachidonic Acid Metabolism

Xiao Yang,Song Xu,Ying Tong,Jichu Luo,Xixing Fang,Jiaxing Du,Changyuan Zhou,Guangnian Hu,Bao Yang,Qisong Zhang

(1) Background: Ulcerative colitis (UC) is a chronic inflammatory bowel disease associated with gut microbiota dysbiosis and metabolic perturbations. Although Liubao tea (LBT) has gastroprotective benefits, the precise mechanisms by which LBT extract (LBTE) alleviates UC by orchestrating microbial and metabolic homeostasis remain poorly understood. (2) Methods: A DSS-induced UC mouse model was used to evaluate LBTE efficacy. Serum pharmacochemistry, untargeted metabolomics, 16S rRNA sequencing, and targeted SCFA metabolomics were integrated to characterize absorbable active constituents, metabolic shifts, and gut microbiota landscapes. SCFA- and arachidonic acid metabolism-related targets were validated by RT-qPCR and Western blotting. PGF models and FMT were used to assess the causal role of gut microbiota in LBTE-mediated efficacy. (3) Results: LBTE preserved colon length and mucosal integrity while reducing IL-6, TNF-α, IL-1β, and oxidative stress. It enriched SCFA-producing genera and increased colonic butyric and valeric acids, activating GPR41/GPR109A signaling, upregulating ZO-1 and occludin, and strengthening the intestinal barrier. LBTE also downregulated PTGS2 and ALOX5, restored PTGS1 and CYP3A11, and inhibited NF-κB signaling. These effects were weakened in PGF mice but reproduced by FMT from LBTE-treated donors, confirming microbiota-dependent protection. (4) Conclusions: LBTE may serve as a complementary strategy for UC prevention and management.
