The effects of FeS on nitrogen removal performance and microbial community of anammox process were studied. During the start-up period, the removal efficiencies of nitrite and total nitrogen were significantly improved by FeS. The addition of FeS increased the content of iron ions in the reactor and promoted the synthesis of heme c , which was involved in the formation of various enzymes. Compared with the control, the abundance of anammox bacteria in the FeS reactor was increased by 29%, and the expression level of the nirS gene (encoding cd 1 type nitrite reductase containing heme) was nearly doubled. The content of nitrite reductase (ammonia-forming) in the community was increased by 26.4%. The difference in functional bacteria and enzyme contents in the microbial community resulted in a difference in nitrogen removal rate (NRR) between the two reactors. High-throughput results indicated that FeS increased the richness and diversity of microbial community and enhanced the metabolic function of the microbial community. The addition of FeS did not change the dominant position of Ca. Kuenenia in both reactors. But the relative abundance of heterotrophic denitrifying bacteria was reduced with FeS, which may be related to the inhibition effect of S 2− produced by FeS.

The anammox bacteria (AnAOB) have been suggested to perform dissimilatory Fe0/Fe2+-dependent nitrate reduction to ammonium (DNRA), with nitrite as intermediate. However, evidence at genetic and transcriptional levels for how AnAOB reduces nitrate is still lacking, making their roles in this process obscure and questionable. The short-term batch assays and long-term operation of bioreactor combined with 16S rDNA sequencing and transcription analysis were used for the first time to investigate the nitrate reduction pathway in anammox consortia. It is shown that AnAOB metabolically well grew with nitrate as the sole nitrogen source. They reduced nitrate to nitrite with highly expressed narG gene. However, the DNRA-enabling gene nrfA was not expressed by AnAOB. Abiotic reduction by Fe0/Fe2+ contributed to the nitrate and nitrite reduction to ammonium. The correlation analysis of microbial community structure and metabolic functions indicated that amino acid and carbohydrate transport and metabolism interactions in coexisting bacteria could facilitate the AnAOB growth with nitrate as the only nitrogen source. Our findings suggest that the contribution of enzymatic DNRA by AnAOB to the observed nitrate reduction has been probably overestimated in previous publications, and it should be questioned until the clear enzymatic mechanism responsible for nitrate reduction is identified.

Excessive ammonia (NH 4 + ) discharge can lead to algal blooms and disrupt water sustainability, so its control is imperative. Although microbiology-triggered anammox process is promising, its application is limited due to time-consuming cultivation of specific microorganisms and need for skilled operation. To bypass these barriers, this study proposed and verified a photo-induced anammox technology that removes NH 4 + and total nitrogen (TN) from water by ultraviolet (UV)/vacuum UV (VUV)/chlorine under anoxic conditions . Under the Cl/N mass ratio of 5:1, the anoxic VUV/UV/chlorine process achieved 66.8% removal of 10 mg-N/L NH 4 + within 10 min along with 57.8% reduction in TN. Besides the evidence from TN loss, this study confirmed nitrogen gas (N 2 ) as the primary degradation product at low dissolved oxygen (DO) concentration of 2.0 mg/L. The selective conversion of NH 4 + into N 2 was mainly attributed to reactive nitrogen species (RNS, 42.5%) and reactive chlorine species (RCS, 57.5%). The TN removal efficiency was insensitive to certain variations of pH (7.0–9.0), NH 4 + concentration (1–30 mg-N/L), chloride (50–125 mg/L), and sulfate (25–100 mg/L), but sensitive to DO and bicarbonate (25–100 mg/L). Given its robustness and high efficiency, the anoxic VUV/UV/chlorine technology may serve as a potentially promising alternative for NH 4 + and TN alleviation in wastewater.

The red color is the most striking character of anaerobic ammonium-oxidizing bacteria (AnAOB) which has been used to estimate the anammox activity roughly. However, the quantitative relationship between the color and activity of anammox sludge still remains unknown. In this study, the chromaticity, activity and their correlation were systematically investigated at different steady-state nitrogen loading rates. The chromaticity of anammox sludge was digitalized by the CIE L*a*b* color space. The results revealed that the average chroma value was found to be significantly correlated with specific anammox activity ( r = 0.940, p < 0.01) and the cluster centers of chromaticity coordinates (a*, b*) of anammox sludge were established to define the typical working states of anammox system. The visible spectra of anammox sludge were proved to originate from the cytochrome c. The correlation between chroma and heme c concentration of anammox sludge was consistent with the fully-reduced cytochrome c and the chroma was determined by both content and redox ratio of cytochrome c. The chromaticity of anammox sludge was able to be linked with the anammox activity via reduced cytochrome c content. The gene abundance of cytochrome c synthetase linked the chromaticity with AnAOB quantity via total cytochrome c content, while the enzyme activity of octaheme hydrazine dehydrogenase linked the chromaticity with AnAOB activity via reduced cytochrome c ratio. Moreover, the redundancy analysis proved that heme c, as the key component of cytochrome c, was the most important explanatory variable accounting for the maximum 69.6% of the total variation of the anammox community, which correlated positively with the relative abundance of dominant AnAOB ( Candidatus Kuenenia). This work aimed at demonstrating the chromaticity of anammox sludge could be developed as an alternative intuitive anammox activity indicator which will promote the monitoring and optimization of anammox process.

In this study, waste iron scraps (WIS) were exerted to alleviate sulfide inhibition on anammox bacteria and promote anammox nitrogen removal from sulfide-containing wastewater. Short-term batch experiments showed that WIS-addition led to the anammox bacteria activity increasing by 124.8 % at an initial sulfide concentration of 40 mgS/L. During the long-term experiments, the nitrogen removal rate (NRR) reached to 8.76 kg/(m 3 ·d) in the WIS-added reactor, while the maximum NRR was only 3.77 ± 0.31 kg/(m 3 ·d) in the non-WIS reactor. In contrast to anammox bacteria development in the non-WIS reactor, the relative abundance of Candidatus Kuenenia (1.4–3.7 %) declined significantly in the WIS-added reactor, but novel potential anammox bacteria Brocadiaceae_unclassified (60.1 %-78.6 %) were highly enriched. Overall, the experimental evidence suggested that WIS-addition not only mitigated the sulfide inhibition on anammox bacteria, but also promoted novel anammox bacteria proliferation. The findings of this work provide a promising solution for wide engineering applications of anammox treating sulfide-containing wastewater.

Anaerobic ammonium oxidation is a critical pathway for nitrogen (N) loss in terrestrial environments, and its coupled interactions with As(V) reduction has recently been recognized. While Fe-bearing minerals are suspected to regulate this coupling, the underlying mechanisms remain elusive. This study demonstrates that Fe-bearing minerals (ferrihydrite, goethite, montmorillonite, illite, and kaolinite) actively mediate anammox-coupled As(V) reduction by facilitating electron transfer and promoting Fe redox cycling. The presence of these minerals increases As(V) reduction by 1.2–2.4-fold and NH 4 + removal by 1.65–3.15-fold, with ferrihydrite exhibiting the strongest effect. In addition to accelerating reaction rates, these minerals reshape N partitioning and microbial community structure: clay minerals primarily influence early-stage NH 4 + dynamics via adsorption, whereas Fe (hydr)oxides sustain long-term coupling by suppressing nitrite oxidizers, limiting NO 3 - accumulation, enriching anammox and Fe-reducing bacteria, and enhancing microbial network stability. Redox-equivalent analysis further shows that these minerals increase the electron utilization efficiency of NH 4 + oxidation coupled with Fe(III) and/or As(V) reduction from 17% to 19–75%. Furthermore, As(V) reduction rates correlate strongly with the mineral specific surface area and functional genes including hzsA , omcS , arsC and arrA . This highlights the combined roles of surface-mediated electron transfer, enhanced Fe(III)/Fe(II) cycling, and coordinated regulation of N transformation and respiratory As(V) reduction. These findings identify Fe-bearing minerals as electron-buffering and -channeling phases that organize interfacial electron flow and strengthen N-Fe-As redox couples, providing a mechanistic framework for mineral-driven biogeochemical interactions in As-contaminated soils and sediments.

Iron nanoparticles (FeNPs) generated from industrial activities could end up into the sewer system, and potentially affect wastewater treatment processes. The impact of FeNPs on anammox process is getting increasing attention. However, the resistance mechanism of anammox granular sludge (AnGS) to FeNPs has not been fully elucidated. The current study investigated the metabolic and morphological response of AnGS to acute and chronic FeNPs exposure. Results showed that nitrogen removal efficiencies were elevated at 1–4 mM FeNPs concentrations compared to 0–0.5 mM FeNPs. Extracellular protein and tyrosine-like and tryptophan-like fluorophore secretions of AnGS were stimulated by FeNPs, which largely contributed to the adsorption of FeNPs on AnGS surface. FeNPs exposure triggered higher necrotic fraction of AnGS compared with no FeNPs condition. Highly absorbed particles appeared inside the bacterial cells of AnGS, soft X-ray imaging illustrated that anammox bacteria maintained intact cellular and anammoxosome structures whereas non-anammox bacterial structures were damaged under FeNPs exposure. Anammox bacterial abundance increased from 4.84% to 20.64%, when FeNPs concentrations increased from 0 mM to 4 mM, and anammoxosome membrane ensured anammox bacterial metabolism under FeNPs exposure. This study extended fundamental understanding of AnGS resistance mechanisms to FeNPs.

The zero-valent iron-anaerobic ammonium oxidation (ZVI-anammox) system has received widespread attention due to its excellent nitrogen removal performance and user-friendly operation. However, its disadvantages include a short service life, high ZVI consumption, and poor system stability. The use of ultrasound as a physical method is increasing in various water treatment processes. In this study, a series of batch tests were conducted to obtain the best ultrasonic parameter and explore the comprehensive effects of ultrasound on a ZVI-anammox system. The highest specific anammox activity of the ZVI-anammox system was found to be 2.88 mg total nitrogen/g of volatile suspended solids/h after ultrasonic treatment (0.2 w/mL, 5 min), which was 37.85% higher than a control group. Additionally, the service life of ZVI extended by 28.57% and the total nitrogen removal efficiency changed from 58.03–72.08 to 63.92–78.33% under ultrasonic irradiation. These phenomena were related to the mechanical force and cavitation of ultrasonic waves. Judging from the characteristics of sludge and ZVI, ultrasound can promote anammox sludge granulation, ease ZVI passivation, and enhance the stability of the entire system. This paper also briefly discusses the impact mechanisms of ultrasound on the ZVI-anammox system. In brief, ultrasound destroys the surface structure of ZVI and thus provides numerous attachment points for microorganisms that improve the performance of the entire system. The proposed ultrasound combined with ZVI is a novel method that has potential for use in large-scale engineering applications in actual sewage treatment after comprehensive analysis.

Iron nanoparticles (FeNPs) are commonly used in various industrial processes, leading to their release into the environment and eventual entrance into wastewater treatment plants (WWTPs). FeNPs undergo dissolution, migration, and transformation in WWTPs, which can potentially affect the stable operation of anaerobic ammonia oxidation (anammox) systems and may be discharged with wastewater or biomass. To better understand the fate of FeNPs in anammox systems, exposure experiments were conducted using anammox granular sludges (AnGS) and FeNPs. Results demonstrated that FeNPs released Fe 2+ upon contact with water, with a portion being bound to functional groups in extracellular polymeric substances (EPS) and the rest entering the bacteria to form highly absorbable substances. A significant amount of FeNPs was observed to cover the surface of AnGS or aggregate and deposit at the bottom of the reactor, eventually converting into Fe 3 O 4 and stably existing within the anammox system. The findings of this study clarify the fate of FeNPs in anammox systems and provide important insights into the stable operation of anammox systems under FeNPs exposure.

The increasing use of engineered nanoparticles (NPs) poses an emerging challenge to biological wastewater treatment. The long-term impact of CuNPs on anaerobic ammonium oxidation (anammox) process was firstly investigated in this study. The nitrogen removal capacity of anammox reactor was nearly deprived within 30 days under the stress of 5.0 mg L −1 CuNPs and the relative abundance of anammox bacteria (Ca. Kuenenia ) was decreased from 29.59% to 17.53%. Meanwhile, copper resistance genes associated with the Cus, Cop and Pco systems were enriched to eliminate excess intracellular copper. After the withdrawal of CuNPs from the influent, the nitrogen removal capacity of anammox biomass recovered completely within 70 days. Overall, anammox biomass showed susceptibility, resistance and resilience to the stress of CuNPs. Therefore, the potential impacts of ENPs on anammox-based processes should be of great concern.

To achieve the in situ capacity expansion of the post-denitrification biological aerated filter (BAF-DN), the integration of BAF with the anammox process (BAF/AX) was proposed. With the objective of maximizing retaining ammonia nitrogen, the operational optimization of BAF was achieved by two distinct strategies. The treatment performance of BAF demonstrated that the removal efficiencies of chemical oxygen demand (COD) and ammonia nitrogen (NH4+-N) was 66.3~67.3% and 4~12%, respectively, under conditions of low aeration intensity (0.4 m3·m−2·h−1) or a shortened empty bed residence time (EBRT) of 30 min. ResidualNH4+-Nin the BAF effluent served as the ammonia substrate for the subsequent anammox process, which was successfully launched by using ceramic particles and sponges as carriers. Notably, the sponge carrier facilitated a shorter start-up period of 41 to 44 days. Furthermore, the sponge-based anammox reactor exhibited a superiorNH4+-Nremoval capacity (≥85.7%), under operations of a shorter EBRT of 40 min, low influentNH4+-Nconcentrations (≤30 mg/L), and COD levels of ≤67 mg/L. In addition, a comprehensive evaluation of the BAF/AX process was conducted, which considered performance, cost-effectiveness, and engineering feasibility. The performance results illustrated that the effluent quality met the standard well (with a COD level of ≤ 50 mg/L, and a TN of ≤3.1~10.5 mg/L). Following a comparison against the low aeration intensity operation, it was recommended to operate BAF at a low EBRT within the BAF/AX process. Consequently, the treated volume was double the volume of the standalone BAF-DN, synchronously achieving low costs (0.413 yuan/m3).

Acute toxicities of anaerobic ammonia oxidation (ANAMMOX) substrates and four antibiotics from pharmaceutical wastewaters on ANAMMOX process were reported. Individual and joint acute toxicity assays were performed using 50% inhibitory concentration (IC 50 ). Results showed that IC 50 values and their 95% confidence interval of ammonium chloride (A), sodium nitrite (B), penicillin G-Na (C), polymyxin B sulfate (D), chloramphenicol (E) and kanamycin sulfate (F) were 2708.9 (2247.9–3169.9), 1475.4 (1269.9–1680.9), 5114.4 (4946.4–5282.4), 10.2 (1.8–18.6), 409.9 (333.7–486.1) and 5254.1 (3934.4–6573.8) mg L −1 respectively, suggesting toxicities were in the order of D > E > B > A > C > F. Joint acute toxicities of bicomponent mixtures A and B, C and D, C and F, D and F were independent; D and E, E and F were additive while C and E were synergistic. Joint acute toxicities of multicomponent mixtures were synergistic or additive. Luminescent bacteria test is an easy and robust method for forecasting the feasibility of ANAMMOX process for pharmaceutical wastewater treatment .
