Trimethoprim degradation by Fenton and Fe(II)-activated persulfate processes

Shizong Wang,Jianlong Wang

Trimethoprim is a pollutant ubiquitous in the environment due to its extensive application, and it cannot be effectively removed by conventional wastewater treatment processes . In this study, the Fenton and the Fe(II)-activated persulfate processes were employed to degrade trimethoprim in an aqueous solution. The results showed that the concentration of persulfate, H 2 O 2 and Fe(II) a had significant influence on the degradation of trimethoprim in both processes. De-ionized water spiked with trimethoprim resulted in the complete degradation of trimethoprim (0.05 mM) by the mineralization of 54.9% of Fenton's reagent when the concentrations of H 2 O 2 and Fe(II) were 1 mM and 0.05 mM, respectively. In contrast, 73.4% of trimethoprim was degraded by the mineralization of 40.5% of the Fe(II)-activated persulfate process when the concentration of persulfate and Fe(II) were each 4 mM. Intermediate compounds with different m / z were detected for the Fenton and the Fe(II)-activated persulfate processes, indicating alternative degradation pathways. In the actual wastewater spiked with trimethoprim, the removal efficiency of trimethoprim decreased to 35.8% and 43.6%, respectively, for the Fenton and the Fe(II)-activated persulfate processes. In addition, the decomposition efficiencies for hydrogen peroxide and persulfate were 43.8% and 92.5%, respectively, which was lower than those in the de-ionized water system. These results demonstrated that wastewater components had a negative influence on trimethoprim degradation and the decomposition of the oxidants (persulfate and H 2 O 2 ). In summary, the Fe(II)-activated persulfate process could be used as an alternative technology for treating trimethoprim-containing wastewater.

Anaerobic biodegradation of trimethoprim with sulfate as an electron acceptor

Liang Bin,Kong Deyong,Qi Mengyuan,Yun Hui,Li Zhiling,Shi Ke,Chen E.,Vangnai Alisa S.,Wang Aijie

Trimethoprim (TMP) is an antibiotic frequently detected in various environments. Microorganisms are the main drivers of emerging antibiotic contaminant degradation in the environment. However, the feasibility and stability of the anaerobic biodegradation of TMP with sulfate as an electron acceptor remain poorly understood. Here, TMP-degrading microbial consortia were successfully enriched from municipal activated sludge (AS) and river sediment (RS) as the initial inoculums. The acclimated consortia were capable of transforming TMP through demethylation, and the hydroxyl-substituted demethylated product (4-desmethyl-TMP) was further degraded. The biodegradation of TMP followed a 3-parameter sigmoid kinetic model. The potential degraders ( Acetobacterium, Desulfovibrio, Desulfobulbus , and unidentified Peptococcaceae) and fermenters ( Lentimicrobium and Petrimonas ) were significantly enriched in the acclimated consortia. The AS- and RS-acclimated TMP-degrading consortia had similar core microbiomes. The anaerobic biodegradation of TMP could be coupled with sulfate respiration, which gives new insights into the antibiotic fate in real environments and provides a new route for the bioremediation of antibiotic-contaminated environments.

Degradation of trimethoprim by gamma irradiation in the presence of persulfate

Zhonglei Zhang,Qi Yang,Jianlong Wang

The degradation and mineralization of trimethoprim (TMP) by gamma irradiation was investigated in the presence of persulfate (PS). The TMP was degraded at initial concentration of 20   mg/L in aqueous solution with addition of 0, 0.5, 1, 1.5, 2   mM persulfate respectively. The effect of pH values (6.5, 7.5 and 8.5) on TMP degradation was also determined. The experimental results showed that the degradation and mineralization of TMP could be significantly enhanced by persulfate at acidic condition (pH=6.5). Several intermediate products generated during gamma irradiation process through hydroxylation, demethylation and cleavage were identified using liquid chromatography with tandem mass spectrometry (HPLC-MS). The degradation pathway of TMP was tentatively proposed based on the identification of intermediate products.

Aged polyamide microplastics enhance the adsorption of trimethoprim in soil environments

Li Jia,Yu Songguo,Cui Min

Microplastics (MPs) in the environment typically age. However, the influence of aged MPs on the adsorption of antibiotics in soil remains unknown. In this study, the adsorption behavior of trimethoprim (TMP) on soil and soil containing aged polyamide (PA) was investigated using batch and stirred flow chamber experiments. The adsorption of TMP on the tested soil with and without PA was fast, with the k a values ranging from 50.5 to 55.6 L (mg min) −1 . The adsorption of TMP on aged PA was more than 20 times larger than that on the tested soil, which resulted in an “enrichment effect.” Furthermore, aged PA altered the pH of the reaction system, thereby enhancing the adsorption of TMP. Consequently, the K d values of TMP for soil, soil containing 5%, and 10% aged PA were 5.64, 12.38, and 23.65 L kg −1 , respectively. The effect of aged PA on the adsorption of TMP on soil depended on pH values. However, TMP adsorption on soil containing 10% aged PA was constantly higher ( p  < 0.01) than that on soil with NaCl concentrations ranging from 0 to 50 mmol L −1 . These findings provide new insights into the effect of environmental MPs on the fate and transport of antibiotics in soil environments.

Efficient biodegradation of trimethoprim and transformation mechanism using the photoelectrocatalytic system

Shun Zhang,Yujie Feng,Bin Liang,Pengfei Si,Dengfei Li,Yanjuan Gao,Aijuan Zhou,Xiuping Yue

In this study, the coupling of a three-electrode photocatalytic microbial fuel cell system (PMFC) was established for trimethoprim (TMP) degradation, to achieve efficient degradation and electrical energy production simultaneously. The photocatalytic bioanode, degradation performance, electrochemical characteristics and microbial structure were analyzed to explore the degradation of TMP in PMFC. The results showed that rapid degradation and enhanced mineralization of TMP were obtained in the PMFC, which were 1.7 and 1.4 times higher than those of traditional microbial fuel cells (MFCs), respectively. Intermediates in the degradation process were identified in both systems, and three degradation pathways were inferred in the PMFC. The electricity characteristics were investigated and superior electrical production and higher electron transfer efficiency were observed in the PMFC, resulting in a maximum power density of 75.1 mW/m 2 and a maximum voltage of 0.225 V. Additionally, the cyclic voltammetry test and electrochemical impedance spectra were used for electrochemical performance characterization, and the results showed the stronger oxidation ability of PMFC with half the charge transfer resistance of MFCs. The microbial community structure demonstrated that the dominant species were enriched and coexisted, involving degradation bacteria, exoelectrogens, and hydrocarbon removal bacterial. Furthermore, canonical correspondence analysis (CCA) revealed a mutual promotion of TMP degradation and electricity production with the involvement of these active genera. In conclusion, this novel technology is an excellent alternative in practical applications for the treatment of wastewater containing antibiotics.

Effects of CYP2C9 genetic polymorphism and drug-drug interactions on trimethoprim metabolism

Hailun Xia,Jun Wu,Yingying Hu,Peiqi Wang,Lu Cao,Ruibin Li,Haoxin Fu,Ren-ai Xu,Saili Xie

The aim of the study was to investigate how CYP2C9 genetic polymorphism and drug-drug interactions with psychiatric drugs affect trimethoprim metabolism. We conducted in vitro enzymatic incubation assays using rat liver microsomes (RLM), human liver microsomes (HLM) and recombinant human CYP2C9 variants, and carried out in vivo pharmacokinetics experiments using Sprague-Dawley rats. The analytes were analyzed using ultra performance liquid chromatography tandem mass spectrometry (UPLC-MS/MS). We determined the kinetic parameters of 12 CYP2C9 variants using trimethoprim as a substrate. The results showed significant decreases in catalytic activity for CYP2C9.3, CYP2C9.16, CYP2C9.42 and CYP2C9.48 compared to CYP2C9.1. Notably, CYP2C9.42 exhibited the lowest catalytic activity, only 28.19 % of CYP2C9.1. Fluoxetine was found to be the most potent among the 17 investigated psychiatric drugs in inhibiting trimethoprim metabolism, with a half-maximal inhibitory concentration (IC 50 ) values of 7.12 μM in RLM and 50.61 μM in HLM, respectively. The combination of trimethoprim and fluoxetine increased the AUC (0-∞) and AUC (0-t) of trimethoprim by 1.97- and 2.23-fold, respectively. Mechanistically, fluoxetine exhibited a competitive inhibition in RLM and a combination of competitive and non-competitive inhibition in HLM on trimethoprim metabolism. Our findings highlighted the significant impacts of CYP2C9 gene polymorphisms and fluoxetine on trimethoprim metabolism, providing insights for the precise administration of trimethoprim.

Antibacterial efficacy and pharmacokinetics of a poly(mPEG-b-PCEA)–trimethoprim conjugate

Ming Niu,Zhenghua Zhang,Rourou Wang,Yaxin Zhou,Jing Zhou,Lumei Pu,Weibing Xu

Research into antimicrobials is crucial for the sustainable development of the livestock industry. Trimethoprim (TMP) is one of the most widely used antimicrobial agents in the world. However, its efficacy is often limited by poor water solubility and dose-limiting properties. In this work, amphiphilic block polymer carriers are prepared using the oxygen-induced reversible addition–fragmentation chain transfer polymerization reaction. TMP is linked to carriers in different ratios to form poly(mPEG-b-PCEA)–TPM conjugates. The conjugates are characterized using various methods including 1H NMR, FT-IR, XRD and SEM. The results of in vitro antimicrobial experiments show that poly(mPEG-b-PCEA)–TMP with different ratios has more prominent antimicrobial effects than the same dosage of pure TMP. poly(mPEG-b-PCEA)–TMP80 shows the most significant results, inhibiting E. coli by 87.19% and S. aureus by 97.02%, with a hemolysis rate of <5% and good biocompatibility. In vivo pharmacokinetics show an increase in Cmax from 4.81 μg mL−1 to 6.57 μg mL−1 and AUC0–48 from 78.55 h μg mL−1 to 89.11 h μg mL−1 for poly(mPEG-b-PCEA)–TMP80 compared to commercially available TMP tablets. This strategy can effectively improve the antibacterial activity and pharmacokinetics and increase the bioavailability of TMP.

Synergistic Control of Trimethoprim and the Antimicrobial Resistome in Electrogenic Microbial Communities

Yaoli Wei,Liying Zhang,Bin Liang,Hanlin Cui,Ke Shi,Zhihong Liu,Aijuan Zhou,Xiuping Yue

Synergistic control of the risks posed by emerging antimicrobials and antibiotic resistance genes (ARGs) is crucial for ensuring ecological safety. Although electrogenic respiration can enhance the biodegradation of several antimicrobials and reduce ARGs accumulation, the association mechanisms of antimicrobial biodegradation (trimethoprim, TMP) with the fate of the antimicrobial resistome remain unclear. Here, the biotransformation pathway of TMP, microbial associations, and functional gene profiles (e.g., degradation, antimicrobial resistance, and electron transfer) were analyzed. The results showed that the microbial electrogenic respiration significantly enhanced the biodegradation of TMP, especially with a cosubstrate sodium acetate supply. Electroactive bacteria enriched in the electrode biofilm positively correlated with potential TMP degraders dominated in the planktonic communities. These cross-niche microbial associations may contribute to the accelerated catabolism of TMP and extracellular electron transfer. Importantly, the evolution and dissemination of overall ARGs and mobile genetic elements (MGEs) were significantly weakened due to the enhanced cometabolic biodegradation of TMP. This study provides a promising strategy for the synergistic control of the water ecological risks of antimicrobials and their resistome, while also highlighting new insights into the association of antimicrobial biodegradation with the evolution of the resistome in an electrically integrated biological process.

Response surface methodology optimization and antimicrobial activity of berberine modified trimethoprim carboxymethyl cellulose

Gui Fu,Zhenzhen Chen,Yingping Qi,Hua Li

In order to solve the problem of bacterial resistance, increase the antibacterial performance and delay the aging of paper, the berberine modified trimethoprim carboxymethyl cellulose was prepared using trimethoprimethylate carboxymethyl cellulose as raw material and trimethoprimethylate lactate (TMP) as antibacterial modifier by Schiff Base reaction. At the same time, berberine (BBR) was used as graft monomer to prepare a multifunctional protective solution integrating strengthening, antibacterial and good biocompatibility. The optimal process conditions were determined by single factor experiment and response surface method. The structure and properties of the product were characterized by FT-IR, XRD, TG and SEM and the antibacterial properties were evaluated by AGAR disk diffusion method. The results showed that when the mass ratio of dialdehyde carboxymethyl cellulose and modified berberine was 7:15, reaction temperature was 40 °C, and pH was 6.0, the optimum synthetic conditions was obtained. And the minimum inhibitory concentration of the product against Staphylococcus aureus and Escherichia coli were 0.2 mg/mL, minimum diameter of antibacterial zone of 6.7 mm and 7.1 mm, respectively, with good antibacterial effect. This paper lays a foundation for further research on the safe and effective antiquities antibacterial agent, and provides a reference for its further development in paper protection.

Preparation and characterization of charcoal from feathers and its application in trimethoprim adsorption

Cheng Cheng,Jian Zhang,Chenglu Zhang,Hai Liu,Weifeng Liu

Feathers were utilized to prepare charcoal which could be used for the adsorption of the antibiotic trimethoprim (TMP). The physicochemical properties of the feather-derived charcoal were determined. The feather-derived charcoal had a surface area of 805.4 m2/g and a well-developed microporous structure. The contents of acidic and basic functional groups on feather-derived charcoal were 1.36 and 1.76 mmol/g, respectively, and pHpzc = 7.52. We studied the influences of adsorbent dose, initial TMP concentration, agitation time, temperature, pH, and ionic strength on the adsorption. The desorption properties of TMP-sorbed feather-derived charcoal were also tested. The adsorption kinetics accorded with the pseudo-second-order kinetic equation, and the overall rate of adsorption was mainly controlled by intra-particle diffusion. The adsorption isotherms for the TMP-feather-derived charcoal system are explained better by the Freundlich isotherm at lower temperature (293 K) while the Langmuir isotherm gave a better fit at higher temperature (313 K). It is considered that a combination of hydrophobic interactions and ion exchange as well as electrostatic interaction was the adsorption mechanisms.

Direct oxidation of antibiotic trimethoprim by unactivated peroxymonosulfate via a nonradical transformation mechanism

Xuerui Yang,Xi Ding,Lei Zhou,Qing Zhao,Yuefei Ji,Xingbao Wang,Jean-Marc Chovelon,Guangli Xiu

Application of activated peroxymonosulfate (PMS) to generate sulfate radical or hydroxyl radical is a promising strategy for wastewater treatment, while our knowledge on the background reaction, namely, the direct interaction between PMS and target contaminants is quite limited. In this contribution, the degradation kinetics, stoichiometry, products and mechanism of the reaction between unactivated PMS and trimethoprim (TMP), one of the most commonly detected micro-pollutants in the aquatic system were investigated systematically. The results indicated that TMP was susceptible to degradation by direct PMS oxidation via a non-radical process. By recording the decay of two reactants simultaneously, the stoichiometric ratio between PMS and TMP was estimated to be approximately 1. Higher PMS levels exhibited a promotion effect on PMS decay. And the degradation was pH-dependent, basic conditions were favorable for TMP degradation, which could be well modeled based on the species-specific reactions. The two amine groups on the pyrimidine ring were identified as the reactive sites. After direct attacks by PMS, they would be oxidized to form hydroxylamine-products, namely, N 8 –OH-TMP and N 9 –OH-TMP. These two hydroxylamine-products were quite stable and resistant to further oxidation by PMS, preventing the formation of more toxic nitroso- and nitro-products. The new findings in the present work would provide beneficial information on the strategy choice for the elimination of specific pollutants, like TMP, as PMS also exhibits relatively high reactivity towards them.

Amorphous PAA-stabilized solid dispersion for enhanced antibacterial activity and bioavailability of trimethoprim

Zhang Zhenghua,Lu Ping,Niu Ming,Long Haitao,Zhao Jie,Xu Weibing,Hang Zhaojia

The clinical application of trimethoprim (TMP) is limited by the low water solubility, short half-life and low oral bioavailability. Polyacrylic acid-stabilized trimethoprim amorphous solid dispersant (PAA/TMP ASD) are prepared by spray drying technology. The PAA/TMP ASD forms microspheres with diameters from hundreds of nanometers to tens of micrometers. The payload of TMP in the ASD can be regulated by adjusting the feeding ratio. During the continuous 7-day test, the variation range of particle size and Zeta potential is both less than 10%. In gastric juice, the ASD show low cumulative release (13.1% and 30.2%) with an initial explosive phase, whereas in PBS and intestinal fluid, the equilibrium release arrives to 64.3% and 41.5%. At a concentration of 0.097 mg/mL, the in vitro inhibition rate against E. coli can reach more than 98%. The inhibition rate of the biofilm formation is 82% at the concentration of 0.2 mg/mL. The TMP/PAA ASD shortens the peak time from 3.333 to 2.0 h, increases the peak concentration from 1.012 to 1.133 µg/mL, and significantly reduces the absorption half-life from 1.387 to 0.191 h. The TMP/PAA ASD formulation prolongs the elimination half-life from 26.033 to 34.699 h, along with increases AUC and MRT values, and demonstrates higher drug exposure and a longer duration of action in vivo. This work develops an important method to enhance the oral utilization rate of TMP.
