概述
黏液物质染色有多种方法，如AB-PAS染色、黏液HID-AB染色、标准阿利新蓝染色等，以上方法大多是利用阿利新蓝 (Alcian)属于阳离子染料可与酸性基团结合，也即阿尔辛蓝与组织内含有的阴离子基团如羧基和硫酸根形成不溶性复合物这一原理。
黏蛋白染色液 (天青A异染法)是利用天青的异染性这一特性，染色后组织或组织成分着染的颜色与染料复合物的原有颜色明显不同而且可形成鲜明的对比，染色后切片处理对异染染色的稳定性非常重要，宜采用乙醇保存。该试剂仅用于科研领域，不适用于临床诊断或其他用途。
自备材料：
1、系列乙醇、蒸馏水、去离子水、二甲苯或环保脱蜡透明液、封片胶
2、显微镜
操作步骤(仅供参考)：
1、 切片二甲苯或脱蜡液 (D292661)脱蜡至水。
2、 黏蛋白染色液 (天青A异染法)染色10min，去离子水冲洗5min。
3、 梯度乙醇脱水，二甲苯或脱蜡液 (D292661) 透明，混合封片剂封片。
染色结果:
酸性黏蛋白和蛋白多糖：红色至紫色 背景：蓝色 |

There are various methods for staining mucus substances, such as AB-PAS staining, mucus HID-AB staining, and standard Alcian Blue staining. Most of these methods utilize the principle that Alcian Blue, a cationic dye, can bind to acidic groups-specifically, Alcian Blue forms insoluble complexes with anionic groups (e.g., carboxyl groups and sulfate groups) present in tissues.
The Mucin Staining Solution (Azure A Metachromatic Method) leverages the metachromatic property of Azure A. After staining, the color of the stained tissue or tissue components differs significantly from the original color of the dye complex, creating a distinct contrast. The post-staining treatment of sections is crucial for the stability of metachromatic staining, and ethanol is recommended for preservation. This reagent is for research use only and not intended for clinical diagnosis or other purposes.
Materials to Be Prepared by the User
1. A series of ethanol solutions, distilled water, deionized water, xylene or environment-friendly dewaxing and clearing solution, and mounting medium
2. Microscope
Operating Procedures (for Reference Only)
1. Dewax sections to water using xylene or dewaxing solution (Cat. No. D292661).
2. Stain with Mucin Staining Solution (Azure A Metachromatic Method) for 10 minutes, then rinse with deionized water for 5 minutes.
3. Dehydrate with gradient ethanol solutions, clear with xylene or dewaxing solution (Cat. No. D292661), and mount sections with mixed mounting medium.
Staining Results
Acidic mucins and proteoglycans: Red to purple
Background: Blue
Precautions
1. Once the reagent is opened, use it as soon as possible to avoid affecting the subsequent experimental results.
2. For your safety and health, wear a lab coat and disposable gloves during operation.
