概述

孔雀石绿又称孔雀绿，是人工合成的染料，CAS号为569-64-2，分子量为364.92。孔雀石绿水溶液(1%)主要由孔雀石绿、去离子水组成，可用于布氏杆菌染色或其他用途，可以与其他染料合用，对真菌浸入后的病理切片染色效果较好。该试剂仅用于科研领域，不宜用于临床诊断或其他用途。

自备材料：

载玻片、盖玻片、酒精灯、光学显微镜。

操作步骤 (仅供参考)：

1、涂片：载玻片编号，于载玻片中央滴加2- 3滴孔雀石绿水溶液 (1 %) 染色2 ~ 3min。

2、加盖洁净的盖玻片，轻轻按压制成压片。

3、光学显微镜在低倍、高倍或油镜下观察细菌形态。

注意事项：

1、涂片之前应事先在背面做好圆圈标记，以便判断后续试验的位置。

2、细菌培养时间会影响染色，阳性菌培养时间过长或3死亡或细菌溶解，都常呈阴性反应。

3、试剂开封后请尽快使用，以防影响后续实验效果。

Malachite green, also known as China green, is a synthetic dye with a CAS number of 569-64-2 and a molecular weight of 364.92. The 1% aqueous solution of malachite green is mainly composed of malachite green and deionized water. It can be used for Brucella staining or other applications, and can be combined with other dyes to achieve excellent staining results for pathological sections invaded by fungi. This reagent is for research use only and is not suitable for clinical diagnosis or any other purposes.

Self-prepared Materials:

Glass slides, coverslips, alcohol lamps, optical microscopes.

Operating Procedures (For Reference Only):

1. Smear Preparation: Number the glass slides. Place 2–3 drops of 1% aqueous malachite green solution in the center of a slide and stain for 2–3 minutes.

2. Cover the smear with a clean coverslip and press gently to make a squash preparation.

3. Observe the bacterial morphology under a low-power, high-power, or oil immersion objective lens of an optical microscope.

Precautions:

1. Mark a circle on the back of the slide before preparing the smear to determine the position for subsequent observation.

2. Bacterial culture duration affects staining results. For positive bacteria, an excessively long culture period, cell death, or bacterial lysis often leads to false-negative reactions.

3. Use the reagent as soon as possible after opening to avoid compromising the results of subsequent experiments.
