阿拉丁生产的LumiDye™ 马来酰亚胺活化牛血清白蛋白 (SMCC-BSA)，即SMCC活化牛血清白蛋白，是由4-(N-马来酰亚胺基甲基)环已烷-1-羟酸琥珀酰亚胺酯(Succinimidyl-4-(N-maleimidomethyl) cyclohexane-1-carboxylate, SMCC)修饰牛血清白蛋白(Bovine Serum Albumin, BSA)形成，携带马来酰亚胺基团，用于含半胱氨酸多肽以及其他巯基化抗原的半抗原-载体偶联。
牛血清白蛋白(BSA)是常用血清蛋白，广泛应用于ELISA、免疫印迹、免疫组化等生化实验。作为高丰度血浆蛋白，BSA稳定性强、水溶性好；分子量67 kDa，分子结构复杂，具备优良免疫原性。每个 BSA 分子可偶联多个半抗原分子，以此提升抗原免疫原性，提高免疫动物针对目标表位产生高滴度抗血清的概率，因此常作为载体蛋白，与半抗原、弱免疫原偶联，用于动物免疫制备特异性抗体。
本产品为BSA与SMCC的连接产物，用 20 摩尔马来酰亚胺 / 摩尔牛血清白蛋白进行活化后，经纯化去除过量的SMCC，溶于含3%海藻糖的PBS-EDTA中，SMCC-BSA最终以冻干粉的形式提供，长期储存也不损失巯基反应性，保证了SMCC-BSA与其它含巯基分子等半抗原偶联的最佳性质。
使用方法
本产品最佳使用浓度为10mg/mL，加入200 uL 纯水，摇晃瓶体使SMCC-BSA完全溶解后可进行标记。

保存条件
4ºC避光保存，12个月内有效，切勿-20ºC或更低温度保存。
注意事项
1. 避免在反应过程中使用含有伯胺和巯基的缓冲液，以减少不必要的竞争反应。
2. 与之偶联的半抗原需含游离巯基。
3. 本产品的偶联产物，须避光保存，请勿冷冻。

Introduction
LumiDye™ SMCC Activated Bovine Serum Albumin (SMCC-BSA) is formed by modifying Bovine Serum Albumin (BSA) with Succinimidyl-4-(N-maleimidomethyl) cyclohexane-1-carboxylate (SMCC). It contains a maleimide group that reacts with thiol groups of target molecules, such as haptens.
Bovine serum albumin (BSA) is a serum albumin protein that has numerous biochemical applications including ELISAs (Enzyme-Linked Immunosorbent Assay), immunoblots, and immunohistochemistry. Like most abundant plasma proteins, it is very stable and soluble. In addition, the 67 kDa protein is sufficiently large and complex to be fully immunogenic. Several hapten molecules can be conjugated to each BSA molecule to increase their immunogenicity and likelihood of eliciting immunized animals to produce high-titer antisera to the epitopes of interest. Consequently, BSA is a popular carrier protein for conjugation to haptens and other weak antigens to make them more immunogenic for antibody production.
This product is a purified conjugate of BSA and SMCC. It is activated with 20 moles of maleimide per mole of bovine serum albumin, then purified to remove excess SMCC. Formulated in PBS‑EDTA containing 3% trehalose, SMCC‑BSA is ultimately supplied as a lyophilized powder. The long-term storage of this product does not affect its thiol reactivity, ensuring the optimal properties for conjugation with other thiol-containing molecules such as haptens.
Instruction for use
The optimal concentration for this product is 10 mg/mL. Add 200 μL of ultrapure water, gently swirl the bottle to ensure complete dissolution of the protein, then proceed with labeling.
Note: The SMCC-BSA solution must be prepared fresh before each use.

Storage
Store at 4℃ in the dark for up to 12 months. Do not store at -20℃ or lower temperatures.
Matters needing attention
1. Avoid using buffers containing primary amines and thiols during the reaction to minimize unnecessary competitive reactions.
2. The hapten to be conjugated must contain free sulfhydryl groups.
3. Products conjugated with the SMCC-BSA should be stored at 4℃ in the dark. Do not freeze.
4. During operation, always wear a lab coat, disposable gloves, and protective equipment.
