1. 手术台上取正常健康新生儿脐带,浸泡于含双抗的无菌生理盐水或 PBS 中常温或 4℃保 存;
2. 超净台内取出脐带,用生理盐水或者 PBS 洗去脐带残留血液,剪成 3-4cm 小段;
3. 用生理盐水或者 PBS 再次清洗,用组织镊去除脐带中的 2 条脐动脉;
6. 放置 5-10 分钟后,根据所选培养容器添加相应量的培养基,放置于 37℃、5%CO2 培养 箱中培养;
7. 培养 3 天后,镜下观察有无细菌污染,进行半量换液;
8. 细胞培养至 7-10 天可见长梭形细胞从组织块内爬出,7 天进行全换液,培养至 12-15 天可见大量细胞爬出,此时爬出的细胞记为 P0;
9. 待组织块周边细胞密度达到 80%-90%左右时,重组胰酶消化细胞进行传代;
1. 待细胞融和度达到约 80%后进行传代(细胞不能太密集,否则容易分化),吸弃瓶中的培 养基,加适量 DPBS 轻轻冲洗 1 次;
将细胞团块悬浮于适量无血清干细胞专用冻存液中,冻存液 重悬细胞密度为 5×10⁵-5 ×10⁶cells/ml;
2. 本产品启封前若有少量细小的凝聚物,是培养基成分的析出,属于正常现象,可轻轻混匀 后使用;

Mesenchymal Stem Cell Serum-Free Medium is a serum‑free, xeno‑free medium designed for the primary isolation and subculture of mesenchymal stem cells (MSCs) derived from various sources (e.g., bone marrow, adipose tissue, umbilical cord tissue, and dental pulp). It maintains the multi‑lineage differentiation potential of MSCs while supporting their robust proliferation rate.
Product Application
Suitable for the growth and expansion of human MSCs from multiple sources.
Instructions for Use
Preparation of MSC Serum‑Free Complete Medium:
Thaw the stem cell culture supplement completely at 2–8 °C. Add it to the MSC serum‑free basal medium at a ratio of 1:20, and mix thoroughly to obtain complete human MSC medium. Store at 4 °C and use within 2 weeks. For primary (P0) isolation, it is recommended to add penicillin‑streptomycin (dual antibiotics) to prevent contamination.
Primary Isolation of MSCs (Tissue Explant Method)
1.Collect normal healthy neonatal umbilical cord during surgery and immerse it in sterile physiological saline or PBS containing dual antibiotics, store at room temperature or 4 °C.
2.In a laminar flow hood, remove the cord and wash off residual blood with saline or PBS, then cut it into 3–4 cm segments.
3.Wash again with saline or PBS, and use tissue forceps to remove the two umbilical arteries from the cord.
4.Open the cord along the umbilical vein, scrape away the venous tissue with forceps, wash the remaining tissue 2–3 times in saline or PBS, and cut into pieces of about 1.5 mm.
5.Evenly distribute the minced tissue pieces onto a culture dish, leaving gaps of about 5 mm between them.
6.After letting stand for 5–10 min, add an appropriate volume of medium according to the culture vessel used, and place in a 37 °C, 5 % CO₂ incubator.
7.After 3 days of culture, check under a microscope for bacterial contamination and perform a half‑volume medium change.
8.Within 7–10 days of culture, spindle‑shaped cells will begin to migrate out of the tissue explants. Perform a full medium change at day 7. By days 12–15, a large number of cells will have migrated out; these cells are designated as P0 (primary).
9.When the cell density around the explants reaches about 80–90 % confluence, dissociate the cells with recombinant trypsin for subculture.
10.The cells after the first passage are designated P1, and so on.
Subculture and Cryopreservation of Human MSCs
1.Subculture when cell confluence reaches approximately 80 % (avoid over‑confluence, which may induce differentiation). Aspirate the medium, and rinse gently once with an appropriate volume of DPBS.
2.Add a gentle cell dissociation solution according to the flask size (e.g., 3–5 mL for a T175 flask) and incubate at room temperature for 1–3 min (tap the flask gently to help detach cells).
3.Under a microscope, confirm that cells have completely detached, then gently pipette to break up clumps. Centrifuge at 1000 rpm for 3–5 min.
4.Carefully aspirate the supernatant, resuspend the cell pellet in an appropriate volume of complete medium, and mix well.
5.Seed the cells at a density of 6000–8000 cells/cm² into a T175 flask containing 25 mL of cell suspension. Gently rock to distribute evenly, then place in a 37 °C, 5 % CO₂ incubator.
6.Replace the medium every 2–3 days. When confluence reaches about 80 %, subculture or harvest the cells for cryopreservation.
Cryopreservation
Resuspend the cell pellet in an appropriate volume of serum‑free, stem cell‑specific cryopreservation medium to achieve a final density of 5×10⁵–5×10⁶ cells/mL.
Transfer the suspension into cryovials, place them in a –80 °C freezer for 24 h, then transfer to liquid nitrogen for long‑term storage.
Thawing of MSCs
1.Pre‑warm the complete human MSC medium. In a biosafety cabinet or laminar flow hood, pipette an appropriate volume (e.g., 10–15 mL for a T75 flask) of complete medium along the upper surface of the culture flask, avoiding direct contact with the bottom surface. Then gently lay the flask flat and place it in a 37 °C, CO₂ incubator for 5–10 min before use. This allows the attachment‑promoting substances in the medium to coat the bottom, enhancing MSC adhesion and reducing the occurrence of "growth voids."
2.Remove the cryovial from liquid nitrogen and quickly thaw it in a 37 °C water bath.
3.Disinfect the outer surface of the cryovial with 70–75 % ethanol. In a biosafety cabinet, open the vial and slowly transfer the cell suspension into a centrifuge tube containing 5–10 mL of pre‑warmed complete medium.
4.Centrifuge at 1200 rpm for 5 min, discard the supernatant, and resuspend the cell pellet in 2 mL of complete medium. Count the cells and seed them at 6000–8000 cells/cm² into the culture flask that was pre‑incubated in step 1. When adding the cells, dispense the suspension directly onto the bottom surface—do not flush against the bottom.
5.Gently rock the flask to distribute the cells evenly. Place the flask in a 37 °C, 5 % CO₂ incubator with saturated humidity for static culture.
6.After 2–3 days, replace with fresh complete medium.
Precautions
1.Thaw the stem cell culture supplement at 2–8 °C. Avoid repeated freeze‑thaw cycles; it is recommended to aliquot and store at –20 °C in the dark.
2.If minor fine precipitates appear before opening, this is normal due to medium component precipitation; mix gently before use.
3.Strictly follow aseptic techniques when using this product.
Storage Conditions
MSC serum‑free basal medium: store at 2–8 °C, protected from light. Shelf life: 12 months.
