ICDHm catalyzes the oxidative decarboxylation of isocitrate to produce α-ketoglutarate, accompanied by the reduction of NAD⁺ to NADH. α-ketoglutarate exhibits a characteristic absorption peak at 505 nm, and the activity of ICDHm can be quantitatively characterized by measuring the absorbance change at this wavelength.
