甲基蓝 (Methyl Blue)，部分文献俗称棉蓝，分子量为 799.8，常被用作生物染色剂，用于动物组织学中原生动物活体、细菌、神经细胞的染色。该试剂仅用于科研领域，不适用于临床诊断或其他用途。
操作步骤 (仅供参考)：
1、样品处理：
①对于石蜡切片： 二甲苯或 脱蜡透明液中脱蜡 5～10 min。 更换新鲜的二甲苯或脱蜡透明液，再次脱蜡 5～10 min。 无水乙醇 5 min。 90% 乙醇 2 min。 70% 乙醇 2 min。 蒸馏水洗涤 2 min。
②对于冰冻切片： 蒸馏水洗涤 2 min。
③对于培养细胞： 用 4% 多聚甲醛固定 10 min 以上 (或采用如下步骤：每 25 cm² 加入 PBS / 甲醇溶液 5 mL，静置 2 min，弃液；加入新鲜甲醇 5 mL，静置 10 min，弃液)。蒸馏水洗涤 2 min，更换新鲜蒸馏水再次洗涤 2 min。
2、染色：
①Methyl Blue Stain (1%) 染色 3～10 min (可根据染色效果与实验需求调整染色时间)。
②用蒸馏水充分洗涤，随后进行镜检与拍照。
染色结果：

注意事项：
1、首次使用本试剂，建议选取 1～2 个样品开展预实验。
2、为保障实验安全，请穿戴实验服、一次性手套进行操作。
3、试剂开封后建议尽快使用，以免影响后续实验效果。
Methyl Blue, referred to as cotton blue in some literatures, has a molecular weight of 799.8. It is widely used as a biological stain for staining living protozoa, bacteria and nerve cells in animal histology. This reagent is for research use only and not intended for clinical diagnosis or other applications.
Procedure (For reference only):
1. Sample treatment:
① Paraffin sections: Deparaffinize in xylene or deparaffinizing clearing solution for 5～10 min. Replace with fresh xylene or deparaffinizing clearing solution and deparaffinize again for 5～10 min. Absolute ethanol 5 min. 90% ethanol 2 min. 70% ethanol 2 min. Wash with distilled water for 2 min.
② Frozen sections: Wash with distilled water for 2 min.
③ Cultured cells: Fix with 4% paraformaldehyde for more than 10 min (or follow the alternative procedure: add 5 mL PBS/methanol solution per 25 cm², incubate for 2 min and discard the solution; add 5 mL fresh methanol, incubate for 10 min and discard the solution). Wash with distilled water for 2 min, replace with fresh distilled water and wash again for 2 min.
2. Staining:
① Stain with Methyl Blue Stain (1%) for 3～10 min (the staining time can be adjusted according to staining effect and experimental requirements).
② Wash thoroughly with distilled water, then perform microscopic examination and photography.
