概述
神经元细胞体包括一个具有皱褶核膜的大细胞核、稀疏的染色质和一个明显的核仁，在细胞体中细胞质是尼氏颗粒，即能够代表粗面内质网并在很多神经元中产生特异的斑点状嗜碱性表现的嗜碱性颗粒。尼氏颗粒可以用很多染色来显示如中性红、亚甲基蓝、甲苯胺蓝和甲基紫等，染色的变异、pH和分化的时间使一些染色既可以仅突出尼氏物质，也可以显示神经元的细胞核和神经胶质。尼氏体 (Nissl body)或称尼氏小体是分布于神经细胞胞质内的三角形或椭圆形小块状物质，能被碱性染料如硫堇、亚甲蓝、甲苯胺蓝和焦油紫等染料染成紫蓝色，各种神经细胞内都含有尼氏体，但其形状、数量、分布位置常常不同，尼氏体也存在于树突中，但不在于轴突和胞体的轴丘；尼氏体会因为生理状态的变化而变化，尼氏体是神经元内合成蛋白质的重要部位，当神经元受到刺激后，胞体内的尼氏体会明显减少。
尼氏染色液(Nissl Stain，焦油紫法)采用焦油紫 (Cresyl violet)作为核心染料，焦油紫具有感光作用，能够很好地显示尼氏体的变化，主要优点是操作简便、染色稳定、适用范围广，可以用于石蜡组织切片的尼氏物质、神经元等的染色，尼氏体的存在和消失是神经细胞是否受损的重要指标，当发生脑炎、脑缺血、轴突反应等情况时，尼氏体会发生溶解甚至消失。该试剂仅用于科研领域，不适用于临床诊断或其他用途。
自备材料：
1、系列乙醇、蒸馏水
2、恒温箱、酒精灯、显微镜
操作步骤(仅供参考)：
1、新鲜组织固定于乙醇、Carnoy固定液或10%中性福尔马林溶液后，常规脱水包埋。
2、切片厚6-8µm，常规二甲苯或脱蜡透明液脱蜡至水。
3、切片入Cresyl Violet Stain，将染色缸置于恒温箱56℃浸染1h或37℃浸染2-3h；如果染色效果不佳，可考虑酒精灯上加温使切片冒气泡为止 (大约10min)，冷蒸馏水冲洗。
4、入Nissl Differentiation分化1-3min，在显微镜下观察至背景接近于无色为止。
5、无水乙醇迅速脱水，常规二甲苯或脱蜡透明液透明，中性树胶封固。
染色结果：
尼氏体: 紫色
背景: 接近于无色或浅蓝色
注意事项：
1、尼氏体离体后容易溶解，所以组织取出后应立即固定，否则难以着色。
2、组织固定起着非常重要的作用，固定可采用乙醇、Carnoy固定液或中性福尔马林溶液。
3、该染色液对石蜡组织切片的尼氏染色效果较好。
4、石蜡切片厚度6-8µm或25µm (皮质神经元密度的评估要用25µm厚的切片)。
5、染色后的标本务必避光保存，否则容易褪色。

The neuronal cell body contains a large nucleus with a folded nuclear membrane, sparse chromatin, and a distinct nucleolus. Within the cell body, the cytoplasm is filled with Nissl granules—basophilic granules that represent the rough endoplasmic reticulum and produce a characteristic spotted basophilic appearance in many neurons. Nissl granules can be visualized using various stains, such as neutral red, methylene blue, toluidine blue, and methyl violet. Variations in staining protocols, pH values, and differentiation times allow some stains to either specifically highlight Nissl substance or simultaneously visualize neuronal nuclei and neuroglia. Nissl bodies, also known as Nissl substance, are triangular or oval granular structures distributed in the cytoplasm of neurons. They can be stained purple-blue by basic dyes including thionine, methylene blue, toluidine blue, and cresyl violet. All types of neurons contain Nissl bodies, but their shape, quantity, and distribution often vary. Nissl bodies are also present in dendrites but are absent from axons and the axon hillock of the cell body. Nissl bodies undergo changes in response to the physiological state of neurons: as important sites for protein synthesis in neurons, their volume decreases significantly when neurons are stimulated.
Nissl Staining Solution (Cresyl Violet Method) uses cresyl violet as the core dye. Cresyl violet is light-sensitive and can well demonstrate the changes of Nissl bodies. It has the main advantages of simple operation, stable staining, and broad applicability. It is suitable for staining Nissl substance and neurons in paraffin-embedded tissue sections. The presence or disappearance of Nissl bodies is a crucial indicator of neuronal damage. Nissl bodies may dissolve or even disappear under pathological conditions such as encephalitis, cerebral ischemia, and axonal reaction. This reagent is for research use only and not intended for clinical diagnosis or other purposes.
Materials to Be Prepared by User
1. Graded ethanol, distilled water
2. Incubator, alcohol lamp, microscope
Procedure (for reference only)
1. Fix fresh tissues in ethanol, Carnoy's fixative, or 10% neutral formalin solution, followed by routine dehydration and embedding.
2. Cut sections with a thickness of 6–8 μm, then perform routine dewaxing to water using xylene or environment-friendly dewaxing clearing solution.
3. Immerse the sections in Cresyl Violet Stain, place the staining jar in an incubator and incubate at 56℃ for 1 hour or at 37℃ for 2–3 hours. If the staining result is unsatisfactory, heat the sections over an alcohol lamp until bubbles appear (approximately 10 minutes), then rinse with cold distilled water.
4. Differentiate the sections in Nissl Differentiation Solution for 1–3 minutes, and observe under a microscope until the background is nearly colorless.
5. Dehydrate rapidly with anhydrous ethanol, clear routinely with xylene or dewaxing clearing solution, and mount with neutral mounting medium.
Staining Results
Nissl bodies: Purple
Background: Nearly colorless or pale blue
Precautions
1. Nissl bodies are prone to dissolution after being isolated from the body. Therefore, tissues must be fixed immediately after collection; otherwise, effective staining cannot be achieved.
2. Tissue fixation plays a vital role. Ethanol, Carnoy's fixative, or neutral formalin solution can be used as fixatives.
3. This staining solution yields favorable results for Nissl staining of paraffin-embedded tissue sections.
4. Prepare paraffin sections with a thickness of 6–8 μm or 25 μm (25 μm-thick sections are required for evaluating the density of cortical neurons).
5. Stained specimens must be stored in the dark to prevent fading.
6. For your safety and health, please wear a lab coat and disposable gloves during operation.
7. Please use the reagent as soon as possible after opening to avoid affecting subsequent experimental results.
