中性粒细胞碱性磷酸酶染色液（NAP）采用偶氮偶联法（同时偶联法），专门用于血液涂片或骨髓细胞涂片的中性粒细胞碱性磷酸酶（NAP）组织化学染色。碱性磷酸酶（ALP/AKP）是一种广泛分布于哺乳动物多种组织中的磷酸酯酶，最适 pH 为 9.2–9.8，NAP 作为成熟中性粒细胞的标志酶之一，主要定位于胞膜及分泌囊泡中。 该染色液利用碱性磷酸酶在碱性条件下水解 AB-BI 磷酸盐底物，释放萘酚与磷酸，萘酚随即与重氮盐偶联生成不溶性有色沉淀，定位于酶活性部位。阳性反应呈现鲜明的蓝色，染色结果稳定可靠，较传统金属盐沉淀法（如钙-钴法）具有操作简便、染色时间短、背景清晰等优势，适用于科研实验中对 NAP 阳性率及积分的判读分析。 本品含重氮盐组分，具有一定刺激性，操作时请佩戴手套，做好个人防护；废液请按有害化学品规范处置。仅限科研使用，不可用于临床诊断。
Neutrophil Alkaline Phosphatase Staining Solution (NAP) employs the azo coupling method (simultaneous coupling method) and is specifically designed for the histochemical staining of neutrophil alkaline phosphatase (NAP) in blood smears or bone marrow cell smears. Alkaline phosphatase (ALP/AKP) is a phosphatase widely distributed in various mammalian tissues, with an optimal pH range of 9.2–9.8. As one of the marker enzymes of mature neutrophils, NAP is primarily localized on the plasma membrane and in secretory vesicles. This staining solution utilizes the enzymatic hydrolysis of the AB-BI phosphate substrate by alkaline phosphatase under alkaline conditions, releasing naphthol and phosphate. The released naphthol immediately couples with a diazonium salt to form an insoluble colored precipitate at the enzyme activity site. Positive reactions appear as a distinct blue coloration. The staining results are stable and reliable, offering advantages over traditional metal salt precipitation methods (such as the calcium-cobalt method), including simpler operation, shorter staining time, and clearer background. It is suitable for research applications involving the assessment of NAP positivity rates and scores. This product contains diazonium salt components and is irritant.
