概述
组织保存液是款即用型低温运输保存液，可用于人与动物新鲜组织样本的短期存储与运输，可有效避免样本在储存或运输过程中的细胞凋亡、坏死等现象，保证细胞的功能性和活力。本产品可在2-8℃条件下保存离体组织72小时，并兼容以下实验：
- 原代细胞提取与培养
- 单细胞悬液制备（用于流式分析、单细胞测序等）
- RNA提取与测序（保障RNA完整性，RIN值≥8）
- 组织病理学切片制备
保存说明
未开封保存液：-20度避光保存，有效期12个月。
开封后保存液：4℃密封保存，建议7天内用完。
组织保存条件：离体组织需完全浸没于保存液中，4℃冷藏，最长保存72小时。
组织处理
离体组织，先用75%酒精浸泡5分钟，无菌PBS洗后尽快放入保存液（体积≥组织体积的5倍），避免干燥。若组织较大，建议切割为厚度≤5mm的小块，确保保存液充分渗透。
保存与运输
4℃静置保存，避免剧烈震动或反复冻融。长途运输需使用冰盒或冷链箱（维持4℃）。
后续实验处理
1. 原代细胞提取：取出组织后，用预冷无菌PBS冲洗2次，按标准消化流程操作。
2. 单细胞提取：机械或酶解法处理前，需去除残留保存液。
3. RNA提取：直接转移至裂解液中，或液氮速冻后-80℃长期保存。
注意事项
1. 禁止冷冻保存组织，以免冰晶破坏细胞结构。
2. 保存时间超过72小时可能导致细胞活性下降或RNA降解，建议尽快处理。
3. 若用于RNA测序，操作全程需佩戴手套并使用RNase-free耗材。
4. 组织长期保存（＞72小时）建议更换新鲜保存液或分装冻存（需验证兼容性）。
5. 本产品仅限于专业人员的科学研究用，不得用于临床诊断或治疗，不得用于食品或药品。

Tissue Storage Solution is a ready‑to‑use low‑temperature transport and storage solution for short‑term storage and transport of fresh human or animal tissue samples. It effectively prevents apoptosis, necrosis, and other damage during storage or transport, thereby maintaining cell functionality and viability. This product preserves excised tissues at 2–8 °C for up to 72 hours and is compatible with the following applications:
Primary cell isolation and culture
Preparation of single‑cell suspensions (for flow cytometry, single‑cell sequencing, etc.)
RNA extraction and sequencing (ensures RNA integrity, RIN ≥ 8)
Histopathological sectioning
Storage Instructions
Unopened solution: Store at –20 °C, protected from light. Shelf life: 12 months.
Opened solution: Store sealed at 4 °C; recommended for use within 7 days.
Tissue Preservation Conditions: Excised tissue must be completely submerged in the preservation solution and stored at 4 °C for a maximum of 72 hours.
Tissue Handling
Immediately after excision, immerse the tissue in 75% ethanol for 5 minutes, rinse with sterile PBS, and then place it into the preservation solution as soon as possible (volume of solution ≥ 5× tissue volume). Avoid drying. For large tissues, cut into pieces no thicker than 5 mm to ensure adequate penetration of the preservation solution.
Preservation and Transport
Store statically at 4 °C, avoiding vigorous shaking or repeated freeze‑thaw cycles. For long‑distance transport, use ice packs or a refrigerated box (maintain 4 °C).
Subsequent Experimental Procedures
Primary cell isolation: After removing the tissue, rinse twice with pre‑chilled sterile PBS, then proceed with standard digestion protocols.
Single‑cell isolation: Remove residual preservation solution before mechanical or enzymatic dissociation.
RNA extraction: Transfer the tissue directly into lysis buffer, or snap‑freeze in liquid nitrogen and store at –80 °C for long‑term preservation.
Precautions
Do not freeze the tissue in the preservation solution, as ice crystals will damage cell structure.
Storage for more than 72 hours may reduce cell viability or cause RNA degradation. Process the tissue as soon as possible.
When used for RNA sequencing, always wear gloves and use RNase‑free consumables throughout the procedure.
For long‑term tissue preservation (>72 hours), it is recommended to replace with fresh preservation solution or aliquot and freeze (compatibility must be verified).
