This product is a ready-to-use, sterile-filtered concentrate of Phytohemagglutinin-L (PHA-L). PHA is a mixture of lectins isolated from red kidney beans (i.e., Phaseolus vulgaris). PHA-L is a component of PHA that exhibits strong leukoagglutination activity and mitogenic activity, but has little erythroagglutination activity. This reagent is designed for in vitro activation of human and mouse white blood cells.
Usage Protocol
This is a pre-titrated 500× solution and can be diluted to 2 µL per mL of culture medium.
Primary Applications
Nonspecific activation and proliferation assays of T lymphocytes.
Culture of peripheral blood mononuclear cells (PBMCs) and induction of cytokine secretion.
Lymphocyte transformation tests.
Positive control stimulant in immune cell functional studies (e.g., ELISPOT, flow cytometric cytokine detection).

Aliquot upon receipt and store frozen at -20°C(12 months). Avoid repeated freeze-thaw cycles.
Human peripheral blood lymphocytes were stimulated with Phytohemagglutinin-L (PHA-L) Solution (500X) (P1511171) for 3 days. The cells were then stained with Recombinant IL-2 Receptor alpha/CD25 Antibody (AF647) (Ab230822) (red) compared to an isotype control (blue). Blue - Mouse IgG (AF647) (Ab190077). Black - Unlabeled control, cells without incubation with primary antibody. Unstimulated PBMCs were used as control (left panel).
