Under certain conditions, sulfhydryl groups undergo a nucleophilic reaction, reacting with 5,5'-dithiobis-2-nitrobenzoic acid (DTNB) to form yellow 2-nitro-5-mercaptobenzoic acid (TNB). This product exhibits a characteristic absorbance peak at 412 nm. By measuring the change in absorbance at this wavelength, the protein sulfhydryl content can be quantitatively determined.
