This plant tissue reactive oxygen species (ROS) assay kit employs the classic nitroblue tetrazolium (NBT) histochemical staining method. The core principle is that the pale‑yellow NBT, which possesses tissue permeability, is selectively reduced by superoxide anion (O₂⁻) in plant tissues to generate an insoluble blue formazan precipitate that is deposited in situ. By analyzing the intensity and distribution of the blue precipitate, the accumulation and dynamics of superoxide anions in different tissues or cellular structures can be visually and qualitatively assessed.
