5.磁珠混匀:阿拉丁多糖纯化磁珠使用前需充分混匀。可采用振荡、涡旋或短暂水浴超声处理,至磁珠完全重悬、底部无沉淀;
6.磁珠清洗:取适量阿拉丁多糖纯化磁珠于离心管中,磁分离弃上清。加入1 mL 结合液重悬洗涤,磁分离去上清,重复3次。末次洗涤后,以结合液定容至10 mg/mL,混匀备用;
7.粗多糖粉末样品推荐用结合液溶解;若样品为溶液状态,可用结合液稀释、超滤置换,或0.1 M HCl/NaOH调节 pH 至5–7范围;
8.取1 mL 粗多糖样品于离心管中,加入清洗后磁珠悬液,涡旋振荡10 s 充分混匀,于25°C 连续旋转或振荡孵育30 min;
注:初次实验推荐用量:2 mL,即 20 mg 磁珠;用户可根据初次纯化效果对磁珠用量进行调整;
9.孵育结束后,将离心管置于磁力架上,磁分离2-3 min(或至上清完全澄清);
1.磁珠取用前应充分混匀,防止取用改变磁珠浓度,避免长时间超声对磁珠表面破坏;
2.磁珠在磁分离后应待溶液完全澄清后,吸弃上清,避免损失磁珠;
Natural polysaccharides exhibit a wide range of biological activities, including immunomodulatory, antioxidant, antiviral, antidepressant, prebiotic, and metabolic regulatory effects, and have therefore attracted considerable attention in the fields of functional foods and pharmaceuticals. However, high purity is a prerequisite for structural characterization and bioactivity evaluation, as residual proteins and pigments in crude polysaccharide preparations are major sources of interference that can compromise analytical accuracy.
Aladdin Polysaccharide Purification Magnetic Beads enable the simultaneous one-step removal of proteins and pigments. The purification process is mild and efficient, with a high polysaccharide recovery rate, making the beads suitable for the high-quality preparation of natural polysaccharides from a variety of sources and providing a convenient and reliable purification solution for polysaccharide research and development.
Reference Procedure
A. Materials Preparation
1. Magnetic beads: Aladdin Polysaccharide Purification Magnetic Beads
2. Magnetic rack: A magnetic rack suitable for magnetic bead separation
3. Sample: Crude polysaccharide sample
4. Binding buffer: 20 mM acetic acid-sodium acetate buffer, pH 4.0
B. Magnetic Bead Preparation
5. Bead mixing: Thoroughly mix the Aladdin Polysaccharide Purification Magnetic Beads before use. The beads may be resuspended by shaking, vortexing, or brief water-bath sonication until the magnetic beads are completely resuspended with no sediment at the bottom;
6. Bead washing: Transfer an appropriate amount of Aladdin Polysaccharide Purification Magnetic Beads into a centrifuge tube, perform magnetic separation, and discard the supernatant. Add 1 mL of binding buffer to resuspend and wash the beads, perform magnetic separation, and remove the supernatant. Repeat 3 times. After the final wash, adjust the volume with binding buffer to a final bead concentration of 10 mg/mL and mix well for use;
C. Crude Polysaccharide Sample Preparation
7. Crude polysaccharide powder samples are recommended to be dissolved in binding buffer; if the sample is in solution form, it may be diluted with binding buffer, subjected to buffer exchange by ultrafiltration, or adjusted to pH 5–7 with 0.1 M HCl/NaOH;
D. Polysaccharide Purification
8. Transfer 1 mL of crude polysaccharide sample into a centrifuge tube, add the washed magnetic bead suspension, vortex for 10 s to mix thoroughly, and incubate at 25°C with continuous rotation or shaking for 30 min;
Note: Recommended amount for the initial experiment: 2 mL, corresponding to 20 mg of magnetic beads; users may adjust the amount of magnetic beads according to the initial purification results;
9. After incubation, place the centrifuge tube on a magnetic rack and perform magnetic separation for 2–3 min (or until the supernatant becomes completely clear);
10. Carefully transfer the supernatant to a clean centrifuge tube. The resulting solution is the purified polysaccharide sample after removal of impurities.
Note: If protein removal after a single adsorption does not meet expectations, collect the supernatant, add freshly washed magnetic beads, and repeat steps 8–9 for a second adsorption.
Precautions
1. Thoroughly mix the magnetic beads before use to avoid changes in bead concentration during sampling, and avoid prolonged sonication to prevent damage to the bead surface;
2. After magnetic separation, wait until the solution becomes completely clear before aspirating and discarding the supernatant to avoid loss of magnetic beads;
3. Avoid freeze-thaw cycles during use and storage of the magnetic beads.
