RPMI 1640 medium was developed by Moore et al. in 1967 at the Roswell Park Memorial Institute (RPMI), New York. It is a modified formulation of McCoy's 5A medium and employs a bicarbonate buffering system. Unlike most mammalian cell culture media, the original formula was designed at pH 8.0; however, this product is adjusted to pH 7.0–7.4. Initially developed for lymphocyte culture, RPMI 1640 is now widely used for a variety of normal and cancer cell types, particularly for suspension cells, and remains one of the most extensively used media.
Antibiotic Supplementation
This product contains a penicillin‑streptomycin mixture (commonly referred to as "dual antibiotics"), the most frequently used combination for preventing microbial contamination in in vitro culture. Penicillin interferes with bacterial cell wall synthesis and is particularly effective against Gram‑positive bacteria; streptomycin binds to the 30S ribosomal subunit, inhibiting protein synthesis, and is effective against both Gram‑negative and Gram‑positive bacteria, with special efficacy against Gram‑negative organisms. The combination effectively prevents most bacterial contamination.
Composition and Usage
The medium contains a variety of amino acids, vitamins, inorganic salts, and other essential nutrients for cell growth. However, it does not contain proteins, lipids, or any growth factors; therefore, supplementation with serum or a serum‑free additive is required for use.
