操作简便高效:一步均质反应,无需洗板、裂解、有机溶剂溶解,兼容 96/384 孔高通量筛选;
对于大多数细胞,通常孵育1~4 h内即可得到理想的检测效果;延长孵育时间,较低数量的细胞的信号强度能得到显著改善,从而使检测灵敏度提高,但是较高数量的细胞的信号会逐渐达到平台期不再呈现线性关系。例如96孔板中,对于5万个细胞/孔的HeLa细胞孵育1 h即可得到理想的检测效果;

Resazurin Cell Viability Assay Kit, also known as Alamar Blue Kit, is developed based on the resazurin chromogenic system. It serves as an integrated detection kit that enables quantitative measurement of cell proliferation and in vitro cytotoxicity evaluation of compounds simultaneously. Its core active ingredient is high-purity resazurin redox indicator, which is compatible with the vast majority of adherent and suspension cell lines. The core detection principle relies on the redox reaction mediated by mitochondrial dehydrogenase in viable cells. Oxidized resazurin is dark blue with no endogenous fluorescence and can freely penetrate cell membranes to enter intracellular compartments. Metabolically active viable cells reduce resazurin to pink, highly fluorescent resorufin via the mitochondrial respiratory chain. Both fluorescence intensity and absorbance value at 573 nm show a strong linear positive correlation with the number of living cells. Dead cells with impaired metabolic capacity cannot trigger this reduction reaction and produce no fluorescent or chromogenic signals. As a metabolic-responsive probe, high-purity resazurin stabilizes the redox state of the dye and reduces background interference from culture media, with no need for additional electron carriers, lysis buffers or cosolvents. Similar to mainstream CCK-8 kits on the market, this product falls into the same category of cell proliferation and cytotoxicity detection reagents and can be used interchangeably in experiments for identical testing purposes. However, they differ fundamentally in chemical system and detection mode. CCK-8 uses WST-8 tetrazolium salt as the core substrate, which is reduced by cells to form water-soluble orange formazan products and only supports single-channel absorbance colorimetric detection at 450 nm. In contrast, this kit adopts a redox dye system to realize dual-channel detection (fluorescence + absorbance) for quantitative reading via a fluorescence microplate reader. Compared with traditional tetrazolium salt assays such as CCK-8 and MTT, this kit delivers multiple core advantages: First, detection sensitivity is greatly improved, with the fluorescent method stably detecting as low as 50–100 cells per well, ideal for trace cell samples. Second, the reagent exhibits extremely low cytotoxicity, allowing 1–24 hours of prolonged incubation for dynamic real-time cell monitoring. Third, it features a homogeneous one-step assay without washing steps, cell lysis or organic solvent dissolution, making it fully compatible with high-throughput screening in 96-well and 384-well plates. It is widely applicable to various experimental scenarios, including high-throughput cytotoxicity screening of small-molecule drugs and natural products, dynamic monitoring of cell proliferation curves, basic biological research on stem cells and tumor cells, combined apoptosis assessment, and quantitative activity detection of primary cells and microorganisms. The Alamar Blue detection technology based on the resazurin redox chromogenic system has decades of mature research application history. This standardized commercial kit is an internationally recognized standard solution for quantifying in vitro cellular metabolic activity. It overcomes the technical limitations of traditional tetrazolium salt methods like CCK-8, which are limited to endpoint quantification, poor performance with trace cell samples, and inability to track long-term dynamic cell status. It addresses pain points such as insufficient data accuracy, cumbersome experimental workflows and high sample consumption in drug development and basic cell research. For segmented research applications, this kit works with all common adherent and suspension cells. Equipped with a fluorescence microplate reader, it can accurately quantify the killing effect of chemotherapeutic drugs, candidate small molecules and environmental toxicants on tumor cells and normal primary cells, and stably output key pharmacodynamic data including IC50 values and cell proliferation inhibition rates. In stem cell and tissue engineering research, it enables long-term non-invasive tracking of cell viability to support the efficacy evaluation of scaffold materials and induction/differentiation systems. In combined models of cellular apoptosis and oxidative stress, it allows simultaneous co-detection of cell viability and apoptotic pathway markers to fully elucidate the mechanism of drug action. It is also adaptable to rapid activity screening of microorganisms and fungi, expanding the evaluation scope of in vitro toxicology and anti-infective drugs. As a stable, low-interference and highly reproducible quantitative detection tool for cell biology, tumor pharmacology, regenerative medicine and drug screening, this kit efficiently generates accurate and repeatable data on cellular metabolic activity. It provides reliable experimental evidence for efficacy assessment of drug candidates, analysis of cellular physiological mechanisms and construction of in vitro disease models, accelerates the transformation of basic research achievements and preclinical drug safety evaluation, and promotes the standardized development of in vitro research on oncology, degenerative diseases, anti-infection and other fields.
Application Scope
Growth curve measurement of adherent/suspension cells, monitoring of stem cell proliferation and differentiation, IC50 determination of anti-tumor drugs, antibacterial agents and natural products, biosafety evaluation of biomaterials, research on oxidative stress and radiation damage, co-staining with apoptotic probes in the same well, gene editing experiments, and optimization of primary cell culture conditions.
Product Features
1.Dual detection modes: Oxidized resazurin is reduced by viable cells to fluorescent resorufin, supporting dual-channel quantitative detection via fluorescence and absorbance with a microplate reader.

3.Low cytotoxicity: Minimal damage to cells; incubation can be extended up to 24 hours. Cells after detection can be harvested for subsequent experiments such as Western Blot, qPCR and enzyme activity assays.
4.Simple and efficient operation: One-step homogeneous reaction without washing, lysis or organic solvent dissolution, compatible with high-throughput screening on 96-well and 384-well plates.
5.Excellent comprehensive performance: No organic solvent dissolution required compared with MTT to avoid crystal interference; wider linear range compared with CCK-8; directly reflects viable cell metabolic activity with higher sensitivity compared with the LDH release assay.
Precautions
Prolonged exposure of this kit to light will elevate background fluorescence during detection; always store and operate away from light.
Aliquot the reagent upon first use to avoid repeated freeze-thaw cycles.
The capacity of different cell lines to reduce resazurin into resorufin depends on their metabolic activity and incubation duration with resazurin.
Optimal detection results can generally be obtained after 1–4 hours of incubation for most cell types. Extending incubation time significantly boosts signal intensity for low cell densities and improves detection sensitivity, while signals from high cell concentrations will gradually reach a plateau and lose linear correlation. For example, ideal signals can be achieved after 1-hour incubation for HeLa cells at 50,000 cells per well in a 96-well plate.
Black 96-well or 384-well cell culture plates are required for detection to prevent signal cross-talk between adjacent wells.
Colorimetric detection can also be performed at 570 nm with 600 nm set as the reference wavelength; however, fluorescence detection is recommended for higher sensitivity.
For most cell types, linearity will be lost when cell density reaches 50,000 or 100,000 cells per well, though fluorescence readings will continue to rise. For a small subset of cell lines, fluorescence signals may slightly decline as cell density further increases.

II. Operating Procedures
1. Cell Culture
(1) Suspension cells
Use black 96-well plates, add 100 μL cell suspension per well, and control the cell density below 30,000 cells per well. Wells filled with culture medium only serve as blank controls. Complete cell culture, drug treatment and cell gradient seeding according to your experimental protocol.
(2) Adherent cells
Digest adherent cells in advance and seed them into black 96-well plates; perform drug treatment as designed after full cell attachment.
a. Add an appropriate amount of trypsin solution to dissociate cells.
Note: The dosage, digestion duration and temperature of trypsin shall be adjusted according to laboratory routine experience.
b. Transfer cell suspension to suitable centrifuge tubes, centrifuge at 1000 rpm for 5 min at room temperature, then aspirate and discard the supernatant.
c. Seed an appropriate number of cells into black 96-well plates; the cell confluency after attachment should not exceed 80%–85%.
d. Conduct drug treatment following the experimental design once cells adhere fully.
Note: The optimal seeding density varies by cell type. For first-time experiments, set a cell concentration gradient to select the density with an ideal fluorescence range for subsequent tests.
If using 384-well plates, seed 25 μL cell suspension per well (volume varies by plate type). The cell number per well should be controlled below 8,000. Multichannel pipettes or automated cell culture systems are recommended for seeding.
Note: Refer to Section 3 of Pre-experiment Preparation for control group configuration.
2. Cell Viability Detection
(1) Equilibrate thawed Resazurin Solution to room temperature and take an appropriate volume as needed.
(2) Mix the detection reagent thoroughly, add it to each well at a volume ratio of 10% relative to culture medium: 10 μL per well for 96-well plates, or 2.5 μL per well for 384-well plates. Shake the plate gently for uniform mixing.
|
Note: If the initial culture volume per well of a 96-well plate is 200 μL, add 20 μL of reagent; the dosage can be scaled proportionally for other volumes accordingly. To achieve optimal detection performance, the volume ratio between Resazurin assay reagent and cell culture medium can be moderately adjusted according to cell line type, incubation duration and expected linear range.
(3) Incubate the plate for 1–4 hours under standard cell culture conditions (37 °C, 5% CO₂). The incubation time can be flexibly extended within 1–24 hours based on the actual color development signal.
Note: Incubation for 1–2 hours is sufficient for most routine experiments. The exact incubation period depends on cell type, cell seeding density and other experimental parameters. For preliminary trials, measure fluorescence signals at 1 h, 2 h and 4 h respectively using a fluorescence microplate reader, then select the time point with the most appropriate fluorescence intensity range for follow-up formal experiments.
(4) After incubation completion, set excitation wavelength at 560 nm and emission wavelength at 590 nm to measure fluorescence intensity. The efficacy of drug treatment can be calculated by comparing RFU (Relative Fluorescence Units) between treatment groups and control groups.
3. Detection Operation
(1) Read the plate with a fluorescence microplate reader.
Note: Operate the instrument strictly in accordance with the manufacturer’s official manual.
Note: Colorimetric absorbance detection is alternatively available at 570 nm with 600 nm set as the reference wavelength. Fluorescence detection is highly recommended for superior
sensitivity.
