生化机理
Vascular endothelial growth factor C (VEGF-C) and VEGF-D constitute a subfamily of the angiogenic VEGF angiogenic factors. VEGF-C is synthesized as a 58 kDa molecule that consists of a VEGF homolgy domain (VHD) flanked by N- and C-terminal propeptides. The proprotein undergoes covalent homodimerization and stepwise proteolytic processing to generate ligands with increasing affinity for VEGF R3/Flt-4. Fully processed VEGF-C containing just the 21 kDa VHD can additionally bind and activate VEGF R2/KDR/Flk-1. Fully processed human VEGF-C shares 98% amino acid sequence identity with mouse and rat VEGF-C. VEGF-C interactions with VEGF R3 are critical for lymphangiogenesis. VEGF-C and VEGF R3 are usually co-expressed at sites with lymphatic vessel sprouting, in the embryo, and in various pathological conditions. Over-expression of VEGF-C in tumor cells induces tumoral lymphatic hyperplasia, resulting in enhanced lymph flow and metastasis to regional lymph nodes. It also induces physiological and intratumoral neoangiogenesis and vessel sprouting through interactions with VEGF R2.
Post-translational: Undergoes a complex proteolytic maturation which generates a variety of processed secreted forms with increased activity toward VEGFR-3, but only the fully processed form could activate VEGFR-2. VEGF-C first form an antiparallel homodimer linked by disulfide bonds. Before secretion, a cleavage occurs between Arg-227 and Ser-228 producing a heterotetramer. The next extracellular step of the processing removes the N-terminal propeptide. Finally the mature VEGF-C is composed mostly of two VEGF homology domains (VHDs) bound by non-covalent interactions.

Recombinant Human VEGF-C Protein (rp181290) - SDS-PAGE
3 μg/lane of Recombinant Human VEGF-C Protein was resolved with SDS-PAGE under reducing (R) and non-reducing (N) conditions and visualized by Coomassie® Blue staining, showing a band at 20.1-27.7 kDa under reducing conditions and 25.1 & 27.6 & 32.2-40.6 kDa under non-reducing conditions.
Members of the VEGF family are typically antiparallel, cystine-knot polypeptide dimers that are covalently linked by two intermolecular disulfide bonds. In VEGF-C, this VEGF homology domain is flanked by C- and N-terminal propeptides that are sequentially cleaved, giving rise to VEGF homologs with distinct functions (PMID: 20145116).
