阿拉丁重组蛋白A/G是一种基因工程蛋白，包含5个蛋白A的IgG结合区和2个蛋白G的IgG结合区。该重组A/G-Cys蛋白已去除了细胞壁结合区、细胞膜结合区和白蛋白结合区，以确保最大程度的特异性IgG结合。该重组蛋白A/G是制备用于纯化多克隆或单克隆IgG抗体的亲和凝胶的理想选择。蛋白A/G能结合多种人、小鼠和大鼠的IgG亚类（例如：人IgG1, IgG2, IgG3, IgG4；小鼠IgG2a, IgG2b, IgG3；大鼠IgG2a, IgG2c）。它还能结合牛、山羊、绵羊、马、兔、豚鼠、猪、狗和猫的总IgG。

重组蛋白A/G在最佳条件下与进行偶联 LumiDye™ AF488，并经纯化去除未结合的游离染料。该偶联物可用于流式细胞术和免疫荧光染色中的免疫球蛋白检测，以及检测方法的开发。绿色荧光探针最大吸收/发射波长约为490nm和525nm。

Aladdin recombinant Protein A/G is a genetically engineered protein containing 5 IgG-binding regions of protein A and 2 of protein G. Cell wall binding region, cell membrane binding region and albumin binding region have been removed from the recombinant A/G-Cys to ensure the maximum specific IgG binding. The recombinant Protein A/G is ideal for making affinity gel to purification of polyclonal or monoclonal IgG antibodies. Protein A/G binds to various human, mouse and rat IgG subclasses (e.g., human IgG1, IgG2, IgG3, IgG4; mouse IgG2a, IgG2b, IgG3; rat IgG2a, IgG2c). It also binds to total IgG from cow, goat, sheep, house, rabbit, guinea pig, pig, dog and cat.

We have detected the binding data of different IgG subtypes with recombinant protein A/G, as shown in the Graph1.

Recombinant Protein A/G was conjugated with LumiDye™ AF488 under optimal conditions and subsequently purified to remove unconjugated free dyes. This conjugate can be used for immunoglobulin detection in Flow Cytometry and IF and for assay development. Green Fluorescence. Excitation/Emission wavelength= 490 nm/525 nm.

生化机理

Plays a role in the inhibition of the host innate and adaptive immune responses. Possesses five immunoglobulin-binding domains that capture both the fragment crystallizable region (Fc region) and the Fab region (part of Ig that identifies antigen) of immunoglobulins. In turn, Staphylococcus aureus is protected from phagocytic killing via inhibition of Ig Fc region. In addition, the host elicited B-cell response is prevented due to a decrease of antibody-secreting cell proliferation that enter the bone marrow, thereby decreasing long-term antibody production. Inhibits osteogenesis by preventing osteoblast proliferation and expression of alkaline phosphatase, type I collagen, osteopontin and osteocalcin. Acts directly as a pro-inflammatory factor in the lung through its ability to bind and activate tumor necrosis factor alpha receptor 1/TNFRSF1A.

IF analysis of EpCAM (green) in HT-29 cells. The cells were fixed with 4% Paraformaldehyde Fix Solution (P395744) for 15 minutes, and blocked with 5% BSA for 1 hour at room temperature. Cells were stained with Citatuzumab (anti-EpCAM) (Ab209902; Isotype: Human IgG1) at 1/1000 dilution in blocking buffer overnight at 4°C, and then incubated with Recombinant Protein A/G, AF488 conjugate (rp303275) at 1/500 dilution for 1 hour at room temperature in the dark (green). PBS instead of the primary antibody was used as the secondary antibody only control. Cells were counterstained with Hoechst 33342 (B113845) (blue) at 1/1000 dilution for 5 minutes. Images were taken on the confocal laser scanning microscope. .
