线粒体是细胞能量的来源，其形态可以有较大的变化 (从杆状到圆形)。线粒体体积很小，只有借助显微镜才能观察到，母系遗传 DNA，其数量、大小和形状根据动物细胞类型而有差异。观察线粒体的最佳方式是用显微镜，组织病理学方法如 Altmann 技术是有帮助的，用组织化学方法成功显示线粒体取决于以下几个因素：组织必须新鲜固定，切片薄 (2～3μm)，苯胺‑酸性品红饱和溶液属于 Altmann 线粒体染色法，但需注意控制分化的程度。该试剂仅用于科研领域，不适用于临床诊断或其他用途。
操作步骤 (仅供参考)：
1、组织固定，推荐使用 Champy 固定液，但 Helly 固定液效果也较好。
2、切片置于二甲苯或脱蜡透明液脱蜡，经梯度乙醇复水至水。
3、将切片浸泡于苯胺‑酸性品红饱和溶液，稍微加热切片至有热气出现，放置 5 min。
4、蒸馏水冲洗切片。
5、用苦味酸乙醇溶液分化，应显微镜下控制染色程度。
6、无水乙醇快速脱水 2 次。
7、二甲苯或脱蜡透明液进行透明处理，非水溶性封片剂封片。

Mitochondria are the energy source of cells and vary greatly in morphology (ranging from rod‑shaped to spherical). Mitochondria are tiny and can only be observed under a microscope. They contain maternally inherited DNA, and their quantity, size and shape differ depending on the type of animal cell. Microscopy represents the optimal way to observe mitochondria, and histopathological techniques such as the Altmann method are also applicable. Successful visualization of mitochondria by histochemical methods depends on the following factors: fresh tissue fixation and thin sections (2～3 μm). Aniline‑Acid Fuchsin Saturated Solution is used for Altmann mitochondrial staining, and strict differentiation control is required. This reagent is for research use only and not intended for clinical diagnosis or other applications.
Procedure (For reference only):
1. Tissue fixation: Champy’s fixative is recommended, while Helly’s fixative also yields satisfactory results.
2. Place sections in xylene or dewaxing clearing solution for dewaxing and clearing.
3. Immerse sections in Aniline‑Acid Fuchsin Saturated Solution, heat gently until steam appears, and incubate for 5 min.
4. Rinse sections with distilled water.
5. Differentiate with picric acid ethanol solution; monitor staining intensity under a microscope.
6. Rapidly dehydrate twice with absolute ethanol.
7. Clear with xylene or dewaxing clearing solution, and mount with non‑aqueous mounting medium.

1. Differentiate carefully to obtain a yellow background.
