概述

棕榈酸钠BSA高脂细胞添加剂的有效成分是棕榈酸钠 (Sodium palmitate)，浓度是6 mM。本产品为无菌 (0.22μm 过滤) 溶液，可与完全培养基直接互溶用于实验。本产品所用溶剂不含NaOH或有机溶剂，对细胞无毒性。本产品可与油酸钠 (Sodium oleate) 共同模拟高自由脂肪酸环境，也可用于诱发细胞脂毒性、诱导内质网应激、氧化应激、线粒体损伤、细胞凋亡等。

使用说明：
1、针对不同细胞种类及实验目的，本产品所用浓度不同，一般与细胞培养基按1: 60-1: 10倍比稀释。
2、对于首次使用本产品，建议采用浓度梯度处理细胞，摸索针对特定细胞、达到特定效果的适宜浓度。
3、待细胞生长至80-100%汇合度时，换成新鲜培养基，按适当浓度滴加本产品，对照组加入等量体积溶剂对照，轻柔混匀后将细胞放入细胞孵箱中继续培养。
4、加入本品20-48h后，收集细胞，进行下一步分析。
注意事项：
1、收到产品后可分装后于-20℃长期保存；可在2-8℃短期保存3个月。
2、由于溶剂 BSA 影响吸光度，使用CCK-8（Aladdin货号C266180）检测细胞毒性时，在弃掉高脂细胞添加剂与培养基混液后，需要用PBS清洗细胞2-3次，再加入CCK-8工作液。

The active ingredient of Sodium Palmitate-BSA High-Fat Cell Supplement is sodium palmitate at a concentration of 6 mM. This product is a sterile solution filtered through a 0.22 μm membrane, which can be directly mixed with complete medium for experiments. The solvent adopted in this product contains no NaOH or organic solvents and exhibits no cytotoxicity to cells. This product can be combined with sodium oleate to simulate a high free fatty acid environment. It can also be applied to induce cellular lipotoxicity, endoplasmic reticulum stress, oxidative stress, mitochondrial damage, cell apoptosis and other related phenotypes.

Instructions for Use：

1. The working concentration varies according to cell types and experimental purposes. Generally, dilute the product with cell culture medium at a volume ratio ranging from 1:60 to 1:10.

2. For first-time use, it is recommended to treat cells with a concentration gradient to explore the optimal concentration for specific cells and experimental outcomes.

3. When cells reach 80%–100% confluence, replace with fresh medium, add the product dropwise at the appropriate concentration, and add an equal volume of solvent vehicle to the control group. Mix gently and place the cells back into the incubator for continuous culture.

4. Collect cells 20–48 h after supplementation for subsequent analysis.

Precautions：

1. Upon receipt, aliquot the product for long-term storage at -20 °C; short-term storage at 2–8 °C is permitted for up to 3 months.

2. The solvent BSA may interfere with absorbance readings. When detecting cytotoxicity using CCK-8 (Aladdin Cat. No. C266180), discard the mixture of high-fat supplement and medium, wash the cells 2–3 times with PBS, then add CCK-8 working solution.
