本方法采用苯酚-硫酸法测定总糖含量：样品中糖经热水提取后，用硫酸脱水生成糠醛或其衍生物，再与苯酚缩合生成橙黄色物质，于480 nm处有最大吸收峰，吸光度与糖浓度成正比，通过标准曲线即可定量总糖。
This method employs the phenol‑sulfuric acid assay for the determination of total sugar content. Sugars in the sample are extracted with hot water, then dehydrated by sulfuric acid to generate furfural or its derivatives, which subsequently condense with phenol to form an orange‑yellow colored substance with a maximum absorption peak at 480 nm. The absorbance is directly proportional to the sugar concentration, and the total sugar content can be quantified using a standard curve.
